An alternative and efficient protocol for the synthesis of methylated pyrrolo‐tetrathiafulvalenes have been developed. The key intermediate in this new synthetic protocol is 4,6‐dimethyl‐N‐tosyl‐(1,3)‐dithiolo[4,5‐c]pyrrole‐2‐one, which can be obtained in high yield in a three‐step synthetic procedure from 5‐dimethyl‐3,4‐dithiocyanopyrrole.  相似文献

A general procedure is reported for the synthesis of cis ring fused azapolycyclic compounds bearing an all‐carbon quaternary stereocenter at the ring fusion and an adequate functionalization for the assembly of new rings leading to advanced synthetic intermediates for Daphniphyllum alkaloid synthesis. The key carbon?carbon bond‐forming step in this approach is a radical cyclization of an N‐cycloalkenyl trichloroacetamide derivative involving a tetrasubstituted enamide to achieve polyfunctionalized lactams.  相似文献

An efficient two‐step synthetic procedure for the preparation of numerous variations of N‐protected α,β‐unsaturated α‐amino acids and their corresponding esters from N‐protected glycine and either aliphatic or aromatic aldehydes was developed. A new three‐step synthetic pathway to generate polycyclic annulated hydantoins via rarely investigated heterocyclic imines is described. This procedure includes a one‐pot reaction forming imines as precursor structures (e.g., Asinger reaction), followed by an Ugi reaction to build up a bisamide structure that allows a ring‐closing reaction to the targeted hydantoins via substitution. This pathway leads to a multiplicity of substances with a potential pharmacological activity.  相似文献

A five‐step procedure for the synthesis of cis‐1‐tosyl‐2‐tosyloxymethyl‐3‐(trifluoromethyl)aziridine was developed, starting from 1‐ethoxy‐2,2,2‐trifluoroethanol, involving imination, aziridination, ester reduction, hydrogenation, and N‐,O‐ditosylation steps. Further synthetic elaborations revealed a remarkable difference in the reactivity of cis‐1‐tosyl‐2‐tosyloxymethyl‐3‐(trifluoromethyl)aziridine with respect to aromatic sulfur and oxygen nucleophiles, thus enabling the selective deployment of this versatile substrate as a building block for the synthesis of functionalized aziridines, azetidines, and benzo‐fused dithianes, oxathianes, dioxanes, and (thio)morpholines.  相似文献

An efficient two‐step synthesis of 3,6‐diaminopyridazine from 3,6‐dichloropyridazine is reported. In this synthetic procedure, 4‐methoxybenzylamine was used as a nitrogen source to substitute the chloro groups of 3,6‐dichloropyridazine to form N,N′‐bis‐(4‐methoxybenzyl)‐pyridazine‐3,6‐diamine. The 4‐methoxybenzyl groups were then removed by treatment with hydrochloric acid to provide the target 3,6‐diaminopyridazine with an overall yield of 78%.  相似文献

A new synthetic approach is described for building the coumarin scaffold through the Lewis acid‐promoted cyclization of novel aryl 3‐(dimethylamino)prop‐2‐enoates 2a – 2f . The latter precursors were prepared via aminomethylenation of the corresponding aryl acetates 4a – 4f with the Bredereck reagent. This approach was used for the synthesis of biologically active natural compounds 1a – 1f , through a three‐step procedure starting from the corresponding phenols.  相似文献

A two step synthetic procedure towards model coil‐rod‐coil triblock copolymers was developed. In the first step, α,ω‐bromo‐functionalized oligophenylenes (rod part) were modified to anionic bifunctional initiators. In the second step, flexible chains were grown from both ends of the rod part by an anionic polymerization procedure leading to polystyrene‐rod‐polystyrene and/or poly(2‐vinyl pyridine)‐rod‐poly(2‐vinyl pyridine) triblock copolymers. Characterization of the final products by gel permeation chromatography, NMR spectroscopy, light scattering, Raman spectroscopy and differential scanning calorimetry demonstrated that well‐defined coil‐rod‐coilblock copolymers can be synthesized.  相似文献

Several novel N′-[(1-ferrocenylethylidene)amino]-N′-β-D-glycopyranosylthioureas have been efficiently synthesized via a six-step procedure, utilizing three monosaccharides (D-glucose, D-galactose and D-xylose) as starting materials, respectively. They have been preliminarily determined to display biological activity against certain fungi. The target compounds have been unambiguously characterized by spectroscopic method and elemental analysis. An initially alternative synthetic route, which was seemingly reasonable, has been excluded due to its inefficiency.  相似文献

Tetracyclic skeletons combining an imidazo[1,2‐a]pyridine moiety with a quinoline framework such as pyrido[2′,1′:2,3]imidazo[4,5‐b]quinoline are stimulating increasing interests since they are close isosteres of a series of powerful antiproliferative compounds. In this paper, we report a novel methodology for the synthesis of pyrido[2′,1′:2,3]imidazo[4,5‐c]quinolines through one‐pot sequential reactions of commercially available or readily obtainable 2‐aminopyridines, 2‐bromophenacyl bromides, aqueous ammonia, and aldehydes. Moreover, dihydropyrido[2′,1′:2,3]imidazo[4,5‐c]quinolines could also be obtained in a similar manner by using various ketones as the substrates in place of aldehydes. Notably, the whole procedure combines condensation/amination/cyclization reactions in one pot to give complex compounds in a simple and practical manner. Compared with literature methods, the synthetic strategy reported herein has the advantages of readily available starting materials, structural diversity of products, good functional group tolerance, and obviation of step‐by‐step operations.  相似文献

An ortho‐selective rhodium‐catalyzed direct C?H arylation of 1,1′‐bi‐2‐naphthol (BINOL), to deliver the widely used but not easily available 3,3′‐diaryl BINOL, has been developed. This highly efficient one‐step synthetic approach is the shortest route to date and is greatly facilitated by the newly developed ligand system comprising tBu2PCl, Ph2‐cod, and Cy3P?HBF4. In addition, the same procedure can facilitate the challenging syntheses of 3‐bulkyaryl BINOLs in good to excellent yields.  相似文献

In this communication an extended “in–out” polymerization method is presented, which leads to the synthesis of novel heteroarm star block terpolymers of the type An(B‐b‐C)n. A four step/one‐pot synthetic procedure is pursued using anionic polymerization under an inert atmosphere. The resulted star‐shaped terpolymer consists of a divinyl benzene nodule bearing pure polystyrene and poly(hexyl methacrylate)‐block‐poly(methyl methacrylate) diblock copolymer arms. It is shown that this kind of star terpolymers can self‐assemble in the bulk forming lamellae mesophase by arm and block segregation. The mechanical properties of the terpolymer have been examined in detail. Finally, the proposed synthetic procedure can be easily employed in other controlled polymerization methods.

Pentenyl phenyl acrylic acid is a structural unit of pepticinnamin E, a natural product and a bisubstrate inhibitor of FPTase. In this article, a new synthetic strategy was developed to prepare pentenyl phenyl acrylic acid with high stereoselectivity and high overall yield of 78.6%. The method used in producing these effects involved the application of a five‐step procedure. Pentenyl phenyl acrylic acid was synthesized starting from 2‐iodo‐benzyl alcohol through an E‐selective Wittig–Horner reaction, and then the Sonogashira reaction was used to produce 2‐(1‐pentynyl)‐E‐ethyl‐cinnamoylate, which was quantitatively hydrogenated, catalyzed by Lindlar catalyst.  相似文献

A novel approach to fabricate polymer brushes on the surface of carbon nanotubes (CNTs) is proposed. Carboxyl groups on the surface of chemically oxidized CNTs were reacted with hexamethylene diisocyanate, followed by a reaction with methacrylamide to give terminal vinyl groups‐functionalized CNTs, so called “CNT‐mer.” The synthetic procedure was investigated step‐by‐step and the synthesized CNT‐mer was used to grow polystyrene (PS) from CNTs by a simple in situ polymerization in the presence of a thermal initiator. By employing 1H NMR, X‐ray photoelectron spectroscopy, thermogravimetric analysis, scanning electron microscopy, transmission electron microscopy, and light scattering, the experimental results were verified. Using this approach, 45% PS with respect to CNTs are grafted on the surface of CNTs with about 4.0 nm thickness. This novel technique would provide a facile route to prepare tailor‐made polymer brushes on the surface of CNTs. © 2006 Wiley Periodicals, Inc. A series of new 2‐aryl‐7‐cyano/ethoxycarbonyl‐6‐methylthio‐1H‐imidazo[1,2‐b]pyrazoles ( 5 ) have been synthesized in moderate to good yields, via a two‐step cyclocondensation procedure of 5‐amino‐4‐cyano/ethoxycarbonyl‐3‐methylthio‐1H‐pyrazole ( 1 ) and α‐bromoacetophenones ( 3 ) or α‐tosyloxyacetophenones ( 2 ), which were prepared by the reactions of acetophenones with [hydroxy(tosyloxy)iodo]benzene (HTIB). The intermediates, 5‐amino‐1‐(aroylmethyl)‐4‐ cyano/ethoxycarbonyl‐3‐methylthio‐1H‐pyrazoles ( 4 ), have been isolated, serving as evidence for the regioselectivity. When utilizing α‐tosyloxy‐acetophenones, the reactions were more eco‐friendly, the reaction time was significantly reduced and the synthetic procedure was more convenient and easier to manipulate. Surprisingly, using potassium carbonate to displace sodium carbonate in the synthesis of 4 , in the case of 1 (R? CN), two novel cyclocondensation products have been isolated and fully characterized, followed by the proposal of a plausible mechanism.  相似文献

In the last twenty years DMC has been employed as an efficient methylating and methoxycarbonylating agent with several monodentate and bidentate nucleophiles, showing great selectivity and unexpected results. In this short review we report on yet another application of DMC chemistry i.e. the synthesis of 5‐membered N‐ and O‐heterocycles. In these reactions DMC acts as a sacrificial molecule since it is not present in the final products, but only in the reaction intermediates as an halogen‐free leaving group. This DMC‐based synthesis of heterocycles resulted of general application, as it is effective for aliphatic and aromatic 1,4‐diols, incorporating several functionalities (primary, secondary, tertiary, allylic, phenolic), as well as, for bifunctional compounds i.e. 4‐amino‐1‐butanol. This synthetic procedure was also employed for industrially relevant compounds such as (‐)‐norlabdane oxide and isosorbide showing to maintain the chiral integrity of the substrate. In one case intramolecular cyclisation of isosorbide was also observed to achieve a strained tricyclic derivative. Comparing this reaction methodology with a chlorine based procedure, the DMC‐mediated pathway is quantitative, occurs in one step, does not require any chlorine‐based chemical or strong acid and does not produce any chlorinated waste material.  相似文献

Reported herein is the single‐step construction of an anti‐configured deoxypropionate motif by syndiospecific propylene oligomerization catalyzed by a Cs‐symmetric zirconocene complex. After oligomerization, oxidation of the oligomers by oxygen afforded oligopropylene alcohols in a single step. This strategy was applied to the single‐step preparation of rel‐(2R,4S,6R,8S)‐2,4,6,8‐tetramethylundecan‐1‐ol, the racemic mixture of the synthetic fragment of the cuticular hydrocarbons isolated from the cane beetle Antitrogus parvulus.  相似文献

A facile and efficient synthetic methodo‐ logy for the preparation of diethyl α,α‐chlorofluoro‐ alkanephosphonates is described. A wide variety of diethyl α‐hydroxyphosphonates were investigated by a two‐step halogenation procedure, which includes nucleophilic chlorination with PPh3 and CCl4 and electrophilic fluorination with N‐fluorobisbenzene‐ sulfonimide. Aromatic and aliphatic α,α‐chlorofluoro‐ phosphonates could be prepared by this method with acceptable yields. © 2010 Wiley Periodicals, Inc.
