Nucleophilic addition of alkyl- and benzylthiols to benzoquinone diimine (1) gave the corresponding 3-alkylthio- or 3-benzylthio-1,4-phenylenediamines (2-5). However, addition of aryl- or heteroarylthiols to 1 formed 2-arylthio- or 2-heteroarylthio-1,4-phenylenediamines (6-14). The structures of 2-14, obtained in 55-91% yields, were confirmed in CDCl3 or DMSO-d6 solution using 1D (NOE difference, coupled 13C NMR spectra, APT and DEPT) and 2D NMR techniques [DQCOSY, NOESY, HETCOR and heteronuclear multiple bond coherence (HMBC)] that resulted in unambiguous proton and carbon NMR resonance assignments. The substituent-induced 13C NMR chemical shift differences were calculated in 2-14 relative to carbon atoms in the model compound N1-(1,3-dimethylbutyl)-N4-phenyl-1,4-phenylenediamine (DMBPPD) (15) (a reduced form of benzoquinone diimine).  相似文献

(1)H and (13)C NMR data for N-substituted morpholines 1-20 were measured using 1D (DEPT, 1D NOE difference) and 2D NMR spectroscopic methods including (1)H-(1)H COSY, long-range (1)H-(1)H COSY, NOESY, gHMBC and gHMQC experiments. At room temperature the (1)H NMR spectra of protonated compounds 2 and 9 show the chair conformation for the morpholine ring. Spin-spin coupling constants were deduced from the resolution-enhanced proton spectra.  相似文献

The 1H and 13C NMR spectra of compounds 1-11 and 16-22 in CDCl3 and DMSO-d6 solutions allowed structural assignment to regioisomers 1/5 and 2/6 and their regioselective cyclization products 16-18 utilizing one- and two-dimensional NMR techniques (APT, DEPT, NOE difference, COSY, NOESY, HETCOR and gHMQC, gHMBC). Temperature-dependent 1H NMR spectra of 8-anilino-5-(4-methyl-2-pentyl)-2,3-dihydro-1,5-benzothiazepin-4(5H)-one (18) indicated a free energy of activation (deltaG++) of ca 17 kcal mol(-1) for interconversion between rotamers. The 1H and 13C NMR spectra of 20 and 22 containing two chiral centers exhibit duplication of several signals, indicating the existence of two diastereomeric forms. The structure of 4 was unambiguously confirmed by x-ray crystallography.  相似文献

Compounds 1-7 form a novel group of dithiocarbamates, first synthesized from the reaction of a series of primary amines with carbon disulfide and 3-bromo ethyl pyruvate in the presence of anhydrous potassium phosphate. Structure elucidation of this group of compounds was accomplished using extensive 1D and 2D NMR spectroscopic studies, including (1)H, (13)C, COSY, NOESY, HSQC, and gHMBC experiments. The distinction between the linear structures I, II and the cyclic structure III was made mainly on the basis of the analysis of the cross peak between H-2 and H-4a in the COSY spectra, in combination with the long-range correlation between H-2 and C-4, 6 in the gHMBC spectra.  相似文献

The structure elucidations and complete (1)H and (13)C NMR assignments are reported for two new natural products: 3-benzylidene-8,8a-dihydroxy-2-methyl-hexahydro-pyrrolo[1,2-a]pyrazine-1,4-dione(1) and 4-hydroxy-6-(hydroxy-phenyl-methyl)-N-(3-methyl-butyryl)-nicotinamide (2). Both of these secondary metabolites were isolated from the fermentation medium of a Mangrove endophytic fungus. High resolution electron impact mass spectrometry (HREIMS), FT-IR Spectroscopy and NMR experiments including gCOSY, gHMQC, gHMBC and NOE were used for determination of the structures and assignments of the amide alkaloids.  相似文献

The structure of 3beta-hydroxyballotinone, a new labdane diterpenoid isolated from Ballota undulata, has been established by NMR spectroscopic studies. In addition, complete and unambiguous assignments of the (1)H and (13)C NMR spectra of three other already known labdanes (ballotinone, ballonigrin and ballonigrinone) isolated from the same source have been achieved. The assignments are based on 2D shift-correlated (1)H--(1)H COSY, (1)H--(13)C gHSQC [(1)J(C,H)] and (1)H--(13)C gHMBC [(n)J(C,H) (n = 2 and 3)], and NOE experiments.  相似文献

Three new dihydrophenanthrofurans, pleionesins A–C (1–3), together with two known dihydrophenanthrenes (4–5) were isolated from the tubers of Pleione yunnanensis (Rolfe). The complete 1H and 13C NMR spectra assignments of these compounds were carried out using 1D and 2D NMR experiments (1H, 13C, selective 1D NOE, HSQC and HMBC). Three monocyclic polyether dilactams, 17,18-dihydro-5H, 9H-dibenzo[e,n]1,4,10,7,13trioxadiazacyclopentadecine-6,10(7H,11H)-dione (1); 9,10,20,21-tetrahydro-5H, 12H-dibenzo[e,q]1,4,10,13,7,16tetraoxadiazacyclooctadecine-6, 13(7H,14H)-dione (2); and 6,7,9,10-tetrahydro-16H, 20H-dibenzo[h,q]1,4,7,13, 10,16tetraoxadiazacyclooctadecine-17, 21(18H,22H)-dione (3) were isolated during the synthesis of several benzoannelated cryptands. The complete assignments of the 1H and 13C NMR spectra of 1, 2 and 3 in CDCl3 were made using gCOSY, gHMBC, gHMQC, HMQC, HSQC, and NOESY 1D techniques. The ortho (H2) benzene protons show significant downfield shifts (1.16-1.43 ppm) that are consistent with an exodentate orientation for the amide carbonyl groups. The X-ray crystal structures of 1, 2 and 3 show that the carbonyl groups adopt an exodentate conformation in the solid state.  相似文献

The temperature-dependent (1)H and (13)C NMR spectra of 2-(2-butynyl)-10-methyl-1,2,3,4-tetrahydropyrazino[1,2-a]indole (4) (as a representative example of 1-9) in CFCl(3) + CD(2)Cl(2) solution are described and discussed. Below 183 K, the hexahydropyrazine ring inversions become slow on the NMR time-scale and 4 exists in principle as two conformational diastereomers. In fact, only one was observed with the N-2 substituent in an equatorial position as shown by a low-temperature NOESY experiment. The energy barrier for conformational interchange was calculated from NMR data to be 8.3 kcal mol(-1) (1 kcal = 4.184 kJ), in agreement with quantum chemical calculations. Unambiguous assignments for all proton and carbon resonances of 1-9 were made using 1D (APT, DEPT, NOE difference) and 2D (COSY, NOESY, gHMQC, gHMBC) NMR techniques.  相似文献

We report the unambiguous assignments of the (1)H and (13)C NMR spectra of two new natural products, namely, 1,4,5,6,7,9-hexahydroxy-2-methoxy-7-methyl-5beta,9beta,8abeta, 6alpha,10aalpha-hexahydroanthracen-10 (10aH)-one (1) and 1,4,6-trihydroxy-2-methoxy-7-methylanthracene-9, 10-dione (2), together with three known anthraquinones. These compounds were all isolated from the marine endophytic fungus No. 1403 collected from the South China Sea. Compounds 3 and 4 were isolated from the marine fungus for the first time. The structures were elucidated by the spectroscopic methods 1D and 2D NMR including COSY, HMQC, HMBC and NOE, and HREIMS. In our cytotoxicity assays, compound 5 showed cytotoxicity toward KB and KBv-200 cells with IC(50) of 1.40 and 2.58 microg/ml, respectively. In addition, the plausible biogenic relationship of compounds 1, 2, 3 and 4 is discussed.  相似文献

An NMR study of 11 naturally occurring abietane diterpenoids is described. In addition to one‐dimensional NMR methods, including DPFGSE 1D‐NOE spectra, two‐dimensional shift‐correlated experiments [1H,1H COSY, 1H,13C‐gHSQC 1J(C,H) and 1H,13C‐gHMBC nJ(C,H) (n = 2 and 3)] were used for the complete and unambiguous 1H and 13C chemical shift assignments of these substances. The structures of three benzylidenebenzyl butyrolactone lignans (gossypifan, carthamogenin, and savinin) have been established on basis of 1H NMR and 13C NMR spectroscopic data. The 1H NMR and 13C NMR spectra of these lignans have been fully assigned by the use of techniques such as gCOSY, non-edited gHSQC, and gHMBC. Complete assignment and most homonuclear hydrogen coupling constant measurements were performed, also providing enough data for the determination of the relative stereochemistry.  相似文献

Both of these secondary metabolites were isolated from the fermentation culture of the endophytic fungus Xylaria sp. We report the unambiguous assignments of the (1)H and (13)C NMR spectra of one new natural product, namely, 6,8-di-O-methyl versiconol (1) together with one known anthraquinone aversin (2) and two xanthones 5-methoxysterigmatocystin (3) and sterigmatocystin (4). These compounds were all isolated from the mangrove endophytic fungus ZSUH-36 from the South China Sea. 1D and 2D NMR experiments including COSY, HMQC and HMBC were used to elucidate the structures. Variations in the (1)H NMR spectrum of 6,8-di-O-methyl versiconol (1) were also observed in the temperature range 25-75 degrees C. Bioactive cage‐like polycyclic compounds have attracted the attention of several research groups because of their unique appearance and their biological activities. Their structures were established on the basis of 1H NMR and 13C NMR spectroscopic data. The 1H and 13C signal assignments and most homonuclear hydrogen coupling constants were assigned by use of techniques such as 1D 1H and 13C NMR and 2D gCOSY, non‐edited gHSQC and gHMBC. The 1H and 13C NMR spectra of methyl-8-(2-furyl)-5-methyl-1,3-dioxo-3,3a,4,6,8,9,9a,9b-octahydrofuro[3,4-f]isoquinoline-7(1H)-carboxylate (1) and trimethyl 8-methyl-3-phenyl-3,4,4a,7-tetrahydro-2,5,6(1H)-isoquinolinetricarboxylate (2) at room temperature displayed doubling of the majority of signals, suggesting the presence of two rotational conformers (rotamers) in a ratio approximately 1:1.2 (in a mixture of CDCl3 and C6D6), approximately 1:1 (in CD2Cl2) and approximately 1:1.4 (in C6D6). On the basis of the temperature-dependent 1H NMR spectra of 1 and 2, the barrier to interconversion of the rotamers was calculated to be approximately 16 kcal mol(-1). The average chemical shifts and spin-spin coupling constants were analyzed for the resolution-enhanced 300 MHz 1H NMR spectrum of 1 at 333 K in CDCl3 solution. From analysis of the spin-spin coupling constants, it is proposed that the nitrogen-containing ring is in a chair conformation with C-2-H equatorial. Low- and room-temperature NOESY experiments in conjunction with theoretical ab initio calculations supported the hypothesis that the two rotamers interchange via rotation about the C-N bond. Spectral assignments of all proton and carbon resonances were made on the basis of one- and two-dimensional NMR experiments (DEPT, DQCOSY, NOESY, HETCOR and gHMBC).  相似文献
