The reaction of 7,9-dibromo-5-tribromornethyl-2-t-butyl-4-cyano-1,3,6,9b-tetraazaphenalene ( 1a ) with p-toluidine is shown to give 4,6-dibromo-2-t-butyl-8,13-dihydro-13-imino-11-methyl-1,3,7,8,13c-pentaazabenzo[de]naphthacene ( 4 ) in two steps with 7,9 dibromo-2-t-butyl-4-cyano-5-p-toluidino-1,3,6,9b-tetraazaphenalene ( 2b ) as the intermediate product. A related annulation reaction of 1a with N-(5-amino-2,4-dimethylphenyl)trimethylacetamide ( 8 ) leads in two steps to 9,11-dibromo-2,13-di-t-butyl-4,6-dimethyl-7H-1,3,7,8,11b,12,14-heptaazadibenzo[de,hi]naphthacene ( 6 ) with 7,9-dibromo-2-t-butyl-4-cyano-5N-(2,4-dimethyl-5-trimethylacetamidophenyl)amino-1,3,6,9b-tetraazaphenalene ( 2d ) as the intermediate product. In a similar fashion the reaction of 1a with o-phenylenediamine forms 14-amino-4,6-dibromo-2-t-butyl-8H-1,3,7,8,13,14c-hexaazobenzo[4,5]cyclohepta[1,2-a]-phenalene ( 12 ) by way of the intermediate 5-N-(2-aminophenyl)amino-7,9-dibromo-2-t-butyl-4-cyano-1,3,6,9b-tetraazaphenalene ( 2e ). The preparation of N-(2,4-dimethyl-5-nitrophenyl)-trimethylacetamide ( 11 ) and its reduction to N-(5-amino-2,4-dimethylphenyl)trimethylacetamide ( 8 ) is also described.  相似文献

The reaction of N-cyano-N′-(6-amino-2-pyridyl)acetamidine ( 5a ) and homophthalic anhydride followed by ring closure of the 2-[2-(carboxymethyl)phenyl]-5-methyl-1,3,4,6,9b-pentaazaphenalene intermediate ( 4a ) gave 5-methyl-13-oxo-13H-4,6,7,13a,13c-pentaazabenzo[hi]chrysene ( 8a ). An analogous series starting with 3-N-(6-amino-2-pyridyl)amino-2-cyano-2-butenenitrile ( 5b ) in place of 5a gave in two steps 5-methyl-13-oxo-13-H-4,7,13a,13c-tetraazabenzo[hi]chrysene-6-carbonitrile ( 8b ). Elemental analysis, ir and pmr spectra of 8a , 8b and several new model compounds aided in confirming the structures of 8a and 8b. The synthesis of one of these model compounds for 5b and phenylacetic anhydride led surprisingly to 2-methyl-9-phenyl-7H-3,7,-10,10b-tetraazacyclohepta[de]naphthalene ( 10 ) in addition to the expected 2-benzyl-4-cyano-5-methyl-1,3,-6,9b-tetraazaphenalene ( 7b ).  相似文献

The reaction of 2-amino-4-chloro-6-methylpyrimidine ( 3a ) with trimethylacetyl chloride gave 4-chloro-6-methyl-2-trimethylacetamidopyrimidine ( 5 ). This latter compound with excess anthranilonitrile gave in one step 2-t-butyl-5-methyl-1,3,4,7,11c-pentaazabenz[de]anthracene ( 6a ). To prepare 2-t-butyl-5-dimethylamino-1,3,4,6,7,11c-hexaazabenz[de]anthracene ( 6b ) it was found necessary to first react 2-amino-4-chloro-6-dimethylamino -5 -triazine ( 3b ) with anthranilonitrile to yield the intermediate product 2-amino-4(2-cyanoanilino)-6-dimethylamino-s-triazine ( 4 ). Reaction of the latter with trimethylacetyl chloride gave 6b .  相似文献

Coupling the diazonium salt of 3-amino-2-cyano-4,6-dimethylthieno[2,3-b]pyridine 1 with malononitrile 2 gave 2-cyano-3-(hydrazonomalononitrile)-4,6-dimethylthieno[2,3-b]pyridine 3 which then reacted with hydrazine compounds 4a-4h to yield corresponding 2-cyano-3-(3,5-diamino-1-substituted-pyrazol-4-yl)azo-4,6-dimethylthieno[2,3-b]pyridines 5a-5h. The 2-cyano-3-(2-amino-5,7-disubstituted-pyrazolo-[1,5-a]pyrimidine-3-yl)azo-4,6-dimethylthieno[2,3-b]pyridines 7a-7f were obtained in good yield by the cyclocondensation reaction of 2-cyano-3-(3,5-diamino-pyrazol-4-yl)azo-4,6-dimethylthieno[2,3-b]pyridine 5a with the appropriate 1,3-diketones 6a-6f under acidic condition.  相似文献

The reaction of 5a-acetyl-6-ethoxycarbonyl-5a,6a-dihydro-6H-cyclopropa[e]pyrazolo[1,5-a]pyrimidine-3-carbonitrile ( 1a ) with benzylamine gave ethyl l-benzyl-5-cyano-8a,9-dihydro-2-methyl-1H-pyrrolo[3,4-e]-pyrazolo[1,5-a]pyrimidine-8a-carboxylate ( 2a ), in addition to 5-acetyl-3-benzylamino-1-(4-cyanopyrazol-3-yl)- 2-pyridone ( 3 ). Reaction of 1a with aniline gave ethyl 6-acetyl-8-anilino-3-cyano-7,8-dihydro-4H-pyrazolo-[1,5-a][1,3]diazepine-8-carboxylate ( 4 ), in addition to ethyl 3-cyano-7-methyl-6-pyrazolo[1,5-a]pyrimidine-acrylate ( 5 ). On the other hand, the same reactions of 1b with benzylamine or aniline gave 2b or 8b , respectively. Though catalytic hydrogenation of 1a over 5% palladium-carbon proceeded by ring fission of cyclopropane ring to give 9 , 1a (or 1b ) afforded 4,5-dihydro derivatives ( 13 or 15 ) by catalytic hydrogenation over platinum oxide. The reactivity of 5-methoxy-4,5,5a,6a-tetrahydro-6H-cyclopropa[e]pyrazolo[1,5-a]pyrimidine ( 16 ), which are related analogs of 1a,b , is also described.  相似文献

Some new 7,9-disubstituted 7H-1,2,3,4-tetrazolo[1,5-c]pyrrolo[3,2-e]pyrimidines 5 have been synthesized either by diazotization of 4-hydrazino-5,7-disubstituted-7H-pyrrolo[2,3-d]pyrimidines 4 obtained by hydrazinolysis of 4-chloro-5,7-disubstituted-7H-pyrrolo[2,3-d]pyrimidines 3 or via a substitution reaction between 3 and sodium azide. 5,7-Disubstituted-7H-pyrrolo[2,3-d]pyrimidin-4(3H)-ones 2 were obtained by cyclocondensation of 2-amino-3-cyano-1,4-disubstituted pyrroles 1 with formic acid which on chlorination using phosphorus oxychloride afforded 3 . A novel route for the synthesis of 4-amino-5,7-disubstituted-7H-pyrrolo[2,3-d]pyrimidines 6 by the reductive ring cleavage of 5 has been reported.  相似文献

The reaction of 3-amino-4,6-dimethyl-2-thieno[2,3-b]pyridine carbohydrazide ( 1 ) with appropriate 1,3-diketones 2a-2d in glacial acetic acid afforded 3-amino-2-[(3,5-disubstituted-pyrazo)-1-yl)carbonyl]-4,6-dimethylthieno[2,3-b]pyridines 3a-3d. 3-Amino-2-[(5-amino-3,4-disubstituted-pyrazol-1-yl)carbonyl]-4,6-dimethylthieno[2,3-b]pyridines 5a-5h were also prepared by treatment of carbohydrazide 1 with appropriate alkylethoxymethylenes and ketene dithioacetals 4a-4h , respectively.  相似文献

Substituted 3-hydroxypyrazoles, which were prepared based on ethyl esters of substituted 4-(pyridin-2-ylthio)- or 4-(1,4-dihydropyridin-2-ylthio)acetoacetic acids and hydrazine hydrate, were used in the synthesis of 6-amino-4-aryl-5-cyano-3-(pyridin-2-ylthiomethyl)-2,4-dihydropyrano[2,3-c]pyrazoles. The molecular and crystal structure of 6-amino-5-cyano-3-(3-cyano-4,6-dimethylpyridin-2-ylthiomethyl)-4-(2-nitrophenyl)-2,4-dihydropyrano[2,3-c]pyrazole was established by X-ray diffraction analysis.  相似文献

A synthesis of novel series of bis(2-amino-3-cyano-4H-chromene), bis(2-amino-3-cyanopyrano[3,2-c]chromene), bis(6-amino-5-cyano-1,4-dihydropyrano[2,3-c]pyrazole), and bis(2,7-diamino-3,4,6-tricyanopyrazolo[1,5-a]pyridine) derivatives linked to thieno[2,3-b]thiophene core via ether or ester linkages was reported. The formation of the target compounds presumably proceeds through a three component reaction of the appropriate bis(aldehyde) with 2 M of each of malononitrile and the corresponding active methylene compounds namely; dimedone, 4-hydroxycoumarine, 5-methyl-2,4-dihydro-3H-pyrazol-3-one, or cyanomethylpyrazole in dioxane at reflux in the presence of piperdine as a basic catalyst. The chemical structures of the new compounds were elucidated by several spectral tools.  相似文献

A number of imidazo[1,5-a]pyrimidine-8-carboxamides were synthesized by reacting various β-dicarbonyl compounds with 5(4)-aminoimidazole-4(5)carboxamide (AICA, 1 ), the non-ribosylated form of AICAR, a key intermediate in the metabolic pathway of purine biosynthesis. Cyclization of 1 with ethylacetoacetate yielded 2-methylimidazo[1,5-a]pyrimidin-1H-4-one-8-carboxamide ( 2 ). The treatment of 2 with phosphorus oxychloride gave 4-chloro-8-cyano-2-methylimidazo[1,5-a]pyrimidine ( 3 ). Various nucleophiles displaced the 4-chloro substituent of 3 under mild conditions. However, the 4-methylthio group of 8-cyano-2-methyl-4-methylthioimidazo[1,5-a)pyrimidine ( 8a ) was also displaced under very mild conditions. Even more strangely, the 4-diethylamino group of 8-cyano-4-diethylamino-2-methylimidazo[1,5-a]pyrimidine ( 5a ) was displaced by ammonia to give 4-amino-8-cyano-2-methylimidazo[1,5-a]pyrimidine ( 7 ).  相似文献

The reaction of 3-amino-4,6-dimethylthieno[2,3-b]pyridine-2-carbonitrile with ethylenediamine in the presence of a catalytic amount of carbon disulfide afforded 2-(4,5-dihydro-1H-imidazol-2-yl)-4,6-dimethylthieno-[2,3-b]pyridine-3-amine while its reaction with triethyl orthoformate followed by the reaction with hydrazine hydrate gave 4-imino-7,9-dimethylpyrido[3',2':4,5]thieno[3,2-d]pyrimidine-3(4H)-amine. These two derivatives underwent cyclocondensation reactions with commercially available reactants to afford new heterocycles containing the thieno[2,3-b]pyridine moiety. Some of the synthesized derivatives were tested for antimicrobial and antifungal activity.  相似文献

Cyclopentylidene- and cyclohexylidene(cyano)acetamides reacted with malononitrile and cyano-(thioacetamide) according to the Michael pattern with exchange of the methylene components to give substituted 1-amino-2,6,6-tricyano-1,3-cyclohexadienes and thieno[2,3-d]pyrimidine-4(3H)-thiones. Condensation of cyclopentylidene- and cyclohexylidene(cyano)acetamide with 1,3-dicarbonyl compounds afforded 4,6-di-methyl-3-cyanopyridine-2(1H)-thione and morpholinium 4-methyl-6-oxo-3-cyano-1,6-dihydropyridine-2-thiolate which were converted into substituted 2-alkylsulfanylpyridines, thieno[2,3-b]pyridines, thiazolo[3,2-a]pyridine, and 2H-[1,3]thiazino[3,2-a]pyridine.  相似文献

This paper describes the synthesis of a new series of 6-amino-4-aryl-5-cyanopyrazolo[3,4-b]pyridines 4 and 4-aryl-5-cyano-6H-pyrazolo[3,4-b]pyridin-6-ones 5 from the reaction of 5-amino-3-methyl-1-phenylpyrazole 1 with arylidene derivatives of malonodinitrile 2 and ethyl cyanoacetate 3 . The structure of the final compounds was determined on the basis of nmr measurements, especially by 1H, 1H-, 1H, 13C-COSY, DEPT and X-ray diffraction.  相似文献

The three-component reaction of 4-hydroxy-6-methylpyran-2(2H)-one with cyanoacetic acid derivatives and carbonyl compounds in EtOH or in the ionic liquid, viz., 1-butyl-3-methylimidazolinium hexafluorophosphate ([bmim][PF6]), affords substituted 2-amino-7-methyl-5-oxo-4,5-dihydropyrano[4,3-b]pyrans. The yield of substituted pyrano[4,3-b]pyrans in [bmim][PF6] is by 10—14% higher than that in EtOH.  相似文献

The synthesis of the title compounds 6 and 8 has first been accomplished by reaction of O-mesitylsulfonyl-hydroxylamine with 5-t-butyl-3H-v-triazolo[4,5-d]pyrimidine ( 5 ) whose preparation is reported in detail. However the preferred route for the synthesis of the 3-amino derivative 8 is based upon the preparation of 3-benzylideneamino-5-t-butyl-3H-v-triazolo[4,5-d]pyrimidine ( 10 ), followed by the removal of the benzylidene protecting group. This critical step was effected by treatment of 10 with dilute hydrochloric acid, in the presence of 2,4-dinitrophenylhydrazine. The diazotization of 5-amino-4-hydrazino-2-t-butylpyrimidine gave predominantly the tetrazolo[1,5-c]-pyrimidine 13 along with a small amount of compound 8.  相似文献
