A new bipyridine‐functionalized dithioester was synthesized and further used as a RAFT agent in RAFT polymerization of styrene and N‐isopropylacrylamide. Kinetics analysis indicates that it is an efficient chain transfer agent for RAFT polymerization of the two monomers which produce polystyrene and poly(N‐isopropylacrylamide) polymers with predetermined molecular weights and low polydispersities in addition to the end functionality of bipyridine. The bipyridine end‐functionalized polymers were further used as macroligands for the preparation of star‐shaped metallopolymers. Hydrophobic polystyrene macroligand combined with hydrophiphilic poly(N‐isopropylacrylamide) was complexed with ruthenium ions to produce amphiphilic ruthenium‐cored star‐shaped metallopolymers. The structures of these synthesized metallopolymers were further elucidated by UV–vis, fluorescence, size exclusion chromatography (SEC), and differential scanning calorimetry (DSC) as well as NMR techniques. © 2007 Wiley Periodicals, Inc. A hetero‐arm star polymer, poly(ethylene glycol)‐poly(N‐isopropylacrylamide)‐poly(L‐lysine) (PEG‐PNIPAM‐PLys), was synthesized by “clicking” the azide group at the junction of PEG‐b‐PNIPAM diblock copolymer with the alkyne end‐group of poly(L‐lysine) (PLys) homopolymer via 1,3‐dipolar cycloaddition. The resultant polymer was characterized by gel permeation chromatography, proton nuclear magnetic resonance, and Fourier transform infrared spectroscopes. Surprisingly, the PNIPAM arm of this hetero‐arm star polymer nearly lose its thermal responsibility. It is found that stable polyelectrolyte complex micelles are formed when mixing the synthesized polymer with poly(acrylic acid) (PAA) in water. The resultant polyelectrolyte complex micelles are core‐shell spheres with the ion‐bonded PLys/PAA chains as core and the PEG and PNIPAM chains as shell. The PNIPAM shell is, as expected, thermally responsive. However, its lower critical solution temperature is shifted to 37.5 °C, presumably because of the existence of hydrophilic components in the micelles. Such star‐like PEG‐PNIPAM‐PLys polymer with different functional arms as well as its complexation with anionic polymers provides an excellent and well‐defined model for the design of nonviral vectors to deliver DNA, RNA, and anionic molecular medicines. © 2009 Wiley Periodicals, Inc. The syntheses of well‐defined 7‐arm and 21‐arm poly(N‐isopropylacrylamide) (PNIPAM) star polymers possessing β‐cyclodextrin (β‐CD) cores were achieved via the combination of atom transfer radical polymerization (ATRP) and click reactions. Heptakis(6‐deoxy‐6‐azido)‐β‐cyclodextrin and heptakis[2,3,6‐tri‐O‐(2‐azidopropionyl)]‐β‐cyclodextrin, β‐CD‐(N3)7 and β‐CD‐(N3)21, precursors were prepared and thoroughly characterized by nuclear magnetic resonance and matrix‐assisted laser desorption/ionization time‐of‐flight mass spectrometry. A series of alkynyl terminally functionalized PNIPAM (alkyne‐PNIPAM) linear precursors with varying degrees of polymerization (DP) were synthesized via atom transfer radical polymerization (ATRP) of N‐isopropylacrylamide using propargyl 2‐chloropropionate as the initiator. The subsequent click reactions of alkyne‐PNIPAM with β‐CD‐(N3)7 and β‐CD‐(N3)21 led to the facile preparation of well‐defined 7‐arm and 21‐arm star polymers, namely β‐CD‐(PNIPAM)7 and β‐CD‐(PNIPAM)21. The thermal phase transition behavior of 7‐arm and 21‐arm star polymers with varying molecular weights were examined by temperature‐dependent turbidity and micro‐differential scanning calorimetry, and the results were compared to those of linear PNIPAM precursors. The anchoring of PNIPAM chain terminal to β‐CD cores and high local chain density for star polymers contributed to their considerably lower critical phase separation temperatures (Tc) and enthalpy changes during phase transition as compared with that of linear precursors. © 2008 Wiley Periodicals, Inc. Reversible addition‐fragmentation chain transfer (RAFT) polymerization is a more robust and versatile approach than other living free radical polymerization methods, providing a reactive thiocarbonylthio end group. A series of well‐defined star diblock [poly(ε‐caprolactone)‐b‐poly(N‐isopropylacrylamide)]4 (SPCLNIP) copolymers were synthesized by R‐RAFT polymerization of N‐isopropylacrylamide (NIPAAm) using [PCL‐DDAT]4 (SPCL‐DDAT) as a star macro‐RAFT agent (DDAT: S‐1‐dodecyl‐S′‐(α, α′‐dimethyl‐α″‐acetic acid) trithiocarbonate). The R‐RAFT polymerization showed a controlled/“living” character, proceeding with pseudo‐first‐order kinetics. All these star polymers with different molecular weights exhibited narrow molecular weight distributions of less than 1.2. The effect of polymerization temperature and molecular weight of the star macro‐RAFT agent on the polymerization kinetics of NIPAAm monomers was also addressed. Hardly any radical–radical coupling by‐products were detected, while linear side products were kept to a minimum by careful control over polymerization conditions. The trithiocarbonate groups were transferred to polymer chain ends by R‐RAFT polymerization, providing potential possibility of further modification by thiocarbonylthio chemistry. © 2011 Wiley Periodicals, Inc. The vitamin B6‐based macroinitiator was prepared for the first time by the transesterification reaction of pyridoxine with 2‐bromoisobutyryl bromide. A pyridoxine‐based star‐shaped block copolymer with a hydrophilic pyridoxine core and a dual hydrophilic poly(2‐(dimethylamino)ethyl methacrylate)‐block‐poly(N‐isopropylacrylamide) arms was synthesized for the first time via a simplified electrochemically mediated atom transfer radical polymerization, utilizing only 20 ppm of catalyst complex. The rate of the polymerizations was controlled by applying appropriate potential/current values during electrolysis to prevent the possibility of intermolecular coupling of the polymer stars. The asymmetric star polymers showed narrow molecular weight distribution (?  = 1.09–1.13). 1H NMR spectral results confirm the formation of star‐like block (co)polymers. These new vitamin B6‐based eagle‐shaped star (co)polymers may find biomedical and biosensor applications as pH‐sensitive and thermo‐sensitive drug delivery systems. We report on the synthesis of an H‐shaped polymer bonding β‐cyclodextrin (β‐CD) at branch points and influences of attached β‐CD on physical properties. First, a poly(ethylene glycol)(PEG)‐based functional macroinitiator bearing two azidos and four chlorines at chain‐ends (PEG‐2N3(‐4Cl)) was prepared via terminal modification reactions. Then, PEG‐2N3(‐4Cl) was applied to initiate the atom transfer radical polymerization of N‐isopropylacrylamide, leading to the synthesis of an H‐shaped block polymer with PEG as the central chain and poly(N‐isopropylacrylamide) (PNIPAM) as side‐arms (PEG‐2N3(‐4PNIPAM)). Azido groups were at the branch points of the polymer. Finally, the click reaction between PEG‐2N3(‐4PNIPAM) and alkynyl monosubstituted β‐cyclodextrin (β‐CD) afforded another H‐shaped polymer with two β‐CDs bonding at the polymer branch points (PEG‐2CD(‐4PNIPAM)). The glass transition temperature (Tg) and lower critical solution temperature (LCST) of the H‐shaped polymer increased after the attachment of β‐CD. The self‐assembly and thermal responsive behaviors, as well as the encapsulation behaviors of PEG‐2CD(‐4PNIPAM) were also altered. When temperature was below the LCSTs, PEG‐2N3(‐2PNIPAM) dissolved in water molecularly, whereas PEG‐2CD(‐4PNIPAM) could self‐assemble into nano‐sized micelles. After the LCST transitions, PEG‐2N3(‐4PNIPAM) aggregated into micron‐sized unstable particles, whereas PEG‐2CD(‐4PNIPAM) transformed into PNIPAM‐cored nanomicelles. Disulfide‐centered star‐shaped poly(ε‐benzyloxycarbonyl‐l ‐lysine)‐b‐poly(ethylene oxide) block copolymers (i.e., A2B4 type Cy‐PZlys‐b‐PEO) were synthesized by the combination of ring‐opening polymerization and thiol‐yne chemistry. Their molecular structures and physical properties were characterized in detail by FTIR, 1H NMR, gel permeation chromatography, differential scanning calorimetry, wide‐angle X‐ray diffraction, and polarized optical microscope. Despite mainly exhibiting an α‐helix conformation, the inner PZlys blocks within copolymers greatly prohibited the crystallinity of the outer PEO blocks and presented a liquid crystal phase transition behavior in solid state. These block copolymers Cy‐PZlys‐b‐PEO self‐assembled into nearly spherical micelles in aqueous solution, which had a hydrophobic disulfide‐centered PZlys core surrounded by a hydrophilic PEO corona. As monitored by means of DLS and TEM, these micelles were progressively reduced to smaller micelles in 10 mM 1,4‐dithiothreitol at 37 °C and finally became ones with a half size, demonstrating a reduction‐sensitivity. Despite a good drug‐loading property, the DOX‐loaded micelles of Cy‐PZlys‐b‐PEO exhibited a reduction‐triggered drug release profile with an improved burst‐release behavior compared with the linear counterpart. Importantly, this work provides a versatile strategy for the synthesis of the disulfide‐centered star‐shaped polypeptide block copolymers potential for intracellular glutathione‐triggered drug delivery systems. © 2014 Wiley Periodicals, Inc. Poly(N‐isopropylacrylamide)s (PNIPAMs) with cholesteryl or pyrenyl moieties at each chain end (CH‐PNIPAMs or Py‐PNIPAMs) were prepared via end‐group modification of α,ω‐dimercapto poly(N‐isopropylacrylamides), ranging in molecular weight from ~ 7000 to 45,000 g mol?1 with a polydispersity index of 1.10 or lower. The telechelic thiol functionalized PNIPAMs were obtained by aminolysis of α,ω‐di(isobutylthiocarbonylthio)‐poly(N‐isopropylacrylamide)s (iBu‐PNIPAMs) obtained by reversible addition‐fragmentation chain transfer (RAFT) polymerization of N‐isopropylacrylamide in the presence of the difunctional chain transfer agent, diethylene glycol di(2‐(1‐isobutyl)sulfanylthiocarbonylsulfanyl‐2‐methyl propionate) (DEGDIM). A novel six‐arm star block copolymer comprising polystyrene (PS) linked to the center and π‐conjugated poly (3‐hexylthiophene) (P3HT) was successfully synthesized using a combination of atom transfer radical polymerization (ATRP) and click reaction. First, star‐shaped PS with six arms was prepared via ATRP of styrene with the discotic six‐functional initiator, 2,3,6,7,10,11‐hexakis(2‐bromoisobutyryloxy)triphenylene. Next, the terminal bromides of the star‐shaped PS were substituted with azide groups. Afterward, the six‐arm star block copolymer PS‐b‐P3HT was prepared using the click coupling reaction of azide‐terminated star‐shaped PS with alkynyl‐terminated P3HT. Various techniques including 1H NMR, Fourier‐transform infrared and size‐exclusion chromatography were applied to characterize the chemical structures of the intermediates and the target block copolymers. Their thermal behaviors and optical properties were investigated using differential scanning calorimetry and UV–vis spectroscopy. Moreover, atomic force microscopy (AFM) was utilized to observe the morphology of the star block copolymer films. In comparison with two linear diblock copolymer counterparts, AFM results reveal the effect of the star block copolymer architecture on the microphase separation‐induced morphology in thin films. © 2012 Wiley Periodicals, Inc. Poly(N‐isopropylacrylamide) (PNIPAAm) gels are temperature‐responsive polymer gels; and were prepared by redox polymerization of N‐isopropylacrylamide in the presence of N,N′‐methylenebisacrylamide as a crosslinking reagent and core‐shell type bioconjugates, which were core‐crosslinked polyion complex micelles formed from the mixture of bovine pancreas trypsin and poly(ethylene glycol)‐block‐poly(α,β‐aspartic acid). The phase transition temperature of PNIPAAm gels was no change with physically immobilization of bioconjugates. Also, the enzymatic activity of bioconjugates was essentially maintained even in PNIPAAm gels, although enzymatic reaction rate was apparently controlled by temperature, i.e., by the degree of swelling of PNIPAAm gels. Further, the control of enzymatic reaction synchronizing the phase transition of PNIPAAm gels immobilized bioconjugates. PNIPAAm gels could immobilize core‐shell type bioconjugates, and were successfully prepared without interfering with the properties of temperature‐responsive polymer gels and the bionanoreactor. © 2007 Wiley Periodicals, Inc. Self‐assembled thermo‐ and pH‐responsive poly(acrylic acid)‐b‐poly(N‐isopropylacrylamide) (PAA‐b‐PNIPAM) micelles for entrapment and release of doxorubicin (DOX) was described. Block copolymer PAA‐b‐PNIPAM associated into core‐shell micelles in aqueous solution with collapsed PNIPAM block or protonated PAA block as the core on changing temperature or pH. Complexation of DOX with PAA‐b‐PNIPAM triggered by the electrostatic interaction and release of DOX from the complexes due to the changing of pH or temperature were studied. Complex micelles incorporated with DOX exhibited pH‐responsive and thermoresponsive drug release profile. The release of DOX from micelles was suppressed at pH 7.2 and accelerated at pH 4.0 due to the protonation of carboxyl groups. Furthermore, the cumulative release of DOX from complex micelles was enhanced around LCST ascribed to the structure deformation of the micelles. © 2008 Wiley Periodicals, Inc. Two types of temperature‐sensitive biodegradable three‐arm and four‐arm star‐shaped poly(DL ‐lactic acid‐co‐glycolic acid‐b‐ethylene glycol) (3‐arm and 4‐arm PLGA–PEG) were successfully synthesized via the coupling reaction of 3‐arm and 4‐arm PLGA and α‐monocarboxyl‐ω‐monomethoxypoly(ethylene glycol) (CMPEG). In dilute aqueous solutions, star PLGA–PEGs showed the temperature‐ and concentration‐dependent formation and aggregation of micelles over specific concentration and specific temperature. With increasing the molecular weight and the relative hydrophobicity of hydrophobic PLGA block, critical micelle temperature (CMT) decreased. Aqueous solution of 4‐arm PLGA–PEG started to form micelles at lower temperature and showed sharper temperature‐dependent growth in micelle size. These results are due to the enhanced hydrophobicity of PLGA block. On the other hand, at high concentration, two types of 3‐arm and 4‐arm PLGA–PEG showed sol–gel–sol transition behavior as the temperature was increased. The 3‐arm and 4‐arm PLGA–PEG showed sol–gel transition at higher polymer concentrations (above 24 wt %) than the PEG–PLGA–PEG triblock copolymer. As the molecular weight and the relative hydrophobicity of PLGA block increased, the critical gel concentration (CGC) decreased. © 2005 Wiley Periodicals, Inc. Poly(N‐vinylcaprolactam) (PNVCL) star‐shaped polymers with four arms and carboxyl end groups were synthesized by reversible addition–fragmentation chain transfer (RAFT) polymerization of N‐vinylcaprolactam (NVCL) employing a tetrafunctional trithiocarbonate as an R‐RAFT agent. The resulting star polymers were characterized using 1H NMR, FT‐IR, gel permeation chromatography (GPC), and UV–vis. Molecular weight of star polymers were analyzed by GPC and UV–vis being observed that the values obtained were very similar. Furthermore, the thermosensitive behavior of the star polymers was studied in aqueous solution by measuring the lower critical solution temperature by dynamic light scattering. Star‐shaped PNVCL were chain extended with ethyl‐hexyl acrylate (EHA) to yield star PNVCL‐b‐PEHA copolymers with an EHA molar content between 4% and 6% proving the living character of the star‐shaped macroCTA. This article describes a divergent strategy to prepare dendrimer‐like macromolecules from vinyl monomers through a combination of atom transfer radical polymerization (ATRP) and click reaction. Firstly, star‐shaped polystyrene (PS) with three arms was prepared through ATRP of styrene starting from a three‐arm initiator. Next, the terminal bromides of the star‐shaped PS were substituted with azido groups. Afterwards, the azido‐terminated star‐shaped PS was reacted with propargyl 2,2‐bis((2′‐bromo‐2′‐methylpropanoyloxy)methyl)propionate (PBMP) via click reaction. Star‐shaped PS with six terminal bromide groups was afforded and served as the initiator for the polymerization of styrene to afford the second‐generation dendrimer‐like PS. Iterative process of the aforementioned sequence of reactions could allow the preparation of the third‐generation dendrimer‐like PS. When the second‐generation dendrimer‐like PS with 12 bromide groups used as an initiator for the polymerization of tert‐butyl acrylate, the third‐generation dendrimer‐like block copolymer with a PS core and a poly (tert‐butyl acrylate) (PtBA) corona was afforded. Subsequently PtBA segments were selectively hydrolyzed with hydrochloric acid, resulting an amphiphilic branched copolymer with inner dendritic PS and outer linear poly(acrylic acid) (PAA). Following the same polymerization procedures, the dendrimer‐like PS and PS‐block‐PtBA copolymers of second generation originating from six‐arm initiator were also synthesized. © 2007 Wiley Periodicals, Inc. 4μ‐A2B2 star‐shaped copolymers contained polystyrene (PS), poly(isoprene) (PI), poly(ethylene oxide) (PEO) or poly(ε‐caprolactone) (PCL) arms were synthesized by a combination of Glaser coupling with living anionic polymerization (LAP) and ring‐opening polymerization (ROP). Firstly, the functionalized PS or PI with an alkyne group and a protected hydroxyl group at the same end were synthesized by LAP and then modified by propargyl bromide. Subsequently, the macro‐initiator PS or PI with two active hydroxyl groups at the junction point were synthesized by Glaser coupling in the presence of pyridine/CuBr/N,N,N ′,N ″,N ″‐penta‐methyl diethylenetri‐amine (PMDETA) system and followed by hydrolysis of protected hydroxyl groups. Finally, the ROP of EO and ε‐CL monomers was carried out using diphenylmethyl potassium (DPMK) and tin(II)‐bis(2‐ethylhexanoate) (Sn(Oct)2) as catalyst for target star‐shaped copolymers, respectively. A novel primary amine‐containing monomer, 1‐(3′‐aminopropyl)‐4‐acrylamido‐1,2,3‐triazole hydrochloride (APAT), was prepared from N‐propargylacrylamide and 3‐azidopropylamine hydrochloride via copper‐catalyzed Huisgen 1,3‐dipolar cycloaddition (click reaction). Poly(N‐isopropylacrylamide)‐b‐poly(1‐(3′‐aminopropyl)‐4‐acrylamido‐1,2,3‐triazole hydrochloride), PNIPAM‐b‐PAPAT, was then synthesized via consecutive reversible addition‐fragmentation chain transfer polymerizations of N‐isopropylacrylamide and APAT. In aqueous solution, the obtained thermoresponsive double hydrophilic block copolymer dissolves molecularly at room temperature and self‐assembles into micelles with PNIPAM cores and PAPAT shells at elevated temperature. Because of the presence of highly reactive primary amine moieties in PAPAT block, two types of covalently stabilized nanoparticles namely core crosslinked and shell crosslinked micelles with ‘inverted’ core‐shell nanostructures were facilely prepared upon the addition of glutaric dialdehyde at 25 and 50 °C, respectively. In addition, the obtained structure‐fixed micelles were incorporated with gold nanoparticles via in situ reduction of preferentially loaded HAuCl4. High resolution transmission electron microscopy revealed that gold nanoparticles can be selectively loaded into the crosslinked cores or shells, depending on the micelle templates employed. © 2008 Wiley Periodicals, Inc. A hetero‐arm star polymer, polystyrene‐poly(N‐isopropylacrylamide)‐ poly(2‐(dimethylamino)ethylmethacrylate) (PSt‐PNIPAM‐PDMAEMA), was synthesized by “clicking” the alkyne group at the junction of PSt‐b‐PNIPAM diblock copolymer onto the azide end‐group of PDMAEMA homopolymer via 1,3‐dipolar cycloaddition. The resultant polymer was characterized by gel permeation chromatography, proton nuclear magnetic resonance spectroscopy and Fourier transform infrared spectroscopy. PSt‐PNIPAM‐PDMAEMA micelles with PSt block as core and PNIPAM and PDMAEMA blocks as shell were formed when adding the copolymer solution in THF into 10 folds of water. Lower critical solution temperature (LCST) of PNIPAM and PDMAEMA homopolymer is 32 °C for PNIPAM and 40 to 50 °C for PDMAEMA, respectively. Upon continuous heating through their LCSTs, PSt‐PNIPAM‐PDMAEMA core‐shell micelles exhibited two‐stage thermally induced collapse. The first‐stage collapse, from 20 to 34 °C, is ascribed to the shrinkage of PNIPAM chains; and the second‐stage collapse, from 38 to 50 °C, is due to the shrinkage of PDMAEMA chains. The synthesis of multiarm star block (and mixed‐block) copolymers are efficiently prepared by using Cu(I) catalyzed azide‐alkyne click reaction and the arm‐first approach. α‐Silyl protected alkyne polystyrene (α‐silyl‐alkyne‐PS) was prepared by ATRP of styrene (St) and used as macroinitiator in a crosslinking reaction with divinyl benzene to successfully give multiarm star homopolymer with alkyne periphery. Linear azide end‐functionalized poly(ethylene glycol) (PEG‐N3) and poly (tert‐butyl acrylate) (PtBA‐N3) were simply clicked with the multiarm star polymer described earlier to form star block or mixed‐block copolymers in N,N‐dimethyl formamide at room temperature for 24 h. The aqueous solution behavior and thermoreversible gelation properties of pyridine‐end‐functionalized poly(ethylene glycol)–poly(L ‐lactide) (PEG–(PLLA)8–py) star block copolymers in the presence of coordinating transition metal ions were studied. In aqueous solutions, the macromonomers self‐assembled into micelles and micellar aggregates at low concentrations and formed physically crosslinked, thermoreversible hydrogels above a critical gel concentration (CGC) of 8% w/v. In the presence of transition metal ions like Cu(II), Co(II), or Mn(II), the aggregate dimensions increased. Above the CGC, the gel–sol transition shifted to higher temperatures due to the formation of additional crosslinks from intermolecular coordination complexes between metal ions and pyridine ligands. Furthermore, as an example, PEG–(PLLA)8–py hydrogels stabilized by Mn(II)–pyridine coordination complexes were more resistant against degradation/dissolution when placed in phosphate buffered saline at 37 °C when compared with hydrogels prepared in water. Importantly, the stabilizing effect of metal–ligand coordination was noticeable at very low Cu(II) concentrations, which have been reported to be noncytotoxic for fibroblasts in vitro. These novel PEG–(PLLA)8–py metallo‐hydrogels, which are the first systems to combine metal–ligand coordination with the advantageous properties of PEG–PLLA copolymer hydrogels, are appealing materials that may find use in biomedical as well as environmental applications like the removal of heavy metal ions from waste streams. © 2012 Wiley Periodicals, Inc. Novel star‐shaped hard–soft triblock copolymers, 4‐arm poly(styrene)‐block‐poly [poly(ethylene glycol) methyl ethyl methacrylate]‐block‐poly{x‐[(4‐cyano‐4′‐biphenyl) oxy] alkyl methacrylate} (4PS‐PPEGMA‐PMAxLC) (x = 3, 10), with different mesogen spacer length are prepared by atom‐transfer radical polymerization. The star copolymers comprised three different parts: a hard polystyrene (PS) core to ensure the good mechanical property of the solid‐state polymer, and a soft, mobile poly[poly(ethylene glycol) methyl ethyl methacrylate] (PPEGMA) middle sphere responsible for the high ionic conductivity of the solid polyelectrolytes, and a poly{x‐[(4‐cyano‐4′‐biphenyl)oxy]alkyl methacrylate} with a birefringent mesogens at the end of each arm to tuning the electrolytes morphology. The star‐shaped hard–soft block copolymers fusing hard PS core with soft PPEGMA segment can form a flexible and transparent film with dimensional stability. Thermal annealing from the liquid crystalline states allows the cyanobiphenyl mesogens to induce a good assembly of hard and soft blocks, consequently obtaining uniform nanoscale microphase separation morphology, and the longer spacer is more helpful than the shorter one. There the ionic conductivity has been improved greatly by the orderly continuous channel for efficient ion transportation, especially at the elevated temperature. The copolymer 4PS‐PPEGMA‐PMA10LC shows ionic conductivity value of 1.3 × 10?4 S cm?1 (25 °C) after annealed from liquid crystal state, which is higher than that of 4PS‐PPEGMA electrolyte without mesogen groups. © 2013 Wiley Periodicals, Inc.
