在308.15K下,研究了表面活性剂琥珀酸二(2-乙基己基)酯磺酸钠(Aerosol-Ot,简称AOT)的浓度和水的含量不同时,溶解的CO~2对反胶束溶解牛血清蛋白(BSA)的性质和异辛烷中AOT反胶束稳定性的影响。实验表明,在适当条件下,CO~2可以使反胶束溶液中的蛋白质全部析出。本研究对有关机理进行了初步分析。  相似文献

Sodium bis(2-ethylhexyl)sulfosuccinate (AOT) is a surfactant commonly used to encapsulate water soluble proteins within the aqueous core of a reverse micelle. In the context of high-resolution NMR studies of encapsulated proteins the size of the resulting reverse micelle is critically important. We have designed and synthesized a short AOT analogue, 3,3-dimethyl-1-butylsulfosuccinate sodium salt and determined that it is able to form reverse micelles and to encapsulate the protein ubiquitin with high structural fidelity. AOT is often found to significantly destabilize encapsulated proteins, largely through charge-charge interactions between the anionic headgroup and the surface of the protein. Here we demonstrate, for the first time, that proportional mixtures of anionic and cationic surfactants can form reverse micelles that are also capable of protein encapsulation with high fidelity.  相似文献

6-Propionyl-2-(N,N-dimethyl)aminonaphtahalene, PRODAN, is widely used as a fluorescent molecular probe due to its significant Stokes shift in polar solvents. It is an aromatic compound with intramolecular charge-transfer (ICT) states which can be particularly useful as sensors. In this work, we performed absorption, steady-state, time-resolved fluorescence (TRES), and time-resolved area normalized emission (TRANES) spectroscopies on PRODAN dissolved in nonaqueous reverse micelles. The reverse micelles are composed of polar solvents/sodium 1,4-bis-2-ethylhexylsulfosuccinate (AOT)/n-heptane. Sequestered polar solvents included ethylene glycol (EG), propylene glycol (PG), glycerol (GY), formamide (FA), dimethylformamide (DMF), and dimethylacetamide (DMA). The experiments were performed with varying surfactant concentrations at a fixed molar ratio W(S) = [polar solvent]/[AOT]. In every reverse micelle studied, the results show that PRODAN undergoes a partition process between the external solvent and the reverse micelle interface. The partition constants, K(p), are quantified from the changes in the PRODAN emission and/or absorption spectra with the surfactant concentration. The K(p) values depend strongly on the encapsulated polar solvent and correlate quite well with the AOT reverse micelle interface's zones where PRODAN can exist and emits. Thus, the partition toward the reverse micelle interface is strongly favored in DMF and DMA containing micelles where the PRODAN emission comes only from an ICT state. For GY/AOT reverse micelles, the K(p) value is the lowest and only emission from the local excited (LE) state is observed. On the other hand, for EG/AOT, PG/AOT, and water/AOT reverse micelles, the K(p) values are practically the same and emission from both states (LE and ICT) is simultaneously detected. We show here that it is possible to control the PRODAN state emission by simply changing the properties of the AOT reverse micelle interfaces by choosing the appropriate polar solvent to make the reverse micelle media. Indeed, we present experimental evidence with the answer to the long time question about from which state does PRODAN emit, a process that can be controlled using the unique reverse micelle interfaces properties.  相似文献

Polypyrrole (PPy) nanotubes were readily fabricated through chemical oxidation polymerization in sodium bis(2-ethylhexyl) sulfosuccinate (AOT) reverse (water-in-oil) emulsions. The reverse cylindrical micelle phase was characterized, and the key factors affecting the formation of PPy nanotubes were systematically inspected. AOT reverse cylindrical micelles were prepared via a cooperative interaction between an aqueous FeCl3 solution and AOT in an apolar solvent. In the H2O/FeCl3/AOT/apolar solvent system, the aqueous FeCl3 solution played a role in increasing the ionic strength and decreasing the second critical micelle concentration of AOT. As a result, AOT reverse cylindrical micelles could be spontaneously formed in an apolar solvent. In addition, iron cations were adsorbed to the anionic AOT headgroups that were capable of extracting metal cations from the aqueous core. Under these conditions, the addition of pyrrole monomer resulted in the chemical oxidation polymerization of the corresponding monomer at the surface of AOT reverse cylindrical micelles, followed by the formation of tubular PPy nanostructures. In a typical composition (74.0 wt % hexane, 22.4 wt % AOT, and 3.6 wt % aqueous FeCl3 solution at 15 degrees C), the average diameter of PPy nanotubes was approximately 94 nm and their length was more than 2 mum. The PPy nanotube dimensions were affected by synthetic variables such as the weight ratio of aqueous FeCl3 solution/AOT, type of apolar solvent, and reaction temperature. Moreover, the relationship between the diameter and the conductivity of the nanotubes was investigated.  相似文献

A wide investigation of the solubilization of the water-soluble salt Yb(NO3)3 in sodium bis(2-ethylhexyl)sulfosuccinate (AOT) reverse micelles and AOT liquid crystals has been carried out. After saturation of water/AOT/organic solvent w/o microemulsions with pure Yb(NO3)3, the Yb(NO3)3/AOT composites were prepared by complete evaporation under vacuum of the volatile components (water and organic solvent) of the salt-containing microemulsions. It was observed that these composites can be totally dissolved in pure n-heptane or CCl4, allowing the solubilization of a noticeable amount of Yb(NO3)3 in quite dry apolar media. By UV–vis–NIR, FT-IR, and 1H NMR spectroscopies, some information on the state of Yb(NO3)3 within AOT reverse micelles were acquired, whereas by small angle X-ray scattering (SAXS), it has been ascertained that Yb(NO3)3 is quite homogeneously distributed as very small clusters among the reverse micelles. An analysis of SAXS and wide-angle X-ray scattering spectra of Yb(NO3)3/AOT composites leads to the hypothesis that, also in these systems, Yb(NO3)3 is dispersed in the surfactant matrix as very small clusters.  相似文献

Electrochemical extraction of proteins by reverse micelle formation   总被引:1,自引:0,他引:1  
The transfer of proteins by the anionic surfactant bis(2-ethylhexyl) sulfosuccinate (AOT) at a polarized 1,2-dichloroethane/water (DCE/W) interface was investigated by means of ion-transfer voltammetry. When the tetrapentylammonium salt of AOT was added to the DCE phase, the facilitated transfer of certain proteins, including cytochrome c (Cyt c), ribonuclease A, and protamine, could be controlled electrochemically, and a well-defined anodic wave for the transfer was obtained. At low pH values (e.g., pH 3.4), the anodic wave was usually well-separated from the wave for the formation of protein-free (i.e., unfilled) reverse micelles. The anodic wave for the protein transfer was analyzed by applying the theory for facilitated transfer of ions by charged ligands and then supplying information regarding the number of AOT anions reacting with one protein molecule and the total charge carried by the protein transfer. However, controlled-potential electrolyses performed for the transfer of Cyt c, which is red, revealed that the protein-AOT complexes were unstable in DCE and liable to aggregate at the interface when the pH of the W phase was 3.4. At pH 7.0, when formation of unfilled reverse micelles occurred simultaneously, the protein-AOT complexes appeared to be stabilized, probably via fusion with unfilled reverse micelles.  相似文献

The effect of dissolved CO(2) and CHF(3) on the precipitation of bovine serum albumin (BSA) from reverse micelles of sodium bis(2-ethylhexyl) sulfosuccinate (AOT) in 2,2,4-trimethylpentane (iso-octane) has been studied at 308.15 K. It was found that BSA can be precipitated completely by CO(2) while the AOT and water remain in the iso-octane continuous phase. This opens up a possibility for recovery of protein solids directly from reverse micellar solutions. CHF(3) is also an effective agent for precipitation of BSA in the reverse micelles when the expansion coefficient of CHF(3) exceeds 0.4. However, the condition required for complete removal of the BSA using CHF(3) results in some precipitation of AOT and water from the solution. Copyright 2000 Academic Press.  相似文献

Dry reverse micelles of AOT in isooctane spontaneously undergo a microstructural transition to an organogel upon the addition of a phenolic dopant, p-chlorophenol. This microstructural evolution has been studied through a combination of light scattering, small-angle neutron scattering (SANS), NMR, and rheology. Several equilibrium stages between the system of dry reverse micelles of AOT and a 1:1 AOT/p-chlorophenol (molar ratio) gel in isooctane have been examined. To achieve this, p-chlorophenol is added progressively to the dilute solutions of AOT in isooctane, and this concentration series is then analyzed. The dry micelles of AOT in isooctane do not undergo any detectable structural change up to a certain p-chlorophenol concentration. Upon a very small increment in the concentration of p-chlorophenol beyond this "threshold" concentration, large strandlike aggregates are observed which then evolve to the three-dimensional gel network.  相似文献

The activity and stability of Chromobacterium viscosum lipase (glycerolester hydrolase, EC 3.1.1.3)-catalyzed olive oil hydrolysis in sodium bis (2-ethyl-1-hexyl)sulfosuccinate (AOT)/isooctane reverse micelles is increased appreciably when low molecular weight polyethylene glycol (PEG 400) is added to the reverse micelles. To understand the effect of PEG 400 on the phase behavior of the reverse micellar system, the phase diagram of AOT/PEG 400/water/isooctane system was studied. The influences of relevant parameters on the catalytic activity in AOT/PEG 400 reverse micelles were investigated and compared with the results in the simple AOT reverse micelles. In the presence of PEG 400, the linear decreasing trend of the lipase activity with AOT concentration, which is observed in the simple AOT reverse micelles, disappeared. Enzyme entrapped in AOT/PEG reverse micelles was very stable, retaining>75% of its initial activity after 60 d, whereas the half-life in simple AOT reverse micelles was 38 d. The kinetics parameter maximum velocity (V max)exhibiting the temperature dependence and the activation energy obtained by Arrhenius plot was suppressed significantly by the addition of PEG 400.  相似文献

The interior water pool of aerosol OT (AOT) reverse micelles tends toward bulk water properties as the micelle size increases. Thus, deviations from bulk water behavior in large reverse micelles are less expected than in small reverse micelles. Probing the interior water pool of AOT reverse micelles with a highly charged decavanadate (V(10)) oligomer using (51)V NMR spectroscopy shows distinct changes in solute environment. For example, when an acidic stock solution of protonated V(10) is placed in a reverse micelle, the (51)V chemical shifts show that the V(10) is deprotonated consistent with a decreased proton concentration in the intramicellar water pool. Results indicate that a proton gradient exists inside the reverse micelles, leaving the interior neutral while the interfacial region is acidic.  相似文献

Dynamic light scattering and NMR spectroscopic experimental evidence suggest the coexistence of two compositionally different self-assembled particles in solution. The self-assembled particles form in solutions containing water, Aerosol OT (AOT, sodium bis(2-ethylhexyl) sulfosuccinate) surfactant, and cholesterol in cyclohexane. In a similar series of studies carried out in 1-octanol only one aggregate type, that is, reverse micelles, is observed. Dynamic light scattering measurements reveal the presence of two different types of aggregates in the microemulsions formed in cyclohexane, demonstrating the coexistence of two compositionally distinct structures with very similar Gibbs energies. One particle type consists of standard AOT reverse micelles while the second type of particle consists of submicellar aggregates including cholesterol as well as small amounts of AOT and water. In microemulsions employing 1-octanol as the continuous medium, AOT reverse micelles form in a dispersed solution of cholesterol in 1-octanol. Although the size distribution of self-assembled particles is well-known for many different systems, evidence for simultaneous formation of two distinctly sized particles in solution that are chemically different is unprecedented. The ability to form microemulsion solutions that contain coexisting particles may have important applications in drug formulation and administration, particularly as applied to drug delivery using cholesterol as a targeting agent.  相似文献

The micropolarity of sodium bis(2-ethylhexyl) sulfosuccinate (AOT) reverse micelles prepared in supercritical ethane and near-critical propane has been determined in terms of a solvent polarity parameter, E T(30) values, by using absorption probes, 1-ethyl-4-methoxycarbonyl pyridinium iodide and 2,6-diphenyl-4-(2,4,6-triphenylpyridinio)-phenolate as a functions of pressure and the molar ratio of water to AOT, W 0, at a constant temperature of 310 K. The micropolarity comparable to that of ethanol was observed for reverse micelles containing water of W 0 = 2. The micropolarity increased with the water content and became independent of pressure after the system changed to a one-phase reverse micelle solution. For a given W 0 value, no difference in the micropolarity was noticed in the micelles prepared in ethane and propane. Phase behaviour investigations have revealed that complete dissolution of 50 mM AOT occurred at 20 MPa in supercritical ethane, while a much lower pressure of 1 MPa was required in near-critical propane. The amount of water solubilized in reverse micelles formed in supercritical ethane was relatively low, reaching a W 0 value of 7 at 36 MPa. In contrast, the amount of water solubilized in near-critical propane reverse micelles was W 0 = 11 at a much lower pressure of 6 MPa. A higher pressure was required to solubilize larger amount of water in reverse micelles prepared in both ethane and propane. The refolding kinetics of the reduced, denatured hen egg white lysozyme in sodium bis(2-ethylhexyl)sulfosuccinate (AOT)-isooctane-water reverse micelles at different water-to-surfactant molar ratios has been investigated by fluorescence spectroscopy and UV spectroscopy. The oxidative refolding of the confined lysozyme is biphasic in AOT reverse micelles. When the water-to-surfactant molar ratio (omega 0) is 12.6, the relative activity of encapsulated lysozyme after refolding for 24 h in AOT reverse micelles increases 46% compared with that in bulk water. Furthermore, aggregation of lysozyme at a higher concentration (0.2 mM) in AOT reverse micelles at omega 0 of 6.3 or 12.6 is not observed; in contrast, the oxidative refolding of lysozyme in bulk water must be at a lower protein concentration (5 microM) in order to avoid a serious aggregation of the protein. For comparison, we have also investigated the effect of AOT on lysozyme activity and found that the residual activity of lysozyme decreases with increasing the concentration of AOT from 1 to 5 mM. When AOT concentration is larger than 2 mM, lysozyme is almost completely inactivated by AOT and most of lysozyme activity is lost. Together, our data demonstrate that AOT reverse micelles with suitable water-to-surfactant molar ratios are favorable to the oxidative refolding of reduced, denatured lysozyme at a higher concentration, compared with bulk water.  相似文献

The recombination of thiocyanate anion radicals, (SCN) 2 , formed pulse radiolytically within the water pools of reverse micelles stabilized with anionic AOT and nonionic Igepal surfactants, was proved as an indicator reaction to study intermicellar exchange. It was found that the exchange process is slower inIgepal than in AOT reverse micelles with the same water to surfactant ratio. The apparent activation enthalpy and entropy of the exchange process were determined in different alkanes. For the AOT and Igepal reverse micelles the activation parameters increase with the droplet size, but for the AOT systems they do not significantly change with the increase of droplet concentration. For non-percolated systems the activation parameters for Igepal reverse micelles approach those for AOT reverse micelles. This result supports existing suggestions that the mechanism of intermicellar exchange does not differ in principle between reverse micelles stabilized with ionic and nonionic surfactants.  相似文献

Reverse micelles formed by sodium bis(2-ethylhexyl) sulfosuccinate (AOT) in isooctane (IO) and water have long been used as a means to provide a confined aqueous environment for various applications. In particular, AOT reverse micelles have often been used as a template to mimic membrane-water interfaces. While earlier studies have shown that membrane-binding peptides can indeed be incorporated into the polar cavity of AOT reverse micelles where they mostly fold into an alpha-helical structure, the underlying interactions leading to the ordered conformation are however not well understood. Herein, we have used circular dichroism (CD) and infrared (IR) spectroscopies in conjunction with a local IR marker (i.e., the CN group of a non-natural amino acid, p-cyano-phenylalanine) and a global IR reporter (i.e., the amide I' band of the peptide backbone) to probe the conformation as well as the hydration status of an antimicrobial peptide, mastoparan x (MPx), in AOT reverse micelles of different water contents. Our results show that at, w0=6, MPx adopts an alpha-helical conformation with both the backbone and hydrophobic side chains mostly dehydrated, whereas its backbone becomes partially hydrated at w0=20. In addition, our results suggest that the amphipathic alpha-helix so formed orients itself in such a manner that its positively charged, lysine-rich, hydrophilic face points toward the negatively charged AOT head groups, while its hydrophobic face is directed toward the polar interior of the water pool. This picture is in marked contrast to that observed for the binding of MPx to phospholipid bilayers wherein the hydrophobic surface of the bound alpha-helix is buried deeper into the membrane interior.  相似文献

The effect of compressed CO2 on the solubilization of bovine serum albumin (BSA) in water/sodium bis-(2-ethylhexyl) sulfosuccinate (AOT)/isooctane reverse micelles was studied by observing phase behavior and recording UV-visible spectra under different conditions. The pH values within the water cores of reverse micelles at different CO2 pressures were also determined. The solubilization capacity of the reverse micelles for the protein increased considerably as CO2 pressure increased within the low-pressure range, but decreased at higher CO2 pressures, so that the micelles eventually lost their ability to solubilize the protein. The effect of CO2 on the stability of the reverse micelles played an important role in the relationship between pressure and protein solubility. A "multicomplex" model was proposed to explain these effects. The different solublization capacities within different pressure ranges demonstrates the unique advantage of using compressed CO2 in the extraction of proteins with reverse micelles.  相似文献

We report a study on encapsulation of various amino acids into gas-phase sodium bis(2-ethylhexyl) sulfosuccinate (NaAOT) reverse micelles, using electrospray ionization guided-ion-beam tandem mass spectrometry. Collision-induced dissociation of mass-selected reverse micellar ions with Xe was performed to probe structures of gas-phase micellar assemblies, identify solute-surfactant interactions, and determine preferential incorporation sites of amino acids. Integration into gas-phase reverse micelles depends upon amino acid hydrophobicity and charge state. For examples, glycine and protonated amino acids (such as protonated tryptophan) are encapsulated within the micellar core via electrostatic interactions; while neutral tryptophan is adsorbed in the surfactant layer. As verified using model polar hydrophobic compounds, the hydrophobic effect and solute-interface hydrogen-bonding do not provide sufficient driving force needed for interfacial solubilization of neutral tryptophan. Neutral tryptophan, with a zwitterionic structure, is intercalated at the micellar interface between surfactant molecules through complementary effects of electrostatic interactions between tryptophan backbone and AOT polar heads, and hydrophobic interactions between tryptophan side chain and AOT alkyl tails. Protonation of tryptophan could significantly improve its incorporation capacity into gas-phase reverse micelles, and displace its incorporation site from the micellar interfacial zone to the core; protonation of glycine, on the other hand, has little effect on its encapsulation capacity. Another interesting observation is that amino acids of different isoelectric points could be selectively encapsulated into, and transported by, reverse micelles from solution to the gas phase, based upon their competition for protonation and subsequent encapsulation within the micellar core.  相似文献

Water-soluble proteins encapsulated within reverse micelles may be studied under a variety of conditions, including low temperature and a wide range of buffer conditions. Direct high-resolution detection of information relating to protein folding intermediates and pathways can be monitored by low-temperature solution NMR. Ubiquitin encapsulated within AOT reverse micelles was studied using multidimensional multinuclear solution NMR to determine the relationship between protein structure, temperature, and ionic strength. Ubiquitin resonances were monitored by 15N HSQC NMR experiments at varying temperatures and salt concentrations. Our results indicate that the structure of the encapsulated protein at low temperature experiences perturbation arising from two major influences, which are reverse micelle-protein interactions and low-temperature effects (e.g., cold denaturation). These two effects are impossible to distinguish under conditions of low ionic strength. Elevated concentrations of nondenaturing salt solutions defeat the effects of reverse micelle-protein interactions and reveal low-temperature protein unfolding. High ionic strength shielding stabilizes the reverse micelle at low temperatures, which reduces the electrostatic interaction between the protein and reverse micelle surfaces, allowing the phenomenon of cold denaturation to be explored.  相似文献
