The imidazol side group of histidine has two nitrogen atoms capable of being protonated or participating in metal binding. Hence, histidine can take on various metal-bound and protonated forms in proteins. Because of its variable structural state, histidine often functions as a key amino acid residue in enzymatic reactions. Ab initio (HF and MP2) calculations were done in modeling the cation (H+, Li+, Na+, K+, Mg2+, Ca2+) interaction with side chain of histidine. The region selectivity of metal ion complexation is controlled by the affinity of the side of attack. In the imidazol unite of histidine the ring nitrogen has much higher metal ion (as well as proton) affinity. The complexation energies with the model systems decrease in the following order: Mg2+ > Ca2+ > Li+ > Na+ > K+. The variation of the bond lengths and the extent of charge transfer upon complexation correlate well with the computed interaction energies.  相似文献

Sulphoxide ligands in piperidinium based ionic liquid were demonstrated as highly efficient, selective and environmentally benign systems for the extraction of plutonium from acidic aqueous solution. The extraction followed ‘cation-exchange mechanism’ via [Pu(NO3)·L]3+ and [PuO2(NO3)·L]+ species. The extraction efficiency followed the trend: APSO > BPSO > BMSO. The phenyl substituted sulphoxides showed higher affinity for plutonium due to a combination of steric as well as electronic factors. Extraction process was thermodynamically spontaneous for all three solvent systems. Oxalic acid and sodium carbonate were suitable for quantitative stripping of Pu4+ and PuO2 2+, respectively. APSO in ionic liquid showed good radiolytic stability.  相似文献

Summary A highly efficient and an optimized synthesis of [1-14C]lauric acid with high specific activity (50 mCi/mmol) is described. [1-14C]lauric acid was prepared from [1-14C]lauronitrile, in 2 minutes with a mixture of concentrated hydrochloric acid: propionic acid (1: 2 v/v) under microwave irradiation, in quantitative yield.  相似文献

Copper-64 was produced as a by-product of 55Co via 64Ni(p,n)64Cu by 15 MeV proton bombardment of natNi resulting in a thick target yield of 5.31 MBq/μAh (143.5 μCi/μAh) and a radiochemical separation yield of 95% (radionuclide purity >97% after 25 hours of bombardment). Rituximab was successively labeled with [64Cu]-CuCl2. N-succinimidyl-1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid (DOTA-NHS) was prepared at 25 °C using DOTA and N-hydroxy succinimide (NHS) in CH2Cl2 followed by the addition of 1 ml of a Rituximab pharmaceutical solution. Radiolabeling was performed at 37 °C in 3 hours. Radio thin-layer chromatography showed an overall radiochemical purity of 90–95% at optimized conditions (specific activity=30 GBq/mg, labeling efficacy; 82%) using various chromatography systems. The final isotonic 64Cu-DOTA-Rituximab complex was passed through a 0.22 μm filter and checked by gel electrophoresis for radiolysis control. Stability of the final product was checked in the formulation and in presence of human serum at 37 °C.  相似文献

A homemade modular PC-controlled radiochemistry system, used to separate the no-carrier-added 64Cu from irradiated electroplated solid cyclotron target 64Ni layers was developed. Upon irradiation the target layer is dissolved in diluted nitric acid, followed by selective liquid–liquid extraction of the 64Cu. The final purification step is achieved making use of an anion exchange column. All the separation procedures can be carried out remote-controlled with a minimized risk of operator errors within less than 2 h, with excellent processing yield (>95%).  相似文献

Ion-exchange sorption of K+ and Na+ from their mixture on X-ray-amorphous and crystalline Sn(IV) hydrophosphates was studied. These sorbents exhibit a high selectivity for K+. Microwave assisted direct aromatic substitution of 3-bromopyridine with K14CN as the cyanide source and catalytic amount of tetrabutylammonium bromide afforded [3-14C]-cyanopyridine 3 in 90% yield. Microwave assisted hydrolysis of 3 with a mixture of concentrated hydrochloric acid and propionic acid afforded [carboxyl-14C]-nicotinic acid in 95% yield whereas microwave assisted hydrolysis of 3 with a mixture of concentrated sulfuric acid and propionic acid afforded [carbonyl-14C]-nicotinamide in 85% yield.  相似文献

This article presents, 122Sb (T 1/2 = 2.723 days, I β- = 97.59%) was produced via the natSn(p,xn) nuclear process at the AMIRS (Cyclone-30, IBA, Belgium). The electrodeposition experiments were carried out by potassium stannate trihydrate (K2Sn(OH)6) and potassium hydroxide. The optimum conditions of the electrodeposition of tin were as follows: 40 g/L natSn, 20 g/L KOH, 115 g/L K2Sn(OH)6, DC current density of 5 A/dm2 with a bath temperature of 75 °C. The electroplated Tin-target was irradiated with 26.5 MeV protons at current of 180 μA for 20 min. Solvent extraction of no-carrier-added 122Sb from irradiated Tin-natural target hydrochloric solution was investigated using di-n-butyl ether (C8H18O). Yields of about 3.61 MBq/μAh were experimentally obtained.  相似文献

A rapid bioassay for 90Sr was developed involving preconcentration of 90Sr/90Y from human urine samples with a cation exchange polymer (poly–acrylamido–methyl–propanesulfonic acid) coated onto magnetic nanoparticles, followed by selective elution of 90Sr (over 90Y) with phosphate for determination by liquid scintillation analysis. The minimum detectable activity for this method (4.9 ± 0.5 Bq/L) is lower than the required sensitivity of 19 Bq/L for 90Sr in human urine samples, as defined in the requirements for radiation emergency bioassay techniques for the public and first responders based on the dose threshold for possible medical attention recommended by the International Commission on Radiological Protection. The relative bias was 9.2%, the relative precision was 3.2%, and the linear dynamic range covered 12–600 Bq/L. This simple and rapid bioassay method is found to be in compliance with the HPS ANSI N13.30 performance criteria for radiobioassay.  相似文献

Summary An attractive and simple method has been developed for the preparation of ammonium [14C]thiocyanate from [14C]thiourea which eliminates the necessity of handling highly hazardous potassium [14C] cyanide. [14C]thiourea was isomerized to ammonium [14C]thiocyanate by heating the aqueous solution of thiourea (12%) in a sealed tube at 160 °C for 24 hours. The product formed was purified by silica-gel column chromatography. A radiochemical yield of 92.7% was obtained based on [14C]thiourea. The specific activity of the product obtained was 53.3 mCi/mmol (1.97 GBq/mmol) and the radiochemical purity was greater than 99%. This method has not been reported so far for the production of this labeled compound.  相似文献

Effects of La3+ and Eu3+ on outward potassium channels (Kout+) in Vicia guard cells have been studied by patch clamping technique. Extracellular La3+ inhibited Kout+ currents with a half-inhibition concentration (IC50) of 81 μmol·L−1. Interestingly, intracellular La3+ activated Kout+ currents at a free concentration of 1.13 × 10−14 mol·L−1, and inhibited Kout+ currents at a free concentration of 5.86 × 10−14 mol·L−1. Extracellular Eu3+ also activated Kout+ currents at concentrations of 10 μmol·L−1 and 50 μmol·L−1, and inhibited Kout+ currents at concentrations of more than 1 mmol·L−1. The effects of La3+ and Eu3+ on Kout+ currents may contribute to regulation of the plant water status, which may be one of the mechanisms of the biological effect of rare earth elements.  相似文献

A novel quinoline derivative, 2,2′-[(5-chloro-8-hydroxyquinoline-7-yl) methylazanediyl] diacetic acid (CHQMADA) was labeled with 99mTc using SnCl2·2H2O as a reducing agent to give a complex with a labeling yield 94 %. Also [99mTc(H2O)3(CO)3]+ was prepared by heating at 100 °C for 30 min using 2 mg CHQMADA at pH 8 to give a labeling yield >99 %. 99mTc-(CO)3 CHQMADA and 99mTc-Sn(II)-CHQMADA showed tissue uptake (target to non target T/NT = 6.80 ± 0.22) and (T/NT = 5.65 ± 0.34) respectively in Escherichia coli induced infection, which is higher than the commercially available 99mTc-ciprofloxacin (T/NT = 3.80 ± 0.80). In conclusion, both complexes were able to differentiate between septic and aseptic inflammation with superiority of [99mTc-(CO)3 CHQMADA].  相似文献

Rituximab was successively labeled with 177Lu-lutetium chloride. 177Lu chloride was obtained by thermal neutron flux (4 × 1013 n cm−2 s−1) of natural Lu2O3 sample with a specific activity of 2.6–3 GBq/mg. The macrocyclic bifunctional chelating agent, N-succinimidyl-1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid (DOTA-NHS) was prepared at 25 °C using DOTA, N-hydroxy succinimide (NHS) in CH2Cl2. DOTA-rituximab was obtained by the addition of 1 mL of a rituximab pharmaceutical solution (5 mg/mL, in phosphate buffer, pH 7.8) to a glass tube pre-coated with DOTA-NHS (0.01–0.1 mg) at 25 °C with continuous mild stirring for 15 h. Radiolabeling was performed at 37 °C in 24 h. Radio-thin layer chromatography showed an overall radiochemical purity of >98% at optimized conditions (specific activity = 444 MBq/mg, labeling efficacy; 82%). The final isotonic 177Lu-DOTA-rituximab complex was checked by gel electrophoresis for structure integrity control. Radio-TLC was performed to ensure that only one species was present after filtration through a 0.22 μm filter. Preliminary biodistribution studies in normal rats were carried out to determine complex distribution of the radioimmunoconjugate up to 168 h. The biodistribution data were in accordance with other antiCD20 radioimmunoconjugates already reported.  相似文献

Several human tumors such as neuroendocrine tumors, medullary thyroid carcinoma, etc., express somatostatin receptors which specifically bind somatostatin and its analogues such as lanreotide, octreotide, etc. In order to prepare a therapeutic agent for targeting such tumors, attempts were made to prepare 90Y-DOTA-Lanreotide. Lanreotide could be successfully conjugated with the macrocyclic chelating agent DOTA (1,4,7,10-tetraaza cyclododecane tetracetic acid) which forms stable complexes with 90Y. 90Y-DOTA-Lanreotide could be prepared in >98% radiochemical purity and remained stable for 72 hours at room temperature. The tracer showed specific binding to A431 cells. Biodistribution studies in C57BL6 mice bearing melanoma showed ∼1.3% uptake pergram of tumor at 24-hour p.i.  相似文献

A preparation of 125I-celecoxib is carried out via an electrophilic substitution reaction. The reaction parameters studied were celecoxib concentration, reaction temperature, pH of the reaction mixture and kinds of oxidizing agents in order to obtain a high radiochemical yield of the 125I-celecoxib. Using 3.7 MBq of Na125I, 150 μg (3.9 mM (mmol/L)) of celecoxib, and 1.6 mM (mmol/L) of chloramine-T (CAT) as oxidant at pH 4 and 60 °C for 15 min a maximum radiochemical yield of 125I-celecoxib (65%) was obtained. The labeled compound was separated and purified by means of high pressure liquid chromatography (HPLC). The biological distribution in infected mice indicates the suitability of radioiodinated celecoxib as imaging of tumor.  相似文献

It is regarded that the spent resins from the water purification systems of moderator (MOD) and the primary coolant of the Canada deuterium uranium-pressurized heavy water reactor (CANDU-PHWR) are a unique waste, owing to their high 14C and gamma-emitting nuclides. In this work, 14C and 3H contents, anion and cation fractions and the predominant gamma-emitting nuclides of the spent resins from 4 units of CANDUPHWRs, were investigated. Also the chemical species of 14C of the spent resins were determined. For a simultaneous separation of 14C and 3H from the spent resins, the wet oxidation-16 wt% H2SO4 stripping process was utilized. The 14C and 3H activity concentration range of the spent resins of the nuclear power plant (NPP), 4 units of all CANDU-PHWR types, was 2.48E5 Bq/g ∼5.33E6 Bq/g, 1.29E5 Bq/g and ∼2.33E5 Bq/g, respectively. Among the analyzed spent resins, the highest 14C and 3H activity concentration was detected in units 4 and 3, respectively. It was found that more than 92% of the 14C activity concentration was retained on the anion resin and the predominant chemical species was inorganic 14C. It was revealed that the anion resin fraction of the spent resins from unit 1 and unit 2, was about 40% and that of unit 3 and unit 4 was around 60%. More than 80% of the total gamma-radioactivity concentration was associated with the cation fraction of the spent resin. The predominant gamma-emitting nuclide of the spent resin for unit 2 was 137Cs, a fission product, and that for unit 4 was 60Co, a corrosion product.  相似文献

Several procedures for preparation of the 95mTc radiotracer following irradiation of a thin Mo target with deuterons were tested. The procedures consisting of alkaline-oxidative fusion of the irradiated target in a mixture of Na2O2 + NaOH and subsequent liquid–liquid extraction with 2-butanone, and acid decomposition of the target in a mixture of H2SO4 + HNO3 followed by extraction chromatography with PAN-Aliquat 336 composite material appeared suitable for the given purpose.  相似文献

Bleomycin (BLM) has been labeled with various radioisotopes and widely used in therapy and diagnosis. 191Os is a parent radionuclide with 15.4 day half-life and decays by beta emission to 191mIr, which is a radionuclide with 4.96 s half-life. BLM was labeled with 191Os-hexachloro-osmate and its distribution and stability in wild-type mice was determined. The complex was obtained at the pH 2 in normal saline at 90 °C in 48 h. Radio-TLC showed an overall radiochemical yield of 93–97%, radio-chemical purity > 97%. The biodistribution study for 191Os-hexachloro-osmate and 191Os-BLM were carried in wild type-mice up to 14 days. Lungs, liver and spleen uptake increased 24–72 h after administration of 191Os-BLM. 24 h after administration, the radioactivity of the kidney increased and remained constant.  相似文献

3H and 14C Measurements of the dry active waste (DAW), such as the cotton, paper, and vinyl, generated from a nuclear power plant (NPP) were conducted with wet oxidation using open vessel equipment based on simulation results. The recovery efficiency with the simulated samples was around 93% with a relative standard deviation (RSD) of 1–3%. A liquid scintillation counter (LSC) was used for counting and adjusted to a quenching correction curve. The counting value was evaluated for the minimum detectable activity (MDA), which was found to be about 4 × 10−1 Bq/g for 3H and 2 × 10−2 for 14C when approximately 5 g of the samples were measured. The measured DAW samples for the cotton, paper, and vinyl generated from NPP achieved of RSD values of 25, 25, and 60%, respectively, for 3H and 0–50% for 14C.  相似文献

Summary We report here a facile synthesis of [1-14C]-mandelic acid. Benzyl bromide on cyanation with K14CN followed by acid hydrolysis in a microwave oven gave [1-14C] phenylacetic acid. The latter on a-bromination followed by hydrolysis under microwave irradiation for 2 minutes furnished the title compound.  相似文献
