The effect of cyclodextrins (CDs) on water solubility of iodine was investigated. Modified CDs greatly enhanced the solubility of iodine. On the contrary, enhancement by natural CDs was rather moderate whereby the solubility was only doubled at the highest β-CD concentration examined. Desorption experiment of iodine from solution was carried out with addition of various CDs to study the effect of CDs on iodine retention. α-CD was the most efficient in retarding iodine desorption. Later, various concentrations of α-CD were used in the desorption experiment to observe its volatile suppression effect and determine the stability constant of iodine/α-CD complexation. At α-CD concentration of 10.3 mM, no lost of iodine from the solution was detected. A model was developed for desorption of iodine from the solution based on mass transfer theory. The stability constant K given by this model was 3.28×104 M−1 which was in the same order as the value estimated in this study by solubility method and as well those reported by other authors. In release experiments of solid state inclusion complexes, stability of inclusion complex powders decreased in the order of α-CD>β-CD>randomly methylated β-CD (RM-β-CD).  相似文献

The objectives of this research were to prepare and characterize inclusion complex of Ezetimibe (EZE) with cyclodextrins (β-cyclodextrin (β-CD) and hydroxypropyl-β-cyclodextrin (HPβ-CD)) and to study the effect of complexation on the dissolution rate of EZE, a water insoluble drug. Phase solubility curve was classified as A P -type for both cyclodextrins, indicating the 2:1 stoichiometric ratio for β-CD–EZE and HPβ-CD – EZE inclusion complexes. The inclusion complexes in the molar ratio of 2:1 (β-CD–EZE and HPβ-CD–EZE) were prepared by various methods such as kneading, coevaporation and physical mixing. The molecular behaviors of drug in all samples were characterized by fourier-transform infrared (FTIR) spectroscopy, differential scanning calorimetry (DSC) and powder X-ray diffraction (PXRD) studies. The results of these studies indicated that complex prepared by kneading and coevaporation methods showed inclusion of the EZE molecule into the cyclodextrins cavities. The highest improvement in in-vitro dissolution profiles was observed in complex prepared with hydroxypropyl-β-cyclodextrin using co-evaporation method. Mean dissolution time and similarity factor indicated significant difference between the release profiles of EZE from complexes and physical mixtures and from pure EZE.  相似文献

Thermodynamic parameters for formation of the inclusion complexes of α-, β- and γ-cyclodextrin (α-, β- and γ-CD) with ibuprofen (BF) in Tris-HCl buffer solutions (pH=7.0) have been determined by isothermal titration calorimetry (ITC) with nanowatt sensitivity, and the inclusion structures have been investigated by using 1H-NMR spectra at 298.15 K. A theoretical study on the inclusion processes between BF and CDs has been performed with the B3LYP/6-31G*//PM3 method in order to investigate the formation mechanism of the inclusion complexes. An analysis of the thermodynamic data indicates that the stoichiometries of α-, β- and γ-CD with BF are all 1:1 and formation of the inclusion complexes α-CD⋅BF and β-CD⋅BF are driven by enthalpy and entropy, whereas formation of γ-CD⋅BF is an entropy driven process. The 1H-NMR spectra provide clear evidence for the inclusion phenomenon, and show that the isobutyl group and aromatic ring of the guest molecule are trapped inside the cavity of the CDs. Theoretical calculations suggest that the complex formed by the BF molecule entering into the cavity of the CD molecule from the wide side is more stable than that from the narrow side.  相似文献

The extent and mode of solubility enhancement exerted by the cyclodextrins (α-, β-, γ-, and HP-β-CDs) on loratadine (Lort) have been experimentally measured under controlled conditions in buffered aqueous solutions. Rigorous nonlinear regression analysis of the phase solubility diagrams obtained in 0.1 mol⋅L−1 phosphate buffer at pH=7.0 and 25 °C revealed the following: neutral Lort (pK a =4.6) tends to form soluble 1:1 and 1:2 Lort/CD complexes with all four of the examined CDs, where complex stability follows the decreasing order β-CD>HP-β-CD>γ-CD>α-CD. The hydrophobic character of Lort constitutes about 66% of the driving force for complex formation whereas specific interactions contribute 11.2 kJ⋅mol−1 towards the stability of the complexes. Thermodynamic studies showed that Lort/CD complex formation was favored by large enthalpic contributions but was impeded by negative entropic changes. Dissolution studies indicate that the dissolution rate of Lort from the freeze-dried Lort/β-CD complex is significantly higher than that of the corresponding physical mixture. Both DSC studies and molecular mechanical modeling of Lort/β-CD interactions were carried out to explore the possible formation of inclusion complexes.  相似文献

Guest–host interaction of prednisone (PN) with cyclodextrins (CDs) have been investigated using phase solubility diagrams (PSD), differential scanning calorimetry (DSC), X-ray powder diffractometry (XRPD), scanning electron microscopy (SEM) and molecular mechanical modeling (MM). Estimates of the complex formation constant (K 11) show that the tendency of PN to complex with CDs follows the order: β-CD>γ-CD>HP-β-CD>α-CD. At the same pH of 7.0, β-CD forms soluble 1:1 and insoluble 1:2 PN/CD complexes (BS-type PSDs). The thermodynamic functions for 1:1 PN/β-CD estimated at pH = 7.0 (ΔG 11o=−20.8 kJ⋅mol−1) show that complexation is driven by enthalpy (−30.7 kJ⋅mol−1) but retarded by entropy (ΔS 11o=−33.1 J⋅mol−1⋅K−1) changes. The MM modeling study indicates the formation of different isomeric 1:1 complexes with CDs. PSD, DSC, XRPD, SEM and MM studies established the formation of inclusion complexes in solution and the solid state.  相似文献

Several host–guest inclusion compounds of eugenol as a guest molecule and cyclodextrins (α-,β-,γ-CD) and heptakis (2,6-di-O-methyl)-β-cyclodextrin (DMβ-CD) as hosts were investigated in the solid state and in aqueous solution. The one-to-one solid inclusion compounds of eugenol and β-CD or γ-CD were prepared, but those of eugenol with α- or DMβ-CD were not obtained under the same condition. However, the UV-visible absorption spectroscopy data indicated that the liquid guest could form a 1:1 inclusion compound with all four hosts respectively in aqueous solution. The two solid inclusion compounds were characterized by powder X-ray diffraction (XRD), infrared spectroscopy (IR), thermogravimetric analysis (TG), differential scanning calorimetry (DSC) and nuclear magnetic resonance (NMR). The association constants (K), calculated from the modified Benesi–Hidebrand equation, of eugenol with α-, β-, γ- and DMβ-CD is 4.95 × 104, 3.96 × 105, 1.47 × 105 and 9.33 × 104 mol−1 dm3, respectively.  相似文献

As a continuation of our previous investigation, interactions between cyclodextrin (β-CD), γ-cyclodextrin (γ-CD) and alkyl trimethylammonium bromides in aqueous solutions have been studied with titration calorimetry and 1H NMR at 298.15 K. The results are discussed in terms of the amphiphilic interaction of CD with surfactants and the iceberg structure formed by water molecules existing around the hydrophobic tail of surfactant molecules. The stoichiometry of the β-CD–surfactant system is 1:1 whereas that of the γ-CD–surfactant system is 1:2. The corresponding formation enthalpy (negative) of the complexes of the two systems decreases with an increase in the number of carbon atoms (n) in hydrophobic chain of surfactant molecule, C n H2n+1, whereas the entropy increases with the enlargement of n.  相似文献

Guest–host interaction of astemizole (Astm) with cyclodextrins (CDs) has been investigated using phase solubility diagrams (PSD), differential scanning calorimetry (DSC), X-ray powder diffractometry (XRPD), proton nuclear magnetic resonance (1H-NMR) and molecular mechanical modeling (MM+). Estimates of the complex formation constant, K 11, show that the tendency of Astm to complex with CDs follows the order: β-CD>HP-β-CD>γ-CD, α-CD. 1:1 Astm/β-CD complex formation at pH=5.0 was largely driven by the hydrophobic effect (desolvation), which was quantitatively estimated at −16.5 kJ⋅mol−1, whereas specific interactions contribute only −5.3 kJ⋅mol−1 to 1:1 complex stability (ΔG 11o=−22.7 kJ⋅mol−1). The percentage contributions of the hydrophobic effect and specific interactions were therefore 73 and 27%, respectively. Both enthalpic and entropic factors contribute equally well (−11 kJ⋅mol−1 each) to 1:1 Astm/β-CD complex stability. 1H-NMR and MM+ molecular modeling studies indicate the formation of different isomeric 1:1 and 1:2 complexes. The dominant driving force for complexation is evidently van der Waals with very little electrostatic contribution. PSD, 1H-NMR, DSC, XRPD and MM+ studies proved the formation of inclusion complexes in solution and the solid state.  相似文献

The purpose of present investigation was to understand the interactions involved in complexation of Nelfinavir Mesylate (NM)—a protease inhibitor, used in the treatment of HIV/AIDS with Beta-cyclodextrin (β-CD) and its subsequent effect on its absorption properties and bioavailability. Milling method was used for complexation. The inclusion complexes were characterized by 2D NOESY NMR and ITC studies. The feasibility of findings was further confirmed by using Cerius2 software of Tripos Inc. using Silicon Graphics O2. Pharmacokinetic studies were carried out in rabbits and data was treated by Student’s t Test. 2D NOESY NMR studies showed very intricate behavior showing interactions amongst drug and β-CD molecule as well as amongst β-CD–β-CD molecules. This fact of formation of molecular aggregates was further confirmed by ITC studies. Computer simulation studies further supported the finding of forming shallow complex. The percent relative bioavailability of complex at the dose of 400 mg/kg in rabbits was 185.37 as compared to the plain NM at 400 mg/kg dose. The studies were conducted at low dose of 200 mg/kg of drug in the form of complex in rabbit does not show statistically significant difference in AUC, T 1/2 and Kel. as compared to plain drug at 400 mg/kg of rabbit.  相似文献

In pH 7.3 buffers, the interactions of a cationic porphyrin, tetrakis(4-N-methylpyridyl)porphyrin (TMPyP), with cyclodextrins (CDs) and disodium phthalate (DSP) have been examined by means of absorption, fluorescence, and induced circular dichroism spectroscopy. α-CD, β-CD, and γ-CD form a 1:1 inclusion complex with a TMPyP monomer, which dimerizes in solution without CD. TMPyP also forms a 1:1 organic cation–organic anion complex with DSP. The 1:1 TMPyP–DSP complex forms a ternary CD–TMPyP–DSP inclusion complex with α-, β-, and γ-CD, in which a DSP molecule is not incorporated into the CD cavity. From the fluorescence intensity change, the␣equilibrium constants have been evaluated for the formation of the inclusion complexes and the organic cation–anion complexes.  相似文献

The electronic absorption spectra and fluorescence spectra of 4-(2-naphthyl)pyridine (1), 2-(4-methyl-2-pyridyl)-4-(2-naphthyl)pyridine (2), and 4-(2-naphthyl)-2-phenylpyridine (3) in solutions and in complexes with β-cyclodextrin (β-CD) and well water-soluble hydroxy-propyl-β-cyclodextrin (HP-β-CD) were studied. Fluorescence near 475 nm observed in aqueous solutions of compounds 1–3 arises from protonated forms of these compounds produced in the excited state. Results of DFT quantum chemical calculations show an increase in proton affinity energies of excited-state naphthylpyridines 2 and 3. The formation of inclusion complexes with cyclodextrins makes protonation of compounds 2 and 3 more difficult, which manifests in large hypsochromic shifts of fluorescence band maxima. The stability constants of the complexes 1·HP-β-CD and 2·HP-β-CD determined from their fluorescence spectra are 3425 and 3760 L mol−1, respectively. The stability constant of the complex 3·HP-β-CD (5500±600 L mol−1) was found from the changes in the solubility of naphthylpyridine 3 in water upon complexation. Semiempirical quantum chemical calculations of the molecular structures and thermodynamic characteristics of pseudorotaxane inclusion complexes of trans-2, cis-2, and trans-2·H2O with HP-β-CD were carried out. The inclusion of vanadocene dichloride (VDC) and 1,1′-dimethyl vanadocene dichloride (MeVDC) into cyclodextrines (α-CD, β-CD and γ-CD) was studied by EPR spectroscopy. It was found that VDC and MeVDC with β-CD and γ-CD form true inclusion compounds, but with α-CD, VDC and MeVDC gave only fine dispersion mixtures. The inclusion was validated by anisotropic EPR spectra of solid samples. In addition, the antimicrobial was validated by anisotropic EPR spectra of solid samples. In addition, the antimicrobial behavior (against E. coli) of each of the complexes was determined. It was established that not only did VDC and MeVDC cause elongation of E. coli, but also the new vanadocene inclusion complexes were effective in this regard.  相似文献

In phosphate buffer solution of pH5.4, the interaction of meso-tetrakis(2-thienyl)porphyrin(H2TTP) and Cu-meso-tetrakis(2-thienyl)porphyrin(Cu-TTP) with α-cyclodextrin(α-CD), β-CD, γ-CD, heptakis(2,3,6-tri-O-methyl)-β-CD(TM-β-CD) has been studied by means of UV-vis, fluorescence and 1HNMR spectroscopy, respectively. The H2TTP and Cu-TTP can form 1:2 inclusion complexes with TM-β-CD and 1:1 inclusion complexes with the other three cyclodextrins. In this paper, the inclusion constants (K) of H2TTP and Cu-TTP for the formation of the inclusion complexes have been estimated from the changes of absorbance and fluorescence intensity in phosphate buffer solution. The inclusive capabilities of different kinds of cyclodextrins are compared. The result shows that the inclusion ability of α-CD with H2TTP and Cu-TTP is the strongest among the three native CDs. The inclusion ability of modified β-CD with H2TTP and Cu-TTP is stronger, compared to the native β-CD, which indicates that the capacity matching plays a crucial role in the inclusion procedure except for the hydrophobic effect. In addition 1HNMR spectra supports the inclusion conformation of the TM-β-CD-Cu-TTP inclusion complex, indicating the interaction mechanism of inclusion processes.  相似文献

Guest–host interactions were examined for neutral diclofenac (Diclo) and Diclofenac sodium (Diclo sodium) with each of the cyclodextrin (CD) derivatives: α-CD, β-CD, γ-CD and 2-hydroxypropyl-β-cyclodextrin (HP-β-CD), all in 0.05 M aqueous phosphate buffer solution adjusted to 0.2 M ionic strength with NaCl at 20 °C, and with β-CD at different pHs and temperatures. The pH solubility profiles were measured to obtain the acid–base ionization constants (pK as) for Diclo in the presence and absence of β-CD. Phase solubility diagrams (PSDs) were also measured and analyzed through rigorous procedures to obtain estimates of the complex formation constants for Diclo/CD and Diclo sodium/CD complexation in aqueous solutions. The results indicate that both Diclo and Diclo sodium form soluble 1:1 complexes with α-, β-, and HP-β-CD. In contrast, Diclo forms soluble 1:1 Diclo/γ-CD complexes, while Diclo sodium forms 1:1 and 2:1 Diclo/γ-CD, but the 1:1 complex saturates at 5.8 mM γ-CD with a solubility product constant (pK sp = 5.5). Therefore, though overall complex stabilities were found to follow the decreasing order: γ-CD > HP-β-CD > β-CD > α-CD, some complex precipitation problems may be faced with aqueous formulations of Diclo sodium with γ-CD, where the overall concentration of the latter exceeds 5.8 mM γ-CD. Both 1H-NMR spectroscopic and molecular mechanical modeling (MM+) studies of Diclo/β-CD indicate the possible formation of soluble isomeric 1:1 complexes in water.  相似文献

This paper describes the formation of polymer inclusion complexes(polymer-CD-ICs) between β-cyclodextrin(β-CD) and aliphatic-aromatic poly(Schiff base)s. Fourier transform infrared(FTIR) spectroscopy, 1H nuclear magnetic resonance spectroscopy(1H-NMR), thermogravimetric analysis(TGA) and X-ray diffraction(XRD) have been used to observe the formation of polymer-CD-ICs. In FTIR spectra, the characteristic peaks of β-CD at 3391 cm−1 shifted to 3418 cm−1 and the intense peak at 1602 cm−1 due to the –C = N– stretching vibration diminished after formation of inclusion complexes. Compared the 1H-NMR of polymer-CD-ICs with β-CD, the chemical shift of the protons H-3, H-5 have shifted to higher field after the formation of inclusion complexes, which is perhaps due to the interaction of these protons with polymers. The TGA analysis revealed that the polymer-CD-ICs had better thermal stability than β-CD, suggesting that the polymer increased the stability of β-CD. The X-ray diffraction patterns displayed that the strong peak for both polymer-CD-ICs at approximately 20.0° (2θ) may confirm their IC formation.  相似文献

The interaction of two benzocycloheptanes namely, pizotifen (Pizo) and ketotifen (Keto), with cyclodextrins (CDs: α-, β-, γ-, and HP-β-CDs) has been investigated by several techniques including phase solubility, X-ray powder diffractometry, 1H-nuclear magnetic resonance and molecular mechanical modeling. The effects of CD type, pH, ionic strength and temperature on complex stability were also explored. The complex formation constant (K 11) values for the Pizo/CD system follows the decreasing order β-CD > γ-CD > HP-β-CD > α-CD. However, for the Keto/CD system it follows the decreasing order γ-CD > β-CD > HP-β-CD > α-CD. The tendency of Pizo and Keto to complex with β-CD is driven to the extent of 70% by the hydrophobic effect. Complex formation of Keto and Pizo was substantially driven by entropy (>100 J⋅mol−1⋅K−1) but slightly retarded by enthalpy (3–8 kJ⋅mol−1). 1H-NMR and MM+ studies indicate multimodal inclusion of the methylpiperadine, thiophene and phenyl moieties into the β-CD cavity.  相似文献

The purpose of present investigation was to investigate the effect of complexation of Nelfinavir Mesylate (NM) – an Anti-HIV drug with Beta-cyclodextrin (β-CD) on its dissolution characteristics and subsequent effect on its absorption properties and bioavailability. Phase solubility studies were conducted to find the interaction of NM with β-CD. Physical mixing and milling method were used for complexation. The inclusion complexes were characterized by X-ray diffractometry, FT-IR and NMR studies and further studied by in-vitro dissolution testing. The plain NM and complex was subjected to intestinal absorption studies by using Everted intestinal sac model. Data was treated statistically by Mann–Whitney U test. Pharmacokinetic studies were carried out in rabbits using cross over design and data was treated by Student’s t test. Phase solubility studies confirmed 1:1 complex formation of NM with β-CD with stability constant of 204.84 M−1. In-vitro dissolution studies of inclusion complexes of NM with β-CD prepared by milling method (T 90=60.89 min) showed better dissolution rate kinetics in distilled water in comparison with plain NM (T 90=374.31). The increased solubility with decreased crystallinity is attributed by inclusion of NM in the cavity of β-CD, which was further confirmed by instrumental studies. Intestinal absorption studies further supports these findings by showing 2.13 times enhancement in the absorption rate of complex as compared to plain NM. The percent relative bioavailability of complex in rabbits was 185.37 as compared to the plain NM.  相似文献

Complex stability constants (K S), standard molar enthalpy changes (ΔH°) and entropy changes (TΔS°) for the inclusion complexation of native β-cyclodextrin (β-CD) (1) and some modified β-CDs, i.e., mono(6-ethylenediamino-6-deoxy)-β-CD (3), mono[6-diethylenetriamino-6-deoxy]-β-CD (4) and their corresponding copper complexes 5 and 6, with four representative bile acid guests, i.e., cholate (CA), deoxycholate (DCA), glycocholate (GCA) and taurocholate (TCA), were determined at 25 °C in aqueous phosphate buffer solution (pH 7.20) by means of isothermal titration microcalorimetry (ITC). The stoichiometry of resulting inclusion complexes between CDs and bile acids was demonstrated by UV and conductivity as well as ITC experiments, showing 1:1 binding model upon all inclusion complexation except for metal-mediated dimer 5. The complex stability constants for modified β-CD 24 are dramatically magnified with the extended length of amino tether. As compared with 3 and 4, copper(II) complexes 5 and 6 significantly enhance not only binding ability but also molecular selectivity toward bile guest molecule CA through multipoint recognition, but decreased complexes stability toward TCA could be attributed to the decreased hydrophobic microenvironment of CDs cavity due to the introduction of copper(II) coordination center. Thermodynamically, the resulting complexes between hosts and bile guests are driven absolutely by enthalpy, accompanied by entropy gain or loss. Using the present data and those previously reported for mono(6-amino-6-deoxy)-β-CD (2), thermodynamic behavior and enhanced molecular selectivity could be discussed from the viewpoint of hydrophobic interactions, electrostatic cooperation and van der Waals between the hosts and guests.  相似文献

The inclusion complexes of a series of bis-quarternary ammonium surfactants, (C n N)2Cl2 (where n = 12, 14, 16) and sodium bis(2-ethylhexyl) sulfosuccinae (AOT), with α-cyclodextrin (α-CD), β-cyclodextrin (β-CD) and γ-cyclodextrin (γ-CD) in aqueous solutions were investigated by using isothermal titration calorimetry (ITC) at 298.15 K. The stability constants, stoichiometry, and formation enthalpies, entropies and Gibbs energies for the complexes in aqueous solutions have been derived from the calorimetric data. The values of the binding constant, K i , are very large, which indicates that these complexes are quite stable in their aqueous solutions. The enthalpy changes (ΔH ) for all of the inclusion processes are negative, showing that the complex process is enthalpy driven. The entropy effect (TΔS ) is negative, so the inclusion process is entropically unfavorable. The large negative Gibbs energy changes indicate that formation of host-guest inclusion complexes is generally a spontaneous process. The thermodynamic parameters are discussed in the light of the different structures of the host and guest molecules.  相似文献

Drugs with poor water solubility were co-ground with cyclodextrins (CDs) to create nanoparticles with improved solubility characteristics. Indomethacin (IDM), furosemide (FRM) and naproxen (NAP) were co-ground with β-CD at the molar ratio of 2:1 (CD:drug). Co-grinding of a drug with CD resulted in not only the formation of drug nanoparticles but also the solubilization of the drug by inclusion complex formation with CD in aqueous media. The nanoparticle fraction of IDM, and FRM from ground mixtures prepared with β-CD was as high as 60–70% while the solubilization fraction was less than 10%. In contrast, β-CD–NAP ground mixture showed a large fraction, 48%, for drug solubilization and only 4% for nanoparticle formation. Furosemide ground mixtures prepared with α-CD, β-CD and γ-CD showed comparatively high nanoparticle fraction while the solubilization fraction was around 10%. Both the nanoparticle fraction and the solubilization fraction were greater in the IDM–β-CD system than those in γ-CD and α-CD systems. The nanoparticle formation of NAP depended on the types of CD used as a co-grinding additive. Naproxen nanoparticles could be prepared by co-grinding NAP and α-CD, while the solubilization of NAP tended to improve when β-CD or γ-CD was used.  相似文献
