A peptide has been designed so that its chelating affinity for one type of metal ion regulates its affinity for a second, different type of metal ion. The prochelator peptide (PCP), which is a fusion of motifs evocative of calcium loops and zinc fingers, forms a 1 : 2 Zn : peptide complex at pH 7.4 that increases its affinity for Zn2+ ∼3-fold in the presence of Tb3+ (log β2 from 13.8 to 14.3), while the 1 : 1 luminescent complex with Tb3+ is brighter, longer lived, and 20-fold tighter in the presence of Zn2+ (log K from 6.2 to 7.5). This unique example of cooperative, heterometallic allostery in a biologically compatible construct suggests the possibility of designing conditionally active metal-binding agents that could respond to dynamic changes in cellular metal status.  相似文献

Fluorescence imaging is a powerful tool for the visualization of biological molecules in living cells, tissue slices, and whole bodies, and is important for elucidating biological phenomena. Furthermore, zinc (Zn2+) is the second most abundant heavy metal ion in the human body after iron, and detection of chelatable Zn2+ in biological studies has attracted much attention. Herein, we present a novel, highly sensitive off–on fluorescent chemosensor for Zn2+ by using the internal charge transfer (ICT) mechanism. The rationale of our approach to highly sensitive sensor molecules is as follows. If fluorescence can be completely quenched in the absence of Zn2+, chemosensors would offer a better signal‐to‐noise ratio. However, it is difficult to quench the fluorescence completely before Zn2+ binding, and most sensor molecules still show very weak fluorescence in the absence of Zn2+. But even though the sensor shows a weak fluorescence in the absence of Zn2+, this fluorescence can be further suppressed by selecting an excitation wavelength that is barely absorbed by the Zn2+‐free sensor molecule. Focusing on careful control of ICT within the 4‐amino‐1,8‐naphthalimide dye platform, we designed and synthesized a new chemosensor ( 1 ) that shows a pronounced fluorescence enhancement with a blueshift in the absorption spectrum upon addition of Zn2+. The usefulness of 1 for monitoring Zn2+ changes was confirmed in living HeLa cells. There have been several reports on 4‐amino‐1,8‐naphthalimide‐based fluorescent sensor molecules. However, 1 is the first Zn2+‐sensitive off–on fluorescent sensor molecule that employs the ICT mechanism; most off–on sensor molecules for Zn2+ employ the photoinduced electron transfer (PeT) mechanism.  相似文献

A highly sensitive fluorescent sensor ZnDN was designed, synthesized and used for tracking intracellular zinc ions in various living cells and direct imaging of prostatic tissue in mice. ZnDN was prepared from the heterocyclic-fused naphthalimide fluorophore, and the zinc receptor, N,N-bis(2-pyridylmethyl)ethylenediamine (BPEN). Upon addition of Zn2+ to the solutions of ZnDN, a remarkable fluorescence enhancement was observed, which could be attributed to the photo-induced electron transfer (PET) mechanism. Since ZnDN exhibited high sensitivity toward Zn2+ in phosphate buffer solution, with a limit of detection of 4.0 × 10−9 mol/L, it was further applied for the imaging of exogenous and endogenous Zn2+ in different living cells. Living cells imaging experiments suggested that ZnDN could image the changes of intracellular free zinc ions, and could be used for two-photon imaging. Moreover, flow cytometry suggested that ZnDN could distinguish cancerous prostate cells from normal cells. Animal experiments indicated that ZnDN had the potential in imaging prostate tissue in vivo.  相似文献

A PVC membrane electrode based on bis(2-nitrophenyl)disulfide carrier exhibits a very good response for Zn2+ in a wide concentration range (from 2.9×10−7 to 3.2×10−2 mol l−1) with a slope of 29.9±0.4 mV per decade of Zn2+ concentration. The response time of the sensor is about 10 s and the membrane can be used for more than 3 months without any observed divergence in potentials. The proposed sensor exhibits very good selectivity for Zn2+ over many cations and can be used in a wide pH range (2-9). This assembly served also as an indicator electrode in potentiometric titrations involving zinc ions and direct potentiometric determination of zinc ions in real and synthetic samples.  相似文献

Zn2+ is an essential regulator of coagulation and is released from activated platelets. In plasma, the free Zn2+ concentration is fine-tuned through buffering by human serum albumin (HSA). Importantly, the ability of HSA to bind/buffer Zn2+ is compromised by co-transported non-esterified fatty acids (NEFAs). Given the role of Zn2+ in blood clot formation, we hypothesise that Zn2+ displacement from HSA by NEFAs in certain conditions (such as type 2 diabetes mellitus, T2DM) impacts on the cellular and protein arms of coagulation. To test this hypothesis, we assessed the extent to which increasing concentrations of a range of medium- and long-chain NEFAs reduced Zn2+-binding ability of HSA. Amongst the NEFAs tested, palmitate (16 : 0) and stearate (18 : 0) were the most effective at suppressing zinc-binding, whilst the mono-unsaturated palmitoleate (16 : 1c9) was markedly less effective. Assessment of platelet aggregation and fibrin clotting parameters in purified systems and in pooled plasma suggested that the HSA-mediated impact of the model NEFA myristate on zinc speciation intensified the effects of Zn2+ alone. The effects of elevated Zn2+ alone on fibrin clot density and fibre thickness in a purified protein system were mirrored in samples from T2DM patients, who have derranged NEFA metabolism. Crucially, T2DM individuals had increased total plasma NEFAs compared to controls, with the concentrations of key saturated (myristate, palmitate, stearate) and mono-unsaturated (oleate, cis-vaccenate) NEFAs positively correlating with clot density. Collectively, these data strongly support the concept that elevated NEFA levels contribute to altered coagulation in T2DM through dysregulation of plasma zinc speciation.

Herein the synthesis and characterization of new, lipophilic highly Zn2+-selective fluorescent probes are reported. High affinity for zinc (Kd 1.1–8.0 nM) over other biologically relevant metals and mixtures of metals was observed. Excitation at 360 nm afforded an emission spectrum with maximum at 530 nm for the zinc bound complex. The linear relationship between fluorescence intensity and zinc concentration indicates that FZnA-probes can be used for quantification. The probes have been synthesized in 28–45% overall yield and the feasibility for further functionalization with biologically relevant side chains has been demonstrated. In vitro studies using PC12 cells and 10 μM of one of the novel probes (FZnA-Ada) visualized endogenous labile Zn2+ after 45 min incubation time.  相似文献

Monitoring labile Zn2+ homeostasis is of great importance for the study of physiological functions of Zn2+ in biological systems. Here we report a novel ratiometric fluorescent Zn2+ sensor, CPBT, which was constructed based on chelation-induced alteration of FRET efficiency. CPBT was readily cell membrane permeable and showed a slight preferential localization in the endoplasmic reticulum. With this sensor, 3D ratiometric Zn2+ imaging was first realized in the head of zebra fish larvae via Z-stack mode. CPBT could track labile Zn2+ in a large number of cells through ratiometric flow cytometric assay. More interestingly, both ratiometric fluorescence imaging and flow cytometric assay demonstrated that the labile Zn2+ level in MCF-7 cells (cisplatin-sensitive) decreased while that in SKOV3 cells (cisplatin-insensitive) increased after cisplatin treatment, indicating that Zn2+ may play an important role in cisplatin induced signaling pathways in these cancer cells.

A Zn2+ sensor exhibiting 3D ratiometric imaging and flow cytometric ability was constructed based on the FRET mechanism, and cisplatin-induced endogenous labile Zn2+ fluctuations were monitored in real time.  相似文献

A series of cobalt-inserted copper zinc ferrites, Cu0.6CoxZn0.4−xFe2O4 (x = 0.0, 0.1, 0.2, 0.3, 0.4) having cubic spinel structure were prepared by the coprecipitation method. Various characterization techniques, including XRD, FTIR, UV-vis and I–V were used to investigate structural optical and electrical properties, respectively. The lattice constant was observed to be decreased as smaller ionic radii Co2+ (0.74 Å) replaced the higher ionic radii Zn2+ (0.82 Å). The presence of tetrahedral and octahedral bands was confirmed by FTIR spectra. Optical bandgap energy was determined in the range of 4.44–2.05 eV for x = 0.0 to 0.4 nanoferrites, respectively. DC electrical resistivity was measured and showed an increasing trend (5.42 × 108 to 6.48 × 108 Ω·cm) with the addition of cobalt contents as cobalt is more conductive than zinc. The range of DC electrical resistivity (108 ohm-cm) makes these nanomaterials potential candidates for telecommunication devices.  相似文献

A new bipyridyl derivative 1 bearing rhodamine B as visible fluorophore was designed, synthesized and characterized as a fluorescent and colorimetric sensor for metal ions. Interaction with Cu2+, Zn2+, Cd2+, Hg+, and Hg2+ ions was followed by UV/Vis and emission spectroscopy. Upon addition of these metal ions, different colorimetric and fluorescent responses were observed. “Off-on-off” (Cu2+, Zn2+, and Hg2+) and “off-on” (Hg+ and Cd2+) systems were obtained. Probe 1 was explored to mimic XOR and OR logic operations for the simultaneous detection of Hg+–Cu2+ and Hg+–Zn2+ pairs, respectively. DFT calculations were also performed to gain insight into the lowest-energy gas-phase conformation of free receptor 1 as well as the atomistic details of the coordination modes of the various metal ions.  相似文献

An efficient quinoline-based fluorescent chemosensor (QLNPY) was successfully developed for the detection of zinc ions (Zn2+). This novel chemosensor displayed higher sensitivity and selectivity toward Zn2+ over other competitive metal ions accompanying with obvious fluorescence enhancement. The QLNPY-Zn2+ complex can be further used as a new fluorescent “turn-off” sensor for pyrophosphate (PPi) and sulfur ion (S2?) via a Zn2+ displacement approach. The limits of detection were calculated to be 3.8 × 10?8 M for Zn2+, 3.7 × 10?7 M for PPi and 4.9 × 10?7 M for S2?. The binding mechanism of QLNPY and Zn2+ was investigated through NMR, HR-MS analysis and further studied by crystallographic analysis. Additionally, further application of QLNPY for sequential bioimaging of Zn2+ and PPi was studied in HepG2 cells, suggesting that the quinoline-based chemosensor possesses great potential applications for the detection of intracellular Zn2+ and PPi in vivo.  相似文献

Poly(acrylp-aminobenzenesulfonamideamidine-p-aminobenzenesulfonylamide) chelating fiber containing "S", "N", and "O" elements was synthesized from polyacrylonitrile fiber and p-aminobenzene sulfonamide and used to enrich and separate trace Bi(III), Hg(III), Au(III), and Pd(IV) ions from wastewater and ore sample solution. The enrichment acidity, flow rate, elution conditions, reuse, interference ions, saturated adsorption capacity, constant of adsorption rate, analytical accuracy, and actual samples on chelating fiber were investigated by means of inductively coupled plasma optical emission spectrometry (ICP-OES) with satisfactory results. Solutions of 100 ng mL–1 of Bi(III), Hg(III), Au(III), and Pd(IV) ions can be enriched quantitatively by this chelating fiber at a rate of 1.0 mL min–1 at pH 4 and desorbed quantitatively with 20 mL of 0.25 M HCl and 2% CS(NH2)2 solution at 50 °C (with recovery 97%). When the chelating fiber was reused for 20 times, the recoveries of the analyzed ions enriched by the fiber were still over 95% (except for Hg(III)). One thousand-fold excesses of Mn2+, Ca2+, Zn2+, Mg2+, Fe3+, Cu2+, Ni2+, Al3+, and Ba2+ ions and thousands-fold excesses of Na+ and K+ cause little interference in the pre-concentration and determination of the analyzed ions. The saturated adsorption capacity of Bi(III), Hg(III), Au(III), and Pd(IV) was 4.850×10–4, 3.235×10–4, 2.807×10–4, and 3.386×10–4 mol g–1, respectively. The constants of adsorption rate were 0.409 min–1 for Bi, 0.122 min–1 for Hg, 0.039 min–1 for Au, and 0.080 min–1 for Pd. The relative standard deviations (RSDs) for the enrichment and determination of 10 ng mL–1 Bi(III), Hg(III), Au(III), and Pd(IV) were lower than 2.3%. The results obtained for these ions in actual samples by this method were basically in agreement with the given values with average errors of less than 1.0%. FT-IR spectra shows that the existence of –SO2–Ar, –H2N–Ar, O=C–NH–, HN=C–NH–, and –HN–SO2 functional groups are verified in the chelating fiber. From the FT-IR spectroscopy, we can see that Hg(III), Au(III), and Pd(IV) are mainly combined with nitrogen and sulfur (or oxygen), and Bi(III) is mainly combined with nitrogen (or oxygen) of the groups to form a chelating complex.  相似文献

Ratiometric fluorescent chemosensors based on the position of ring annulation of the naphthol–thiazole moiety for quantification of zinc ions in aqueous ethanol were synthesized and investigated. It was found that sensor 3 exhibited a remarkably large red shift of 140 nm in emission upon complexation with Zn2+. A TD-B3LYP/6-31G (d,p) calculation was performed to characterize the nature of the fluorescence behavior of sensor 3 upon Zn2+ complexation. The combination of experimental and computational analyses provides a more complete understanding of the molecular level origin of these unique photophysical properties of this type of chemosensor.  相似文献

One-dimensional coordination polymers (1D-CPs) tend either to dissociate into constitutive ligands and metals readily in solution, or to aggregate randomly and amorphously, which prevents them from widespread application. In the present research, 1D-CPs comprising bridging dipyrrin ligands and divalent metal ions (Zn2+, Ni2+, and Cu2+) are synthesized. A liquid/liquid interfacial reaction gives rise to single crystals suitable for X-ray diffraction analysis: A dichloromethane solution of the ligand is layered with aqueous metal(ii) acetate, such that the coordination reaction proceeds at the liquid/liquid interface. Isolated single fibers of the zinc coordination polymer may be exfoliated from the single crystal or bulk solid upon ultrasonication. Atomic force microscopy (AFM) detects the isolated fibers with lengths of more than several μm. The exfoliated 1D-CP wires feature good processability, realizing a conjugate with single-wall carbon nanotubes (SWCNTs), and a thin film on a transparent SnO2 electrode. The processed materials show electric conversion ability: For example, the modified SnO2 electrode serves as a photoanode for a photoelectric conversion system. The designability and tunability of the present 1D-CPs is demonstrated by a ligand modification, affording a luminescent property and an extension of the photoelectric conversion response to longer wavelengths.  相似文献

New photo-induced electron transfer (PET) probes OMOX and OBOX, carrying an additional binding site in the form of ‘oxadiazole nitrogen’ have been designed to evaluate binding interactions with biologically significant Li+, Na+, K+, Ca2+, Mg2+, and Zn2+ including environmentally toxic Ba2+ and Cd2+ using optical spectral techniques. While Li+, Na+, and K+ did not appreciably perturb either the absorption or emission spectra, Ba2+, Ca2+, Mg2+, Zn2+, and Cd2+ induced slight red shifts (2-8 nm) in the UV-visible spectra as well as pronounced chelation induced enhanced fluorescence (CHEF). Both OMOX and OBOX exhibited the highest CHEF in contact with the zinc ion, whereas Ba2+, Ca2+, Mg2+, and Cd2+ induced relatively less emission enhancements. OBOX, which is a poorer emitter (Φf=0.0062) than OMOX (Φf=0.015), showed highly promising 160-fold emission enhancement in the presence of Zn2+. Potential, therefore is available in OBOX to function as a selective luminescent ‘off-on’ sensor for Zn2+ in the presence of coordinatively competing Ba2+, Ca2+, Mg2+, and Cd2+ ions.  相似文献

The three dimensional structure of Ferric uptake regulation protein dimer from E. coli, determined by molecular modeling, was docked on a DNA fragment (iron box) and Zn2+ ions were added in two steps. The first step involved the binding of one Zn2+ ion to what is known as the zinc site which consists of the residues Cys 92, Cys 95, Asp 137, Asp141, Arg139, Glu 140, His 145 and His 143 with an average metal-Nitrogen distance of 2.5 Å and metal-oxygen distance of 3.1–3.2 Å. The second Zn2+ ion is bound to the iron activating site formed from the residues Ile 50, His 71, Asn 72, Gly 97, Asp 105 and Ala 109. The binding of the second Zn2+ ion strengthened the binding of the first ion as indicated by the shortening of the zinc-residue distances. Fe2+, when added to the complex consisting of 2Zn2+/Fur dimer/DNA, replaced the Zn2+ ion in the zinc site and when a second Fe2+ was added, it replaced the second zinc ion in the iron activating site. The binding of both zinc and iron ions induced a similar change in Fur conformations, but shifted residues closer to DNA in a different manner. This is discussed along with a possible role for the Zn2+ ion in the Fur dimer binding of DNA in its repressor activity. A simple, low cost and sensitive voltammetric sensor was developed for the simultaneous detection of Pb2+, Cd2+, and Zn2+ based on a disposable carbon fiber rod (CFR). The important factors to enhance the sensing property were creation of a clean surface by dealing with CFR at a high potential and electrochemical deposition of Bi film to improve the accumulation of heavy metal ions.  相似文献

A series of Zn2+‐selective two‐photon fluorescent probes (AZnM1−AZnN) that had a wide range of dissociation constants (KdTP=8 nm‐ 12 μM ) were synthesized. These probes showed appreciable water solubility (>3 μM ), cell permeability, high photostability, pH insensitivity at pH>7, significant two‐photon action cross‐sections (86–110 GM) upon complexation with Zn2+, and can detect the Zn2+ ions in HeLa cells and in living tissue slices of rat hippocampal at a depth of >80 μm without mistargeting and photobleaching problems. These probes can potentially find application in the detection of various amounts of Zn2+ ions in live cells and intact tissues.  相似文献

Chemosensors are developed to image zinc ions. Fluorescence enhancement due to Zn2+ binding is an excellent way to detect its presence. A chemosensor for Zn2+ based on dipicolylamine (DPA) groups connected by a pyridyl amide backbone has been synthesized. Addition of 2-chloroacetyl chloride to 2,6-diaminopyridine affords 2,6-bis(chloroethylamido)pyridine, which is converted to the sensor BADPA-P by 2,2′-dipicolylamine displacement of chlorine. This compound along with two others, the mono-DPA, ADPA-P and the benzyl in place of pyridyl, BADPA-B, present three potential Zn2+ sensors. It was found that BADPA-P in the presence of Zn2+ shows a large increase in fluorescence, whether in polar organic or aqueous environments. Its fluorescence in the presence of Cd2+, unlike with Zn2+, is not enhanced when excited at longer wavelengths. Proton NMR measurements, indicate two Zn2+ ions bind to BADPA-P. Also, Zn2+ enhances fluorescence even when other metal ions are present.  相似文献

This paper reports a series of comparative experiments on the activity of carbon- and oxygen-centred radical species in a model reaction of the radical addition of THF to imines mediated by a series of zinc alkyl/air reaction systems. The study strongly contradicts the notion that generally R˙ radicals are the initiating species in organic reactions mediated by RnM/air systems, and simultaneously demonstrates that oxygen-centred radical species are the key intermediates responsible for the initiation process. In addition, a new efficient RZn(L)/air initiating system for radical organic reactions exampled by a model reaction of radical addition of THF to imines is developed. Moreover, the isolation and structural characterization of the first zinc alkylperoxide supported by a carboxylate ligand, [Zn43-OOtBu)34-O)(O2CEt)3]2, as well as the novel octanuclear zinc oxo(alkoxide) aggregate with entrapped O–THF species, [Zn44-O)(μ3-2-O–THF)(O2CEt)5]2, provide clear mechanistic signatures for the mode of function of the RZn(O2CR′)/air system.  相似文献

The interaction of zinc(II) complex of N,N′‐bis(guanidinoethyl)‐2,6‐pyridinedicarboxamide (Gua) with DNA was studied by CD spectroscopy and agarose gel electrophoresis analysis. The results indicate that the DNA binding affinity of Zn2+‐Gua is stronger than that of Gua and the Zn2+‐Gua can promote the cleavage of phosphodiester bond of supercoiled DNA under a physiological condition, which is ~106 times higher than DNA natural degradation. The hydrolysis pathway was proposed as the possible mechanism for DNA cleavage promoted by the Zn2+‐ Gua. The acceleration is due to cooperative catalysis of the zinc cation center and the functional groups (bisguanidinium groups).  相似文献
