A new building block strategy for the synthesis of fluorine-containing 1,2,4-trisubstituted furan was reported. The 1,2,4-trisubstituted furan was constructed through [3+2] cycloaddition reaction of fluoroacetyl-containing diazocompound and aromatic alkyne, catalyzed by Rh2(OAc)4.  相似文献

The reaction of Baylis-Hillman acetates and ethyl mercaptoacetate in the presence of DBU in DMF produced 2,3,4-trisubstituted tetrahydrothiophenes at room temperature as a diastereomeric mixture via the sequential SN2′ and Michael addition. Aromatization of tetrahydrothiophenes by DDQ oxidation produced 2,3,4-trisubstituted thiophenes in good yields.  相似文献

Pd(II)-catalyzed intramolecular Wacker-type cyclizations of o-trisubstituted allylphenols were studied. The chelation-induced axially chiral catalytic system, Pd(CF3COO)2-8 (1:1 molar ratio), showed excellent catalytic activities and enantioselectivities in the Wacker-type cyclizations of o-trisubstituted allylphenols with up to 94% ee.  相似文献

Isothiocyanate 6, obtained from vinyliminophosphorane 4 with CS2, reacted with various aliphatic primary amines to afford directly the 1,4,5-trisubstituted imidazolidine-2-thiones 5. However, the reaction of isothiocyanate 6 with aliphatic secondary amines provided 2,4,5-trisubstituted thiazoles 10 in good yields.  相似文献

Abstract

3,4,5-Trisubstituted piperidine-containing compounds displayed a range of biological activities, are potential drug candidates to treatment of Alzheimer’s disease, sexual dysfunctions or obesity, heart and kidney insufficiency. However, the inherent difficulty in obtaining a variety of 3,4,5-trisubstituted piperidines has greatly hindered their systematic medicinal chemistry studies for them. In this paper, we disclose an efficient synthesis of racemic and chiral 3,4,5-trisubstituted piperidines via SN2’ reaction.  相似文献

Palladium(0)/copper iodide catalyzed Sonogashira cross-coupling of 2-aryl-3-iodo-4-(phenylamino)quinolines with terminal alkynes afforded series of 1,2,4-trisubstituted 1H-pyrrolo[3,2-c]quinolines in a single-step operation. Conversely, the 4-(N,N-allylphenylamino)-2-aryl-3-iodoquinoline derivatives were found to undergo PdCl2(PPh3)2/CuI catalyzed intramolecular Heck reaction to yield the corresponding 1,3,4-trisubstituted 1H-pyrrolo[3,2-c]quinolines.  相似文献

Regioselectivity of the addition of the highly functionalized zinc-copper reagents to (η3-allyl)Fe(CO)4 cationic salts was studied. For 1,1-disubstituted allyl cation 1, the zinc-copper reagents added predominantly at the unsubstituted terminus. For 1,1,2-trisubstituted allyl cation 2, reactive zinc-copper reagents attacked mainly at the unsubstituted terminus while less reactive zinc-copper reagents added to a coordinated CO ligand. For 1,1,3-trisubstituted allyl cation 3, the addition occurred at both the less substituted allyl terminus and a coordinated CO ligand.  相似文献

A method for the synthesis of tri-substituted furans has been developed based on Cu(I)-catalyzed reaction of terminal alkynes with β-keto α-diazoesters. This method for the synthesis of 2,3,5-trisubstituted furans is operationally simple and applicable to wide substrate scope. Moreover, this synthesis employs cheap Cu(MeCN)4PF6 as the catalyst and no additional ligand is needed. Similar reaction has also been applied to ethyl (E)-2-diazo-3-(methoxyimino)butanoate for the synthesis of 2,3,5-trisubstituted N-methoxypyrroles with limited success.  相似文献

Carbon nucleophiles were used to open a 2,2,3-trisubstituted ethynylaziridine. A cyanide nucleophile opened the ring at the more substituted carbon, proceeding regioselectively with inversion of configuration. In an attempt to expand upon the scope of the reaction, Normant cuprates were reacted with a 2,2,3-trisubstituted ethynylaziridine. This reaction produced chiral allenes via an anti-SN2′ pathway. X-ray analysis of a derivative allowed the absolute stereochemistry of the anti-allenes to be assigned as P.  相似文献

The reaction of imino esters of indole-series acids with acid hydrazides gave N(1)-acylamidrazones, which, during heating, were converted to 3,5-disubstituted 1H-1,2,4-triazoles containing indole fragments. Compounds of this type were also synthesized by the reaction of indolyl-containing 2,5-disubstituted 1,3,4-oxadiazoles with formamide. Condensation of 2,5-disubstituted 1,3,4-oxadiazoles with aniline gave 3,4,5-trisubstituted 4H-1,2,4-triazoles containing indolyl radicals. Novel syntheses of 2,3,8-trisubstituted pyrido[2,3-d]pyridazines and 2,3,5-trisubstituted pyrazino[2,3-d]pyridazines are described. Two complementary routes to pyrido[2,3-d]pyridazines were developed, the first of which began by constructing the pyridine ring, and the second of which started by constructing the pyridazine ring. Pyrazino[2,3-d]pyridazines were prepared in a route employing an aza-Wadsworth-Emmons cyclization as the key step. The resulting compounds were found to be high affinity ligands for the GABAA receptor benzodiazepine binding site.  相似文献

Isomeric six- and five-membered (N2)-aromatics, 6-methylpyridazines and 5-vinylpyrazoles, which energetic topological aromaticity is comparable to that of benzene, are shown to be efficiently produced by sequential isomerization–elimination processes from the corresponding 6-methylidene-1,4,5,6-tetrahydropyridazines and 5-vinylpyrazolines, respectively. The latter precursors are available from the same N-sulfonyl,C-homoallyl-hydrazone substrates by a suitable choice of previously reported conditions for Pd-catalyzed CH-oxidative C,N-ring closing processes. The generality of these cyclization, isomerization, and aromatization reactions, for which detailed mechanisms are proposed, provides a systematic access to wide ranges of 3,4,6-trisubstituted 6-methyl-1,4-dihydropyridazines and 6-methylpyridazines, and their 3,4,5-trisubstituted 5-vinylpyrazole isomers.  相似文献

InCl3·3H2O was found to be a mild and effective catalyst for the efficient, one-pot, three component synthesis of 2,4,5-trisubstituted imidazoles at room temperature. Tetrahydropyran compounds can be directly synthesized from allylbromide and carbonyl compounds by means of one-pot Babier-Prins cyclization promoted by BPyX/SnX′2 or BBIMBr/SnBr2 complex (functionalized RTILs) under solvent-free conditions. 2,6-Homo-bissubstituted- and 2,6,6-trisubstituted, especially 6-(spirocycloalkyl)-, tetrahydropyran compounds can be prepared in good yields.  相似文献

An efficient methodology for the synthesis of 2,2,3-trisubstituted tetrahydroquinolines has been developed, which involves the triphenylphosphine-CCl4-mediated stereospecific rearrangement of α,α-disubstituted indoline-2-methanols 15 to 2,2,3-trisubstituted tetrahydroquinolines 26. The rearrangement precursors 15 are readily prepared by the diastereoselective Grignard addition to 2-acylindolines 13. The total syntheses of (+)-benzastatin E (1) and natural virantmycin (2a) were accomplished utilizing this methodology. This rearrangement reaction might afford some chemical precedent for the biogenetic pathway of the benzastatin family.  相似文献
