The versatile intermediates 2-(2-amino-4,5-dimethoxyphenyl)-6,7-dimethoxy-1,2,3,4-tetrahydroquinoline ( 2a ) and 6-(2-amino-4,5-dimethoxyphenyl)-5,6,7,8-tetrahydro[1,3]dioxolo[4,5-g]quinoline ( 2b ) were used in the preparation of a wide variety of 12,13-dihydro-11bH-quino[1,2-c]quinazolines by reaction with triethyl orthoformate, cyanogen bromide, urea and carbon disulfide in pyridine. Reaction of the thio and keto products with methyl iodide and phosphorus oxychloride, respectively, gave the requisite methylthio and chloro derivatives. Novel Reissert type reactions occurred when the intermediates 2a,b were reacted with acetic anhydride or benzoyl chloride. The attempted dehydrogenation of 12,13-dihydro-2,3,9,10-tetramethoxy-11bH-quino[1,2-c]quinazoline ( 3a ) is also reported.  相似文献

The reaction of methyl 2-bromo-6-(trifluoromethyl)-3-pyridinecarboxylate ( 1 ) with methanesulfonamide gave methyl 2-[(methylsulfonyl)amino]-6-(trifluoromethyl)-3-pyridine-carboxylate ( 2 ). Alkylation of compound 2 with methyl iodide followed by cyclization of the resulting methyl 2-[methyl(methylsulfonyl)amino]-6-(trifluoromethyl)-3-pyridinecarboxylate ( 3 ) yielded 1-methyl-7-(trifluoromethyl)-1H-pyrido[2,3-c][1,2]thiazin-4(3H)-one 2,2-dioxide ( 4 ). The reaction of compound 4 with α,2,4-trichlorotoluene, methyl bromopropionate, methyl iodide, 3-trifluoromethylphenyl isocyanate, phenyl isocyanate and 2,4-dichloro-5-(2-propynyloxy)phenyl isothiocyanate gave, respectively, 4-[(2,4-dichlorophenyl)methoxy]-1-methyl-7-(trifluoromethyl)-1H-pyrido[2,3-c][1,2]thiazine 2,2-dioxide ( 5 ), methyl 2-[[1-methyl-2,2-dioxido-7-(trifluoromethyl)-1H-pyrido[2,3-c][1,2]thiazin-4-yl]oxy]propanoate ( 6 ), 1,3,3-trimethyl-7-(trifluoromethyl)-1H-pyrido[2,3-c][1,2]thiazin-4(3H)-one 2,2-dioxide ( 7 ), 4-hydroxy-1-methyl-7-(trifluoromethyl)-N-[3-(trifluoromethyl)phenyl]-1H-pyrido[2,3-c][1,2]thiazine-3-carboxamide 2,2-dioxide ( 8 ), 4-hydroxy-1-methyl-7-(trifluoromethyl)-N-phenyl-1H-pyrido[2,3-c][1,2]thiazine-3-carboxamide 2,2-dioxide ( 9 ) and N-[2,4-dichloro-5-(2-propynyloxy)phenyl]-4-hydroxy-1-methyl-7-(trifluoromethyl)-1H-pyrido[2,3-c][1,2] thiazine-3-carboxamide 2,2-dioxide ( 10 ).  相似文献

The reactions of the tetracyclic ketone, 1,2-dihydro-11-(trifluoromethyl)-3H,7H-quino[8,1-cd] [1,5]benzoxazepin-3-one ( 1 ) with pyrrolidine, piperazine, N-methylpiperazine, and dimethyl-amine gave the enamines 2, 13, 11 , and 4 . These were reduced with sodium borohydride to the corresponding 3-amino derivatives 3, 14, 12 , and 5 . The 3-(2-hydroxyethylpiperazino) derivative ( 8 ) was obtained from the 3-chloro compound ( 7 ); 7 was prepared from the carbinol ( 6 ). The 3-NH2 derivative ( 10 ) was obtained by reduction of the oxime ( 9 ). In 3, 5, 6, 7, 8, 10, 12 , and 14 , the -OCH2 protons were non-equivalent, since in the pmr spectrum of each of these compounds there was seen a symmetrical, perturbed AB quartet, with a common JAB of 12 cps, that must be attributed to geminal interproton coupling. This phenomenon had not previously been observed with 1, 9 , or the enamines, since in their pmr spectra, the -OCH2 protons had invariably been seen as singlets.  相似文献

Michael addition of 5,11-dihydrodibenz[b,e][1,4]oxazepines to acrylonitrile gave 5,11-di-hydrodibenz[b,e][1,4]oxazepine-5-propionitriles, which were converted to the corresponding methyl 5-propionates. Mild alkaline hydrolysis of these esters furnished the key intermediates, the 5,11-dihydrodibenz[b,e] [1,4]oxazepine-5-propionic acids. In the preferred procedure, these propionic acids were cyclized to the tetracyclic ketones in yields of 60-90%, by the action of one equivalent of trifluoroacetic anhydride in benzene. When ring C was involved in cylcization, 1,2-dihydro-3H,7H-quino[8,1-cd][1,5]benzoxazepin-3-ones were obtained, and when ring A was involved, 1,2-dihydro-3H,8H-quino[1,8-ab] [4,1]benzoxazepin-3-ones were formed. With 1c , both modes of cyclization were observed. In an alternative procedure that gave lower yields, the propionic acids were first converted to the corresponding acid chlorides, and these were cyclized by means of anhydrous stannic chloride; as anticipated, the products by this method were identical with those obtained via the trifluoroacetic anhydride method. Several reactions of these tetra-cylcic ketones, e.g., their ease of oxime formation, and dehydrogenation to the α,β-unsaturated ketones, are described. The α,β-unsaturated ketones do not form oximes. The structures assigned to the ketones are based on interpretations of their pmr, uv, and ir spectra. These assignments were confirmed by an X-ray analysis of a single crystal of one representative ketone, 2b ; the X-ray study was also useful in indicating the thermodynamically preferred conformation of 2b . The structures of all the other compounds followed from similar spectral interpretations. The mass spectra of a few compounds are also discussed.  相似文献

Summary. The 1,3-dipolar intermediates generated by addition of isoquinoline, to dialkyl acetylenedicaboxylates are trapped by N-alkylisatins to produce dialkyl 1,2-dihydro-2-oxo-1-alkylspiro[3H-indol-3,2′-[2H,11bH][1,3]oxazino[2,3-a]isoquinoline]-3′,4′-dicarboxylates in excellent yields. The reaction of isoquinoline, quinoline, or pyridine with dimethyl acetylenedicarboxylate in the presence of ninhydrin led to dimethyl 1,2-dihydro-1,3-dioxospiro[3H-indene-3,2′-[2H,11bH][1,3]oxazino[2,3-a]isoquinoline]-3′,4′-dicarboxylate, dimethyl 1,2-dihydro-1,3-dioxospiro[3H-indene-3,3′[3H,4aH][1,3]oxazino[3,2-a]quinoline]-1,2-dicarboxylate, or dimethyl 1,2-dihydro-1,3-dioxospiro[3H-indene-3,2′-[2H,9aH]pyrido[2,1-b][1,3]oxazino]-3,4-dicarboxylate.  相似文献

Trifluoromethylated molecules, as well as pyrazol-3-one derivatives, are found in a wide range of biologically active compounds. An highly facile, green and mild access to substituted 3-(trifluoromethyl)-4,5-dihydro-1H-furo[2,3-c]pyrazole derivatives by the cascade [3 + 2] Michael/Alkylation of unsaturated pyrazolones with α-bromomalonate has been developed. The reactions can be performed in a easily available and environmentally benign solvent, and the substituted 3-(trifluoromethyl)-4,5-dihydro-1H-furo[2,3-c]pyrazole derivatives can be obtained in good to excellent yields by a simple purification step. The antifungal activities of these functional pyrazole derivatives were also evaluated and they showed high antifungal activity against Alternaria solani.  相似文献

Thirteen 1-(9,10-dihydro-4H-benzo[4,5]cyclohepta[1,2-b]thiophen-4-yl)-3-alkylaminoazetidines 11 have been synthesized in three steps from 4-amino-9,10-dihydro-4H-benzo[4,5]cyclohepta[1,2-b]thiophene ( 6 ), which was obtained from the reduction of either 4-azido 4 or 4-hydroxyimino 5 derivatives. All the compounds have been evaluated as potential antidepressive agents.  相似文献

The Claisen-Schmidt condensation between 1,2-dihydro-11-(trifluoromethyl)-3H,7H-quino-[8,1 -cd][1,5]benzoxazepin-3-one, 1 , and aromatic aldehydes has been investigated. The acid catalyzed reactions yielded the trans-2-benzylidene derivatives, 4 ; the structures and configurations of the group of compounds represented by 4 have been confirmed by pmr in conjunction with the Eu(fod)3 shift reagent. In contrast, catalysis with sodium hydroxide gave the isomeric 2-benzyl-endocyclic α,β-unsaturated ketones, 3. Finally, the 4 could be isomerized to the corresponding 3 by means of sodium hydroxide. The ir, uv, and pmr spectra of these compounds are discussed.  相似文献

The synthesis and reactions of methyl 2-[3-(trifluoromethyl)phenyl]-4H-furo[3,2-b]pyrrole-5-carboxylate (1a) are described. Upon reaction with methyl iodide, benzyl chloride, or acetic anhydride, this compound gave N-substituted products 1b-d. By hydrolysis of compounds 1a-c, the corresponding acids 2a-c were formed, or by reaction with hydrazine-hydrate, the corresponding carbohydrazides 3a-c were formed. By heating 2-[3-(trifluoromethyl)phenly]-4H-furo[3,2-b]pyrrole-5-carboxylic acid (2a) in acetic anhydride, 4-acetyl-2-[3-(trifluoromethyl)phenyl]furo[3,2-b]pyrrole (4) was formed. By hydrolysis of 4, 2-[3-(trifluoromethyl)phenyl]-4H-furo[3,2-b]pyrrole (5a) was formed, and reactions with methyl iodide or benzyl chloride gave N-substituted products 5b-c. The reaction of 4 with dimethyl butynedioate gave substituted benzo[b]furan 6. Compound 3a reacted with triethyl orthoesters giving 7a-c, which afforded with phosphorus (V) sulphide the corresponding thiones 8a-c. The thiones 8a-c reacted with hydrazine hydrate to form hydrazine derivatives 9a-c. The reaction of triethyl orthoformiate with compounds 9a-c led to furo[2′,3′: 4,5]pyrrolo[1,2-d][1,2,4]triazolo[3,4-f][1,2,4]triazines 10a-c. Hydrazones 11a-c were formed from 3a-c and 5-[3-(trifluoromethyl)phenyl]furan-2-carboxaldehyde. The effect of microwave irradiation on some condensation reactions was compared with “classical” conditions. The results showed that microwave irradiation shortens the reaction time while affording comparable yields.  相似文献

The synthesis of the title compounds was carried out by cyclization via isocyanate of (E)-4,5-dihydro-10H-benzo[5,6]cyclohepta[1,2-b]-thiophene-10-ylideneacetic acid and 4,5-dihydro-10H-benzo[5,6]cyclohepta[1,2-b]-thiophene-10-ylacetic acid respectively, which were obtained by the Wadsworth-Emmons modification of the Wittig reaction of 4,5-dihydro-10H-10-oxobenzo[5,6]cyclohepta[1,2-b]thiophene and triethyl phosphonacetate. 5-Amino-lH-1,2,4-triazolylcarbothiohydrazides gave β and γ-oxo-esters in boiling ethanol [1,2,4]triazolo- [1,5-d][1,2,4,6]tetrazepine-5-thiones 3 . Analogously ethyl 2-oxocyclohexanecarboxylate provided a mixture of two diastereomeric spiro derivatives 5 and 6 . At 130°, 2-acetonyl-5-methyl-4,5-dihydro-1,3,4-oxadiazole-5-thione ( 8 ) was formed. Ring closure of 3e (R1 = CH3, R2 = CH2CH2COOEt, Q = morpholino) lead to the isomeric pyrrolo[2,1-g][1,2,4]triazolo[1,5-d][1,2,4,6]tetrazepin-8(11H)-one ( 12 ) and pyrrolo[1,2-f][1,2,4]triazolo-[1,5-d][1,2,4,6]tetrazepin-10(7H)-one ( 13 ) derivatives representing two new ring systems.  相似文献

The reaction of anthranilonitrile with 2-chloroethyl isocyanate yields 2-[3-(2-chloroethyl)ureido]benzo-nitrile ( 6 ) which, upon heating, or treatment with base, undergoes a double cyclization to form 2,6-dihydroimidazo[1,2-c]quinazolin-5-(3H) one ( 8 ) in excellent yield. When heated with hydrochloric acid, 6 is converted initially into 2-(2-chloroethylamino)-4H-[3,1]benzoxazin-4-one ( 18 ) and further into 3-(2-chloroethyl)-2,4-(1H,3H)quinazolinedione ( 15 ). The acid-catalyzed reaction of 2,3-dihydro-5H-oxazolo[2,3-b]quinazolin-5-one ( 14 ) with nucleophilic reagents yields 3-substituted 2,4-(1H,3H)quinazolinediones.  相似文献

The preparation of 7-amino-2,3-dihydro-8-nitro-1H-pyrrolo[1,2-a]benzimidazole from 1,4-diacetamido-2,3-dinitrobenzene is described. Reaction of this compound with 2,5-dimethoxytetrahydrofuran produces 2,3-dihydro-8-nitro-7-N-pyrrolo-1H-pyrrolo[1,2-a]benzimidazole, which can be cyclised to produce two new heterocyclic ring systems, 9,10-dihydro-8H-pyrrolo[1,2-a]pyrrolo(1′,2′:1,2]imidazo[5,4-f]quinoxaline and 9,10-dihydro-8H-pyrrolo[2,1-c]pyrrolo[1′,2′:1,2]imidazo[4,5-h][1,2,4]benzotriazine. The corresponding diamine, 7,8-diamino-2,3-dihydro-1H-pyrrolo[1,2-a]benzimidazole undergoes a variety of condensation reactions to produce several new heterocyclic systems, for example, with formic acid, 1,7,8,9-tetrahydroimidazo-[4,5-e]pyrrolo[2,1-6]benzimidazole is formed and with diacetyl, 9,10-dihydro-2,3-dimethyl-8H-pyrrolo-[1′,2′:1,2]imidazo[5,4-y]quinoxaline is obtained.  相似文献

The transamination of 2-amino-4-imino-2-perfluoropentene with ethylenediamine gives the corresponding 6-fluoro-5,7-bis(trifluoromethyl)-2,3-dihydro-1H-1,4-diazepine. The transamination of 2-amino-4-imino-2-perfluoropentene by diethylenetriamine is accompanied by intramolecular nucleophilic substitution of the α-fluorine atom to form 1,9-bis(trifluoromethyl)-3,4,6,7-tetrahydro-2H-pyrazino[1,2-a]pyrazine whose structure was established by an X-ray structural investigation. Although the previously reported (1,2) chemical reduction of 2,3-dihydro-3-(6-nitroveratry-lidene)-4H-benzopyran-4-one with stannous chloride occurred with cyclization to the 6H-[1]-benzopyrano [4,3-b]quinoline ring system, the present study of the catalytic (palladium/carbon) reduction of 2,3-dihydro-6-methoxy-3-(6-nitroveratrylidene)-4H-benzopyran-4-one ( 1 ) (3) has indicated that other products in addition to the expected benzopyranoquinoline ( 3 ) may be isolated, depending upon the conditions of the reduction. The products of the reduction of 1 , isolated and structurally determined, include 2,9,10-trimethoxy-6H-[1]benzopyrano-[4,3-b]quinoline(3),2,9,10-trimethoxy-6a,7,12,12a-tetrahydro-6H-[1]benzopyrano[4,3-b]-quinoline (2), the N-oxide of 3 (6 ), and 6-hydroxy-2,9,10-trimethoxy-6H-[l]benzopyrano-[4,3-b Jquinoline ( 8 ).  相似文献

The reaction of sulfene with N,N-disubstituted 3-aminomethylene-2,3-dihydro-4-thiochromanones and-2,3,5,6-tetrahydro-4-thiopyranones gave 1,4-cycloadducts which are derivatives of new heterocyclic systems, namely 3,4-dihydro-5H-[1]benzothiopyrano[3,4-e]-1,2-oxathiin and 3,4,7,8-tetrahydro-5H-thiopyrano[3,4-e]-1,2-oxathiin, respectively. The reaction of benzoyl isothiocyanates and methoxycarbonyl isothiocyanate with 4-amino-4,5-dihydro-3-(methylthio)-1,2,4-triazin-5-ones in acetonitrile gave several substituted 4H-1,3,4-thiadiazolo[2,3-c]-1,2,4-triazin-4-ones VIa-h instead of the expected thioureas. Derivatives of two new molecular structures, namely, 7,8-dihydro-6H,10H-[1,2,5]thiadiazolo[3′,4′:4,5]pyrimido[2,1-b][1,3]thiazin-10-one and 6,7-dihydro-9H-thiazolo[3,2-a][1,2,5]thiadiazolo[3,4-d][pyrimidin-9-one, and derivatives of N-substituted sulfamic acid, namely, (8-amino-3,4-dihydro-2H,6H-pyrimido[2,1-b][1,3]thiazin-6-on-7-yl)sulfamic acid and (7-amino-2,3-dihydro-5H-thiazolo[3,2-a]pyrimidin-5-on-6-yl)sulfamic acid, were separated out as by-products in the reduction reaction of 8-amino-3,4-dihydro-7-nitroso-2H,6H-pyrimido[2,1- b][1,3]thiazin-6-one and 7-amino-2,3-dihydro-6-nitroso-5H-thiazolo[3,2-a]pyrimidin-5-one derivatives, respectively, with sodium hydrosulfite. A mechanism of reaction, which hypothesizes the action of sodium hydrosulfite in an asymmetic form, is proposed. The results of single-crystal X-ray investigation on 7,8-dihydro-6H,10H-[1,2,5]thiadiazolo[3′,4′:4,5]pyrimido[2,1-b][1,3]thiazin-10-one (R = 0.032 for 863 reflections) and (8-amino-3,4-dihydro-2H,6H-pyrimido[2,1-b]- [1,3]thiazin-6-on-7-yl)sulfamic acid, sodium salt (R = 0.028 for 3507 reflections) are reported.  相似文献
