Methyl 2‐acetamido‐2‐deoxy‐β‐d ‐glucopyranoside (β‐GlcNAcOCH3), (I), crystallizes from water as a dihydrate, C9H17NO6·H2O, containing two independent molecules [denoted (IA) and (IB)] in the asymmetric unit, whereas the crystal structure of methyl 2‐formamido‐2‐deoxy‐β‐d ‐glucopyranoside (β‐GlcNFmOCH3), (II), C8H15NO6, also obtained from water, is devoid of solvent water molecules. The two molecules of (I) assume distorted 4C1 chair conformations. Values of ϕ for (IA) and (IB) indicate ring distortions towards BC2,C5 and C3,O5B, respectively. By comparison, (II) shows considerably more ring distortion than molecules (IA) and (IB), despite the less bulky N‐acyl side chain. Distortion towards BC2,C5 was observed for (II), similar to the findings for (IA). The amide bond conformation in each of (IA), (IB) and (II) is trans, and the conformation about the C—N bond is anti (C—H is approximately anti to N—H), although the conformation about the latter bond within this group varies by ∼16°. The conformation of the exocyclic hydroxymethyl group was found to be gt in each of (IA), (IB) and (II). Comparison of the X‐ray structures of (I) and (II) with those of other GlcNAc mono‐ and disaccharides shows that GlcNAc aldohexopyranosyl rings can be distorted over a wide range of geometries in the solid state.  相似文献

The hexo­pyran­osyl ring of the title compound, [Hg(CH3)(C9H15O7S)], adopts the 4C1 chair conformation, and the anomeric configuration of the thio­methyl­mercury linkage is β. The compound exists as two symmetry‐independent conformers, A and B, within the unit cell, and each shows an almost linear S—Hg—C arrangement. Most of the bond distances and angles in A and B are similar, although a marked difference exists in the side‐chain conformation. Weak secondary intramolecular (between Hg and ring O) and intermolecular (between A and B conformers) interactions are documented.  相似文献

In the title compound, C23H34O4, which is an intermediate in the synthesis of pregnane derivatives with a modified skeleton that show potent abortion‐inducing activity, the conformation of ring B is close to half‐chair due to the presence of both the C=C double bond and the axial 5β‐methyl group. Rings A and C have conformations close to chair, while ring D has a twisted conformation around the bridgehead C—C bond. Molecules are hydrogen bonded via the hydroxyl and acetoxy groups into infinite chains. Quantum‐mechanical ab initio Roothan Hartree–Fock calculations show that crystal packing might be responsible for the low values of the angles between rings A and B, and between ring A and rings C and D, as well as for a different steric position of the methyl ketone side chain compared to the geometry of the free molecule.  相似文献

The crystal structure of the title compound [systematic name: 2‐(1,3‐dithiolo[4,5‐b][1,4]dithiin‐2‐ylidene)‐6,6‐bis(methoxyethoxymethoxymethyl)‐1,3‐dithiolo[4,5‐b][1,4]dithiepine], C21H30O6S8, a spiro‐substituted BEDT–TTF analogue [BEDT–TTF is bis(ethylenedithio)tetrathiafulvalene], has a strongly bent heterocyclic framework. The seven‐membered ring adopts a pseudo‐chair conformation with notably widened ring bond angles, especially at the methylene C atoms [119.49 (11) and 117.60 (11)°]. The axial side chain adopts an extended conformation, but the equatorial side chain curls back on itself and the O atom nearest the ring system is involved in three short contacts to H atoms (2.45–2.53 Å). The molecules pack in centrosymmetrically related pairs, which are isolated from each other by columns of the polyether side chains. This study emphasizes the ease of distortion of the neutral bis(propylenedithio)tetrathiafulvalene ring structure, and how the need to accommodate side chains can easily override the tendency of these donor systems to form stacks in the crystalline state.  相似文献

Methyl β‐D‐mannopyranosyl‐(1→4)‐β‐D‐xylopyranoside, C12H22O10, (I), crystallizes as colorless needles from water, with two crystallographically independent molecules, (IA) and (IB), comprising the asymmetric unit. The internal glycosidic linkage conformation in molecule (IA) is characterized by a ϕ′ torsion angle (O5′Man—C1′Man—O1′Man—C4Xyl; Man is mannose and Xyl is xylose) of −88.38 (17)° and a ψ′ torsion angle (C1′Man—O1′Man—C4Xyl—C5Xyl) of −149.22 (15)°, whereas the corresponding torsion angles in molecule (IB) are −89.82 (17) and −159.98 (14)°, respectively. Ring atom numbering conforms to the convention in which C1 denotes the anomeric C atom, and C5 and C6 denote the hydroxymethyl (–CH2OH) C atom in the β‐Xylp and β‐Manp residues, respectively. By comparison, the internal glycosidic linkage in the major disorder component of the structurally related disaccharide, methyl β‐D‐galactopyranosyl‐(1→4)‐β‐D‐xylopyranoside), (II) [Zhang, Oliver & Serriani (2012). Acta Cryst. C 68 , o7–o11], is characterized by ϕ′ = −85.7 (6)° and ψ′ = −141.6 (8)°. Inter‐residue hydrogen bonding is observed between atoms O3Xyl and O5′Man in both (IA) and (IB) [O3Xyl...O5′Man internuclear distances = 2.7268 (16) and 2.6920 (17) Å, respectively], analogous to the inter‐residue hydrogen bond detected between atoms O3Xyl and O5′Gal in (II). Exocyclic hydroxymethyl group conformation in the β‐Manp residue of (IA) is gauche–gauche, whereas that in the β‐Manp residue of (IB) is gauche–trans.  相似文献

3‐Deoxy‐3‐fluoro‐d ‐glucopyranose crystallizes from acetone to give a unit cell containing two crystallographically independent molecules. One of these molecules (at site A) is structurally homogeneous and corresponds to 3‐deoxy‐3‐fluoro‐β‐d ‐glucopyranose, C6H11FO5, (I). The second molecule (at site B) is structurally heterogeneous and corresponds to a mixture of (I) and 3‐deoxy‐3‐fluoro‐α‐d ‐glucopyranose, (II); treatment of the diffraction data using partial‐occupancy oxygen at the anomeric center gave a high‐quality packing model with an occupancy ratio of 0.84:0.16 for (II):(I) at site B. The mixture of α‐ and β‐anomers at site B appears to be accommodated in the lattice because hydrogen‐bonding partners are present to hydrogen bond to the anomeric OH group in either an axial or equatorial orientation. Cremer–Pople analysis of (I) and (II) shows the pyranosyl ring of (II) to be slightly more distorted than that of (I) [θ(I) = 3.85 (15)° and θ(II) = 6.35 (16)°], but the general direction of distortion is similar in both structures [ϕ(I) = 67 (2)° (BC1,C4) and ϕ(II) = 26.0 (15)° (C3TBC1); B = boat conformation and TB = twist‐boat conformation]. The exocyclic hydroxymethyl (–CH2OH) conformation is gg (gauchegauche) (H5 anti to O6) in both (I) and (II). Structural comparisons of (I) and (II) to related unsubstituted, deoxy and fluorine‐substituted monosaccharides show that the gluco ring can assume a wide range of distorted chair structures in the crystalline state depending on ring substitution patterns.  相似文献

The title compound, C23H32O4, has a 3β configuration, with the epoxy O atom at 16α,17α. Rings A and C have slightly distorted chair conformations. Because of the presence of the C5=C6 double bond, ring B assumes an 8β,9α‐half‐chair conformation slightly distorted towards an 8β‐sofa. Ring D has a conformation close to a 14α‐envelope. The acetoxy and acetyl substituents are twisted with respect to the average molecular plane of the steroid. The conformation of the mol­ecule is compared with that given by a quantum chemistry calculation using the RHF–AM1 (RHF = Roothaan Hartree–Fock) Hamiltonian model. Cohesion of the crystal can be attributed to van der Waals interactions and weak intermolecular C—H?O interactions, which link the mol­ecules head‐to‐tail along [101].  相似文献

The fact that molecular crystals exist as different polymorphic modifications and the identification of as many polymorphs as possible are important considerations for the pharmaceutic industry. The molecule of N‐benzyl‐4‐hydroxy‐1‐methyl‐2,2‐dioxo‐1H‐2λ6,1‐benzothiazine‐3‐carboxamide, C17H16N2O4S, does not contain a stereogenic atom, but intramolecular hydrogen‐bonding interactions engender enantiomeric chiral conformations as a labile racemic mixture. The title compound crystallized in a solvent‐dependent single chiral conformation within one of two conformationally polymorphic P212121 orthorhombic chiral crystals (denoted forms A and B). Each of these pseudo‐enantiomorphic crystals contains one of two pseudo‐enantiomeric diastereomers. Form A was obtained from methylene chloride and form B can be crystallized from N,N‐dimethylformamide, ethanol, ethyl acetate or xylene. Pharmacological studies with solid–particulate suspensions have shown that crystalline form A exhibits an almost fourfold higher antinociceptive activity compared to form B.  相似文献

In the title compound, [Cu(C15H20N2O4)]n, the copper(II) coordination is square planar. The anionic l ‐tyrosyl‐l ‐leucinate ligand binds in an N,N′,O‐tridentate mode to one CuII cation on one side and in an O‐monodentate mode to a second CuII cation on the other side, thus defining –Cu—O—C—O—Cu′– chains which run along the a axis. The title compound, C16H24O10·0.11H2O, is a key intermediate in the synthesis of 2‐deoxy‐2‐[18F]fluoro‐d ‐glucose (18F‐FDG), which is the most widely used molecular‐imaging probe for positron emission tomography (PET). The crystal structure has two independent molecules (A and B) in the asymmetric unit, with closely comparable geometries. The pyranose ring adopts a 4C1 conformation [Cremer–Pople puckering parameters: Q = 0.553 (2) Å, θ = 16.2 (2)° and ϕ = 290.4 (8)° for molecule A, and Q = 0.529 (2) Å, θ =15.3 (3)° and ϕ = 268.2 (9)° for molecule B], and the dioxolane ring adopts an envelope conformation. The chiral centre in the dioxolane ring, introduced during the synthesis of the compound, has an R configuration, with the ethoxy group exo to the mannopyranose ring. The asymmetric unit also contains one water molecule with a refined site‐occupancy factor of 0.222 (8), which bridges between molecules A and B via O—H...O hydrogen bonds.  相似文献

The title compound, also known as β‐erythroadenosine, C9H11N5O3, (I), a derivative of β‐adenosine, (II), that lacks the C5′ exocyclic hydroxymethyl (–CH2OH) substituent, crystallizes from hot ethanol with two independent molecules having different conformations, denoted (IA) and (IB). In (IA), the furanose conformation is OT1E1 (C1′‐exo, east), with pseudorotational parameters P and τm of 114.4 and 42°, respectively. In contrast, the P and τm values are 170.1 and 46°, respectively, in (IB), consistent with a 2E2T3 (C2′‐endo, south) conformation. The N‐glycoside conformation is syn (+sc) in (IA) and anti (−ac) in (IB). The crystal structure, determined to a resolution of 2.0 Å, of a cocrystal of (I) bound to the enzyme 5′‐fluorodeoxyadenosine synthase from Streptomyces cattleya shows the furanose ring in a near‐ideal OE (east) conformation (P = 90° and τm = 42°) and the base in an anti (−ac) conformation.  相似文献

Fluorine substitutions on the furanose ring of nucleosides are known to strongly influence the conformational properties of oligonucleotides. In order to assess the effect of fluorine on the conformation of 3′‐deoxy‐3′‐fluoro‐5‐methyluridine (RTF), C10H13FN2O5, we studied its stereochemistry in the crystalline state using X‐ray crystallography. The compound crystallizes in the chiral orthorhombic space group P212121 and contains two symmetry‐independent molecules (A and B) in the asymmetric unit. The furanose ring in molecules A and B adopts conformations between envelope (2E, 2′‐endo, P = 162°) and twisted (2T3, 2′‐endo and 3′exo, P = 180°), with pseudorotation phase angles (P) of 164.3 and 170.2°, respectively. The maximum puckering amplitudes, νmax, for molecules A and B are 38.8 and 36.1°, respectively. In contrast, for 5‐methyluridine (RTOH), the value of P is 21.2°, which is between the 3E (3′‐endo, P = 18.0°) and 3T4 (3′‐endo and 4′‐exo, P = 36°) conformations. The value of νmax for RTOH is 41.29°. Molecules A and B of RTF generate respective helical assemblies across the crystallographic 21‐screw axis through classical N—H…O aand O—H…O hydrogen bonds supplemented by C—H…O contacts. Adjacent parallel helices of both molecules are linked to each other via O—H…O and O…π interactions.  相似文献

The title compound, C14H12O4, forms crystals which appear monoclinic but are actually twinned triclinic. The asymmetric unit consists of two similar mol­ecules, which differ only in the conformation of the 3‐oxobutyl side chain. The mol­ecular conformation is characterized by an intra­molecular O—H⋯O hydrogen bond between the hydroxy group and the adjacent carbonyl O atom. The crystal structure is stabilized by O—H⋯O hydrogen bonds connecting the mol­ecules into zigzag chains running along the b axis.  相似文献

In the title compounds, C21H30O4, (I), and C23H34O4, (II), respectively, which are valuable intermediates in the synthesis of important steroid derivatives, rings A and B are cis‐(5β,10β)‐fused. The two molecules have similar conformations of rings A, B and C. The presence of the 5β,6β‐epoxide group induces a significant twist of the steroid nucleus and a strong flattening of the B ring. The different C17 substituents result in different conformations for ring D. Cohesion of the molecular packing is achieved in both compounds only by weak intermolecular interactions. The geometries of the molecules in the crystalline environment are compared with those of the free molecules as given by ab initio Roothan Hartree–Fock calculations. We show in this work that quantum mechanical ab initio methods reproduce well the details of the conformation of these molecules, including a large twist of the steroid nucleus. The calculated twist values are comparable, but are larger than the observed values, indicating a possible small effect of the crystal packing on the twist angles.  相似文献

The title compound, C21H28O4, has a 4‐acetoxy substituent positioned on the steroid α face. The six‐membered ring A assumes a conformation intermediate between 1α,2β‐half chair and 1α‐sofa. A long Csp3—Csp3 bond is observed in ring B and reproduced in quantum‐mechanical ab initio calculations of the isolated molecule using a molecular‐orbital Hartree–Fock method. The water content of the title compound, C13H24O10·3H2O, creates an extensive hydrogen‐bonding pattern, with all the hydroxyl groups of the disaccharide acting as hydrogen‐bond donors and acceptors. The water molecules are arranged in columns along the crystallographic b axis and form, together with one of the hydroxyl groups, infinite hydrogen‐bonded chains. In the 1H NMR spectra of the 1‐vinylpyrroles with amino‐ and alkylsulfanyl groups in 5 and 2 positions, an extraordinarily large difference between resonance positions of the HA and HB terminal methylene protons of the vinyl group is discovered. Also, the one‐bond 1J(Cβ,HB) coupling constant is surprisingly greater than the 1J(Cβ,HA) coupling constant in pyrroles under investigation, while in all known cases, there was a reverse relationship between these coupling constants. These spectral anomalies are substantiated by quantum chemical calculations. The calculations show that the amine nitrogen lone pair is removed from the conjugation with the π‐system of the pyrrole ring so that it is directed toward the HB hydrogen. These factors are favorable to the emergence of the intramolecular C–HB???N hydrogen bonding in the s‐cis(N) conformation. On the other hand, the spatial proximity of the sulfur to the HB hydrogen provides an opportunity of the intramolecular C–HB???S hydrogen bonding in the s‐cis(S) conformation. Presence of the hydrogen bond critical points as well as ring critical point for corresponding chelate ring revealed by a quantum theory of atoms in molecules (QTAIM) approach confirms the existence of the weak intramolecular C–H???N and C–H???S hydrogen bonding. Therefore, an unusual high‐frequency shift of the HB signal and the increase in the 1J(Cβ,HB) coupling constant can be explained by the effects of hydrogen bonding.
