Gold nanoelectrode ensembles were produced by electrodeposition using multiwalled carbon nanotubes (MWNTs) as template. A new third generation amperometric biosensor for hydrogen peroxide was developed based on adsorption of horseradish peroxidase (HRP) at the glassy carbon (GC) electrode modified with Au nanoelectrode ensembles/multiwalled carbon nanotubes/chitosan film. The resulting HRP biosensor offered an excellent detection for hydrogen peroxide at −0.11 V with a linear response range of 2.08 × 10−7 to 7.6 × 10−3 M with a correlation coefficient of 0.998, and response time <5 s. The detection limit was 1.02 × 10−7 M at 3σ. The biosensor displays rapid response, expanded linear response range, and excellent repeatability. The simple and fast fabrication of the sensor makes it superior to other techniques.  相似文献

Abstract

Multiwalled carbon nanotubes (MWNTs) were treated with a mixture of concentrated sulfuric and nitric acid to introduce carboxylic acid groups to the nanotubes. Conducting polymer film was prepared by electrochemical polymerization of neutral red (NR). By using a layer‐by‐layer method, homogeneous and stable MWNTs and poly (neutral red) (PNR) multilayer films were alternately assembled on glassy carbon (GC) electrodes. With the introduction of PNR, the MWNTs/PNR multilayer film system showed synergy between the MWNTs and PNR, with a significant improvement of redox activity due to the excellent electron‐transfer ability of carbon nanotubes (CNTs) and PNR. The electropolymerization is advantageous, providing both prolonged long‐term stability and improved catalytic activity of the resulting modified electrodes. The MWNTs/PNR multilayer film modified glassy carbon electrode allows low potential detection of hydrogen peroxide with high sensitivity and fast response time. As compared to MWNTs and PNR‐modified GC electrodes, the magnitude of the amperometric response of the MWNTs/PNR composite‐modified GC electrode is more than three‐fold greater than that of the MWNTs modified GC electrode, and nine‐fold greater than that of the PNR‐modified GC electrode. With the immobilization of glucose oxidase onto the electrode surface using glutaric dialdehyde, a biosensor that responds sensitively to glucose has been constructed. In pH 6.98 phosphate buffer, nearly interference‐free determination of glucose has been realized at ?0.2 V vs. SCE with a linear range from 50 µM to 10 mM and response time <10s. 基于Pt纳米颗粒修饰直立的碳纳米管电极制备了葡萄糖生物传感器.铂纳米颗粒是利用电位脉冲沉积法修饰到直立碳纳米管上的,可以增强电极对酶反应过程当中产生的过氧化氢的催化行为.用扫描电镜和透射电镜观察了直立碳纳米管在修饰Pt纳米颗粒前后的形态.该酶电极对葡萄糖的氧化表现出很好的响应,线性范围为1×10-5~7×10-3mol/L,响应时间小于5s,并且有很好的重现性.  相似文献

A new amperometric biosensor for hydrogen peroxide was developed based on cross-linking horseradish peroxidase (HRP) by glutaraldehyde with multiwall carbon nanotubes/chitosan (MWNTs/chitosan) composite film coated on a glassy carbon electrode. MWNTs were firstly dissolved in a chitosan solution. Then the morphology of MWNTs/chitosan composite film was characterized by field-emission scanning electron microscopy. The results showed that MWNTs were well soluble in chitosan and robust films could be formed on the surface. HRP was cross-linked by glutaraldehyde with MWNTs/chitosan film to prepare a hydrogen peroxide biosensor. The enzyme electrode exhibited excellent electrocatalytic activity and rapid response for H2O2 in the absence of a mediator. The linear range of detection towards H2O2 (applied potential: −0.2 V) was from 1.67 × 10−5 to 7.40 × 10−4 M with correction coefficient of 0.998. The biosensor had good repeatability and stability for the determination of H2O2. There were no interferences from ascorbic acid, glucose, citrate acid and lactic acid.  相似文献

Abstract

A new nanocomposite was developed by combination of prussian blue (PB) nanoparticles and multiwalled carbon nanotubes (MWNTs) in the matrix of biopolymer chitosan (CHIT). The PB and MWNTs had a synergistic electrocatalytic effect toward the reduction of hydrogen peroxide. The CHIT/MWNTs/PB nanocomposite‐modified glassy carbon (GC) electrode could amplify the reduction current of hydrogen peroxide by ~35 times compared with that of CHIT/MWNTs/GC electrode and reduce the response time from ~60 s for CHIT/PB/GC to 3 s. Besides, the CHIT/MWNTs/PB nanocomposite‐modified GC electrode could reduce hydrogen peroxide at a much lower applied potential and inhibit the responses of interferents such as ascorbic acid (AA) uric acid (UA) and acetaminophen (AC). With glucose oxidase (GOx) as an enzyme model, a new glucose biosensor was fabricated. The biosensor exhibited excellent sensitivity (the detection limit is down to 2.5 µM), fast response time (less than 5 s), wide linear range (from 4 µM to 2 mM), and good selection.  相似文献

利用卟啉(Hemin)具有模拟酶的功能,与多壁碳纳米管(MWCNTs)构建了一种新型的过氧化氢(H2O2)生物传感器。首先,利用Hemin与MWCNTs之间的π-π键作用,在超声分散下制备Hemin/MWCNTs纳米复合物;采用滴涂技术并在nafion的作用下将其固载在电极表面,制得该H2O2生物传感器(nafion/Hemin/MWCNTs/GCE)。采用紫外-可见分光光度法(UV-Vis)对合成的纳米复合物进行了分析;采用扫描电镜(SEM)对电极的表面形貌进行了表征;采用循环伏安法和计时电流法考察了该修饰电极的电化学行为;并对传感器的行为进行了详细的研究。在最优条件下,此修饰电极对H2O2具有明显的催化作用,电流与H2O2的浓度在6.0×10-7~1.8×10-3 mol/L范围内呈现良好的线性关系,检出限达2.0×10-7 mol/L。此传感器制作简单,具有较高的灵敏度和良好的稳定性及重现性。  相似文献

Nanosized Prussian blue (PB) particles were synthesized with a chemical reduction method and then the PB nanoparticles were assembled on the surface of multiwall carbon nanotubes modified glassy carbon electrode (PB/MWNTs/GCE). The results showed that the PB/MWNTs nanocomposite exhibits a remarkably improved catalytic activity towards the reduction of hydrogen peroxide. Glucose oxidase (GOD) was immobilized on the PB/MWNTs platform by an electrochemically polymerized o‐phenylenediamine (OPD) film to construct an amperometric glucose biosensor. The biosensor exhibited a wide linear response up to 8 mM with a low detection limit of 12.7 μM (S/N=3). The Michaelis–Menten constant Km and the maximum current imax of the biosensor were 18.0 mM and 4.68 μA, respectively. The selectivity and stability of the biosensor were also investigated.  相似文献

A sensitive amperometric glucose biosensor based on platinum nanoparticles (PtNPs) combined aligned carbon nanotubes (ACNTs) electrode was investigated. PtNPs which can enhance the electrocatalytic activity of the electrode for electrooxidating hydrogen peroxide by enzymatic reaction were electrocrystallized on 4‐aminobenzene monolayer‐grafted ACNTs electrode by potential‐step method. These PtNPs combined ACNTs' (PtNPs/ACNTs) surfaces were characterized by scanning electron microscopy (SEM) and transmission electron microscopy (TEM). The highly dispersed PtNPs on ACNTs can be obtained. The enzyme electrode exhibits excellent response performance to glucose with linear range from 1×10?5–7×10?3 mol L?1 and fast response time within 5 s. Furthermore, this glucose biosensor also has good reproducibility. It is demonstrated that the PtNPs/ACNTs electrode with high electrocatalytic activity is a suitable basic electrode for preparing enzyme electrodes.  相似文献

Colloidal carbon microspheres (CMS) are dispersed in chitosan (CHIT) solution to form an organic-inorganic hybrid with excellent micro-environment for the immobilization of biomolecules. A novel amperometric biosensor for the determination of hydrogen peroxide (H(2)O(2)) has been constructed by entrapping horseradish peroxidase (HRP) in as-synthesized CMS/CHIT hybrid. The modification of glassy carbon electrode is made by a simple solution-evaporation method. The electrochemical properties of the biosensor are characterized in electrochemical methods. The proposed biosensor shows high sensitive determination and fast response to H(2)O(2) at -0.15 V. The constructed HRP/CHIT/CMS/GC electrode also exhibits a fine linear correlation with H(2)O(2) concentration. The calculated value of the apparent Michaelis-Menten constant, 2.33 mM, suggests that the HRP in CMS/CHIT hybrid keeps its native bioactivity and has high affinity for H(2)O(2).  相似文献

A kind of nanocomposites with good dispersion in water was prepared through noncovalent adsorption of toluidine blue (Tb) on multiwalled carbon nanotubes (MWCNT) for electric communication between horseradish peroxidase (HRP) and electrode. The nanocomposites could be conveniently cast on electrode surface. With the aid of chitosan, HRP was then immobilized on the nanostructure to form a reagentless amperometric sensor for hydrogen peroxide. UV-vis spectroscopy and electrochemical impedance spectroscopy were used to characterize the adsorption of Tb on MWCNT. The presence of both Tb as mediator of electron transfer and MWCNT as conductor enhanced greatly the enzymatic response to the reduction of hydrogen peroxide. The novel biosensor exhibited fast response towards hydrogen peroxide with a detection limit of 1.7x10(-6)M and the linear range extended up to 4x10(-4)M without the interference of ascorbic acid and uric acid. The Michaelis-Menten constant (K'(m)) of the immobilized HRP was evaluated to be 0.16mM.  相似文献

Chitosan was chemically branched with ferrocene moieties and further used as a support for the immobilization of horseradish peroxidase on a glassy carbon electrode. The reagentless biosensor device showed a linear amperometric response toward hydrogen peroxide concentrations between 35 x 10(-6) M and 2.0 x 10(-3) M. The biosensor reached 95% of the steady-state current in about 20 s and its sensitivity was 28.4 x 10(-3) microA x M(-1). The enzyme electrode retained 94% of its initial activity after 2 weeks of storage at 4 degrees C in 50 x 10(-3) M sodium phosphate buffer at pH 7.0.  相似文献

A novel hydrogen peroxide biosensor has been constructed based on the characteristics of the carbon nanotube. The multiwall carbon nanotube (MWNT) was used as a coimmobilization matrix to incorporate horseradish peroxidase (HRP) and electron transfer mediator methylene blue (MB) onto a glassy carbon electrode surface. Cyclic voltammetry and amperometric measurements were employed to demonstrate the feasibility of methylene blue as an electron carrier between the immobilized peroxidase and the surface of glassy carbon electrode. The amperometric response of this resulting biosensor to H2O2 shows a linear relation in the range from 4 μM to 2 mM. The detection limit was 1 μM when the signal to noise ratio is 3. The presence of dopamine and ascorbic acid hardly affects the sensitive determination of H2O2. This biosensor also possesses very good stability and reproducibility.  相似文献

Iron‐phthalocyanines (FePc) are functionalized at multi‐walled carbon nanotubes (MWNTs) to remarkably improve the sensitivity toward hydrogen peroxide. We constructed a highly sensitive and selective glucose sensor on FePc‐MWNTs electrode based on the immobilization of glucose oxidase (GOD) on poly‐o‐aminophenol (POAP)‐electropolymerized electrode surface. SEM images indicate that GOD enzymes trapped in POAP film tend to deposit primarily on the curved tips and evenly disperse along the sidewalls. The resulting GOD@POAP/FePc‐MWNTs biosensor exhibits excellent performance for glucose with a rapid response (less than 8 s), a wide linear range (up to 4.0×10?3 M), low detection limits (2.0×10?7 M with a signal‐to‐noise of 3), a highly reproducible response (RSD of 2.6%), and long‐term stability (120 days). Such characteristics may be attributed to the catalytic activity of FePc and carbon nanotube, permselectivity of POAP film, as well as the large surface area of carbon nanotube materials.  相似文献

A novel approach to construct a amperometric biosensor for determination of H2O2 is described. Horseradish peroxidase (HRP) as a base enzyme was immobilized into the mixture of multiwalled carbon nanotubes (MWNTs) and polyvinyl butyral (PVB). Taking the classical hydroquinone as mediator, cyclic voltammetry and amperometric measurements were used to study and optimize the performance of the resulting H2O2 biosensor. The effect of the concentration of MWNTs, HRP, hydroquinone, solution pH, and the working potential of amperometry on the electrochemical biosensor was systematically studied. The results showed that the fabricated biosensor demonstrated significant electrocatalytic activity for the reduction of hydrogen peroxide with wide linear range from 0.000832 to 0.6 mM, and low detection limit 0.000167 mM (S/N = 3) with fast response time less than 8 s. The apparent Michaelis–Menten constant was determined to be 0.049 mM. Additionally, the biosensor exhibited high sensitivity, rapid response and good long-term stability.  相似文献

Abstract

A new amperometric biosensor for adenosine-5′-triphosphate (ATP) was designed using a platinum-dispersed carbon paste into which glycerol kinase and glycerol-3-phosphate oxidase were incorporated. The biosensor is based on the detection of hydrogen peroxide produced by the enzymatic reaction of ATP with glycerol and the subsequent oxidation of glycerol-3-phosphate. The use of the platinum-dispersed carbon paste electrode lowered the oxidation potential for hydrogen peroxide, permitting the sensitive detection of ATP at 0.4 V vs. Ag/AgCl. A linear response to ATP was observed in the concentration range of 1 x 10?5 to 2.5 x 10?3 M.

采用电沉积方法制备六氰合铁酸铜钴-多壁碳纳米管复合修饰电极(CuCoHCF-MWCNTs/GCE).研究碳纳米管用量、电解液组成对该修饰电极性能的影响.结果表明,与单一的六氰合铁酸铜钴薄膜修饰电极相比,六氰合铁酸铜钴-多壁碳纳米管复合修饰电极具有更优良的电化学特性,以其催化氧化过氧化氢,峰电流与过氧化氢浓度在3.16×10-5~2.92×10-3mol·L-1范围内呈良好的线性关系,线性回归方程为ip(μA)=0.5529+1.1299C(×10-4mol·L-1),相关系数r=0.9966,检出限为1.75×10-5mol·L-1.  相似文献

A carbon nanotubes‐based amperometric cholesterol biosensor has been fabricated through layer‐by‐layer (LBL) deposition of a cationic polyelectrolyte (PDDA, poly(diallyldimethylammonium chloride)) and cholesterol oxidase (ChOx) on multi‐walled carbon nanotubes (MWNTs)‐modified gold electrode, followed by electrochemical generation of a nonconducting poly(o‐phenylenediamine) (PPD) film as the protective coating. Electrochemical impedance measurements have shown that PDDA/ChOx multilayer film could be formed uniformly on MWNTs‐modified gold electrode. Due to the strong electrocatalytic properties of MWNTs toward H2O2 and the low permeability of PPD film for electroacitve species, such as ascorbic acid, uric acid and acetaminophen, the biosensor has shown high sensitivity and good anti‐interferent ability in the detection of cholesterol. The effect of the pH value of the detection solution on the response of the biosensor was also investigated. A linear range up to 6.0 mM has been observed for the biosensor with a detection limit of 0.2 mM. The apparent Michaelis‐Menten constant and the maximum response current density were calculated to be 7.17 mM and 7.32 μA cm?2, respectively.  相似文献

Liquid crystal cubic phase formed with monoolein has been used as immobilizing matrix to host redox protein hemoglobin on glassy carbon electrode surface. The promoted direct electron transfer between hemoglobin and electrode was observed and a large average kinetic electron transfer rate constant k(s) of 3.03(±0.02)s(-1) was estimated. The electrode modified with cubic phase containing hemoglobin retains the bioactivity of hemoglobin and shows excellent bioelectrocatalytic activity to the reduction of hydrogen peroxide with a small apparent Michaelis-Menten constant of 0.25(±0.03)mM. A novel reagentless hydrogen peroxide biosensor was constructed using the hemoglobin-containing cubic phase modified electrode and the proposed hydrogen peroxide biosensor shows a linear range of 7.0-239μM with a detection limit of 3.1(±0.2)μM and good stability and reproducibility.  相似文献

A new biosensor for the amperometric detection of hydrogen peroxide was developed based on the coimmobilization of horseradish peroxidase (HRP) and methylene blue on a beta-type zeolite modified glassy carbon electrode without the commonly used bovine serum albumin-glutaraldehyde. The intermolecular interaction between enzyme and zeolite matrix was investigated using FT-IR. The cyclic voltammetry and amperometric measurement demonstrated that methylene blue co-immobilized with HRP in this way displayed good stability and could efficiently transfer electrons between immobilized HRP and the electrode. The sensor responded rapidly to H2O2 in the linear range from 2.5 x 10(-6) to 4.0 x 10(-3) M with a detection limit of 0.3 microM. The sensor was stable in continuous operation.  相似文献
