The HPLC enantiomeric separation of seven 4-iminoflavans was successfully accomplished in the normal phase mode using six polysaccharide-based chiral stationary phases namely, Chiralcel®OD-H, Chiralcel®OD, Chiralcel®OJ, Chiralpak®AD, Chiralpak®IA and Chiralpak®IB under normal and polar organic phase modes. The resolution depended on nature and concentration of alcoholic modifier. The results demonstrate clearly that the chromatographic system based on the coated and immobilized type Chiralpak®IB and Chiralcel®OD-H CSPs provide a powerful analytical tool for enantiomeric separation of all the 4-iminoflavans used in this study.

A novel chiral stationary phase (CSP) consisting of a silica-based chitosan tris(3-chlorophenylcarbamate) derivative was used for the enantioseparation of nine racemic compounds in the normal phase mode by high-performance liquid chromatography (LC). The influence of the type and percentage of modifier in the mobile phase on the optimization of the resolution was investigated. The resolution of epoxiconazole using this system was compared with those achieved using commercially available Chiralcel® OD-H and Chiralpak® AD-H columns. Among the tested racemates, seven racemic compounds were separated most quickly on the CSP of chitosan tris(3-chlorophenylcarbamate). Baseline or near-baseline separation was achieved for benzoin, penconazole, hexaconazole, and epoxiconazole, while the others were partially separated. The enantioseparation results for epoxiconazole were not inferior to those of commercially available Chiralcel® OD-H and Chiralpak® AD-H columns.  相似文献

The challenging separation of regioisomers and enantiomers of monoacylglycerols (MAGs) of fatty acids has been achieved using a single chiral HPLC column (Chiralpak® IC or Chiralpak® IA) and hexane/2‐propanol eluent mixtures, without previous derivatization of the samples. The efficacy of the method is independent of the chain length and the degree of unsaturation of the fatty acids of the MAG. In addition, the separation method allows monitoring the time course for the loss of enantiomeric purity of the fatty acid‐derived MAGs in polar hydroxylic solvents.  相似文献

Analytical HPLC methods using derivatized cellulose and amylose chiral stationary phases were developed for the separation of the enantiomers of homocamptothecin (hCPT) derivatives which constitute a promising series of potent anticancer agents targeting DNA topoisomerase I. The resolutions were performed using a normal phase methodology with two silica-based celluloses tris-3,5-dimethylphenylcarbamate (Chiralcel OD-H) and tris-methylbenzoate (Chiralcel OJ) or two amyloses tris-3,5-dimethylphenylcarbamate (Chiralpak AD) and tris-(S)-1-phenylethylcarbamate (Chiralpak AS). The mobile phase and the chiral stationary phase were varied to achieve the best resolution. Different types and concentration of aliphatic alcohols in the mobile phase were also tested along with the temperature dependence. An optimal baseline separation (Rs > 1.5) was readily obtained in most cases. The different columns gave complementary results in term of resolution. The limits of detection and quantification were between 0.08–0.40 M and 0.24–1.80 M, respectively and the enantiomeric purity was superior to 99.9%.  相似文献

HPLC method enabling chiral separation and determination of citalopram (CIT), a widely used antidepressant, and its synthetic precursor citadiol in one analysis was developed and validated. Moreover, supercritical fluid chromatography was also tested and was proved to be less effective for this separation purpose. The optimized HPLC system was composed of Chiralcel OD-H column and n-hexane/propane-2-ol/triethylamine 96/4/0.1 (v/v/v) as mobile phase, column temperature 25 °C, flow rate 1.0 mL min?1, UV detection at 250 nm. The effects of amount of propane-2-ol, triethylamine addition, and temperature on enantioselectivity and resolution of the enantiomers were evaluated. The method was found to be suitable for determination of the enantiomeric purity of CIT in bulk drugs. Enantiomers of CIT were determined in two commercially available pharmaceuticals.  相似文献

Summary This paper describes the enantiorecognition of 1-methyl-3-hydroxy-5-aryl-2-pyrrolidinonic systems by high-performance liquid chromatography using two chiral derivatized cellulose stationary phases (CSPs) operated in the normal phase mode. According our results, the tris-(3,5-dimethylphenyl carbamate) cellulose, Chiralcel? OD, is more suitable than the tris-(4-methyl-phenylbenzoate), Chiralcel? OJ. On the first column, the resolution of the pyrrolidinonic compounds depends on the alcoholic modifier percentage. A possible solute-stationary phase recognition mechanism is discussed. The temperature and mobile phase composition have been considered to explain the different contribution for the enantiomeric resolution.  相似文献

A sensitive and accurate LC method was developed and further validated for the determination of enantiomeric purity of GSK962040. Before separation, a pre-column derivatization procedure was performed. Baseline separation with a resolution higher than 1.9 was accomplished within 15 min using a Chiralpak AD-H (250 × 4.6 mm; particle size 5 μm) column, with n-hexane: 2-propanol (85:15 v/v) as mobile phase at a flow rate of 1 mL min?1. The eluted analytes were subsequently detected with a UV detector at 260 nm. The effects of mobile phase components and temperature on enantiomeric selectivity as well as resolution of enantiomers were thoroughly investigated. The calibration curves were plotted within the concentration range between 4 and 200 μg mL?1 (n = 8), and recoveries between 98.15 and 101.48% were obtained, with relative standard deviation (RSD) lower than 1.42%. The LOD and LOQ for the Boc-GSK962040 were 1.23 and 4.15 μg mL?1 and for its enantiomer were 1.38 and 4.76 μg mL?1, respectively. The developed method was also evaluated and validated by analyzing bulk samples with different enantiomeric ratios of GSK962040. It was demonstrated that the method was accurate, robust and sensitive, and also had practical utilities for real analysis.  相似文献

The enantiomers of 1-(α-aminoarylmethyl)-2-naphthol, 1-(α-aminoalkyl)-2-naphthol and 2-(α-aminoarylmethyl)-1-naphthol analogues were separated on tris(3,5-dimethylphenyl)carbamoyl cellulose-based CelluCoat and Chiralcel OD-H chiral stationary phases, with n-heptane/alcohol or n-hexane/alcohol as mobile phase. The experimental data are utilised to discuss the effects of the mobile phase composition, the nature of the alcoholic modifier and the specific structural features of the analytes (1- or 2-naphthol analogues with aryl or alkyl substituents) on the retention and separation. The separation performances of CelluCoat and Chiralcel OD-H columns were compared. Due to its high resolution ability and its effectivity, CelluCoat proved to be a good choice for the enantiomeric separation of aminonaphthol analogues.  相似文献

Radio high pressure liquid chromatography (radio-HPLC) is the method of choice for quality control of radiopharmaceuticals labelled with short lived isotopes. Our preparations of “no carrier added”11C-labelled palmitic acid and L-methionine are both designed to end with a HPLC separation on either silica gel or C-18 reversed phase material. Since the crude reaction mixtures contain milligram amounts of inactive substrate materials, both separations must be carried out at preparative scale. Nevertheless they are performed in less than 10 min. The most critical factor for the separation of11C-palmitic acid from the main by-product pentadecane is the solvent composition: while the11C-L-methionine separation is especially sensitive to pH variations.  相似文献

An alternative green microextraction method based on ultrasound-assisted surfactant-enhanced emulsification microextraction (UASEME) using a low-density extraction solvent coupled with HPLC has been developed for preconcentration and determination of six benzimidazole anthelmintics, namely, oxfendazole, albendazole, mebendazole, flubendazole, fenbendazole, and niclosamide. The separation was achieved within 12 min, using an Inertsil® C18 column (4.6 × 150 mm, 5.0 µm), with a gradient mobile phase of acetonitrile and 0.1 % (v/v) formic acid. Under the optimum UASEME conditions using Tergitol® TMN-6 and 1-octanol as emulsifier and extraction solvent, respectively, linearity was in the range of 0.5–5,000 μg L?1 with the coefficients of determination (R 2) ranging from 0.9959 to 0.9999. Enrichment factors were obtained up to 89, corresponding to limits of detection ranging from 0.50 to 6.00 µg L?1. Intra-day (n = 8) and inter-day (n = 3 × 3) precisions were obtained with relative standard deviations for retention time and peak area of lower than 2 and 15 %, respectively. The proposed method was successfully applied to determine the target benzimidazoles in milk formulae.  相似文献

A simple and accurate chiral liquid chromatographic method was developed for the enantiomeric purity determination of d-nateglinide and quantitative determination of l-nateglinide in bulk drug samples. Good resolution (R s  > 6.0) between d-enantiomer and l-enantiomer of nateglinide were achieved with Chiralpak AD-H (250 × 4.6 mm, 5 μm particle size) column using hexane and ethanol (90:10 v/v) as mobile phase at 25 °C temperature. Flow rate was kept as 1.0 mL min?1 and elution was monitored at 210 nm. The effects of the mobile phase composition, the flow rate and the temperature on the chromatographic separation were investigated. Developed method is capable to detect (LOD) and quantitate (LOQ) l-nateglinide to the levels of 0.3 and 1.0 μg mL?1 respectively, for 10 μL injection volume. The percentage RSD of the peak area of six replicate injections of l-nateglinide at LOQ concentration was 5.2. The percentage recoveries of l-nateglinide from d-nateglinide ranged from 97.9 to 99.7. The test solution and mobile phase was found to be stable up to 24 h after preparation. The developed method was validated with respect to LOD, LOQ, precision, linearity, accuracy, robustness and ruggedness.  相似文献

The high-performance liquid chromatographic separation of enantiomers of N-protected non-protein amino acid esters was investigated by using a cellulose tris(3,5-dimethylphenylcarbamate) chiral stationary phase column (Daicel Chiracel OD). The effect of the N-protecting groups and the ester groups on chiral discrimination was examined. The benzyloxycarbonyl (Z), 4-methoxybenzyloxycarbonyl, and 9-fluorenylmethoxycarbonyl derivatives gave good enantiomeric separations, while the formyl and t-butoxycarbonyl groups marred them. Almost all the alkyl esters examined and the benzyl ester gave enantiomeric separations better than or of the same order as the methyl ester. The N-Z-protected methyl esters of a number of non-protein -amino acids were well resolved using hexane–2-propanol as a mobile phase. The resolution of -amino acid derivatives was inferior to that of the isomeric -amino acid derivatives.  相似文献

Ultraperformance convergence chromatography/tandem triple quadrupole mass spectrometry (UPC2-MS/MS) is a novel tool in separation science that combines the advantages of supercritical fluid chromatography with ultraperformance liquid chromatography/MS/MS technology. The use of nontoxic CO2 fluid and a postcolumn additive to complement MS/MS allows better control of analyte retention for chiral separation and high-sensitivity determination with different chiral stationary phases. This paper reports the stereoselective separation and determination of the chiral neonicotinoid sulfoxaflor in vegetables and soil by UPC2-MS/MS. Baseline resolution (Rs?≥?1.56) of and high selectivity (LOQ?≤?1.83 μg/kg) for the four stereoisomers were achieved by postcolumn addition of 1 % formic acid–methanol to a Chiralpak IA-3 using CO2/isopropanol/acetonitrile as the mobile phase at 40 °C, 2,500 psi, and for 6.5 min in electrospray ionization positive mode. Rearranged Van’t Hoff equations afforded the thermodynamic parameters ΔH ο and ΔS ο, which were analyzed to promote understanding of the enthalpy-driven separation of sulfoxaflor stereoisomers. The interday mean recovery, intraday repeatability, and interday reproducibility varied from 72.9 to 103.7 %, from 1.8 to 9.2 %, and from 3.1 to 9.4 %, respectively. The proposed method was used to study the pharmacokinetic dissipation of sulfoxaflor stereoisomers in soil under greenhouse conditions. The estimated half-life ranged from 5.59 to 6.03 d, and statistically nonsignificant enantioselective degradation was observed. This study not only demonstrates that the UPC2-MS/MS system is an efficient and sensitive method for sulfoxaflor stereoseparation, but also provides the first experimental evidence of the pharmacokinetic dissipation of sulfoxaflor stereoisomers in the environment. A solvent extraction system has been developed for the separation of172Lu from its long-lived172Hf parent. The parent-daughter pair in equilibrium is maintained in a solution of HDEHP, and the heavy lanthanide daughter is extracted into 9M HCl. The separation factor for this generator is approximately 104. The rare-earth activity so obtained is proposed for compound labelling research and animal biodistribution studies in nuclear medicine.  相似文献

A systematic study is presented on the extraction of60Co and65Zn bromides and chlorides from H2SO4 with tri-n-butyl phosphate (TBP) and Amberlite LA-2. It has been found that the presence of a little HBr or HCl enhances the extraction of the two isotopes with both solvents. The extractions of60Co and65Zn have been studied as functions of the acidity (HBr, HCl or H2SO4) in the aqueous phase, and of the diluent and solvent concentrations in the organic phase. The presence of water-miscible alcohols or acetone enhances extraction. A suggested extraction mechanism is presented in the light of the obtained results.  相似文献

Summary A new type of chiral stationary phase has been prepared by coating porous graphite with a near-monolayer of an adsorbed enantiomeric modifier which then acts as an adsorbed stationary phase. The most effective modifiers are L- or D-isomers of N-(2-naphthalene-sulphonyl)-phenylalanine (NS-Phe). Conveniently 80% of monolayer coverages, corresponding to 1.2 mol m–2, are achieved by adsorption of NS-Phe from methanolic solution. Enantiomeric separations by complexation with cupric ion show base-line resolution of -amino and -hydroxy acids with separation factors up to 2. Identical separations are achieved with chiral phases made from the L- and D-isomers of NS-Phe except that the elution orders of enantiomers are inverted. Reduced plate heights are around 5 and the adsorbed chiral phases are extremely stable, retention ratios being unchanged after passage of 7000 column volumes of eluent. A mechanism of retention is proposed, which fits the experimental observations.  相似文献

A simple solvent extraction procedure for the efficient separation of the radioactive tracers95Nb and182Ta from each other in a mixture using di-(2-ethylhexyl)phosphoric acid (HDEHP) as extractant is described. Tantalum was found to be quantitatively extracted from an aqueous madium, which is 1.6N in HCl and 10?2 M in oxalic acid, with a HDEHP solution of 0.1 M concentration. Extractabilities of both niobium and tantalum in mineral acids like HCl, H2SO4 and HNO3 and in some organic acids like oxalic, citric, etc., in HDEHP under the experimental conditions were also studied. The reliability of the separation procedure was verified further by γ-ray spectrometry.  相似文献

Eggshell material was used as low-cost and eco-friendly biosorbent for removal of 134Cs, 60Co and 152+154Eu radionuclides from aqueous solution. The eggshell material was calcined at 500 and 800 °C, and then characterized. Comparative studies on the natural and calcined eggshell for sorption of the three radionuclides were carried out. It was found that, the uptake is in the order: Eu(III) > Co(II) > Cs(I). Further, column chromatography was used in separation of 134Cs, 60Co and 152+154Eu using 0.15, 0.2 and 0.5 mol/l nitric acid, respectively. Eggshell material can be considered as a promising material for separation of radionuclides from radioactive waste solution.  相似文献

High-performance liquid chromatography with chemiluminescence detection based on the reaction with acidic potassium permanganate and formaldehyde was explored for the determination of neurotransmitters and their metabolites. The neurotransmitters norepinephrine and dopamine were quantified in the left and right hemispheres of rat hippocampus, nucleus accumbens and prefrontal cortex, and the metabolites vanillylmandelic acid, 3,4-dihydrophenylacetic acid, 5-hydroxyindole-3-acetic acid and homovanillic acid were identified in human urine. Under optimised chemiluminescence reagent conditions, the limits of detection for these analytes ranged from 2.5?×?10?8 to 2.5?×?10?7 M. For the determination of neurotransmitter metabolites in urine, a two-dimensional high-performance liquid chromatography (2D-HPLC) separation operated in heart-cutting mode was developed to overcome the peak capacity limitations of the one-dimensional separation. This approach provided the greater separation power of 2D-HPLC with analysis times comparable to conventional one-dimensional separations. Figure

A very rapid method for the separation of molybdenum(VI) from neutron irradiated uranium and its fission products is described. The procedure is based on the selective extraction of molybdenum(VI) by a 0.1M solution of 2-hexylpyridine in benzene from 4M HCl+0.04M KSCN. Decontamination factors were estimated to be >104 for the radionuclides of niobium, zirconium, ruthenium, lanthanum, cerium, promethium, yttrium, strontium and barium.  相似文献
