The substrate specificity of the heat-stable stereospecific amidase from Klebsiella oxytoca was investigated. In addition to the original substrate, 3,3,3-trifluoro-2-hydroxy-2-methylpropanamide, the amidase accepted 2-hydroxy-2-(trifluoromethyl)-butanamide and 3,3,3-trifluoro-2-amino-2-methylpropanamide as substrates. Compounds with larger side chains and compounds where the hydroxyl group was substituted with a methoxy group, or in which the CF3 group was substituted by CCl3, were not accepted. The biotransformation is a new synthetic route to (R)-(+)-3,3,3-trifluoro-2-amino-2-methylpropanoic acid, and its related (S)-(−)-amide, and to (R)-(+)-2-hydroxy-2-(trifluoromethyl)-butanoic acid and its related (S)-(−)-amide.  相似文献

Heating the substituted pyridinium and isoquinolinium salts with E-1,2-di(alkylsulfonyl)-1,2-dichloroethenes in either chloroform or acetone in the presence of three-fold excess of Et3N gave high yields of substituted 1,2-di(alkylsulfonyl)indolizines and 1,2-di(alkylsulfonyl) pyrrolo[2,1-a]isoquinolines, respectively. Effects of the structure of 3-substituted pyridinium ylides on the regioselectivity of their reaction with E-1,2-di(alkylsulfonyl)-1,2-dichloroethenes were revealed. It was shown that the presence of electron-releasing and electron-withdrawing substituents in the pyridinium ylide favors the formation of 8-substituted and 6-substituted 1,2-(dialkylsulfonyl)indolizines, respectively.  相似文献

Sonogashira coupling of (E)-α-iodovinylstannanes 1 with (trimethylsilyl)acetylene gave (Z)-1-(trimethylsilyl)-3-(tributylstannyl)alk-3-en-1-ynes 2, which underwent a desilylation reaction to afford (Z)-3-(tributylstannyl)alk-3-en-1-ynes 3 in high yields. (1E,3Z)-1-Halo-3-(tributylstannyl)-substituted 1,3-dienes 5 could be synthesized stereoselectively via hydrozirconation of (Z)-3-(tributylstannyl)alk-3-en-1-ynes 3, followed by trapping with iodine or NBS.  相似文献

(1R)-1-Deuterated alcohols with high enantiomeric excess were prepared via TiCl4/Et3SiD reduction of acetals arising from the reaction of aldehydes with (1S,2S)-3,3,3-trifluoro-1-phenylpropane-1,2-diol 9. Such a chiral auxiliary was synthesized in an enantiomerically pure form starting from l-mandelic acid. Due to its benzylic nature, it was easily removed from the reaction product of the reductive 1,3-dioxolane ring-cleavage to afford the desired α-deuterated alcohol.  相似文献

Cathodic reductions of 2,2′-furils in the presence of N-arylcarbonimidoyl dichlorides lead to 2-arylimino-4,5-di(2-furyl)-1,3-dioxoles in high yields, along with minor amounts of (E)-1,2-di(2-furyl)vinylene bis(N-arylchloroformimidates). HF and B3LYP density functional theory methods have been applied to the determination of molecular geometries and to study the topomerization mechanism of aryliminodioxoles. The molecular structure of (E)-1,2-di(2-furyl)vinylene bis[N-(2-chloro-4-methylphenyl)chloroformimidate] has been determined by X-ray crystallography and compared with the calculated structure.  相似文献

The 3,3,3-trifluoroprop-1-en-2-yl-substituted furans (4) were synthesized via palladium-catalyzed cyclization-isomerization of 1,1,1-trifluoro-2-[(tert-butyldimethylsilyloxy)methyl]-3-alkynylbut-2-en-1-ols (3), which were readily obtained from 1,1,1-trifluoro-2-[(tert-butyldimethylsilyloxy)methyl]-3,3-dibromoprop-2-ene (1) in three steps.  相似文献

New, highly functionalised (1E,3E)-1-(benzoylamino)-4-(dimethylamino)buta-1,3-diene-1,2,3-tricarboxylates proved to be useful and versatile reagents in the formation of highly substituted pyridine, N-aminopyridine, pyrrole and pyrido[3,4-c]pyridazine derivatives. The formation of the particular type heterocyclic system is dependent on the starting (1E,3E)-1-(benzoylamino)-4-(dimethylamino)buta-1,3-diene-1,2,3-tricarboxylate. By an appropriate choice of different ester groups it is possible to drive the reactions towards the formation of either pyridine or pyrrole derivatives.  相似文献

The selective conversion of 2,2,2-trifluoro-1-furan-2-yl-ethanone into (E)- and (Z)-oximes and oxime ethers and subsequent oxazaborolidine-catalyzed enantioselective reduction using different amino alcohols furnished both enantiomers of the important chiral building block 2,2,2-trifluoro-1-furan-2-yl-ethylamine with e.e. of up to 88%. Oxidation of the furan ring afforded both enantiomers of 3,3,3-trifluoroalanine in 91–93% yields.  相似文献

3,7-endo-Disubstituted 2,5-diazabicyclo[2.2.1]heptanes were obtained by iodo-cyclization of N,N′-di[(S)-1-phenylethyl]-(E,E)-4,5-diamino-1,8-diphenyl-1,7-octadiene and substituted N,N′-di[(S)-1-phenylethyl]-1,2-diamino-4-alkenes. Removal of only one N-substituent of the bridged piperazines was achieved by reduction with ammonium formate and Pd/C. Unexpected cleavage of the skeleton of vinyl-substituted bridged piperazines was observed using hydrogen, leading to substituted 3-aminopyrrolidines.  相似文献

Reactions of the title meso forms, (1R,2S)-1,2-di(2-furyl)-1,2-di(3-guaiazulenyl)ethane (1) and (1R,2S)-1,2-di(3-guaiazulenyl)-1,2-di(2-thienyl)ethane (2), with a two molar amount of TCNE in benzene at 25 °C for 5 h (for 1) and 48 h (for 2) under oxygen give new compounds, 2,2,3,3-tetracyano-4-(2-furyl)-8-isopropyl-6-methyl-1,4-dihydrocyclohepta[c,d]azulene (3) and 2,2,3,3-tetracyano-8-isopropyl-6-methyl-4-(2-thienyl)-1,4-dihydrocyclohepta[c,d]azulene (4), respectively, in 74 and 21% isolated yields. Comparative studies on the above reactions as well as the spectroscopic properties of the unique products 3 and 4, possessing interesting molecular structures, are reported and, further, a plausible reaction pathway for the formation of these products is described.  相似文献

The reactions of substituted pyridinium salts with E-1,2-di(alkylsulfonyl)-1,2-dichloroethenes proceed regiospecifically. Heating of these reagents in chloroform in the presence of a threefold excess of Et3N affords substituted 1,2-di(alkylsulfonyl)indolizines in high yields. The structures of the reaction products were confirmed by physicochemical methods, including X-ray diffraction. Dedicated to Academician N. K. 6-(Trifluoromethyl)comanic acid reacts regioselectively with phenylhydrazine in water to give 5-[3,3,3-trifluoro-2-(phenylhydrazono)propyl]-1-phenyl-1H-pyrazole-3-carboxylic acid. Similar reaction in dioxane leads to 3-[3,3,3-trifluoro-2-(phenylhydrazono)propyl]-1-phenyl-1H-pyrazole-5-carboxylic acid. A strong solvent influence on the reaction route was also found for 6-(trifluoromethyl)comanic acid derivatives.  相似文献

The compounds (E)-1,2-diphenyl-2-(phenylimino)ethanol, (E)-1,2-diphenyl-2-(p-tolylimino)ethanol, and (E)-2-((4-chlorophenyl)imino)-1,2-diphenylethanol) were synthesized by reaction of a p-substituted aniline with benzoin then oxidized with chromium trioxide–triethylamine in chloroform to give (E)-1,2-diphenyl-2-(phenylimino)ethanone, (E)-1,2-diphenyl-2-(p-tolylimino)ethanone, and (E)-2-((4-chlorophenyl)imino)-1,2-diphenylethanone in very high yield. Wittig reaction of (E)-3-(3-guaiazulenyl)propenal (11) with (3-guaiazulenylmethyl)triphenylphosphonium bromide (9) in ethanol containing NaOEt at 25 °C for 24 h under argon gives the title new (2E,4E)-1,3-butadiene derivative 4, in 33% isolated yield, which upon treatment with hexafluorophosphoric acid (i.e., 65% HPF6 aqueous solution) in tetrahydrofuran (=THF) at 25 °C for 1 h under aerobic conditions affords a new air (two-electron) oxidation product (E)-ethylene-1,2-bis(3-guaiazulenylmethylium) bis(hexafluorophosphate) (14), quantitatively, and further, zinc-reduction of 14 in trifluoroacetic acid (=CF3COOH) at 0 °C for 1 h under argon reverts 4, quantitatively. Along with the above interesting results, our discovered another preparation method, spectroscopic properties, crystal structure, and electrochemical behavior of 4, which serves as a strong two-electron donor and acceptor, compared with those of the previously reported (E)-1,2-di(3-guaiazulenyl)ethylene (3) are documented in detail.  相似文献

Depending on the reaction conditions, the reactions of (E)-3,3,3-trichloro-1-nitropropene with cyclohexanone enamines led to bicyclo[4.2.0]octanes or trisubstituted enamines, which are the ring-chain tautomers capable of reversible transformations. Diastereoselectivity of the reactions of (E)-3,3,3-trichloro(trifluoro)-1-nitropropenes with cycloalkanone enamines were studied, a series of hitherto unknown CX3-containing nitroalkylated enamines and γ-nitro ketones were synthesized, the structures of novel compounds were determined by NMR spectroscopy and X-ray diffraction.  相似文献

3,3,3-Trifluoropropynyllithium, in situ generated by treatment of 2-bromo-3,3,3-trifluoro-1-propene 1 with 2.0 equiv. of LDA at −78 °C, was trapped with d-glyceraldimine 2 to give trifluoromethylated propargylic amine 4 in 55% yield. Starting from the key intermediate 4, Boc-protected (R)-5,5,5-trifluoronorvaline and (S)-5,5,5-trifluoronorvaline were concisely synthesized over three steps in 62% and 63% yield, respectively.  相似文献

1,1,3,3,3-Pentafluoro-2-pentafluorophenyl-1,2-epoxypropane 1 reacted with trimethylphosphite giving two diastereomers, (Z)- and (E)-3,6-bis(trifluoromethyl)-3,6-bis(pentafluorophenyl)-1,4-dioxan-2,5-dione 2a, b in a 1:1 ratio, cyclodimerisation product of the intermediately generated α-lactone 4. The values of physicochemical properties determine completely the sequence of chromatographic elution of (Z)- and (E)-isomers of 1,2-disubstituted unsaturated compounds and can therefore be used as reference data for their GC-identification. The isomer with the minimum boiling point, relative density, and refractive index has the least retention parameters. By the ratio of the values of these properties the (Z)- and (E)-isomers form two groups: (Z) > (E) and (Z) < (E). This feature is due to the larger polarity of the (Z)-isomers of asymmetrically substituted compounds containing polar substituents at the double bond C=C. The discovered regularities may be extended to the cis- and trans-isomers of 1,2-disubstituted cycloalkanes and even to the position isomers of substituted arenes.  相似文献
