A combined assay for the determination of paclitaxel, docetaxel and ritonavir in human feces and urine is described. The drugs were extracted from 200 μL urine or 50 mg feces followed by high‐performance liquid chromatography analysis coupled with positive ionization electrospray tandem mass spectrometry. The validation program included calibration model, accuracy and precision, carry‐over, dilution test, specificity and selectivity, matrix effect, recovery and stability. Acceptance criteria were according to US Food and Drug Administration guidelines on bioanalytical method validation. The validated range was 0.5–500 ng/mL for paclitaxel and docetaxel, 2–2000 ng/mL for ritonavir in urine, 2–2000 ng/mg for paclitaxel and docetaxel, and 8–8000 ng/mg for ritonavir in feces. Inter‐assay accuracy and precision were tested for all analytes at four concentration levels and were within 8.5% and <10.2%, respectively, in both matrices. Recovery at three concentration levels was between 77 and 94% in feces samples and between 69 and 85% in urine samples. Method development, including feces homogenization and spiking blank urine samples, are discussed. We demonstrated that each of the applied drugs could be quantified successfully in urine and feces using the described assay. A liquid chromatography with tandem mass spectrometry method was developed for the simultaneous screening of 34 drugs and poisons in forensic cases. Blood (0.5 mL, diluted 1:1 with water) or 1.0 mL of urine was purified by solid‐phase extraction. Gastric contents (diluted 1:1 with water) were treated with acetonitrile, centrifuged, and supernatant injected. Detection was achieved using a Waters Alliance 2695/Quattro Premier XE liquid chromatography tandem mass spectrometry system equipped with electrospray ionization, operated in the multiple reaction monitoring modes. The method was validated for accuracy, precision, linearity, and recovery. The absolute recovery of drugs and toxic compounds in blood was greater than 51% with the limit of detection in the range of 0.02–20 ng/mL. The absolute recovery of drugs and toxic compounds in urine was greater than 61% with limit of detection in the range of 0.01–10 ng/mL. The matrix effect of drugs and toxic compounds in urine was 65–117% and 67–121% in blood. The limit of detection of drugs and toxic compounds in gastric content samples were in the range of 0.05–20 ng/mL. This method was applied to the routine analysis of drugs and toxic compounds in postmortem blood, urine, and gastric content samples. A liquid chromatography–tandem mass spectrometric method for the quantification of granisetron and its major metabolite, 7‐hydroxy granisetron in human plasma and urine samples was developed and validated. Respective stable isotopically labeled granisetron and 7‐hydroxy granisetron were used as internal standards (IS). Chromatography was performed using an Xselect HSS T3 analytical column with a mobile phase of 20% acetonitrile in water (containing 0.2 mM ammonium formate and 0.14% formic acid, pH 4) delivered in an isocratic mode. Tandem mass spectrometry operating in positive electrospray ionization mode with multiple reaction monitoring was used for quantification. The standard curves were linear in the concentration ranges of 0.5–100 ng/mL for granisetron and 0.1–100 ng/mL for 7‐hydroxy granisetron in human plasma samples, and 2–2000 ng/mL for granisetron and 2–1000 ng/mL for 7‐hydroxy granisetron in human urine samples, respectively. The accuracies were >85% and the precision as determined by the coefficient of variations was <10%. No significant matrix effects were observed for granisetron or 7‐hydroxy granisetron in either plasma or urine samples. Granisetron was stable under various storage and experimental conditions. This validated method was successfully applied to a pharmacokinetic study after intravenous administration of 1 mg granisetron to a pregnant subject. A highly selective and sensitive liquid chromatography–tandem mass spectrometry (LC MS/MS) method was developed for the quantification of metronidazole (MTZ) in human feces. The analyte was recovered from feces after liquid–liquid extraction with ethyl acetate and separated on Waters Symmetry® C18 (100 × 4.6 mm, 5μm) column using 0.1% formic acid in water and acetonitrile (40:60, v/v) as the mobile phase. A stable‐deuterated internal standard metronidazole‐d4 (MTZ‐d4) was used in the study. Mass analysis was performed on a triple quadrupole mass spectrometer in the positive electrospray ionization mode. A linear response function of MTZ was established in the concentration range of 0.50–250 ng/g, based on dry mass. The mean extraction recovery of MTZ (97.28%) and MTZ‐d4 (96.76%) from spiked feces samples was consistent at higher as well as lower concentrations. Post‐column infusion analysis showed no ion‐suppression/enhancement effects and the mean IS‐normalized matrix factor ranged from 0.986 to 1.013. Spiked feces samples stored at −20 and − 70°C for long‐term stability were stable for at least 3 months, while extracted samples (dry and wet extracts) were stable up to 24 h. The method was applied to determine MTZ in feces of 12 healthy Indian subjects.  相似文献

A facile and sensitive multi‐residue detection approach of pressurized liquid extraction following high‐performance liquid chromatography tandem mass spectrometry was established to detect the residues of adrenergic drugs, steroids, sedative, colorant and antioxidant in feed. The conditions employed for pressurized liquid extraction involved acetonitrile/ethyl acetate (1:1, v/v) as the extracting solvent, the temperature 80°C, two cycles and a static time of 10 min. The extraction was followed by a solid‐phase extraction clean‐up step. The separation of samples was done by C18 column with the mobile phase of 5 mM ammonium acetate solution and acetonitrile with 0.1% formic acid. The limits of quantification ranged from 0.03 to 1 μg/kg, limits of detection were in a range of 0.01–0.5 μg/kg, and average recoveries were 70.4–98.6%. The pressurized liquid extraction procedure was optimized and overall method was validated in terms of sensitivity, linearity, selectivity, matrix effect, accuracy, recovery and stability of the target drugs in the pressurized liquid extraction extracts solution. The screening method was proved to be fast, selective, accurate and sensitive for screening drugs.  相似文献

Three methods were developed and validated for determination of nemonoxacin in human feces and its major metabolite, nemonoxacin acyl‐β‐ d ‐glucuronide, in human urine and feces. Nemonoxacin was extracted by liquid–liquid extraction in feces homogenate samples and nemonoxacin acyl‐β‐ d ‐glucuronide by a solid‐phase extraction procedure for pretreatment of both urine and feces homogenate sample. Separation was performed on a C18 reversed‐phase column under isocratic elution with the mobile phase consisting of acetonitrile and 0.1% formic acid. Both analytes were determined by liquid chromatography–tandem mass spectrometry with positive electrospray ionization in selected reaction monitoring mode and gatifloxacin as the internal standard. The lower limit of quantitation (LLOQ) of nemonoxacin in feces was 0.12 µg/g and the calibration curve was linear in the concentration range of 0.12–48.00 µg/g. The LLOQ of the metabolite was 0.0010 µg/mL and 0.03 µg/g in urine and feces matrices, while the linear range was 0.0010–0.2000 µg/mL and 0.03–3.00 µg/g, respectively. Validation included selectivity, accuracy, precision, linearity, recovery, matrix effect, carryover, dilution integrity and stability, indicating that the methods can quantify the corresponding analytes with excellent reliability. Phenibut (3-phenyl-4-aminobutyric acid) is a γ-aminobutyric acid mimetic drug, which is used clinically as a mood elevator and tranquilizer. In the present work, a rapid, selective and sensitive liquid chromatography–tandem mass spectrometry method for quantification of phenibut in biological matrices has been developed. The method is based on protein precipitation with acidic acetonitrile followed by isocratic chromatographic separation using acetonitrile–formic acid (0.1% in water; 8:92, v/v) mobile phase on a reversed-phase column. Detection of the analyte was performed by electrospray ionization mass spectrometry in multiple reaction monitoring mode with the precursor-to-product ion transition m/z 180.3 → m/z 117.2. The calibration curve was linear over the concentration range 50–2000 ng/mL. The lower limit of quantification for phenibut in rat brain extracts was 50 ng/mL. Acceptable precision and accuracy were obtained over the whole concentration range. 分析检测有机肥料中的兽药残留对于评价土壤微生物群落和人类健康的潜在风险至关重要。该文建立了一种QuEChERS联用高效液相色谱-串联质谱(HPLC-MS/MS)同时检测有机肥料中10种氟喹诺酮类药物残留的分析方法。样品经乙二胺四乙酸二钠(Na2EDTA)-Mcllvaine缓冲溶液(pH=4.0)和乙腈提取,采用Atlantis T3 C18色谱柱(250 mm×4.6 mm,5 μm),以0.2%(体积分数)甲酸水溶液和乙腈为梯度洗脱的流动相,采用电喷雾离子(ESI)源,在多反应监测(MRM)扫描模式下进行检测。10种氟喹诺酮类药物用内标法定量。结果表明,10种氟喹诺酮类药物在10~500 μg/kg范围内线性关系良好,相关系数均大于0.9930。方法检出限为0.5~2.5 μg/kg,定量限为1.7~8.3 μg/kg。该分析方法成功应用于9个有机肥样品的兽药污染调查。结果显示,样品的平均加标回收率为82.5%~117.1%,相对标准偏差为3.4%~10.2%。该方法准确可靠、灵敏度高,能够满足多种兽药残留的同时检测,该方法有望为有机肥料兽药风险评估提供依据。  相似文献

Chemicals are widely used in aquaculture and one of the main recipients of these analytes is the aquatic environment. The aim of this work was to develop and validate a simple and sensitive method for the determination of multiclass chemotherapeutic agents in farmed fish and shellfish using matrix solid‐phase dispersion and liquid chromatography‐tandem mass spectrometry. Residues of azamethiphos, three avermectins, two carbamates, and two benzoylureas were extracted from samples using silica gel as clean‐up adsorbent and 0.5% acetic acid in acetonitrile as elution solvent. The extraction conditions were investigated and optimized using an experimental design. Mass spectrometry detection was carried out in positive electrospray ionization mode with multiple‐reaction monitoring scan (except for benzoylurea family). Matrix‐matched standards were used for the drugs quantification. Good linearity (R2 ≥ 0.996) was observed in the range of 5–500 μg kg?1. Limits of detection were in the range of 1.5–3.7 μg kg?1. Recoveries from salmon samples spiked with veterinary drugs were in the range 84.9–118%. Precision was satisfactory since relative standard deviations were lower than 10.6%. The method can be successfully applied for the analysis of fish and shellfish from aquaculture.  相似文献

A simple sample treatment procedure and sensitive liquid chromatography–tandem mass spectrometry method were developed for the simultaneous quantification of the concentrations of human immunodeficiency virus‐1 integrase strand transfer inhibitors – raltegravir, dolutegravir and elvitegravir – in human plasma and cerebrospinal fluid (CSF). Plasma and CSF samples (20 μL each) were deproteinized with acetonitrile. Raltegravir‐d3 was used as the internal standard. Chromatographic separation was achieved on an XBridge C18 column (50 × 2.1 mm i.d., particle size 3.5 μm) using acetonitrile–water (7:3, v/v) containing 0.1% formic acid as the mobile phase at a flow rate of 0.2 mL/min. The run time was 5 min. Calibration curves for all three drugs were linear in the range 5–1500 ng/mL for plasma and 1–200 ng/mL for CSF. The intra‐ and inter‐day precision and accuracy of all three drugs in plasma were coefficient of variation (CV) <12.9% and 100.0 ± 12.2%, respectively, while those in CSF were CV <12.3% and 100.0 ± 7.9%, respectively. Successful validation under the same LC–MS/MS conditions for both plasma and CSF indicates this analytical method is useful for monitoring the levels of these integrase strand transfer inhibitors in the management of treatment of HIV‐1 carriers.  相似文献

A high‐throughput method based on ultrasonic‐assisted extraction, 96‐well plate thin‐film microextraction was established to determinate 18 antibiotics in animal feed. In this method, the extraction was implemented by ultrasonic‐assisted extraction for 30 min with disodium ethylenediaminetetraacetic acid‐McIlvaine buffer (pH 5) containing 6% sodium chloride w/v, purified by thin‐film microextraction and combined with 96‐well plate system to improve the efficiency. Optimization of thin‐film microextraction conditions was performed by methods of single factor and response surface, and finalized as: condition time: 20 min; adsorption time: 55 min; washing time: 5 s with water; desorption time: 30 min with acetonitrile/water (8:2, v/v) containing 0.1% formic acid v/v. Evaluation of different extractive phases showed that polystyrene‐divinylbenzene‐polyacrylonitrile was the optimum coating. The analysis was performed by ultra‐high performance liquid chromatography with tandem mass spectrometry. Recovery, inter‐ and intraday precision, linearity, limit of detection, and quantitation were evaluated. The average recoveries of 18 antibiotics were 66.6–93.5% at three spiked levels, intraday precision was 1–8.4%, and interday precision was 3.0–16.4%. The linearity was good for r> 0.99. Limits of detection and quantification were found in the range of 1–14 and 4–48 µg/kg, respectively.  相似文献

A simple, sensitive and reliable LC–MS/MS method was developed and validated for the quantification of anemoside B4, a potential antiviral constituent isolated from Pulsatilla chinensis in rat plasma, tissue, bile, urine and feces. All biological samples were prepared by protein precipitation method, and ginsenoside‐Rg1 was chosen as the internal standard (IS). The analyte and IS were separated using a C18 column (2.1 × 50 mm, 1.8 μm) and a mobile phase consisting of 0.1% formic acid in water (v /v) and acetonitrile running at a flow rate of 0.2 mL/min for 5 min. The multiple reaction monitoring transitions were monitored at m /z 1219.5–749.5 for anemoside B4 and 845.4–637.4 for ginsenoside‐Rg1 in electrospray ionization negative mode. The calibration curve was linear in the range of 10–2000 ng/mL for all biological matrices with a lower limit of quantification of 10 ng/mL. The validated method was successfully applied to a pharmacokinetics, tissue distribution and excretion study. These preclinical data will be beneficial for further development of anemoside B4 in future studies.  相似文献

A simple and selective high‐performance liquid chromatography method coupled with fluorescence detection was developed for the simultaneous measurement of trace levels of four estrogens (estrone, estradiol, estriol and 17α‐ethynyl estradiol) in environmental matrices. For feces samples, solid–liquid extraction was applied with a 1:1 v/v mixture of acetonitrile and ethyl acetate as the extraction solvent. For liquid samples (e.g., leachate and groundwater), hydrophobic/lipophilic balanced automated solid‐phase extraction disks were selected due to their high recoveries compared to conventional C18 disks. Chromatographic separations were performed on a reversed‐phase C18 column gradient‐eluted with a 45:55 v/v mixture of acetonitrile and water. The detection limits were down to 1.1 × 10?2 (estrone), 4.11 × 10?4 (estradiol), 5.2 × 10?3 (estriol) and 7.18 × 10?3 μg/L (17α‐ethynyl estradiol) at excitation/emission wavelengths of 288/310 nm, with recoveries in the range of 96.9 ± 3.2–105.4 ± 3.2% (n = 3). The method was successfully applied to determine estrogens in feces and water samples collected at livestock farms and a major river in Northeast China. We observed relatively high abundance and widespread distribution of all four estrogens in our sample collections, implying the urgency for a comprehensive and intricate investigation of estrogenic fate and contamination in our researched area.  相似文献

建立了同时测定水产品中64种兽药残留的超高效液相色谱-四极杆/静电场轨道阱高分辨质谱分析方法。样品经乙腈-水溶液(80∶20,v/v)提取,经乙腈饱和的正己烷和乙二胺-N-丙基硅烷(PSA)吸附剂净化。使用可加热电喷雾(HESI)离子源,全扫描/数据依赖二级扫描(Full MS/ddMS2)Top 1模式检测,外标法定量。结果表明,64种兽药在各自的浓度范围内线性关系良好(r2≥0.996 7),在3种水产品基质(鱼、虾、贝类)中3个不同浓度加标水平的平均回收率为56.2%~126.4%,相对标准偏差为1.3%~29.8%,方法的定量限为0.2~10μg/kg。该方法简便、准确、灵敏,适用于批量水产品中多种兽药残留的快速筛查。  相似文献

A simultaneous determination method using solid‐phase extraction and liquid chromatography with tandem mass spectrometry was developed to detect and quantify the presence of seven multiclass veterinary antibiotics (13 compounds in total) in surface water samples, which included the effluents of livestock wastewater and sewage treatment plants, as well as the reservoir drainage areas from dense animal farms. The pH of all water samples was adjusted to 2 or 6 before solid‐phase extraction using Oasis HLB cartridges. The developed method was fully validated in terms of linearity, method detection limit, method quantitation limit, accuracy, and precision. The linearity of all tested drugs was good, with R2 determination coefficients ≥ 0.9931. The method detection limits and method quantitation limits were 0.1–74.3 and 0.5–236.6 ng/L, respectively. Accuracy and precision values were 71–120 and 1–17%, respectively. The determination method was successfully applied for monitoring water samples obtained from the Yeongsan River in 2015. The most frequently detected antibiotics were lincomycin (96%), sulfamethazine (90%), sulfamethoxazole (88%), and sulfathiazole (50%); the maximum concentrations of which were 398.9, 1151.3, 533.1, and 307.4 ng/L, respectively. Overall, the greatest numbers and concentrations of detected antibiotics were found in samples from the effluents of livestock wastewater, sewage treatment plants, and reservoir drainage areas. Diverse veterinary antibiotics were present, and their presence was dependent upon the commercial sales and environmental properties of the analytes, the geographical positions of the sampling points, and the origin of the water.  相似文献

A rapid, simple and sensitive, liquid chromatography–tandem mass spectrometry (LC‐MS/MS) method was developed and validated for simultaneous determination of bergenin, chlorogenic acid and four flavonoids in a QingGanSanJie preparation in rat plasma. Puerarin was selected as the internal standard (IS). Plasma samples were precipitated with methanol and separated with a reverse phase Agilent Poroshell 120 EC‐C18 column using a gradient mobile phase of methanol–water containing 0.1% formic acid (v/v). A triple quadruple mass spectrometer was used for quantification (limit of detection 0.36–5.55 ng/mL). Intra‐day and inter‐day precisions were within 15% and the average extraction recoveries ranged from 85 to 115% for each analyte. The method allowed simultaneous quantification for the first time of the pharmacokinetics of bergenin, chlorogenic acid and four flavonoids after intragastric administration of a QingGanSanJie extract in Sprague–Dawley rats. It was found that bergenin and chlorogenic acid had typical extravascular administration concentration–time curves; flavonoids had a bimodal distribution improving bioavailability and extending the pharmacodynamics period. A rapid, selective, and sensitive liquid chromatography–tandem mass spectrometry method was developed and validated for the simultaneous determination of unbound sunitinib and its active metabolite N‐desethyl sunitinib in plasma. Plasma and post‐dialysis buffer samples were extracted using a liquid–liquid extraction procedure with acetonitrile–n‐butylchloride (1:4, v/v). Chromatographic separation was achieved on a Waters X‐Terra® MS RP18 column with a mobile phase consisting of acetonitrile and water (60:40, v/v) containing formic acid (0.1%, v/v) using an isocratic run, at a flow‐rate of 0.2 mL/min. Analytes were detected by electrospray tandem mass spectrometry in the selective reaction monitoring mode. Linear calibration curves were generated over the ranges 0.1–100 and 0.02–5 ng/mL for sunitinib and 0.2–200 and 0.04–10 ng/mL for N‐desethyl sunitinib in plasma and in phosphate‐buffered solution, respectively. The values for both within‐day and between‐day precision and accuracy were well within the generally accepted criteria for analytical methods. The analytical range was sufficient to determine the unbound and total concentrations of both analytes. The method was applied for measurement unbound concentrations in addition to total concentrations of sunitinib and its metabolite in plasma of a cancer patient receiving 50 mg daily dose. A multiclass, multiresidue determination method is reported for the detection of ten veterinary drugs, including scopolamine, metoclopramide, acriflavine, berberine, tripelennamine, diphenhydramine, acrinol, triamcinolone, loperamide, and roxithromycin in pork, milk, and eggs. The method involves a simple extraction using 0.1% formic acid in acetonitrile, followed by defatting with n‐hexane, centrifugation, and filtration prior to liquid chromatography with tandem mass spectrometric analysis. As ion suppression and enhancement effects are reported, matrix‐matched calibrations are used for quantification, with determination coefficients ≥0.9765. For the majority of the tested analytes, the intra‐ and interday accuracy (expressed as recovery %) range from 70.6 to 94.6% and from 70.1 to 93.3%, respectively, and the precision (expressed as relative standard deviation) ranges from 0.5 to 19.8% and from 2.8 to 18.4% in all matrices. The limits of quantification range between 0.5 and 10 ng/g. The validated tandem mass spectrometry method is successfully applied to market samples; the target analytes are not detected in any of the tested samples. In terms of accuracy, no extract cleanup is deemed necessary. The developed method is feasible for the simultaneous detection of the tested analytes in pork, milk, and eggs.  相似文献

A simple and rapid high‐performance liquid chromatography–tandem mass spectrometric assay for determination of paclitaxel on rat dried blood spots was developed and validated. The extracted sample was chromatographed without further treatment using a reverse‐phase Oyster ODS3, 4.6 × 50 mm, 3 µm column with mass spectrometry detection. The mobile phase comprised of acetonitrile–water, 60:40 v/v, with a flow rate of 0.4 mL/min was used. The calibration was linear over the range 0.2–20 ng/mL. The limits of detection and quantification were 0.08 and 0.2 ng/mL, respectively. The intra‐ and inter‐day precision (CV%) and accuracy (relative error %) were less than 10 and 12%, respectively. A sensitive and selective liquid chromatography–tandem mass spectrometry method for the determination of pethidine in human plasma was developed and validated over the concentration range of 4–2000 ng/mL. After addition of ketamine as internal standard, liquid–liquid extraction was used to produce a protein‐free extract. Chromatographic separation was achieved on a 100 × 2.1 mm, 5 µm particle, AllureTM PFP propyl column, with 45:40:15 (v/v/v) acetonitrile–methanol–water containing 0.2% formic acid as mobile phase. The MS data acquisition was accomplished by multiple reactions monitoring mode with positive electrospray ionization interface. The lower limit of quantification was 4 ng/mL; for inter‐day and intra‐day tests, the precision (RSD) for the entire validation was less than 7%, and the accuracy was within 95.9–106.5%.
