The present study investigated the rhythmic changes in glutathione status in midgut gland and hemolymph as well as in glutathione reductase (GR) activity in the crayfish Procambarus clarkii. In order to determine the circadian nature of these rhythms different groups of crayfish were submitted to constant-darkness conditions for 24 or 72 h after they had spent 15 days under light-dark 12:12 cycles. The animals of the different batches were killed at 6 h intervals during a 24 h cycle. Reduced glutathione (GSH) and oxidized glutathione (GSSG) in hemolymph and midgut as well as midgut GR activity were determined in midgut gland and hemolymph by fluorometric and spectrophotometric method. Data analysis by chronogram and single Cosinor revealed circadian rhythmicity for GSH and GSSG concentration in both tissues as well as midgut GR activity. The rhythm parameters revealed oxidative stress induced by light. The possible correlation between the glutathione rhythm and other metabolic and behavioral rhythms of crayfish as well as the importance of the glutathione circadian temporal order in the adaptation of crayfish are discussed.  相似文献

This work was carried out to study the antioxidant circadian system of two species of crayfish of different latitude origin. We investigated (1) whether both species possess glutathione circadian rhythms and (2) whether both species' rhythms differ in their ability to synchronize to 24 h cycles. Two batches of Procambarus clarkii and P. digueti were kept in (1) light-dark (LD) 12:12 low irradiance (LI) cycles and then exposed to (2) 72 h of complete darkness, (3) LD 12:12 high irradiance (HI), (4) LD 20:4 LI and (5) LD 20:4 HI for 2 weeks. The midgut and hemolymph were sampled and reduced and oxidized glutathione as well as glutathione reductase and glutathione peroxidase were assayed. Cosinor and analysis of variance revealed differences between both species. Procambarus clarkii robust antioxidant circadian rhythms are able to entrain to all conditions resetting to lights on or off. However, the P. digueti weak circadian glutathione system did not entrain to the LD cycles, showing a random distribution of phases. In this species, LD 12:12 and 20:4 HI evidenced significant daily rhythms indicating a damped circadian antioxidative system that is enhanced by the effect of light. This suggests that each species' photoperiodic history determines the adaptive abilities of the circadian antioxidative mechanisms.  相似文献

The circadian clock at the hypothalamic suprachiasmatic nucleus (SCN) entrains output rhythms to 24-h light cycles. To entrain by phase-advances, light signaling at the end of subjective night (circadian time 18, CT18) requires free radical nitric oxide (NO•) binding to soluble guanylate cyclase (sGC) heme group, activating the cyclic guanosine monophosphate (cGMP)-dependent protein kinase (PKG). Phase-delays at CT14 seem to be independent of NO•, whose redox-related species were yet to be investigated. Here, the one-electron reduction of NO• nitroxyl was pharmacologically delivered by Angeli’s salt (AS) donor to assess its modulation on phase-resetting of locomotor rhythms in hamsters. Intracerebroventricular AS generated nitroxyl at the SCN, promoting phase-delays at CT14, but potentiated light-induced phase-advances at CT18. Glutathione/glutathione disulfide (GSH/GSSG) couple measured in SCN homogenates showed higher values at CT14 (i.e., more reduced) than at CT18 (oxidized). In addition, administration of antioxidants N-acetylcysteine (NAC) and GSH induced delays per se at CT14 but did not affect light-induced advances at CT18. Thus, the relative of NO• nitroxyl generates phase-delays in a reductive SCN environment, while an oxidative favors photic-advances. These data suggest that circadian phase-locking mechanisms should include redox SCN environment, generating relatives of NO•, as well as coupling with the molecular oscillator.  相似文献

A surface sensitive to reactive oxygen species (ROS) was prepared by reduction of a diazonium salt on glassy carbon electrode followed by the chemical coupling of glutathione (GSH) playing the role of an antioxidant species. The presence of active GSH was characterized through spectroscopic studies and electrochemical analysis after labeling of the -SH group with ferrocene moieties. The specific reactivity of GSH vs ROS was evaluated with scanning electrochemical microscopy (SECM) using the reduction of O(2) to superoxide, O(2)(?-), near the GSH-modified surface. Approach curves show a considerable decrease of the blocking properties of the layer due to reaction of the immobilized GSH with O(2)(?-) and the passage of GSH to the glutathione disulfide (GSSG). The initial surface could be regenerated several times with no significant variations of its antioxidant capacity by simply using the biological system glutathione reductase (GR)/NADPH that reduces GSSG back to GSH. SECM imaging shows also the possibility of writing local and erasable micropatterns on the GSH surface by production of O(2)(?-) at the tip probe electrode.  相似文献

The glutathione (GSH) system is one of the most powerful intracellular antioxidant systems for the elimination of reactive oxygen species (ROS) and maintaining cellular redox homeostasis. However, the rapid kinetics information (at the millisecond to the second level) during the dynamic antioxidation process of the GSH system remains unclear. As such, we specifically developed a novel dual-wire nanosensor (DWNS) that can selectively and synchronously measure the levels of GSH and ROS with high temporal resolution, and applied it to monitor the transient ROS generation as well as the rapid antioxidation process of the GSH system in individual cancer cells. These measurements revealed that the glutathione peroxidase (GPx) in the GSH system is rapidly initiated against ROS burst in a sub-second time scale, but the elimination process is short-lived, ending after a few seconds, while some ROS are still present in the cells. This study is expected to open new perspectives for understanding the GSH antioxidant system and studying some redox imbalance-related physiological.  相似文献

Photodynamic therapy removes unwanted or harmful cells by overproduction of reactive oxygen species (ROS). Fractionated light delivery in photodynamic therapy may enhance the photodynamic effect in tumor areas with insufficient blood supply by enabling the reoxygenation of the treated area. This study addresses the outcome of fractionated irradiation in an in vitro photodynamic treatment (PDT) system, where deoxygenation can be neglected. Our results show that fractionated irradiation with light/dark intervals of 45/60 s decreases ROS production and cytotoxicity of PDT. This effect can be reversed by addition of 1,3-bis-(2-chlorethyl)-1-nitrosurea (BCNU), an inhibitor of the glutathione reductase. We suggest that the dark intervals during irradiation allow the glutathione reductase to regenerate reduced glutathione (GSH), thereby rendering cells less susceptible to ROS produced by PDT compared with continuous irradiation. Our results could be of particular clinical importance for photodynamic therapy applied to well-oxygenated tumors.  相似文献

A microchip electrophoresis method was developed for simultaneous determination of reactive oxygen species (ROS) and reduced glutathione (GSH) in the individual erythrocyte cell. In this method, cell sampling, single-cell loading, docking, lysing, and capillary electrophoretic separation with LIF detection were integrated on a microfluidic chip with crossed channels. ROS was labeled with dihydrorhodamine 123 in the intact cell, while GSH was on-chip labeled with 2,3-naphthalene-dicarboxaldehyde, which was included in the separation medium. On-chip electrical lysis, characterized by extremely fast disruption of the cellular membrane (<40 ms), was exploited to minimize enzymatic effects on analyte concentrations during the determination. The microfluidic network was optimized to prevent cell leaking from the sample reservoir (S) into separation during the separation phase. The structure of the S was modified to avoid blockage of its outlet by deposited cells. Detection limits of 0.5 and 6.9 amol for ROS and GSH, respectively, were achieved. The average cell throughput was 25 cells/h. The effectiveness of the method was demonstrated in the simultaneous determination of GSH and ROS in individual cells and the variations of cellular GSH and ROS contents in response to external stimuli.  相似文献

The influence of various phenolic compounds on the lactoperoxidase (LPO)/hydrogen peroxide (H2O2)-catalyzed oxidation of biochemical reductants such as reduced beta-nicotinamide adenine dinucleotide (NADH), reduced beta-nicotinamide adenine dinucleotide phosphate (NADPH) or reduced glutathione (GSH) was investigated by electron spin resonance (ESR) spectroscopy. Micromolar quantities of phenolic compounds such as 17beta-estradiol, phenol, and p-chlorophenol enhanced the LPO/H2O2-catalyzed oxidation of NAD(P)H or GSH to generate a large amount of superoxide radical (O2*-) or glutathione thiyl radical (GS*), while, phenolic compounds such as quercetin and Trolox C greatly suppressed the generation of O2*- and GS*. In order to elucidate the effects of phenolic compounds on the generation of O2*- and GS*, their quenching activities for a stable radical, 1,1-diphenyl-2-picrylhydrazyl (DPPH), were investigated by ESR spectroscopy. 17beta-Estradiol, phenol, and p-chlorophenol showed very weak scavenging activities for DPPH, but quercetin and Trolox C showed strong activities. This suggests that the ability of phenolic compounds to enhance LPO/H2O2-catalyzed oxidation of NAD(P)H or GSH relates inversely to their ability to quench DPPH. That is, phenolic compounds having weak quenching activity against DPPH may enhance the LPO/H2O2-catalyzed oxidation of NAD(P)H or GSH to generate a large amount of O2*- or GS*.  相似文献

Graphitic carbon nitride (g‐C3N4) has been used as photosensitizer to generate reactive oxygen species (ROS) for photodynamic therapy (PDT). However, its therapeutic efficiency was far from satisfactory. One of the major obstacles was the overexpression of glutathione (GSH) in cancer cells, which could diminish the amount of generated ROS before their arrival at the target site. Herein, we report that the integration of Cu2+ and g‐C3N4 nanosheets (Cu2+–g‐C3N4) led to enhanced light‐triggered ROS generation as well as the depletion of intracellular GSH levels. Consequently, the ROS generated under light irradiation could be consumed less by reduced GSH, and efficiency was improved. Importantly, redox‐active species Cu+–g‐C3N4 could catalyze the reduction of molecular oxygen to the superoxide anion or hydrogen peroxide to the hydroxyl radical, both of which facilitated the generation of ROS. This synergy of improved ROS generation and GSH depletion could enhance the efficiency of PDT for cancer therapy.  相似文献

Crystallins in the retina may serve a chaperone-like protective function. In this study we measured mRNA levels for alpha-, beta- and gamma-crystallins in rat retinas following treatment with potentially damaging levels of light. We also determined crystallin protein patterns in photoreceptor cell rod outer segments (ROSs) isolated from rats exposed to intense light. Weanling albino rats were maintained in a dim cyclic light environment or in darkness for 40days. At P60 animals were treated with intense visible light, for as long as 8h, beginning at various times of the day or night. Retinas were excised immediately after light treatment and used for quantitative RT-PCR, or to prepare ROSs for western analysis. Some eyes were frozen in OCT for crystallin immunohistochemistry. Intense light exposure led to increases in mRNA expression for all retinal crystallins and to changes in ROS crystallin immunoreactivity. These light-induced changes were found to depend on the time of day that exposure started, duration of light treatment and previous light rearing history. We suggest that crystallin synthesis in retina exhibits a dependence on both light stress and circadian rhythm and that within photoreceptor cells crystallins appear to migrate in a light-independent, circadian fashion.  相似文献

Biologically, light including ultraviolet (UV) radiation is vital for life. However, UV exposure does not come without risk, as it is a major factor in the development of skin cancer. Natural protections against UV damage may have been affected by lifestyle changes over the past century, including changes in our sun exposure due to working environments, and the use of sunscreens. In addition, extended "day time" through the use of artificial light may contribute to the disruption of our circadian rhythms; the daily cycles of changes in critical bio-factors including gene expression. Circadian disruption has been implicated in many health conditions, including cardiovascular, metabolic and psychiatric diseases, as well as many cancers. Interestingly, the pineal hormone melatonin plays a role in both circadian regulation as well as protection from UV skin damage, and is therefore an important factor to consider when studying the impact of UV light. This review discusses the beneficial and deleterious effects of solar exposure, including UV skin damage, Vitamin D production, circadian rhythm disruption and the impact of melatonin. Understanding these benefits and risks is critical for the development of protective strategies against solar radiation.  相似文献

Abstract— The organization of the circadian timing system in Saimiri sciureus was probed using the phasic (abrupt transition) and tonic (continuous action) effects of light intensity. The behavior of the simultaneously monitored circadian rhythms of feeding behavior, colonic temperature, and urinary potassium excretion was studied in response to the phasic effects of (a) an abrupt 8-h phase delay in the light–dark (LD) cycle and (b) a series of non-24 h LD cycles ( T = 18 to 30 h). These studies demonstrated that the feeding and temperature rhythms were more tightly coupled to the light-dark cycle than was the rhythm of urinary potassium excretion. The tonic effects of constant levels of illumination confirmed this conclusion. In constant light, internal desynchronization spontaneously occurred in 25% of animals with the potassium rhythm exhibiting a period quite different from that of the feeding and colonic temperature rhythms. Thus, the response of the internal circadian timekeeping system to phasic and tonic light inputs shows that the system in this species comprises at least two potentially independent oscillators with differential light sensitivities.  相似文献

A capillary zone electrophoretic (CZE) method coupled with laser-induced fluorescence (LIF) was developed for the simultaneous determination of two important intracellular parameters related to oxidative stress (i.e. reactive oxygen species, ROS, and reduced glutathione, GSH). This rapid and sensitive method was applied to the study of oxidative stress in cultured V79 fibroblast cells. The fluorogenic reagents selected were: (i) dihydrorhodamine-123 (DHR-123) which is converted intracellularly by ROS to the fluorescent rhodamine-123 dye (Rh-123), and (ii) naphthalene-2,3-dicarboxaldehyde (NDA), which reacts quickly with GSH in cell extracts to produce a fluorescent adduct. Separation of Rh-123, GSH-NDA and gamma-glutamylcysteine-NDA adducts was performed using an uncoated fused-silica capillary and a 100 mM borate buffer, pH 9.2, at 20 degrees C and at an applied voltage of 25 kV; LIF detection was operated using an argon laser. The cell line was also tested for its ability to alleviate oxidative stress induced by tert-butylhydroperoxide (t-BuOOH). Exposure to t-BuOOH (up to 3 mm for 2 h) did not affect the intracellular ROS and GSH concentrations. At higher (4-10 mM) t-BuOOH concentrations, an inverse relationship between the concentrations of ROS and GSH was obtained, showing that the present method can readily evaluate the gradual consumption of the primary cellular scavenger of ROS which occurs simultaneously with the increase of oxidative insult.  相似文献

To investigate the effect of a blue light-filtering intraocular lens (IOL) and a UV-absorbing IOL on light-induced damage to retinal pigment epithelial (RPE) cells laden with the lipofuscin fluorophore N -retinylidene- N -retinylethanolamine (A2E), A2E-laden RPE cells were exposed to white light which was filtered by either a blue light-filtering IOL or a UV-absorbing IOL. After 30 min of illumination the cell viability and the level of reactive oxygen species (ROS), free glutathione (GSH), vascular endothelial growth factor (VEGF) and pigment epithelium-derived factor (PEDF) were determined. In the absence of an IOL, the white light exposure decreased cell viability to 37.2% of the nonirradiated control. The UV-absorbing IOL tended to reduce light-induced cell death; however, the decrease was not significant. The blue light-filtering IOL significantly attenuated light-induced cell damage, increasing cell viability to 79.5% of the nonirradiated control. The presence of the blue light-filtering IOL significantly increased GSH and PEDF levels, and decreased ROS and VEGF levels. This study suggests that a blue light-filtering IOL may be more protective against A2E-induced light damage and inhibit more light-induced ROS and VEGF production than a conventional UV-absorbing IOL.  相似文献

The glutathione (GSH) redox reaction is critical for defense against cellular reactive oxygen species (ROS). However, direct and real‐time monitoring of this reaction in living mammalian cells has been hindered by the lack of a facile method. Herein, we describe a new approach that exploits the GSH biosynthetic pathway and heteronuclear NMR. [U‐13C]‐labeled cysteine was incorporated into GSH in U87 glioblastoma cells, and the oxidation of GSH to GSSG by a ROS‐producing agent could be monitored in living cells. Further application of the approach to cells resistant to temozolomide (TMZ), an anti‐glioblastoma drug, suggested a possible new resistance mechanism involving neutralization of ROS. This result was corroborated by the observation of up‐regulation of glutathione peroxidase 3 (GPx3). This new approach could be easily applied to redox‐dependent signaling pathways and drug resistance involving ROS.  相似文献

Spatial and temporal control over chemical and biological processes plays a key role in life and material sciences. Here we synthesized a two‐photon‐activatable glutathione (GSH) to trigger the interaction with glutathione S‐transferase (GST) by light at superior spatiotemporal resolution. The compound shows fast and well‐confined photoconversion into the bioactive GSH, which is free to interact with GST‐tagged proteins. The GSH/GST interaction can be phototriggered, changing its affinity over several orders of magnitude into the nanomolar range. Multiplexed three‐dimensional (3D) protein networks are simultaneously generated in situ through two‐photon fs‐pulsed laser‐scanning excitation. The two‐photon activation facilitates the three‐dimensional assembly of protein structures in real time at hitherto unseen resolution in time and space, thus opening up new applications far beyond the presented examples.  相似文献
