Exposure to artificial UV wavelengths and the UV component of sunlight delays positive phototaxis in the green alga Volvox aureus. Broad band wavelength filters were used to modify the output from UV-B sources (280-320 nm) and natural sunlight. The delay in phototaxis by artificial UV is increased with exposure to shorter UV-B wavelengths. Natural sunlight experiments were performed with exposure to full sunlight and to its UV component only. The UV component present in summer sunlight produced long periods of inhibition in phototaxis and even lethality, while exposure to the total spectrum of sunlight had no significant effects on movement or survival. The data indicate that although this species of alga is well equipped to deal with present levels of UV exposure, increases in the short UV-B wavelengths in sunlight may force an alteration in patterns of photomovement.  相似文献

Abstract— Dry cysts of Colpoda inflata were irradiated with an argon plasma arc lamp, the output of which simulates space radiation in its wavelength distribution although it is of higher intensity. Rapid evacuation reduced the rate of excystment as well as the percentage excysting; low temperature (near that of liquid nitrogen) had little effect on either measurement. All cysts were therefore irradiated at room temperature in a cell flushed with dry nitrogen. Radiation passed for 1 min through a sapphire filter, with a cutoff at 145 nm, prevented excystment. At smaller doses progressively less damaging effects were observed as judged by both the increase in time required for excystment and the percentage excysted. No dark recovery was observed in dry cysts. Exposure of cysts to radiation passed through a series of filters which progressively remove short ultraviolet wavelengths indicate that it is mainly the quartz-transmitted wavelengths (cutoff 185 nm) which damage the cysts. The presumption is that most of the vacuum-ultraviolet wavelengths are superficially absorbed; their lesser effect is also partially attributable to their lower intensity. Inasmuch as 1 min of argon lamp radiation is equivalent to 24 min in space, it is evident that Colpoda cysts would not long survive a journey in space, had they reached there in viable condition.  相似文献

In previous work, we evaluated the effects of ultraviolet (UV = 280-400 nm) radiation on the early life stages of a planktonic Calanoid copepod (Calanus finmarchicus Gunnerus) and of Atlantic cod (Gadus morhua). Both are key species in North Atlantic food webs. To further describe the potential impacts of UV exposure on the early life stages of these two species, we measured the wavelength-specific DNA damage (cyclobutane pyrimidine dimer [CPD] formation per megabase of DNA) induced under controlled experimental exposure to UV radiation. UV-induced DNA damage in C. finmarchicus and cod eggs was highest in the UV-B exposure treatments. Under the same spectral exposures, CPD loads in C. finmarchicus eggs were higher than those in cod eggs, and for both C. finmarchicus and cod embryos, CPD loads were generally lower in eggs than in larvae. Biological weighting functions (BWF) and exposure response curves that explain most of the variability in CPD production were derived from these data. Comparison of the BWF revealed significant differences in sensitivity to UV-B: C. finmarchicus is more sensitive than cod, and larvae are more sensitive than eggs. This is consistent with the raw CPD values. Shapes of the BWF were similar to each other and to a quantitative action spectrum for damage to T7 bacteriophage DNA that is unshielded by cellular material. The strong similarities in the shapes of the weighting functions are not consistent with photoprotection by UV-absorbing compounds, which would generate features in BWF corresponding to absorption bands. The BWF reported in this study were applied to assess the mortality that would result from accumulation of a given CPD load: for both C. finmarchicus and cod eggs, an increased load of 10 CPD Mb(-1) of DNA due to UV exposure would result in approximately 10% mortality.  相似文献

We examine the Breit-Wigner-Fano (BWF) line shape in the Raman spectra of carbon single-wall nanotubes (SWNTs) dispersed in aqueous suspensions. Bundling and electronic effects are studied by comparing undoped SWNTs (C-SWNTs) to boron-doped nanotubes (B-SWNTs) in a variety of different surfactant solutions. For SWNTs dispersed with nonionic surfactants that are less effective in debundling than ionic surfactants, the Raman spectra retain a large BWF feature. However, we demonstrate that even for SWNTs dispersed as isolated nanotubes by ionic surfactants the BWF feature may be present and that the intensity of the BWF is highly sensitive to the specific surfactant. In particular, surfactants with electron-donating groups tend to enhance the BWF feature. Also, modification of the SWNT electronic properties by boron doping leads to enhanced surfactant dispersion relative to undoped C-SWNTs and also to modification of the BWF feature. These observations are in agreement with reports demonstrating an enhancement of the BWF by bundling but also agree with reports that suggest electron donation can enhance the BWF feature even for isolated SWNTs. Importantly, these results serve to caution against using the lack or presence of a BWF feature as an independent measure of SWNT aggregation in surfactant dispersions.  相似文献

We performed in situ experiments during the austral summer of 1998 to quantify the mortality of the fresh-water copepod Boeckella gracilipes as a function of the UV dose. The copepods were exposed to solar radiation at the water-surface for approximately 24-34 h. Long-pass cut-off filters (Schott) were used in the exposure experiments. UV radiation and PAR were measured with an IL-1700 (International Light Inc.) and a PUV-500 radiometer (Biospherical Instruments Inc.). A biological weighting function for UV-induced mortality was calculated by fitting a model based on a logistic curve. Our results show that UV damage in this species is strongly wavelength- and dose-dependent. B. gracilipes was highly vulnerable to both UV-B (290-320 nm) and UV-A radiation (< 360 nm). The shape of the BWF obtained for B. gracilipes resembles more closely the action spectra (AS) for UV-induced erythema, than the AS for naked DNA.  相似文献

The degree of ultraviolet enhanced reactivation (UVR) exhibited by mammalian cells when infected with Herpes simplex virus inactivated by different wavelengths of far ultraviolet (UV) radiation was measured. A wavelength dependence for this effect is presented over the wavelength region 238–297 nm. Within the limits of the deviations obtained, the degree of UVR exhibited is similar at each wavelength. This suggests that virus irradiated with different wavelengths of UV radiation received the same type of damage or that cells repaired the different types of viral damage with the same efficiency.  相似文献

The effect of UV-B radiation (UVBR, 280-315 nm) on growth rate during 72 h of incubation, was measured for two marine microalgae -Dunaliella tertiolecta (Chlorophyceae) and Phaeodactylum tricornutum (Bacillariophyceae). The resulting inhibition of growth rate was analysed by calculating biological weighting functions (BWFs). The growth rate of D. tertiolecta was slightly more inhibited by UVBR (over the whole range of the spectrum) than was the growth rate of P. tricornutum, but the wavelength dependencies were the same. Our results were compared with results from photosynthesis experiments of Andreasson and W?ngberg [1] , where two methods, pulse amplitude modulation (PAM) fluorescence and carbon fixation, were measured for these same algae. The BWF for the growth rate, here, showed more wavelength dependency than the BWF for the previous two photosynthesis measurements - except for the carbon fixation BWF in P. tricornutum, which was closer to the BWF for growth rate. The wavelength dependency of the growth rate inhibition showed less variation between the species than the inhibition of the photosynthesis.  相似文献

The G band in Raman spectra of single-wall carbon nanotube (SWNT) bundles is studied between 3 and 500 K. The G band is best fit with five Lorentzian lines and one Breit–Wigner–Fano (BWF) line, indicating coupling of phonons to the electronic continuum of metallic SWNTs. It is found that the line width of the BWF line decreases with increasing temperature. This temperature-dependent behavior is contrary to that of the Lorentzian lines, where the line width increases with increasing temperature. The coupling constant 1/q of the BWF line is also found to decrease with increasing temperature. These temperature-dependent behaviors of BWF line provide evidence that it is the bundling effect of SWNTs that greatly enhances the BWF coupling.  相似文献

Laser-induced breakdown thresholds for several pure metals were determined using a nanosecond laser. A Q-switched pulsed Nd:YAG laser operating at infrared (1064 nm), visible (532 nm) and ultraviolet (266 nm) wavelengths has been used. The plasma was generated by focusing the Nd:YAG laser on the target in air at atmospheric pressure. The dispersed plasma light was detected using a two-dimensional intensified charge-coupled device (CCD) detector. The studied elements were chosen according to their different thermal and physical properties, particularly boiling point, melting point and thermal conductivity. The effect of wavelength on the plasma threshold has been discussed. Laser fluence thresholds in the ultraviolet were larger than those obtained using visible and infrared radiation, while the energy threshold is larger using infrared radiation. Correlations between the plasma threshold of metal targets and the melting point and boiling point at 266, 532 and 1064 nm have been established. The results indicate that thermal effects have an important influence on the ablation behavior of metals at the three wavelengths used.  相似文献

Photomovement measurements were carried out with swarmers of the brown algae Scytosiphon lomentaria (Lyngb.) Link and Petalonia fascia (O. F. Müll.) as a function of irradiance direction, photon irradiance, spectral composition and ultraviolet radiation (UVR, lambda=280-400 nm) dose. Swarmers from both species showed similar photomovement patterns: negative phototaxis occurred under photon irradiances of 10-90 micromol photons m(-2) s(-1), and no movement was observed at 190 micromol photons m(-2) s(-1). The translocational velocity measured between 10 and 90 micromol m(-2) s(-1) ranged from 100 to 200 microm s(-1). The accumulation of swarmers presented a peak at 450 nm (waveband of 50 nm), and smaller peaks at 400 and 500 nm; no effect was observed at wavelengths of 550 nm and above. The decline in phototactic index (an estimator of photomovement response) of swarmers was linearly correlated with the logarithm of UVR doses. These data were correlated with levels of natural solar radiation in the field. It is hypothesized that motility of swarmers could be a critical factor in the survival of these species under a scenario of increased UVR.  相似文献

Abstract—Light-induced behavioral responses of Euglena gracilis have been investigated in single cells by means of a video system coupled to an optical microscope. Light intensity-effect curves at different wavelengths in the near UV and visible range have been determined. From these curves the action spectrum for the step-down photophobic response of Euglena has been calculated. From a comparison with the results obtained using a population method by means of a phototaxigraph, it is concluded that a single photomotile reaction is responsible for cell accumulation, brought about by trapping in the light spot and possibly by phototaxis towards scattered light from organisms already in the light field.  相似文献

To estimate the inhibitory effect of the changing UVB radiation (UVBR, 280-315nm) on earth's ecosystems, an understanding of its wavelength dependency is needed. The tool used for these estimations is the biological weighting function (BWF), whereby the inhibition of different wavelengths is calculated. BWFs were determined for three algae species from different classes, Phaeodactylum tricornutum (Bacillariophyceae), Dunaliella tertiolecta (Chlorophyceae) and Rhodomonas sp. (Cryptophyceae), using polychromatic irradiation, where the UVBR spectra were varied with cut-off filters. For each alga, BWFs were determined for two photosynthetic parameters; the quantum yield measured as fluorescence from Photo System II in a pulse-amplitude-modulation (PAM) fluorometer, and the fixation of (14)C-labelled carbon dioxide. The BWFs were calculated with the Rundel method, using the radiation data between 270 and 360nm with 1nm resolution. The results show that the UVBR damages were generally higher when using the carbon fixation measurements than when measuring with the PAM technique. When using PAM, P. tricornutum in particular had a sensitivity intermediate between the sensitive Rhodomonas sp. and the more tolerant D. tertiolecta, but was as sensitive as, or even more sensitive, than Rhodomonas sp. when using carbon fixation. D. tertiolecta was shown to be less sensitive when using both techniques and the inhibition of its photosynthesis was almost as high when using PAM as when using carbon fixation. We concluded that, although the PAM technique has advantages such as being cleaner and easier to use, it is unable to substitute the carbon fixation measurements. Not only are the algae less sensitive when measured with PAM than they are when measured as carbon fixation, the relationship between the effects on the algae measured with the two techniques also differs. As fixation of carbon dioxide integrates a larger part of the photosynthetic machinery, it should be favoured as a measure of photosynthesis.  相似文献

The effect of different wavelengths of ultraviolet (UV) radiation on Herpes simplex virus when assayed on mammalian cells (measured by plaque forming ability) was investigated. The wavelength dependence of viral inactivation was obtained for 11 different wavelengths over the region 238–297 nm. The resulting action spectrum does not closely follow the absorption spectrum of either nucleic acid or protein. The most effective wavelengths for viral inactivation are over the region 260–280 nm.  相似文献

The unicellular flagellate Euglena gracilis shows positive phototaxis at low fluence rates (≤10 W m 2) and negative phototaxis at high fluence rates (≥100 W m 2). Currently, retinal or flavins/pterins are discussed as chromo-phores of the photoreceptor. When grown in the presence of 4 mM nicotine, a retinal inhibitor, for several generations, the cells still showed both responses, indicating that retinal is unlikely to be the chromophoric group of the photoreceptor responsible for phototaxis. The native flavin(s) can be substituted by growing the cells in roseo-flavin dissolved in the medium. The absorption spectrum of roseoflavin extends well beyond the action spectrum for phototaxis (up to 600 nm). Excitation at wavelengths >550 nm does not cause phototactic orientation in control cells but causes both positive and negative phototaxis in roseoflavin-grown cells, indicating an uptake and assembly of the chromophore in the photoreceptor complex. The white mutant strain 1224-5/1f, induced by streptomycin treatment, lacks flavins as indicated by fluorescence spectroscopy. The phototaxis-deficient pheno-type cannot be complemented by the addition of external riboflavin. Fluorescence spectra of intact paraxonemal bodies (PAB) indicate that both pterins and flavins are involved in photoperception and that the excitation energy is efficiently funneled from the pterins to the flavins. This energy transfer is disrupted by solubilization of the PAB. In intact PAB flavins are not accessible to reducing or oxidizing substances, indicating that they are located inside the structure, while pterins are accessible, so that their localization can be assumed to be on the surface. The results described above are discussed with regard to the potential involvement of flavins and pterins as well as retinal in photoperception.  相似文献

Abstract —Ultraviolet (UV) action spectra were obtained for lethality and mutagenesis (reversion to tryptophan independence) in Escherichia coli WP2s for wavelengths 254–405 nm with detailed analysis in the UVB region (290–320 nm). Parallel chemical assay yields of pyrimidine dimers in DNA of E. coli RT4 were determined at the same wavelengths. Spectral regions isolated from a Xe arc and resonance lines from a high-pressure Hg-Xe arc lamp were both used for irradiation. In all cases, precise energy distributions throughout the isolated Xe bands regions were defined.
Lethality, mutagenesis, and dimer induction all decreased in efficiency in a similar fashion as the wavelengths of the radiation increased. Between 300 and 320 nm, all characteristics measured showed differences of about two and a half orders of magnitude. Between these wavelengths, the values of the three end points used either coincide with or parallel the absorption spectrum of DNA. The mutagenesis action spectrum coincides closely with the absorption spectrum of DNA. The lethality spectrum is closely parallel to the mutagenicity spectrum; the points, however, consistently occur at about 2 nm longer wavelengths. A calculation derived from the slope of the UVB spectra reveals that a 1-nm shift of the solar UV spectrum to shorter wavelengths would result in a 35% increase in its mutagenic potential. At 325 nm, both biological action spectra show sharp decreases in slope. In addition, above 325 nm the spectra for lethality. mutagenicity, and dimer formation diverge sharply; lethalities at these UVA wavelengths were approximately tenfold greater relative to mutagenicity than at shorter wavelengths. The relative yield of dimer formation by 365 nm radiation is intermediate between the yields for lethality and mutagenesis.  相似文献

The UVC-induced synthesis of hydrogen peroxide (H2O2) by cultured rose (Rosa damascena Mill, cv Gloire de Guilan) cells is inhibited by broad-band radiation containing UVA, UVB and blue wavelengths. In order to understand the basis for this inhibition, we tested the effects of broad-band radiation and of monochromatic radiation on NADH- and NADPH-dependent O2- synthases of purified plasma membranes. Both activities were inhibited by wavelengths from 254 to 435 nm, with the NADPH-dependent enzyme being more sensitive than the NADH-dependent enzyme. Incubation in the presence of flavin adenine dinucleotide had a small but significant ameliorative effect on the inhibition of the O2- synthase activities of plasma membranes by 300 nm radiation. The data support the hypothesis that the NADH- and NADPH-dependent activities are catalyzed by different enzymes. The NADPH-dependent activity, but not the NADH-dependent activity, was sufficiently sensitive to account for the inhibition of H2O2 synthesis by the broad-band radiation.  相似文献

Photoreceptor pigments have been isolated biochemically from the freshwater dinoflagellate Peridinium gatunense, and characterized spectroscopically. At least four different chromoproteins can be detected in the crude extract and the membrane fraction isolated from the cells absorbing at 580, 638, 667 and 710 nm, which correspond with the maxima in the action spectrum for phototaxis in this organism. Light energy absorbed by shorter wavelength pigments is emitted as fluorescence at wavelengths which are absorbed by pigments with maxima at longer wavelengths. Protein separation on a MonoQ anion exchanger column using fast liquid chromatography resulted in a non-bound fraction and four bound fractions eluted by an NaCI gradient, which differed in their pigment composition.  相似文献

We describe a general method of finding the transport properties of molecules in polymer films by photolysis. Poly(methyl methacrylate) samples held at different temperatures are exposed to UV radiation at discrete wavelengths, and the time evolution of the volatile photoproducts are detected with a quadrupole mass spectrometer. A diffusion model is used to fit the experimental data and deduce diffusion coefficients for the main photoproduct methyl formate. The average value at room temperature was equal to 1.9 x 10 (-11) cm (2) s (-1) at all wavelengths investigated. Together with the values derived at other temperatures, an Arrhenius plot was obtained and the activation energy for methyl formate diffusion within the polymeric thin film calculated from the slope of the graph. We envision that this new method will find application to a variety of problems involving the mass transport of molecules through boundary layers of single or multilayer thin film structures.  相似文献
