Compression-induced apoptosis of nucleus pulposus (NP) cells plays a pivotal role in the pathogenesis of intervertebral disc degeneration (IVDD). Recent studies have shown that the dysregulation of mitochondrial fission and fusion is implicated in the pathogenesis of a variety of diseases. However, its role in and regulatory effects on compression-induced apoptosis of NP cells have not yet been fully elucidated. Heat shock protein 70 (HSP70) is a major cytoprotective heat shock protein, but its physiological role in IVDD, especially its effect on mitochondrial fission and fusion, is still unknown. Herein, we found that compression could induce mitochondrial fission, which ultimately trigger apoptosis of NP cells via the mitochondrial apoptotic pathway. In addition, we identified the cytoprotective effects of HSP70 on NP cells, and we found that promoting the expression of HSP70 could protect NP cells from abnormal mechanical loading in vitro and in vivo. Finally, we showed that HSP70 inhibited compression-induced mitochondrial fission by promoting SIRT3 expression, thereby attenuating mitochondrial dysfunction and the production of reactive oxygen species and ultimately inhibiting the mitochondrial apoptotic pathway in NP cells. In conclusion, our results demonstrated that HSP70 could attenuate compression-induced apoptosis of NP cells by suppressing mitochondrial fission via upregulating SIRT3 expression. Dipeptidyl peptidase-4 (DPP-4) inhibitors are used for the treatment of type 2 diabetes mellitus (DM). Recent studies have shown that beyond their effect in lowing glucose, DPP-4 inhibitors mitigate DM-related microvascular complications, such as diabetic retinopathy. However, the mechanism by which pathological retinal neovascularization, a major clinical manifestation of diabetic retinopathy, is inhibited is unclear. This study sought to examine the effects of evogliptin, a potent DPP-4 inhibitor, on pathological retinal neovascularization in mice and elucidate the mechanism by which evogliptin inhibits angiogenesis mediated by vascular endothelial growth factor (VEGF), a key factor in the vascular pathogenesis of proliferative diabetic retinopathy (PDR). In a murine model of PDR, an intravitreal injection of evogliptin significantly suppressed aberrant retinal neovascularization. In human endothelial cells, evogliptin reduced VEGF-induced angiogenesis. Western blot analysis showed that evogliptin inhibited the phosphorylation of signaling molecules associated with VEGF-induced cell adhesion and migration. Moreover, evogliptin substantially inhibited the VEGF-induced activation of adenosine 5′-diphosphate ribosylation factor 6 (Arf6), a small guanosine 5′-triphosphatase (GTPase) that regulates VEGF receptor 2 signal transduction. Direct activation of Arf6 using a chemical inhibitor of Arf-directed GTPase-activating protein completely abrogated the inhibitory effect of evogliptin on VEGF-induced activation of the angiogenic signaling pathway, which suggests that evogliptin suppresses VEGF-induced angiogenesis by blocking Arf6 activation. Our results provide insights into the molecular mechanism of the direct inhibitory effect of the DPP-4 inhibitor evogliptin on pathological retinal neovascularization. A high cholesterol/cholate diet induced hypercholesterolemia and fatty liver in both spontaneously hypertensive rats (SHR) and normotensive control rats (WKY). However, in contrast to previous concepts, the levels of cholesterol ester, triacylglycerol and phosphatidylcholine in plasma as well as triacylglycerol in liver were higher in WKY than in SHR fed a normal diet. The high cholesterol/cholate diet elevated the levels of plasma cholesterol, plasma cholesterol ester and hepatic triacylglycerol, and the extent of elevation was significantly higher in WKY than in SHR. Increases both in monoene/saturated ratios, an indication of elevated delta 9-desaturase activity, and in linoleate/arachidonate ratios, a possible indication of impaired desaturation-elongation activity, were observed in hepatic and plasma lipids of both strains fed the high cholesterol/cholate diet. The increases in monoene/saturated ratios were similar in both strains, but the increases in the linoleate/arachidonate ratios were higher for the plasma cholesterol esters of WKY than of SHR. The n-6/n-3 ratios of plasma and hepatic lipids were higher in WKY than in SHR throughout the experiments. These diet-induced changes observed in hepatic and plasma lipids were not reflected in the aortic lipids. Thus, hypertension per se does not promote the development of hyperlipemia and fatty liver induced by a high cholesterol/cholate diet. Our results also suggest that the metabolism of polyenoic fatty acids is different between SHR and WKY.  相似文献

alpha-Linolenic acid (18:3n-3) is known to autoxidize several fold faster than linoleic acid (18:2n-6). Feeding a high alpha-linolenate or a high linoleate diet to rats resulted in significant changes in the n-3/n-6 ratios of 20 and 22 carbon highly unsaturated fatty acids in erythrocytes. However, the rates of hemolysis observed in N2- or O2-atmosphere were similar between the two dietary groups. No significant amounts of conjugated dienes were detected and no measurable changes in the fatty acid compositions were observed during the incubations, indicating that the hemolysis occurred without involving significant lipid peroxidation. When stimulated with a free radical initiator, [2,2'-azobis-(2-amidinopropane)dihydrochloride] (AAPH), hemolysis occurred more rapidly, conjugated dienes formed and unsaturated/saturated ratios of phospholipid fatty acids decreased. However, no statistically significant difference was observed in these parameters of the two dietary groups. These results indicate that hemolysis occurs without involving lipid peroxidation but is accelerated by free radicals through lipid peroxidation, and that the difference in autoxidizabilities of alpha-linolenate and linoleate is not reflected in the rates of hemolysis and autoxidation in rat erythrocytes.  相似文献

Ischemia–reperfusion (I/R) injury accelerates the cardiomyocytes (CMs) death by oxidative stress, and thereby deteriorates cardiac function. There has been a paradigm shift in the therapeutic perspective more towards the prevention or amelioration of damage caused by reperfusion. Cardiac microvascular endothelial cells (CMECs) are more vulnerable to reperfusion injury and play the crucial roles more than CMs in the pathological process of early I/R injury. In this study, we investigate that CU06-1004, as a vascular leakage blocker, can improve cardiac function by inhibiting CMEC’s hyperpermeability and subsequently reducing the neutrophil’s plugging and infiltration in infarcted hearts. CU06-1004 was delivered intravenously 5 min before reperfusion and the rats were randomly divided into three groups: (1) vehicle, (2) low-CU06-1004 (1 mg/kg, twice at 24 h intervals), and (3) high-CU06-1004 (5 mg/kg, once before reperfusion). CU06-1004 treatment reduced necrotic size and cardiac edema by enhancing vascular integrity, as demonstrated by the presence of intact junction proteins on CMECs and surrounding pericytes in early I/R injury. It also decreased the expression of vascular cell adhesion molecule 1 (VCAM-1) on CMECs, resulting in reduced infiltration of neutrophils and macrophages. Echocardiography showed that the CU06-1004 treatment significantly improved cardiac function compared with the vehicle group. Interestingly, single high-dose treatment with CU06-1004 provided a greater functional improvement than repetitive low-dose treatment until 8 weeks post I/R. These findings demonstrate that CU06-1004 enhances vascular integrity and improves cardiac function by preventing lethal myocardial I/R injury. Congenital heart disease (CHD) is the most common type of birth defect, but its underlying molecular mechanisms remain unidentified. Previous studies determined that Homo sapiens LYR motif containing 1 (LYRM1) is a novel nucleoprotein expressed at the highest level in adipose tissue and in high levels in heart tissue. The LYRM1 gene may play an important role in the development of the human heart. This study was designed to identify the biological characteristics of the LYRM1 gene in heart development. On the basis of expression-specific differentiation markers identified with quantitative real-time RT-PCR and the morphology of LYRM1-overexpressing cells during differentiation, ectopic expression was not found to significantly affect differentiation of P19 cells into cardiomyocytes. MTT assays and cell cycle analysis showed that LYRM1 dramatically increases the proliferation of P19 cells. Furthermore, data from annexin V-FITC binding and caspase-3 activity revealed that LYRM1 can inhibit the apoptosis of P19 cells. Our data suggest that LYRM1 might have the potential to modulate cell growth, apoptosis, and heart development.  相似文献

This study presents a simple and rapid solvent exchange procedure using a hollow fiber. Antioxidants (Irganox 1010, Irganox 1076 and Irgafos 168) and solvents such as tetrahydrofuran (THF), carbon tetrachloride and toluene were selected as model compounds and sample solvents, respectively. After injection of the sample solution into the hollow fiber and solvent evaporation, the precipitated analytes in lumen and pores of the fiber were washed with methanol (the mobile phase for separation and determination by HPLC-diode array detection) and good chromatographic peaks were obtained. The effect of different parameters such as fiber length, volumes of sample and washing solvents were investigated and the optimum conditions were selected. The repeatability of the method was tested and it was found that the relative standard deviation (R.S.D.) was less than 10% for all analytes. Also enrichment factors of 3.03, 2.21 and 1.19 times were obtained for Irganox 1010, Irganox 1076 and Irgafos 168, respectively, when 200 μL sample and 50 μL methanol (washing solvent) were used.  相似文献

Vinyl pivalate (VPi) was polymerized in bulk by ultraviolet-ray initiation at low temperatures using 2,2′-azobis(2,4-dimethylvaleronitrile) (ADMVN) and 2,2'-azobis(isobutyronitrile) (AIBN) as photoinitiators. High molecular weight (HMW) poly(vinyl pivalate) (PVPi), having a number-average degree of polymerization (Pn) of 13,000–28,000, was obtained at conversions below 30% and converted by saponification to a syndiotacticity-rich HMW poly(vinyl alcohol) (PVA) microfibrillar fiber with Pn of 7300–18,300, syndiotactic diad (S-diad) and triad contents of ∼ 64% and ∼ 39%, respectively, and crystal melting temperature (Tm) of ∼ 249°C. ADMVN gave higher Pn than AIBN. On the other hand, conversion was smaller with the former than with the latter, and it was found that the initiation rate of ADMVN was lower than that of AIBN. Pn of PVA was constant while Pn of the precursor PVPi increased with increasing conversion. The syndiotacticity, Tm and thermal stability of PVA obtained from PVPi were much superior to those of PVA derived from poly(vinyl acetate) prepared under the same polymerization conditions. Polymerization of VPi at lower temperatures gave PVA with higher syndiotacticity. © 1997 John Wiley & Sons, Inc.  相似文献

Two polyimides, PI(DAT-6FDA) and PI(DAPT-6FDA), from N-(2,4-diaminophenyl)-N,N-diphenylamine (DAT) or N-(4-(2′,4′-diaminophenoxy)phenyl-N,N-diphenylamine (DAPT) and 4,4′-(hexafluoroisopropylidene)diphthalic anhydride (6FDA) were prepared to clarify the structural effect on the resulting memory properties. The memory device based on PI(DAT-6FDA) showed an unstable volatile behavior, while the device based on PI(DAPT-6FDA) with a more bulky donor (D) unit exhibited a stable non-volatile FLASH type memory characteristic with a long retention time over 104 s. The theoretical simulation based on the density functional theory (DFT) suggested that the greater distinct charge separation between the ground and charge transfer (CT) states led to a highly stable memory behavior. Also, it was clarified that PI(DAPT-6FDA) had a highly twisted conformation compared to PI(DAT-6FDA) in the ground state, and a more twisted dihedral angle between the D and acceptor (A) units was induced in the CT state, which led to the non-volatile memory characteristic.  相似文献

In this work, aligned and molecularly oriented bone‐like PLLA semihollow fiber yarns were manufactured continuously from an optimized homogeneous polymer‐solvent‐nonsolvent system [PLLA, CH2Cl2, and dimethyl formamide (DMF)] by a single capillary electrospinning via self‐bundling technique. Here, it should be emphasized that the self‐bundling electrospinning technique, a very facile electrospinning technique with a grounded needle (which is to induce the self‐bundling of polymer nanofibers at the beginning of electrospinning process), is used for the alignment and molecular orientation of the polymer fiber, and the take‐up speed of the rotating drum for the electrospun fiber yarn collection is very low (0.5 m/s). PLLA can be dissolved in DMF and CH2Cl2 mixed solvent with different ratios. By varying the ratios of mixed solvent system, PLLA electrospun semihollow fiber with the porous inner structure and compact shell wall could be formed, the thickness of the shell and the size of inner pores could be adjusted. The results of polarized FTIR and wide angle X‐ray diffraction investigations verified that as‐prepared PLLA semihollow fiber yarns were well‐aligned and molecularly oriented. Both the formation mechanism of semihollow fibers with core‐shell structure and the orientation mechanism of polymer chains within the polymer fibers were all discussed. The as‐prepared self‐bundling electrospun PLLA fiber yarns possessed enhanced mechanical performance compared with the corresponding conventional electrospun PLLA fibrous nonwoven membranes. © 2010 Wiley Periodicals, Inc. Electricity production from brewery wastewater using dual-chamber microbial fuel cells (MFCs) with a tin-coated copper mesh in the anode was investigated by changing the hydraulic retention time (HRT). The MFCs were fed with wastewater samples from the inlet (inflow, MFC-1) and outlet (outflow, MFC-2) of an anaerobic digester of a brewery wastewater treatment plant. Both chemical oxygen demand removal and current density were improved by decreasing HRT. The best MFC performance was with an HRT of 0.5 d. The maximum power densities of 8.001 and 1.843 μW/cm2 were obtained from reactors MFC-1 and MFC-2, respectively. Microbial diversity at different condi-tions was studied using PCR-DGGE profiling of 16S rRNA fragments of the microorganisms from the biofilm on the anode electrode. The MFC reactor had mainlyGeobacter,Shewanella, andClostridium species, and some bacteria were easily washed out at lower HRTs. The fouling characteristics of the MFC Nafion membrane and the resulting degradation of MFC performance were examined. The ion exchange capacity, conductivity, and diffusivity of the membrane decreased significantly after foul-ing. The morphology of the Nafion membrane and MFC degradation were studied using scanning electron microscopy and attenuated total reflection-Fourier transform infrared spectroscopy.  相似文献

Preoxidation process is usually needed in the treatment of arsenic-containing drinking water because arsenite (i.e. As[III]) is less easily removed by adsorption. Nano-scale titanium dioxide is an efficient photocatalyst for arsenite oxidation but its application in water treatment is limited due to the difficulty of separation or packed-bed application of the tiny particles. This study synthesized a composite photocatalyst by loading titanium dioxide onto activated carbon fiber (TiO2/ACF). The effects of calcination temperature, photocatalyst dosage, pH, initial concentration of As(III) and common anions on the oxidation of As(III) were studied. Photocatalytic oxidation of As(III) took place in minutes and followed first-order kinetics. 0.80 mg L−1 of As(III) could be entirely oxidized to As(V) within 30 min in the presence of 3.0 g L−1 photocatalyst and under UV-light irradiation. The oxidation of As(III) occurred in a wide range of pH as examined from 2 to 10 with the oxidation efficiency increasing markedly with pH. The presence of phosphate and silicate significantly decreased As(III) oxidation at pH 7, while the effect of sulfate and chloride was small.  相似文献

A method is described for the identification of a mixture of carbendazim and linuron. It is based on adsorptive stripping voltammetry at a carbon fiber ultramicroelectrode. Conditions for the determination of carbendazim in a mixture were optimized and the method was applied to soil samples. Summary A general method for the analysis of lipid classes by liquid chromatography has been developed using a multivariate optimization strategy to target optimal system conditions. The method was validated using a ruggedness test in the form of a Plackett-Burman design thereby exploring the immediate region around the optimum to ensure stable analytical conditions. Detection and quantitation were optimized by a factorial design in the light scattering detector parameters thus ensuring maximum detector response. This method was found to be suitable for a broad range of lipid sources from vegetable and animal origin, examples of the separation achieved are given for oat kernel, soybean and bovine milk lipids.  相似文献

A low cost, high performance ultraviolet digester was developed and tested in our laboratory. Its main features include limited costs (around 600 € for all the parts) and high efficiency toward total organic carbon (TOC) abatement. As a result the TOC of typical natural waters is reduced to less than 10% of its initial value with half an hour treatment. Easiness of use, temperature control and flexibility in sample size (up to 25 mL) and number (up to twelve 12 mL test tube) are other important characteristics of the developed UV digester.This apparatus was tested as a pretreatment method for the determination of the total content of trace elements such as copper, uranium and platinum in natural waters by cathodic adsorptive stripping voltammetry. The voltammetric technique was chosen because it is strongly interfered by the presence of even low levels of natural organic complexing molecules. As a result, a 30 minute UV treatment yielded results non distinguishable from ICP-MS data.  相似文献

A screening method based on liquid chromatography/electrospray tandem mass spectrometry was developed in order to control the illegal use of corticosteroids as growth promoters in cattle. The objective was the detection of low residue levels of corticosteroids or metabolites in biological matrices. Relative to other studies published on this subject, the present work focused on enhancing specificity and sensitivity. Firstly, fragmentation of corticosteroids by collision-induced dissociation was studied. In positive mode, the losses of H(2)O for each hydroxyl group fixed on the molecule, as well as the loss of HF or HCl for halogenated compounds, were observed. For higher collision energy, fragmentations in the B, C and D rings were induced. The negative mode was found to be more specific, inducing a cleavage of the C(20)-C(21) bond with concomitant loss of formaldehyde (CH(2)O). Secondly, three acquisition methods in the negative mode were studied and evaluated, recorded signals being the parent ion [M + acetate](-) and the two daughter ions, [M - H](-) and [M - H - CH(2)O](-). For dexamethasone, MS/MS instrumental detection limits of fragment ion and neutral loss scans, and of multiple reaction monitoring (MRM), were 250, 20 and 5 pg injected, respectively. The MRM method was then evaluated with the objective of use for the detection of corticosteroid residues in biological samples (urine, hair, muscle) and for a metabolism study.  相似文献

A comprehensive, fully automated strategy is demonstrated for HPLC-UV chromatographic method development using ChromSword optimization software. The strategy involves: (1) the automated screening of various column and mobile phase combinations, (2) rational selection of the best starting conditions; and (3) subsequent automated method development to generate optimized separation methods. Pharmaceutical compounds were applied to solve problematic drug impurity separations. ChromSword software automates the screening, optimization, and documentation steps thus reducing the method development time. The strategy was compared to a manual method development approach showing the automated method strategy affords better selectivity in a shorter time.  相似文献

采用溶胶-凝胶方法研制的聚甲基苯基乙烯基硅氧烷/羟基硅油复合涂层的固相微萃取探头及顶空固相微萃取-气相色谱联用技术(HS-SPME-GC),测定了制漆厂排放的工业废水中的甲苯和二甲苯。研究了影响该方法分析灵敏度的各种条件因素:萃取时间和温度,解吸时间和温度及离子强度等。结果表明新探头性能优于商用聚二甲基硅氧烷探头。方法的检出限为0.01μg/L-0.1μg/L,相对标准偏差小于6%;除苯以外线性范围达3个数量级。  相似文献
