Methyl (1S,2R,4S,5R)-7-aza-5-bromo-bicyclo[2.2.1]heptane-2-carboxylate was synthesized in high yield in short time from methyl (1R,2S,4R,5R)-2-amino-4,5-dibromocyclohexanecarboxylate through intramolecular cycloamination under microwave-assisted conditions. The following substitution reaction by trifluoro-acetate anion also took place in microwave-assisted conditions to afford methyl (1S,2R,4S,5S)-7-aza-5-hydroxy-bicyclo[2.2.1]heptane-2-carboxylate. In the acyloxylation reaction, unusual endo-selectivity was observed owing to 7-azabicyclo[2.2.1]heptane skeleton.  相似文献

5-n-Alkylcycloalkane[d]pyrazolo[3,4-b]pyridines were prepared by a microwave-assisted cyclocondensation reaction between 5-aminopyrazoles and cyclic ketones under solvent-free conditions and catalyzed by iodine. This procedure provides a simple one-step and environmentally friendly methodology with good yields for the synthesis of these compounds.  相似文献

A three-component domino reaction between a 3-formylchromone, an isocyanide, and 1,3-dimethylalloxan which affords novel 5-(furo[3,4-b]chromenyl)-5-hydroxybarbiturate derivatives is reported. The reaction sequence consists of an initial [4+1] cycloaddition followed by an activated electrophilic heteroaromatic substitution on the furan ring to afford the products. This novel cascade reaction sequence represents an atom-economic route to biologically interesting molecules.  相似文献

A series of the novel 5-alkynyl- and furo[2,3-d]pyrimidine derivatives in which the sugar moiety is replaced by a methoxymethyl (MOM) group is synthesised using the Sonogashira cross-coupling reaction under both conventional and microwave conditions, in good to excellent yields. The 5-endo-dig cyclisation of 5-alkynylpyrimidine derivatives promoted by a Pd-catalyst or base gives the corresponding furo[2,3-d]pyrimidines in good yields.  相似文献

μ-(η55-4,5-Dihydro-as-indecenyl)bis(η5-cyclopentadienyliron) (I) was synthesized from 2-[(dimethylamino)methyl]biferrocene via 2-[(dimethylamino)methyl]biferrocene methiodide. The latter was converted to 2-(cyanomethyl)biferrocene, which was subjected to base hydrolysis to give (2-biferrocenyl)acetic acid. The acid was cyclized with trifluoroacetic anhydride to [4-μ-(η55-as-indecenyl)bis(η5-cyclopentadienyliron)]trifluoroacetate ester. Hydrolysis of the latter followed by reduction gave compound I.  相似文献

A novel bis-calix[5]arene (1) with divergent cavities was prepared in four steps starting from p-methylcalix[5]arene. This molecule is composed of two calix[5]-bis-crown-3 units held together, via their narrow rims, by a N,N″-1,4-phenylene-bis[N′-(2-ethenyloxyethyl)]diureido bridging moiety. Owing to the anion-binding ability of the ureido moieties embedded in its framework, bis-calix[5]arene 1 acts as a heterotetratopic monomer and, in the presence of 1,8-octanediyldiammonium dichloride, self-assembles into AA/BB-type polycapsular oligomers. Extensive 1H NMR investigations have demonstrated that the nature and stoichiometry of these supramolecular oligomers are strongly dependent on the concentration and the ratio of the two monomers.  相似文献

Pharmacologically relevant 5-thioxo-6H-imidazo[1,2-c]quinazolines and 5-oxo-6H-imidazo[1,2-c]quinazolines were prepared by sequential reactions of α-aminoketones with 2-isothiocyanatobenzonitrile (ITCB) and 2-isocyanatobenzonitrile (ICB), respectively. The functionalization of the thioxo moiety allowed the synthesis of 5-amino-6H-imidazo[1,2-c]quinazolines and of 1,2,4-triazolo[4,3-a]-imidazo[1,2-c]quinazolines.  相似文献

3,6-Disubstituted 1,2,4-triazolo[3,4-b][1,3,4]thiadiazoles together with the unknown systems 2-(3-aryl-5-methyl-1H-[1,2,4]triazol-1-yl)-5-aryl-1,3,4-thiadiazoles were obtained by anodic oxidation, under aprotic conditions, of aryl aldehyde N-(5-aryl-1,3,4-thiadiazol-2-yl) hydrazones. Azafulvenium methides generated by the thermal extrusion of SO2 from 1-methyl- and 1,1-dimethyl-1H,3H-pyrrolo[1,2-c]thiazole-2,2-dioxides undergo [1,8]H sigmatropic shifts to give vinylpyrroles. Pillar[5]arenes, designed and prepared by Ogoshi et al. in 2008 initially, refer to fifth classical macrocyclics. Among a wide range of pillar[5]arenes, rim-differentiated pillar[5]arenes containing five identical substituents on one rim and five different identical groups on the other rims are considered the most noteworthy type of pillar[5]arenes. As compared with the perfunctionalized pillar[5]arene, the self-assembly properties of rim-differentiated pillar[5]arenes have more varieties. On the other hand, in comparison with other types of pillar[5]arenes, the rim-differentiated pillar[5]arenes exhibit a more rigid symmetrical structure. In the present review, the synthetic methods, host-guest interactions, self-assembly properties and applications of rim-differentiated pillar[5]arenes are summarized. Hopefully, this review will be conducive to researchers in macrocyclic supramolecular chemistry.  相似文献

The reaction of 2-aminothiazoles with ethyl acetoacetate in acetic or polyphosphoric acid gave a series of 5H-[1,3]thiazolo[3,2-a]pyrimidin-5-one derivatives which were nitrated with a mixture of nitric and sulfuric acid to 6-nitro-5H-[1,3]thiazolo[3,2-a]pyrimidin-5-ones, and the latter were reduced to the corresponding amines.  相似文献

An efficient one-pot synthetic method for the highly substituted 5H-[1,3,4]thiadiazolo[3,2-a]pyrimidine-6-carboxylate derivatives has been accomplished via a microwave irradiation of MCRs of various aldehydes, 2-aminothiadiazole, and acetoacetate without any catalyst in acetic acid. This approach provides a convenient one pot method for the synthesis of 5H-[1,3,4]thiadiazolo[3,2-a]pyrimidine-6-carboxylate derivatives.  相似文献

The Smiles rearrangement was successfully applied to 4-hydroxybenzo[b]thiophene furnishing a facile entry to the 4-amino derivative. The rearrangement was extended to 5-methoxy-4-methoxycarbonyl[1]benzothieno[2,3-b]pyridine obtained via aza-Wittig/electrocyclization reaction of novel N-(4-methoxybenzothiophen-2-yl)iminomethyldiphenylphosphorane with methyl trans-4-oxo-2-pentenoate. The preparation of a novel 5-amino-4-methoxycarbonyl[1]benzothieno[2,3-b]pyridine, which is of interest as a potential secondary peptide structure mimic, was successfully achieved.  相似文献

A facile synthesis of novel 2-amino-6-arylmethyl-7-carboxamido-7,8-dihydropyrimido[5,4-f][1,4]thiazepin-5-ones is described. The synthesis was developed on solid phase and was applied to provide a series of analogs in good yield. The key reactions are acylation of a cysteine derivative with 2,4-dichloropyrimidine-5-carbonyl chloride followed by cyclization to generate a 6-arylmethyl-7-carboxamido-2-chloro-7,8-dihydropyrimido[5,4-f][1,4]thiazepin-5-one, which is further derivatized with an amine to give the desired 2-amino-6-arylmethyl-7-carboxamido-7,8-dihydropyrimido[5,4-f][1,4]thiazepin-5-one.  相似文献

A new, environmentally benign two-step synthesis of 5H[1,3]-thiazolo[2,3-b]quinazoline-3,5-(2H)-dione and 5H-thiazolo[2,3-b]quinazolin-5-one derivatives has been accomplished stepwise. The substituted 2-aminobenzoic acid upon condensation with thiourea in 1-butyl-3-methylimidazolium bromide at moderate temperature under nitrogen atmosphere yielded 2-thioxo-1H-4-quinazolinones. The resulting intermediate 2-thioxo-1H-4-quinazolinones, when reacted with 2-chloroethanoic acid/2-chloropropanal underwent cyclization to yield the desired product in excellent yields.  相似文献
