A series of 4‐aryl‐3,7,7‐trimethyl‐1‐phenyl‐7,8‐dihydro‐1H‐pyrazolo[3,4‐b]quinolin‐5(6H)‐ones were synthesized via the three‐component reaction of aromatic aldehydes, 5,5‐dimethyl‐1,3‐cyclohexandione and 5‐amino‐3‐methyl‐1‐phenylpyrazole in ionic liquid without using any catalyst. This protocol has the advantages of easier work‐up, milder reaction conditions, short reaction time, and environmentally benign procedure. J. Heterocyclic Chem., (2011).  相似文献

The synthesis of ethyl 6‐aryl‐4‐oxo‐4,6‐dihdro‐1(12)(13)H‐pyrimido[2′,1′:4,5][1,3,5]triazino[1,2‐a]‐benzimidazole‐3‐carboxylates ( 4a‐p ) was described via pyrimidine ring annulation to 4‐aryl‐3,4‐dihydro[1,3,5]triazino[1,2‐a]benzimidazole‐2‐amines ( 2a‐p ) which were obtained from 2‐guanidinobenzimidazole ( 1 ). Tautomerism in the prepared compounds was investigated using nmr spectroscopy. Compounds 2a‐p were found to be present in dimethyl sulfoxide solution predominantly as 3,4‐dihyhydro tautomeric form. Compounds 4a‐p existed in dynamic equilibrium of 1‐, 12‐ and 13H‐forms. It was found that methylation of 4a‐d led to 13‐methyl substituted derivatives 9a‐d exclusively.  相似文献

A series of 4‐aryl‐3,7,7‐trimethyl‐1‐phenyl‐7,8‐dihydro‐1H‐pyrazolo[3,4‐b]quinolin‐ 5(6H)‐ones were synthesized by the three‐component reaction of aromatic aldehydes, 3‐methyl‐1‐phenyl‐1H‐pyrazol‐5‐amine, and 5,5‐dimethyl‐1,3‐cyclohexandione in the presence of sodium 1‐dodecanesulfonate in aqueous medium. Compared to the previous methods, this new protocol has the advantages of convenient operation, higher yields, lower cost, and environmentally benign procedure. J. We previously reported that reactivity towards the Suzuki cross‐coupling reaction of 3‐iodoimidazo[1,2‐a]pyridines substituted at C(2) is largely influenced by the nature of this 2‐substituent. Hence, with the aim to expand the scope of this coupling process to the 6‐position of this series, it seemed important to similarly determine the influence of the nature of the 2‐substituent (H, alkyl, or aryl) on the rate of coupling. From this work, the Suzuki‐type cross‐coupling was shown to proceed efficiently on 6‐bromo‐2‐methyl‐ and 2‐(4‐fluorophenyl)imidazo[1,2‐a]pyridines, whereas the 6‐Br derivative unsubstituted at C(2) appeared to be poorly reactive. By modifying the reaction conditions in terms of catalyst and base, and the nature of the halogen, the reactivity of the unsubstituted series was largely enhanced. Finally, this work led us to establish efficient and convenient Suzuki reaction conditions for the 6‐(hetero)arylation of 6‐halogenoimidazo[1,2‐a]pyridines depending on the nature of the 2‐substituent and boronic acid.  相似文献

The cycloaddition reaction of cyclic imidates, 2‐benzyl‐5,6‐dihydro‐4H‐1,3‐oxazines 1a , 1b , 1c , 1d , 1e , 1f , with dimethyl acetylenedicarboxylate 2 , trimethyl ethylenetricarboxylate 4 , or dimethyl 2‐(methoxymethylene)malonate 6 afforded new fused heterocyclic compounds, such as methyl (6‐oxo‐3,4‐dihydro‐2H‐pyrrolo[2,1‐b]‐1,3‐oxazin‐7‐ylidene)acetates 3a , 3b , 3c , 3d , 3e , 3f (71–79%), dimethyl 2‐(6‐oxo‐3,4,6,7‐tetrahydro‐2H‐pyrrolo[2,1‐b]‐1,3‐oxazin‐7‐yl)malonates 5b , 5c , 5d , 5e , 5f (43–71%), or methyl 6‐oxo‐3,4‐dihydro‐2H,6H‐pyrido[2,1‐b]‐1,3‐oxazine‐7‐carboxylates 7a , 7b , 7c , 7d , 7e , 7f (32–59%), respectively. In these reactions, 1a , 1b , 1c , 1d , 1e , 1f (cyclic imidates, iminoethers) functioned as their N,C‐tautomers (enaminoethers) 2 to α,β‐unsaturated esters 2 , 4, and 6 to give annulation products 3 , 5 , and 7 following to the elimination of methanol, respectively. A novel and efficient isocyanide‐based multicomponent reaction between alkyl or aryl isocyanides 1 , 2,3‐diaminomaleonitrile ( 2 ), naphthalene‐2,3‐diamines ( 6 ) or benzene‐1,2‐diamine ( 9 ), and 3‐oxopentanedioic acid ( 3 ) or Meldrum's acid ( 4 ) or ketones 7 was developed for the ecologic synthesis, at room temperature under mild conditions, of 1,6‐dihydropyrazine‐2,3‐dicarbonitriles 5a – 5f in H2O without using any catalyst, and of 3,4‐dihydrobenzo[g]quinoxalin‐2‐amine and 3,4‐dihydro‐3,3‐dimethyl‐quinoxalin‐2‐amine derivatives 8a – 8g and 10a – 10e , respectively, in the presence of a catalytic amount of p‐toluenesulfonic acid (TsOH) in EtOH, in good to excellent yields (Scheme 1).  相似文献

Novel 11‐amino‐6‐aryl‐6,7‐dihydroindeno[1,2‐e] pyrimido[4,5‐b][1,4]diazepin‐5(5aH)‐ones 4a‐f were prepared regioselectively by the tricomponent reaction of 4,5,6‐triaminopyrimidine 1, 1,3‐indandione 2 and aromatic aldehydes 3a‐f. The bicomponent approach, using 2,4,5,6‐tetraaminopyrimidine 5 and 2‐aryl‐ideneindandiones 6a‐f as reagents, afforded 9,11‐diamino‐6‐aryl‐6,7‐dihydroindeno[1,2‐e]pyrimido[4,5‐b]‐[1,4]diazepin‐5(5aH)‐ones 7a‐f in good yields and the regioisomeric 8,10‐diamino derivatives 8a‐c in lower yields. Both, bi‐ and tricomponent approaches were performed by microwave irradiation and all products were fully characterized by detailed NMR measurements.  相似文献

A series of 6‐substituted‐2‐(4‐methyl‐2‐substituted phenylthiazol‐5‐yl)H‐imidazo[1,2‐a]pyridine derivatives 4a , 4b , 4c , 4d , 4e , 4f , 4g , 4h , 4i , 4j , 4k , 4l is described. The antitubercular activity of the synthesized compounds was determined against Mycobacterium smegmatis MC2 155 strain. From the activity result, it was found that the phenyl or 4‐fluorophenyl group at 2 position of thiazole nucleus and bromo substituent at 6 position of imidazo[1,2‐a]pyridine showed good antitubercular activity.  相似文献

One‐pot reaction of 3‐aryl‐5‐methyl‐1,3,4‐oxadiazolin‐2‐ones 1a‐g with ethanolamine yielded the 4‐(2‐hydroxyethyl)‐2‐aryl‐5‐methyl‐2,4‐dihydro‐3H‐1,2,4‐triazolin‐3‐ones 2a‐g which were converted to the azido compounds 6a‐g . These azides on 1,3‐dipolar cycloaddition with DMAD afforded the dimethyl‐1‐[2‐(2‐aryl‐5‐methyl‐3‐oxo‐1,2,4‐triazol‐4‐yl)ethyl]‐1H‐1,2,3‐triazol‐4,5‐dicarboxylates 7a‐g which on conversion to bishydrazides 8a‐g and further cyclisation with 2,5‐hexanedione afforded the title compounds 9a‐g . This new short route for the so far unkown bis‐(triazolinone‐triazole)ethanes involves mild and convergent 1,3‐dipolar cycloaddition reaction yielding overall good yields of the products.  相似文献

A novel method is reported for the synthesis of 9,9‐disubstituted 9H‐pyrrolo[1,2‐a]indoles. Cyclization of 1‐[2‐(1‐aryl(or methyl)ethenyl)phenyl]‐1H‐pyrroles, which can be easily prepared from 2‐(1‐aryl(or methyl)ethenyl)anilines, proceeds smoothly, in general, at 0° in the presence of a catalytic (or an equimolar) amount of HI in MeCN to provide the desired products.  相似文献

Treatment of 3‐methylamino‐5‐phenylthiophene with α,β‐unsaturated esters, i.e., methyl acrylate, (E)‐methyl crotonate, diethyl fumarate, diethyl maleate and ethyl propiolate, in tetrahydrofuran for several days at reflux gave 1‐methyl‐3,4‐dihydrothieno[2,3‐e]pyridin‐2‐ones 4 and/or 1‐methylthieno[2,3‐e]pyridin‐2‐ones 5 , depending on the structure of the esters. On the other hand, the same reactions with α,β‐unsaturated nitriles such as acrylonitrile and tetracyanoethene, gave the corresponding thiophenes 7 and 10 bearing 2‐cyanoethyl and 1,2,2‐tricyanoethenyl groups at C‐2, respectively. The reaction with (Z)‐1,2‐dicyanoethene under the same conditions produced the corresponding thiophene 9 bearing the 1,2‐dicyanoethenyl group and 1,2‐dicyano‐5‐methylaminobiphenyl.  相似文献

In this study, 10 different substituted aromatic bis‐benzaldehydes were synthesized by treating hydroxy benzaldehydes with various dihaloalkanes. Bis aldehydes 5a , 5b , 5c , 5d , 5e , 5f , 5g , 5h , 5i , 5j were treated with 2‐(5‐phenyl‐1H‐tetrazole‐1‐yl)acetohydrazide ( 3 ) in acidic medium and in the presence of ammonium acetate to yield a series of new isomeric bis(2‐(5‐((5‐phenyl‐1H‐tetrazol‐1‐yl)methyl)‐4H‐1,2,4‐triazol‐3‐yl)phenoxy)alkanes ( 6a , 6b , 6c , 6d , 6e , 6f , 6g , 6h , 6i , 6j ) in excellent to good yield. The newly synthesized compounds were characterized by the available spectroscopic analysis.  相似文献

The photochemical reactions of 2‐substituted N‐(2‐halogenoalkanoyl) derivatives 1 of anilines and 5 of cyclic amines are described. Under irradiation, 2‐bromo‐2‐methylpropananilides 1a – e undergo exclusively dehydrobromination to give N‐aryl‐2‐methylprop‐2‐enamides (=methacrylanilides) 3a – e (Scheme 1 and Table 1). On irradiation of N‐alkyl‐ and N‐phenyl‐substituted 2‐bromo‐2‐methylpropananilides 1f – m , cyclization products, i.e. 1,3‐dihydro‐2H‐indol‐2‐ones (=oxindoles) 2f – m and 3,4‐dihydroquinolin‐2(1H)‐ones (=dihydrocarbostyrils) 4f – m , are obtained, besides 3f – m . On the other hand, irradiation of N‐methyl‐substituted 2‐chloro‐2‐phenylacetanilides 1o – q and 2‐chloroacetanilide 1r gives oxindoles 2o – r as the sole product, but in low yields (Scheme 3 and Table 2). The photocyclization of the corresponding N‐phenyl derivatives 1s – v to oxindoles 2s – v proceeds smoothly. A plausible mechanism for the formation of the photoproducts is proposed (Scheme 4). Irradiation of N‐(2‐halogenoalkanoyl) derivatives of cyclic amines 5a – c yields the cyclization products, i.e. five‐membered lactams 6a , b , and/or dehydrohalogenation products 7a , c and their cyclization products 8a , c , depending on the ring size of the amines (Scheme 5 and Table 3).  相似文献

The reaction of 4‐amino‐5,5‐dimethyl‐5H‐1,2‐oxathiole 2,2‐dioxide ( 1 ) with 2‐(arylidene)malononitriles 2 in ethanol, at reflux, using piperidine as catalyst, afforded 5‐amino‐3,3‐dimethyl‐7‐aryl‐3H‐[1,2]oxathiolo[4,3‐b]pyridine‐6‐carbonitrile 1,1‐dioxides ( 3 ) in moderate chemical yields.  相似文献

Desymmetrization of the divinyl carbinol 1,4‐pentadien‐3‐ol was accomplished by the asymmetric 1,3‐dipolar cycloaddition of azomethine imines based on a magnesium‐mediated, multinucleating chiral reaction system utilizing diisopropyl (R,R)‐tartrate as the chiral auxiliary. The corresponding optically active trans‐pyrazolidines, each with three contiguous stereogenic centers, were obtained with excellent regio‐, diastereo‐, and enantioselectivity, with results as high as 99 % ee. This reaction was shown to be applicable to both aryl‐ and alkyl‐substituted azomethine imines. The use of a catalytic amount of diisopropyl (R,R)‐tartrate was also effective when accompanied by the addition of MgBr2.  相似文献

Three α‐phenylmalonamides have been prepared by the selective nucleophilic cleavage of 5,7‐dimethyl‐2‐phenyl‐1‐oxo‐1H‐pyrazolo[1,2‐a]pyrazol‐4‐ylium‐3‐olate in solventless microwave syntheses. The three weak nucleophiles employed were aniline, p‐chloroaniline and m‐toluidine. The α‐phenylmalonamides of these three aniline derivatives could not be prepared using the previously reported solvent syntheses via 3‐oxopyrazolo[1,2‐a]pyrazol‐8‐ylium‐1‐olates. All products were characterised using, infrared spectroscopy, 1H nmr and electrospray mass spectrometry. The single crystal X‐ray structures of the starting pyrazolo‐[1,2‐a]pyrazole and α‐phenylmalon‐m‐toluidide are also reported.  相似文献

In both 2,5‐dimethyl‐6,7‐dihydrobenzo[h]pyrazolo[1,5‐a]quinazoline, C16H15N3, (I), and 2‐tert‐butyl‐5‐methyl‐6,7‐dihydrobenzo[h]pyrazolo[1,5‐a]quinazoline, C19H21N3, (II), which crystallizes with Z′ = 2 in the space group P, the non‐aromatic carbocyclic rings adopt screw‐boat conformations. The molecules of (I) are linked into chains of rings by a combination of C—H...N and C—H...π(arene) hydrogen bonds, while in (II) there are no hydrogen bonds of any kind.  相似文献

The principal direction of conventional photolysis of the regioisomeric 2,2‐dimethyl‐5,5‐diphenyl‐ and 5,5‐dimethyl‐2,2‐diphenyl‐substituted 4‐diazodihydrofuran‐3(2H)‐ones 1a and 1b , respectively, is the Wolff rearrangement, while other photochemical processes, which are giving rise to the formation of C? H‐insertion, 1,2‐alkyl‐ or ‐aryl‐shifts, as well as H‐atom‐abstraction products occur to a much lower degree (Schemes 2 and 3). The ratio of similar reaction products from both regioisomers 1a and 1b is essentially independent of their structure, and a substantial effect of the relative position of the Ph and diazo group to each other on the yield of C? H‐insertion products does not occur. Based on stereochemical considerations, the Wolff rearrangement of diazodihydrofuran‐3(2H)‐ones apparently proceeds in a concerted manner, whereas the appearance in the reaction mixture of 1,2‐shift and H‐atom‐abstraction products points to the parallel generation during photolysis of singlet and triplet carbenes (Schemes 4 and 5).  相似文献

Flash vacuum pyrolysis (FVP) of 1,7‐bis‐(3‐aroylideneamino)‐4,6,10,12‐tetramethyl‐2,8‐dioxo‐1,7‐diazacyclododeca‐3,5,9,11‐tetraene‐3,9‐dicarbonitriles 11a‐c at 650°C and 0.02 Torr yielded 5,7‐dimethyl‐3‐(4‐methylbenzoyl)‐pyrazolo[1,5‐a]pyridine‐4‐carbonitrile 14 , 4,6‐dimethyl‐2‐oxo‐1,2‐dihydropyridine‐3‐carbonitrile 16 and 3‐aryl‐3‐oxo‐propionitriles 17a,b .
