3',6'-Bis(diphenylphosphinyl)fluorescein (PF-1) was synthesized as a highly selective and sensitive fluorescent probe for imaging O(2) (.-) in living cells. The design strategy for the probe was based on the nucleophilic mechanism of O(2) (.-) to mediate deprotection of this probe to give fluorescein. Upon reaction with O(2) (.-), the probe exhibits a strong fluorescence response and high selectivity for O(2) (.-) over other reactive oxygen species and some biological compounds. The phosphinate-based probe, as a new fluorescent reagent, is cell-permeable and can detect micromolar changes of O(2) (.-) concentrations by using confocal microscopy in living cells. The unique combination of good selectivity, high sensitivity, good water solubility, and rapid reactivity establishes the potential value of the probe for facilitating investigations of the generation, metabolism, and mechanisms of superoxide-mediated cellular homeostasis and injury.  相似文献

以4-(二乙氨基)水杨醛为原料,经7-二氨基-3-羧酸香豆素合成了一种新型酰胺类Cu2+荧光探针1。本实验具有合成原料经济易得,合成方法简单易行的特点。并且该探针对Cu2+的检测具有良好的离子选择性,抗干扰性能和较高的检测灵敏度,探针1的浓度为10-5 mol/L时检测限达到2.4×10-7mol/L。同时该探针可以与Cu2+离子1:2的摩尔比络合。  相似文献

A novel two-photon excited fluorescent probe for cadmium (named as TPCd) was designed and synthesized utilizing a prodan (6-acetyl-2-methoxynaphthalene) derivative as the two-photon fluorophore and an o-phenylenediamine derivative as the Cd(2+) chelator, which possessed favorable photophysical properties and good water-solubility. The probe was designed with a photoinduced electron transfer (PET) mechanism and thus was weakly fluorescent itself. After binding with Cd(2+) which blocked the PET process, the fluorescence intensity of the probe was enhanced by up to 15-fold under one-photon excitation (OPE) and 27-fold under two-photon excitation (TPE), respectively. The two-photon action cross-section (Φδ) of the TPCd-Cd complex at 740 nm reached 109 GM compared to 3.6 GM for free TPCd, indicating the promising prospect of the probe in two-photon application. TPCd chelated Cd(2+) with 1 : 1 stoichiometry, and the apparent dissociation constant (K(d)) was 6.1 × 10(-5) M for the one-photon mode and 7.2 × 10(-5) M for the two-photon mode. The probe responded to Cd(2+) over a wide linear range from 0.1 to 30 μM with a detection limit of 0.04 μM. High selectivity of the probe towards Cd(2+) was acquired in Tris-HCl/sodium phosphate buffer. The probe was pH-independent in the biologically relevant pH range and non-toxic to living cells at reasonable concentration levels, warranting its in vivo applications. Through two-photon microscopy imaging, the probe was successfully applied to detect Cd(2+) uptake in living HepG2 cells.  相似文献

In this paper, a simple and sensitive small-molecule fluorescent probe, 2,5-dihydroxy-4'-dimethylaminochalcone (DHDMAC), was designed and synthesized for the detection of human serum proteins via hydrophobic interactions after polyacrylamide gel electrophoresis (PAGE). This probe produced lower fluorescence emission in the absence of proteins, and the emission intensity was significantly increased after the interaction with serum proteins. To demonstrate the imaging performance of this probe as a fluorescent dye, a series of experiments was conducted that included sensitivity comparison and 2D-PAGE. The results indicated that the sensitivity of DHDMAC staining is comparable to that of the most widely used fluorescent dye, SYPRO Ruby, and more protein spots (including thyroxine-binding globulin, angiotensinogen, afamin, zinc-α-2-glycoprotein and α-1-antichymotrypsin) were detected after 2D-PAGE. Therefore, DHDMAC is a good protein reporter due to its fast staining procedure, low detection limits and high resolution.  相似文献

A small molecule fluorescent probe, 4-[2-(4-chlorophenyl)-4,5-diphenyl-1H-imidazol-1-yl]aniline(L) for detecting Ag+ ion was gently synthesized via one-pot multi-component reaction catalyzed by H3[PW12O40] under solvent-free microwave irradiation. When the concentration of Ag+ ion changed from 0 to 8.0×10−6 mol/L in the solution of H2O/CH3OH(19:1, volume ratio), the fluorescence emission spectrum was blue-shifted and accompanied by a gradual increase in fluorescence intensity with a low detection limit of 3.0×10−11 mol/L. Moreover, UV-Vis absorption titration experiment demonstrated a 1:1 stoichiometric ratio and an association constant of (9.95±0.44)×105 L/mol between probe L and Ag+ ion, and thus their complexation mechanism was also proposed and verified. More importantly, this fluorescent probe was remarkably specific for Ag+ ion under the interference of other metal ions and exhibited a wide pH application range of 4.0-8.0. Additionally, preliminary application of this probe was also carried out and satisfactory results were shown.  相似文献

A highly selective and sensitive fluorescent Zn(2+) sensor, 2,6-bis(2-hydroxy-benzoic acid hydrazide)-4-methylphenol (1), was designed and synthesized. In aqueous THF (4 : 6 v/v) ligand 1 induces a 2 : 1 complex formation with respect to Zn(2+) at physiological pH. This probe features visible light excitation(390 nm) and emission (490 nm) profiles, excellent selectivity responses for Zn(2+)over other competing biological metal ions with K(d) < 1 pM(2), LOD < 1 ng L(-1) and about 680 fold enhancement in fluorescent intensity upon Zn(2+) binding. It also exhibits cell permeability and intracellular Zn(2+) sensing in A375 human melanoma cancer cell.  相似文献

Two new compounds,4-(2-bromophenxl-4,5-diphenyl-imidazol-1-yl)ani 1 ine(probe 1)and 4-[2,4,5-tris(4-bromopheny1)-1H-imidazol-1-y1]aniline(probe 2),were synthesized via a soft and high-efficiency one-pot microwave-assisted method under solvent-free conditions.Their sensing to different metal ions was detected by UV spectrophotometry and fluorescence spectrometry.Probe 2 revealed highly selective and sensitive UV and fluorescence response to Fe^3+ion.Upon the addition of Fe^3+ion,probe 2 showed obvious color change of the solution,conspicuous absorbance enhancement and relatively quick fluorescence quenching.The detection limit for Fe^3+ion was respectively calculated to be 0.72μmol/L(fluorescent detection)and 0.48μmol/L(UV-spectnim detection).Also,probe 2 was bound by Fe^3+ion to tonn a 1:1 complex.Moreover,preliminary application of probe 2 tor detecting Fe^3+ion in aqueous solution was attempted,and satisfying results were obtained.  相似文献

A new fluorogenic method for selective and sensitive determination of beryllium using 2,6-diphenyl-4-benzo-9-crown-3-pyrane (DBCP) was developed. The proposed fluorescent probe undergoes fluorescent emission intensity enhancement upon binding to beryllium ions in MeOH/H(2)O (70:30, v/v) solution. The fluorescence enhancement of DBCP is attributed to a 1:1 complex formation between DBCP and Be(2+) ion, which has been utilized as the basis for selective detection of Be(2+) ion. With the optimum condition described, the fluorescence enhancement at 531 nm was linear to the concentration of beryllium in the range of 1.6×10(-8)-1.6×10(-7) M and a detection limit of 1.5×10(-9) M. The fluorescent probe exhibits high selectivity for Be(2+) ion over the other common mono, di- and trivalent cations.  相似文献

2-(2′,5′-Dihydroxy-phenyl)-4(3H)-quinazolinone (DHPQ), a new fluorescent dye that exhibits excited state intramolecular proton transfer (ESIPT) reaction and possesses good photophysical properties, is synthesised and used as fluorescent probe for detection of Hg2+. Mercuric ions can be detected and quantitated by measuring the fluorescent intensity decrease of the probe. The decrease of fluorescence intensity of DHPQ upon the addition of Hg2+ was attributed to the blocking of ESIPT reactions of DHPQ and quenching its fluorescence. The analytical performance characteristics of the proposed Hg2+ probe were investigated. The probe can be applied to the quantification of Hg2+ with a concentration range covering from 8.0?×?10?7 to 2.0?×?10?4?mol?L?1, with a working pH range of 5.5–6.5. It shows excellent selectivity for Hg2+ over other transition metal cations. The proposed method was testified for the Hg2+ assay in river water samples with satisfying recoveries.  相似文献

A new two-photon fluorescent Cd(2+) probe APQ is developed by introducing a N(1),N(1)-dimethyl-N(2)-(pyridin-2-ylmethyl)ethane-1,2-diamine binding group and a 4-methoxyphenylvinyl conjugation-enhancing group to the 2- and 6-positions of quinoline. This probe shows a large red shift and good emission enhancement under Cd(2+) binding. It also exhibits a high ion selectivity for Cd(2+) (especially over Zn(2+)) and a large two-photon absorption cross section at 710 nm. Two-photon microscopy imaging studies reveal that the new probe is non-toxic and cell-permeable and can be used to detect intracellular Cd(2+) under two-photon excitation.  相似文献

Copper is the third most abundant essential transition metal ion in the human body. It's responsible for important activities in many living things, but excessive intake of Cu2+ can lead to a range of diseases. A colorimetric and turn-off fluorescent probe (E)-2-(5-(benzothiazol-2-yl)-2-(diethylamino)-4-hydroxybenzylidene)-N-phenylhydrazine-1-carbothioamide ( ZTR ) was designed and synthesized by thiosemicarbazone Schiff base as a specific complexes site strategy to achieve highly specific Cu2+ detection. The fluorescence of the probe ZTR solution fell dramatically when Cu2+ was added, and its appearance changed from dazzling blue to nearly colorless. The simple structure and readily available fluorescent probe provide a novel approach for the quantitative detection of Cu2+ in the linear range from 0 to 0.12 μM, with a detection limit down to 16 nM, and with high selectivity for Cu2+ over 15 other metal ions. Job’s plot analysis showed that probe ZTR and Cu2+ formed a 1:1 coordination complex. In addition, because of its low detection limits and fast response time, the created fluorescent molecule was effectively used to study the target ions on test paper strips and in water samples.))  相似文献

Two highly selective OFF-ON green emitting fluorescent thiol probes (1 and 2) with intense absorption in the visible spectrum (molar extinction coefficient ε is up to 73?800 M(-1) cm(-1) at 509 nm) based on dyads of BODIPY (as electron donor of the photo-induced electron transfer, i.e.PET) and 2,4-dinitrobenzenesulfonyl (DNBS) (as electron acceptor of the PET process) were devised. The single crystal structures of the two probes were determined. The distance between the electron donor (BODIPY fluorophore) and the electron acceptor (DNBS) of probe 2 is larger than that of probe 1, as a result the contrast ratio (or the PET efficiency) of probe 2 is smaller than that of probe 1. However, fluorescence OFF-ON switching effects were observed for both probe 1 and probe 2 in the presence of cysteine (the emission enhancement is 300-fold for probe 1 and 54-fold for probe 2). The fluorescence OFF-ON sensing mechanism is rationalized by DFT/TDDFT calculations. We demonstrated with DFT calculations that DNBS is ca. 0.76 eV more potent to accept electrons than the maleimide moiety. The probes were used for fluorescent imaging of cellular thiols.  相似文献

In this paper, 2-carboxybenzaldehyde rhodamine B thiohydrazine(1) was synthesized and developed as a fluorescent probe to recognize Hg2+in DMF/H2 O(1:9, v/v) solution with high selectivity. The probe can be applied to the quanti?cation of Hg2+with a linear concentration range covering from 1.0×10-7mol/L to 1.0×10-5mol/L(R2= 0.9985) and a detection limit of 4.2×10-8mol/L. The experiment results show that the response of probe 1 to Hg2+is pH-independent in a wide range from 4.0 to 9.0. Moreover, the probe 1 exhibits excellent selectivity toward Hg2+over other common metal cations. Most importantly, the probe can be employed to monitor Hg2+in living cells using fluorescent imaging technique with satisfied results.  相似文献

A new rhodamine B-based dual-function chromo- and fluorogenic probe for Cu(2+) and ClO(-) has been designed, synthesized, and characterized. The probe comprises a spectroscopic unit of rhodamine B and a Cu(2+)-specific chelating unit of pyridinecarboxamide as well as a ClO(-)-specific reactive moiety of diacylhydrazine, and is a highly selective and extremely sensitive fluorescent and colorimetric sensor for Cu(2+) and ClO(-) in different pH conditions. 为了选择性检测小分子生物硫醇,以具有优良荧光性能的喹啉酮为荧光团,依据依布硒啉中Se—N键易与硫醇分子反应的性质,将喹啉酮组块(E)-3-(5-巯基-1,3,4-恶二唑-2-基)-N-(4-甲基-2-氧代-1,2-二氢喹啉-7-基)丙烯酰胺(MQ5)与依布硒啉2-(4-溴苯基)苯并[d][1,2]硒唑-3(2H)-酮(SQ6)对接,设计合成了一种新型荧光探针(E)-N-(4-甲基-2-氧代-1,2-二氢喹啉-7-基)-3-(5-((4-(3-氧代苯并[d][1,2]硒烯唑-2 (3H)-基)苯基)硫基)-1,3,4-噁二唑-2-基)丙烯酰胺(MNQ)。 通过傅里叶变换红外光谱(FT-IR)、核磁共振波谱仪(NMR)和高分辨质谱(HRMS)和荧光光谱等测试手段对其进行了结构表征,探究了其荧光性能。 结果表明,MNQ对谷胱甘肽(GSH)有明显的荧光猝灭,在其他氨基酸等干扰时,探针具有良好的抗干扰能力,可作为识别检测GSH的荧光猝灭型探针。 检测限为2.99×10-8 mol/L,响应时间在35 s可完成,有望作为检测谷胱甘肽的荧光探针。  相似文献

在乙醇体系中构建三元配合物的镝荧光探针,并利用盐酸四环素(TC)对镝荧光探针具有荧光猝灭作用,提出了一种检测牛奶中四环素残留的新方法.首先确定体系的激发波长为305 nm,发射波长为574 nm.在配比、加料顺序、时间等方面对镝荧光探针进行了条件优化,确定了镝离子(Dy3+)、磺基水杨酸(SSA)、三正辛基氧化磷(TOPO)的最佳配比为1:2:0.1和30 min最佳检测时间.其次建立了检测盐酸四环素的线性曲线,并获得检测范围为10-6~2×10-5 mol/L.最后,利用此荧光探针对处理的牛奶样品进行加样回收检测,回收率在96.9 %~104.4 %.实验证明建立的方法科学可行,对盐酸四环素具有高选择性.  相似文献
