[reaction: see text] A new asymmetric halo aldol reaction has been developed by reacting cyclopropyl carbonyl derived enolates with aldehydes. The absolute structure was unambiguously confirmed by X-ray structural analysis. Eight examples were reported with good yields and up to complete control of diastereomeric excesses. These halo aldol products have been readily cyclized in the presence of weak bases to produce chiral 2,3-disubstituted tetrahydrofuran derivatives in good yield without any observed epimerization.  相似文献

[reaction: see text] NaIO(4) oxidizes alkali metal halides efficiently in aqueous medium to halogenate alkenes and aromatics and produce the corresponding halo derivatives in excellent regio and stereoselectivity. The system also demonstrates the asymmetric version of bromo hydroxylation using beta-cyclodextrin complexes, resulting in moderate ee.  相似文献

A novel generation of 1-iodo-3-siloxy-1,3-butadienes has been developed by reacting trimethylsilyl iodide (TMS-I) with alpha, beta-unsaturated ketones in dichloromethane at 0 degrees C without the use of any catalyst. The halo aldol reaction of these butadiene intermediates with aldehydes was efficiently carried out by using magnesium iodide as the catalyst. Twelve beta-iodo-alpha,beta-unsaturated-beta'-hydroxyketones (halo aldols) have been synthesized under the new condition with excellent geometric selectivity and good chemical yields (>80% chemical yields for 11 examples).  相似文献

[reaction: see text] A new three-component halo aldol reaction has been discovered for the tandem formations of I-C/C-C bonds by activating the alpha',beta-positions of alpha,beta-acetylenic ketones. The key intermediates, 1-iodo-3-siloxy-1,3-butadienes, were generated from allenolates and directly monitored by (1)H NMR spectroscopic analysis. Excellent geometric selectivity (>95%) and good yields (65-82%) have been achieved for 10 examples.  相似文献

The development of a general catalytic asymmetric aldehyde alpha-alkylation reaction constitutes a major challenge in organic synthesis. Here, we report the first and successful approach toward its solution: (S)-alpha-methyl proline catalyzes the intramolecular alkylation of various halo aldehydes to the corresponding formyl cyclopentanes, -cyclopropanes, or -pyrrolidines in excellent yields and enantioselectivities. Most remarkably, racemization, aldolization, or catalyst alkylation do not occur to any significant extend, further illustrating the power, mildness, and profound selectivity of enamine catalysis.  相似文献

Protonated pyrrolidine based small organic molecules have been designed and evaluated for the asymmetric direct aldol reaction in water. The designed organocatalysts are multifunctional in nature and exploit the combined effect of hydrogen bonding and hydrophobic interactions for enantioselective catalysis in water. As a result a unique direct asymmetric aldol reaction in water catalyzed by a small organic molecule having an amide linkage has been developed. The developed catalyst affords chiral β-hydroxyketones in good yields (93%) and enantioselectivities (upto 62%) in water.  相似文献

The organopalladium complex containing ortho-metalated (S)-[1-(dimethylamino)ethyl]phenylene as the chiral auxiliary has been used as the chiral template to promote the asymmetric cycloaddition reaction between diphenylvinylphosphine and 3,4-dimethyl-1-phenylarsole. A diphenylphosphino-substituted asymmetrical heterobidentate arsanorbornene (As-P) ligand was obtained stereoselectively on the chiral palladium template in moderate yield. The chiral benzylamine auxiliary could be removed chemoselectively from the template by treatment with HCl to produce the neutral complex [(As-P)PdCl2]. In contrast to their reported P-P analogue, the arsenic donor in the dichloro complex could be eliminated stereospecifically under mild reaction conditions to generate the corresponding 1-(diphenylphosphino)-3,4-dimethyl-2,4-cyclohexadiene, which remained as a bidentate ligand at the PdCl2 unit via phosphorus and the eta2-C4-C5 double bond. The arsenic-elimination process was found to be influenced by the halo ligand in [(As-P)PdX2]. A similar process was observed with the analogous dibromo complex, but the corresponding diiodo species did not show similar reactivity. All of the novel As-Pd complexes have been characterized by X-ray crystallography.  相似文献

A reaction sequence involving halogenation and replacement of the halo substituent by a nitro group using dinitrogen tetraoxide has led to a general, convenient route to 5-nitrooxazoles. Reaction schemes employing both bromine and iodine as the halo substituent have been investigated; however, the method using iodine preceded by a mercuration step affords a better overall yield in the range of 20–50%. Both 2- and 4-nitro-oxazoles can be prepared by this latter sequence, though in lower overall yields (4–12%).  相似文献

The synthesis has been achieved of a series of 9-substituted derivatives of 6-(4-nitroimidazol-5-yl)- and 6-(5-nitroimidazol-4-yl)thiopurines by the reaction of 6-(nitroimidazolyl)thiopurines with alkyl and aralkyl halides, haloalcohols, haloacids and their esters, halo ketones, halo aldehyde acetals, and arylsulfonate esters using potassium carbonate in DMF.  相似文献

A Novel Hunsdiecker‐Borodin reaction (HBR) has been carried out efficiently with α,β‐unsaturated aliphatic and aromatic carboxylic acids by using N‐halo succinimides such as N‐chloro succinimide (NCS), N‐bromo succinimide (NBS), and N‐iodo succinimide (NIS) under micellar media. The reaction with α,β‐unsaturated aromatic carboxylic acids afforded β‐halo styrenes in excellent yield while α,β‐unsaturated aliphatic carboxylic acids underwent decarboxylation and to give corresponding halo derivatives. The reactions are dramatically accelerated in micellar media. This procedure works efficiently in CTAB (cetyl trimethyl ammonium bromide), SDS (sodium dodecyl sulfate), and TX (Triton‐X‐100) media under stirred conditions at room temperature. At reflux temperatures the yield of reaction products were further enhanced from good to excellent.  相似文献

The reaction of a racemic reagent on a mixture of enantiomers with small ee (ee=enantiomeric excess) has been studied for amine acylation. A substantial asymmetric amplification could be realized, for example, from 67 to >95.5 ee. The combination of asymmetric amplifications is subsequently discussed. Two sequential asymmetric amplifications, one using a racemic reagent and another using a positive nonlinear effect allowed us to start from 1.5 % ee and end with a large amount of a product of 97 % ee.  相似文献

The ionic Diels-Alder reaction, whereby an alpha,beta-unsaturated acetal in combination with a Lewis or Bronsted acid forms an equilibrium concentration of an activated dienophile, has been developed to provide an enantioselective synthesis of cis-decalins. Cyclohex-2-enone type chiral acetals of (2R,3R)-butane-2,3-diol have been screened against Lewis and Br?nsted acids with a variety of dienes and are efficient for the synthesis of a limited subset of cis-decalin structures. Diastereoselectivities of 73% and 82% have been found for the asymmetric ionic Diels-Alder reaction between the chiral acetal derivatives of cyclohex-2-enone (6) and 2-methylcyclohex-2-enone (18) with 2,3-dimethyl-1,3-butadiene (7). Terminal substituents on the diene partner in general render the system unreactive. However a synthetically useful cis-decalin 31, derived from the reaction of 2-methylcyclohex-2-enone and Z-3-t-butyldimethyl-silyloxypenta-1,3-diene has been prepared in enantiomerically pure form in 74% isolated yield using this asymmetric ionic Diels-Alder protocol.  相似文献

In the presence of a catalytic amount of chiral binaphthylthiophosphoramide L2 (6 mol %) and Cu(I) (3 mol %), the asymmetric addition of diethylzinc to N-sulfonylimines could be achieved in good yields with moderate to high ee (63-93% ee) at 0 degrees C in toluene. A novel chiral binaphthylthiophosphoramide ligand system for this asymmetric addition reaction has been explored.  相似文献

The asymmetric catalytic aza-Morita-Baylis-Hillman reaction of N-sulfonated imines with α,β-unsaturated ketones has been successfully conducted by using chiral bifunctional phosphine amides as catalysts. A series of new chiral bifunctional phosphine amides were designed, synthesized, and systematically studied for this asymmetric reaction. The corresponding aza-MBH adducts were obtained in good yields (75-99%) and up to very good enantiomeric excesses (51-95% ee) under mild conditions.  相似文献

A biphasic catalytic system with water-soluble rhodium complexes of sulfonated (R)-2,2′-bis(diphenylphosphino)-1,1′-binaphthyl (labeled as (R)-BINAPS) in ionic liquid BMI·BF4 has been developed for the asymmetric hydroformylation of vinyl acetate under mild conditions. The corresponding ruthenium complexes have been investigated for the biphasic asymmetric hydrogenation of dimethyl itaconate. The biphasic asymmetric hydroformylation of vinyl acetate provided 28.2% conversion and 55.2% enantiomeric excess when BMI·BF4–toluene was used as the reaction medium at 333 K and 1.0 MPa for 24 h. The biphasic asymmetric hydrogenation of dimethyl itaconate in BMI·BF4iPrOH at 333 K and 2.0 MPa afforded 65% enantiomeric excess with an activity similar to the homogenous analogs. Both biphasic catalytic systems with (R)-BINAPS ligand could be reused several times without significantly decrease in the activity, enantio- and regio-selectivities. The effects of properties of ionic liquid, molar ratio of ligand to rhodium, temperature, pressure and reaction time have been discussed.  相似文献

A new set of phosphorus-containing chiral ligands has been synthesized and used with silver(I) for the catalytic asymmetric 1,3-dipolar cycloaddition reaction of azomethine ylides. One of these ligands (POFAM6) was found to produce an effective catalyst for the asymmetric 1,3-dipolar cycloaddition reaction of azomethine ylides with electron deficient dipolarophiles to form pyrrolidines in up to 97% yield and 89% ee.  相似文献

Chiral BINOL-derived Br?nsted acids catalyze the enantioselective asymmetric Morita-Baylis-Hillman (MBH) reaction of cyclohexenone with aldehydes. The asymmetric MBH reaction requires 2-20 mol % of the chiral Br?nsted acid 2e or 2f and triethylphosphine as the nucleophilic promoter. The reaction products are obtained in good yields (39-88%) and high enantioselectivities (67-96% ee). The Br?nsted-acid-catalyzed reaction is the first example of a highly enantioselective asymmetric MBH reaction of cyclohexenone with aldehydes.  相似文献

New bifunctional primary amine-squaramides catalyzed asymmetric Michael addition reaction of α,α-disubstituted aldehydes to maleimides has been developed. This organocatalytic asymmetric reaction provides easy access to functionalized succinimides with a broad substrate scope. 本文报道用环加成-芳构化串连反应制备多取代芳香族化合物的方法。将1-氯-2-卤-3-苯硒基-1,3-丁二烯(5)与丁炔二酸二甲酯进行Diels-Alder反应时,能直接生成多取代的芳香族化合物4-苯硒基-5-卤代邻苯二甲酸二甲酯(4)。若用2-卤素-3-苯硒基-1,3-丁二烯(1)与丁炔二酸二甲酯进行Diels-Alder反应,只得到正常的加成产物4-苯硒基-5-卤素-1,4-环己二烯-1,2-二甲酸二甲酯(3),需再用DDQ处理,才得到多取代芳香族化合物(4)。  相似文献
