Nitropyrazoles     
A method of synthesizing nitro derivatives of lH,4H-pyrazolo[4,3-c]pyrazole is developed, and some of its transformations are studied. 3-Methyl-6-nitro-, 3-carboxy(methoxy-carbonyl, carbamoyl)-6-nitro-, 3-amino-6-nitro-, 3-nitro-, 3,6-dinitro-, 1,4-diacetonyl-3,6-dinitro-, and lH,4H-3-aminopyrazolo[4,3-c]pyrazoles were obtained from 1H,4H-3 -methylpyrazolo[4,3-c]pyrazole. Unsubstituted 1H,4H-pyrazolo[4,3-c]pyrazole, the first member of this series, was obtained for the first time. The compounds prepared were characterized by1H,13C,14N, and15N NMR spectroscopy. NH-Acidity and basicity of the series of pyrazolo[4,3-c]pyrazoles synthesized is studied and the effect of the fused pyrazole ring on these properties is examined.Deceased March 18, 1989.Translated fromIzvestiya Akademii Nauk. Isomeric 4-acetyl-5-amino-3-methyl- and 4-acetyl-3-amino-5-methylpyrazoles (2, 3) were formed in the reaction of hydrazines with 3-[amino(methylthio)methylene]pentan-2,4-dione (1) (diacetylketeneN,S-acetal). Pyrazolo[3,4-d]pyrimidines (5a,b) were synthesized by condensation of 4-acetyl-5-amino-1,3-dimethylpyrazole (2a) with amide dimethylacetals followed by treatment with ammonium acetate. Z-3-Ferrocenylmethylene-2-methylenecamphane andE-2-ferrocenylmethylene-3-methylenequinuclidine were synthesized by isomerization of the corresponding isomericE- andZ-1,3-dienes in an acidic medium. The dienes obtained form [4+2]-cycloadditionendo-adducts withN-phenylmaleimide, do not form cyclodimers upon thermal or acid-catalyzed [4+2]-cyclodimerization, and addZ-3-ferrocenylmethylene-1,2,7,7-tetramethylbicyclo[2.2.1]hept-2-ylium andE-2-ferrocenylmethylene-3-methyl-1-azoniabicyclo[2.2.1]oct-3-ylium salts, respectively, at the terminal methylene group to give linear addition products. The first representative of perhydro-1,3-dioxa-6-azocines, 8,10-dimethyl-7,13-dioxa-10-azaspiro[5.7]tridecane, was synthesized by the reaction of 4-methyl-1-oxa-4-azaspiro[4.5]decane with propylene oxide. Summary A new series of thiophenic isosters of thioxanthones, namely: 2-substituted-4H-thieno[2,3-b][1]benzothiopyran-4-ones and 5-substituted-2-nitro-8-methyl-4H-thieno[2,3-b][1]benzothiopyran-4-ones were synthesized as potential schistosomicidal agents. The synthesized compounds were characterized by their1H-NMR data.

Zusammenfassung Es wurde eine neue Serie von thiophenischen Isosteren des Thioxanthons, nämlich 2-substituierte 4H-Thieno[2,3-b][1]benzothiopyran-4-one und 5-substituierte 2-Nitro-8-methyl-4H-thieno[2,3-b][1]benzothiopyran-4-one als potentielle schistosomicide Wirkstoffe synthetisiert. Die synthetisierten Verbindungen wurden mittels ihrer1H-NMR Daten charakterisiert.

Carboxy- and carbomethoxy derivatives of resorcinols with two benzotriazole substituents have been synthesized. 2(2,4-Dihydroxyphenyl)2H-1,3-bis-(4-carboxybenzotriazole) [2,4-bis(2H-4-carboxybenzotriazole-2-yl)-1,3-dihydroxybenzene] (DCBDH) was prepared by azo coupling of 4-carboxy-2-nitrobenzene diazonium chloride with resorcinol followed by reductive cyclization. 2(2,4-Dihydroxyphenyl)2H-1,3-bis(4-carbomethoxy-benzotriazole) [2,4-bis-(2H-4-carbomethoxy-benzotriazole-2-yl)-1,3-dihydroxybenzene] (DCMBDH) was obtained by esterification of the free dicarboxylic acid with methanol. The compounds were characterized by their elemental analyses and melting points, and by their IR, UV,1H NMR, and13C NMR spectra.Copolycondensations were carried out to incorporateDCBDH orDCMBDH into polyamides or polyesters. Summary A number of condensed quinazolines have been synthesized by the reaction of 5-phenyl-3-hydrazino-1,2,4-triazolo[4,3-c]quinazoline with appropriate phenyl isothiocyanates, arylcarbaldehydes, ethyl orthoformate, acetic anhydride, and nitrous acid. A general method for the synthesis of 5-substituted indolizidines based on intramolecular cyclization oftrans- andcis-2-allyl-6-R-1,2,3,6-tetrahydropyridines, obtained from pyridine and triallylborane, has been elaborated. The closure of the five-membered ring is carried out by hydroboration-oxidation followed by cyclization of the resulting δ-amino alcohols in the presence of the Ph3P−CBr4−Et3N system. (Pr2BH)2 and Pr3B are used as the hydroborating reagents, and H2O2 in an acid medium is used for the oxidation of 2-[3-(dipropylboryl]-Δ2-piperideines formed. This method has been used for the synthesis of two natural alkaloids: indolizidine 209D (cis-5-hexylindolizidine) and itstrans-isomer were prepared fromcis- andtrans-2-allyl-6-hexyl-1,2,3,6-tetrahydropiridine, respectively; indolizidine 167B andtrans-5-propylindolizidine were synthesized fromcis- andtrans-2,6-diallyl-1,2,3,6-tetra-hydropyridine, respectively. O-Ethyl alkynylphosphonites [EtOP(O)(H)CэCR] (1 and2) react withp-bromobenzaldehyde and benzylamine to form the usual acyclic products of the Kabachnik-Fields reaction or phosphorus-containing heterocycles depending on the nature of the substituent R at the β-carbon atom of the acetylene fragment of phosphonite. In the case of R=But,O-ethyl (α-benzylamino-p-bromobenzyl)(3,3-dimethylbut-1-ynyl)phosphinate (3) was obtained. In the case of R=Me, 1-benzyl-2-p-bromophenyl-3-ethoxy-5-methyl-3-oxo-Δ4-1,3-λ5-azaphospholine (4) was formed. Compounds3 and4 were also synthesized by the reactions of the above-mentioned phosphonites with the corresponding azomethines. Dedicated to the memory of Academician M. I. Methods for the synthesis of [2,2]paracyclophane derivatives containingo- andm-carboranyl substituents in position 4, separated from the [2,2]paracyclophane system by one or two C atoms (alcohols and ketones) were developed. The Clemmensen reduction of a number of ketones of the [2,2]paracyclophane ando-carbonane series occurs abnormally. Dialkyl β-(2-aroxyethylamino)ethylphosphonates,N-[β-(2-methoxyphenoxy)ethyl]-N′-[β-(diethoxyphosphoryl)ethyl]-α,ω-diaminoalkanes, and diethyl β-(N-arylpiperazino)ethyl-phosphonates were synthesized by the reactions of dialkyl vinylphosphonates with β-aroxyethylamines,N-[β-(2-methoxyphenoxy)ethyl]-α,ω-diaminoalkanes, andN-aryl-piperazines, respectively. Abstract

Two flavindogenides, 3-(2-chlorobenzylidene)-flavanone and 3-(2,4-dichloro- benzylidene)-flavanone reacted with six 5-substituted-2-aminobenzenethiols, the substituents being fluoro, chloro, bromo, methyl, methoxyl, and ethoxyl, to give respective 12 new compounds, 10-substituted-7-(2-chlorophenyl/2,4-dichlorophenyl)- 6H-6a,7-dihydro-6-phenyl[1]benzopyrano[3,4?c][1,5]benzothiazepines (5a–l) in 60–70% yields. The products were characterized on the basis of microanalytical data for elements and IR, 1 H, and 13 C NMR and mass spectral studies. All the synthesized compounds were evaluated for their antimicrobial activity against the bacteria,Escherichia coli and GFC,and the fungi,Aspergillus niger, Aspergillus flavus,and Curvularia lunata.  相似文献

3-Acyl-6-methyl-5H-thiazolo[2,3-c][1,2,4]triazines were synthesized by reaction of 1-acyl-2-bromoacetylenes with 6-methyl-3-thioxo-1,2,4-triazin-5-one in methanol in the presence of triethylamine at 20 °C. The structure of 3-benzoyl-6-methyl-5H-thiazolo[2,3-c][1,2,4]triazine was established by X-ray structural analysis. Substituted 3-(2-acetylvinylthio)1,2,4-triazin-5-ones were obtained by the reaction of -acetylenic ketones with 6-methyl-3-thioxo-1,2,4-triazin-5-one under the same conditions. Arsenic is a type 1 carcinogen and its toxicity is critically dependent on chemical speciation. However, after decades of research, the biogenesis of at least fifty naturally occurring arsenic species is still not well understood.Here, based on experimental work, it is proposed a set of pathways for the formation of multiple arsenic species that might help to clarify the present situation.These are focused on the thiol protein arsenic bond and on its interaction with reactive metabolites. In fact, arsenic bound to glutathione interacting with sulfur adenosyl methionine (SAM), MethylCB12 and AdoCB12, forms a number of complexes that might be key intermediates in arsenic biochemistry. These include dimethylarsino glutathione (DMAG) m/z 412 [M + H]+, synthesized non-enzymatically from glutathione and cacodylate. Trimethylarsonio glutathione (TMAG) m/z 426 [M]+ synthesized from DMA, GSH and SAM, apparently by a classical Challenger methylcarbonium attack. Tetramethyl arsonium ion m/z 135 [M]+ is formed in a third step, apparently by carbanion methylation. The presence of trimethylarsine oxide (TMAO) m/z 137 [M + H]+ is attributed to the hydrolysis of TMAG or TMA, or to carbanion methylation of dimethylarsinoyl glutathione (m/z 428 [M]+) formed from cacodylate and GSH. Cantoni type attacks of DMAG on SAM were unsuccessful, eventually due to competition of the trivalent S+ atom of SAM for the AsIII atom attack. The presence of dimethylarsonio diglutathione (DMADG m/z 717 [M]+), is suggested to result from a GS- attack on dimethylarsenoyl glutathione (m/z 428 [M + H]+). The presence of dimethylarsenoyladenosine (m/z 372 [M + H]+), trimethylarsenosugar adenine (m/z 370 [M]+), and dimethylthioarsenosugar adenine (m/z 388 [M + H]+), is explained by the synthesis of the pecursor dimethylarsonio-adenosine glutathione DMAAG (m/z 661 [M]+), a likely source of oxo-and trimethylated arsenosugars, as well as of thio-arsenosugars by the cleavage of its S-C bond. The results gathered suggest that cell vacuoles might play a major role in arsenic metabolism, and that the dominance of oxo-As sugars, in algae extracts, may be supported by a mechanism of synthesis independent of DMAAG (m/z 661).They also offer an explanation for the reason why arsenobetaine, and tetramethylarsonium are loosely bound to biotic tissues. Four arsenic species new to science, to the best of our knowledge, and a number of known arsenic compounds were synthesized in this work, identified by HPLC–ESI-MSn and FTICR–ESI-MS, and suggestions regarding their mechanisms of synthesis were advanced. These results provide a framework for arsenic biochemistry which may explain the origin of a significant part of arsenic known metabolites.  相似文献

Summary 2,3-Dihydro-3-oxo-benzo[b]thiophen-1,1-dioxide (1) reacts as CH-acidic component with amidines of orthoesters and anilines resp. to give the anilinomethylene derivates3, 4, and5. With triethyl orthoformiate the hydroxymethylene-compound7 is obtained. Anilino- and phenylhydrazino derivates8 and9 prove the carbonyl activity of1, azo-coupling leads to10, whereas treatment of1 with sulfur and malononitrile yields the benzo[b]thieno[2,3-d]thiophenes11. Introduction of substituents with active NH-functions in position 2 of the dicyanomethylene-product2, such as azocoupling, reaction with phenyl isocyanate and formation of enamines, leads to ring closure reactions between a nitrile and the NH-group. Thereby the phenyl-benzo[4,5]thieno[2,3-c]pyridazines12, the phenyl-1H-benzo[4,5]thieno[2,3-c]pyridines13 and the phenylamino-benzo[4,5]thieno[2,3-c]pyridines14 are obtained.13C and15N-NMR spectroscopy was used as proof of the ring closure reactions.

(Methylchlorosilyl)methyldichlorophosphines have been synthesized by the reaction of [dimethy(diethylamino)silyl]- or [methyl-bis(diethylamino)silyl]methylmagnesium chlorides with PCl3 in ether at –40÷–20 °C and subsequent treatment of the reaction mixture with dry HCl.
