Both 6‐(1H‐indol‐3‐yl)‐3‐methyl‐4‐(4‐methylphenyl)‐1‐phenyl‐1H‐pyrazolo[3,4‐b]pyridine‐5‐carbonitrile and 6‐(1H‐indol‐3‐yl)‐3‐methyl‐4‐(4‐methoxyphenyl)‐1‐phenyl‐1H‐pyrazolo[3,4‐b]pyridine‐5‐carbonitrile crystallize from dimethylformamide solutions as stoichiometric 1:1 solvates, viz. C29H21N5·C3H7NO, (I), and C29H21N5O·C3H7NO, (II), respectively; however, 6‐(1H‐indol‐3‐yl)‐3‐methyl‐1‐phenyl‐4‐(3,4,5‐trimethoxyphenyl)‐1H‐pyrazolo[3,4‐b]pyridine‐5‐carbonitrile, C31H25N5O3, (III), crystallizes in the unsolvated form. The heterocyclic components of (I) are linked by C—H...π(arene) hydrogen bonds to form cyclic centrosymmetric dimers, from which the solvent molecules are pendent, linked by N—H...O hydrogen bonds. In (II), the heterocyclic components are linked by a combination of C—H...N and C—H...π(arene) hydrogen bonds into chains containing two types of centrosymmetric ring, and the pendent solvent molecules are linked to these chains by N—H...O hydrogen bonds. Molecules of (III) are linked into simple C(12) chains by an N—H...O hydrogen bond, and these chains are weakly linked into pairs by an aromatic π–π stacking interaction.  相似文献

Six derivatives of 4‐amino‐1,5‐dimethyl‐2‐phenyl‐2,3‐dihydro‐1H‐pyrazol‐3‐one (4‐aminoantipyrine), C11H13N3O, (I), have been synthesized and structurally characterized to investigate the changes in the observed hydrogen‐bonding motifs compared to the original 4‐aminoantipyrine. The derivatives were synthesized from the reactions of 4‐aminoantipyrine with various aldehyde‐, ketone‐ and ester‐containing molecules, producing (Z)‐methyl 3‐[(1,5‐dimethyl‐3‐oxo‐2‐phenyl‐2,3‐dihydro‐1H‐pyrazol‐4‐yl)amino]but‐2‐enoate, C16H19N3O3, (II), (Z)‐ethyl 3‐[(1,5‐dimethyl‐3‐oxo‐2‐phenyl‐2,3‐dihydro‐1H‐pyrazol‐4‐yl)amino]but‐2‐enoate, C17H21N3O3, (III), ethyl 2‐[(1,5‐dimethyl‐3‐oxo‐2‐phenyl‐2,3‐dihydro‐1H‐pyrazol‐4‐yl)amino]cyclohex‐1‐enecarboxylate, C20H25N3O3, (IV), (Z)‐ethyl 3‐[(1,5‐dimethyl‐3‐oxo‐2‐phenyl‐2,3‐dihydro‐1H‐pyrazol‐4‐yl)amino]‐3‐phenylacrylate, C22H23N3O3, (V), 2‐cyano‐N‐(1,5‐dimethyl‐3‐oxo‐2‐phenyl‐2,3‐dihydro‐1H‐pyrazol‐4‐yl)acetamide, C14H14N4O2, (VI), and (E)‐methyl 4‐{[(1,5‐dimethyl‐3‐oxo‐2‐phenyl‐2,3‐dihydro‐1H‐pyrazol‐4‐yl)amino]methyl}benzoate, C20H19N3O3, (VII). The asymmetric units of all these compounds have one molecule on a general position. The hydrogen bonding in (I) forms chains of molecules via intermolecular N—H...O hydrogen bonds around a crystallographic sixfold screw axis. In contrast, the formation of enamines for all derived compounds except (VII) favours the formation of a six‐membered intramolecular N—H...O hydrogen‐bonded ring in (II)–(V) and an intermolecular N—H...O hydrogen bond in (VI), whereas there is an intramolecular C—H...O hydrogen bond in the structure of imine (VII). All the reported compounds, except for (II), feature π–π interactions, while C—H...π interactions are observed in (II), C—H...O interactions are observed in (I), (III), (V) and (VI), and a C—O...π interaction is observed in (II).  相似文献

The dimethyl sulfoxide hemisolvate of perindoprilat [systematic name: (1S)‐2‐((S)‐{1‐[(2S,3aS,7aS)‐2‐carboxyoctahydro‐1H‐indol‐1‐yl]‐1‐oxopropan‐2‐yl}azaniumyl)pentanoate dimethyl sulfoxide hemisolvate], C17H28N2O5·0.5C2H6OS, an active metabolite of perindopril, has been synthesized, structurally characterized by single‐crystal X‐ray diffraction and compared with its ethanol disolvate analogue [Pascard et al. (1991). J. Med. Chem. 34 , 663–669]. Both compounds crystallize in the orthorhombic P212121 space group in the same zwitterionic form, with a protonated alanine N atom and an anionic carboxylate group at the n‐alkyl chain. The three structural units present in the unit cell (two zwitterions and the solvent molecule) are held together by a rich system of O—H...O, N—H...O and C—H...O hydrogen‐bond contacts.  相似文献

A concise and efficient synthesis of a series of amino‐substituted benzimidazole–pyrimidine hybrids has been developed, starting from the readily available N4‐(2‐aminophenyl)‐6‐methoxy‐5‐nitrosopyrimidine‐2,4‐diamine. In each of N5‐benzyl‐6‐methoxy‐4‐(2‐phenyl‐1H‐benzo[d]imidazol‐1‐yl)pyrimidine‐2,5‐diamine, C25H22N6O, (I), 6‐methoxy‐N5‐(4‐methoxybenzyl)‐4‐[2‐(4‐methoxyphenyl)‐1H‐benzo[d]imidazol‐1‐yl]pyrimidine‐2,5‐diamine, C27H26N6O3, (III), 6‐methoxy‐N5‐(4‐nitrobenzyl)‐4‐[2‐(4‐nitrophenyl)‐1H‐benzo[d]imidazol‐1‐yl]pyrimidine‐2,5‐diamine, C25H20N8O5, (IV), the molecules are linked into three‐dimensional framework structures, using different combinations of N—H…N, N—H…O, C—H…O, C—H…N and C—H…π hydrogen bonds in each case. Oxidative cleavage of 6‐methoxy‐N5‐(4‐methylbenzyl)‐4‐[2‐(4‐methylphenyl)‐1H‐benzo[d]imidazol‐1‐yl]pyrimidine‐2,5‐diamine, (II), with diiodine gave 6‐methoxy‐4‐[2‐(4‐methylphenyl)‐1H‐benzo[d]imidazol‐1‐yl]pyrimidine‐2,5‐diamine, which crystallized as a monohydrate, C19H18N6O·H2O, (V), and reaction of (V) with trifluoroacetic acid gave two isomeric products, namely N‐{5‐amino‐6‐methoxy‐6‐[2‐(4‐methylphenyl)‐1H‐benzo[d]imidazol‐1‐yl]pyrimidin‐2‐yl}‐2,2,2‐trifluoroacetamide, which crystallized as an ethyl acetate monosolvate, C21H17F3N6O2·C4H8O2, (VI), and N‐{2‐amino‐6‐methoxy‐4‐[2‐(4‐methylphenyl)‐1H‐benzo[d]imidazol‐1‐yl]pyrimidin‐5‐yl}‐2,2,2‐trifluoroacetamide, which crystallized as a methanol monosolvate, C21H17F3N6O2·CH4O, (VIIa). For each of (V), (VI) and (VIIa), the supramolecular assembly is two‐dimensional, based on different combinations of O—H…N, N—H…O, N—H…N, C—H…O and C—H…π hydrogen bonds in each case. In each of ethyl N‐{2‐amino‐5‐formyl‐6‐[methyl(phenyl)amino]pyrimidin‐4‐yl}glycinate, C16H19N5O3, (I), N‐{2‐amino‐5‐formyl‐6‐[methyl(phenyl)amino]pyrimidin‐4‐yl}glycinamide, C14H16N6O2, (II), and ethyl 3‐amino‐N‐{2‐amino‐5‐formyl‐6‐[methyl(phenyl)amino]pyrimidin‐4‐yl}propionate, C17H21N5O3, (III), the pyrimidine ring is effectively planar, but in each of methyl N‐{2‐amino‐6‐[benzyl(methyl)amino]‐5‐formylpyrimidin‐4‐yl}glycinate, C16H19N5O3, (IV), ethyl 3‐amino‐N‐{2‐amino‐6‐[benzyl(methyl)amino]‐5‐formylpyrimidin‐4‐yl}propionate, C18H23N5O3, (V), and ethyl 3‐amino‐N‐[2‐amino‐5‐formyl‐6‐(piperidin‐4‐yl)pyrimidin‐4‐yl]propionate, C15H23N5O3, (VI), the pyrimidine ring is folded into a boat conformation. The bond lengths in each of (I)–(VI) provide evidence for significant polarization of the electronic structure. The molecules of (I) are linked by paired N—H...N hydrogen bonds to form isolated dimeric aggregates, and those of (III) are linked by a combination of N—H...N and N—H...O hydrogen bonds into a chain of edge‐fused rings. In the structure of (IV), molecules are linked into sheets by means of two hydrogen bonds, both of N—H...O type, in the structure of (V) by three hydrogen bonds, two of N—H...N type and one of C—H...O type, and in the structure of (VI) by four hydrogen bonds, all of N—H...O type. Molecules of (II) are linked into a three‐dimensional framework structure by a combination of three N—H...O hydrogen bonds and one C—H...O hydrogen bond.  相似文献

In the title complex salt, [Ir(C5H4O)(C16H22N6)(CO)](CF3O3S), the IrIII centre adopts a distorted octahedral geometry with a facial coordination of the tris(3,5‐dimethyl‐1H‐pyrazol‐1‐yl)methane ligand. The C—C distances of the iridacycle are in agreement with its iridacyclohexa‐2,5‐dien‐4‐one nature, which presents a nonsymmetric boat‐like conformation with the C—Ir—C vertex more bent than the C—C(=O)—C vertex. The supramolecular architecture is mainly directed by CO...CO and CO...π and Csp3—H...O interactions, the arrangement of which depends on the anion.  相似文献

Pharmaceutical cocrystals are crystalline solids formed by an active pharmaceutical ingredient and a cocrystal former. The cocrystals 2,6‐diaminopyridine (DAP)–piracetam [PIR; systematic name: 2‐(2‐oxopyrrolidin‐1‐yl)acetamide] (1/1), C5H7N3·C6H10N2O2, (I), and 2,6‐diaminopyridine–theophylline (TEO; systematic name: 1,3‐dimethyl‐7H‐purine‐2,6‐dione) (1/1), C5H7N3·C7H8N4O2, (II), were prepared by the solvent‐assisted grinding method and were characterized by IR spectroscopy and powder X‐ray diffraction. Cocrystal (I) crystallized in the orthorhombic space group Pbca and showed a 1:1 stoichiometry. The DAP and PIR molecules are linked by an N—H…O hydrogen‐bond interaction. Self‐assembly of PIR molecules forms a sheet of C (4) and C (7) chains. Cocrystal (II) crystallized in the monoclinic P 21/c space group and also showed a 1:1 stoichiometry. The DAP and TEO molecules are connected by N—H…N and N—H…O hydrogen bonds, forming an R 22(9) heterosynthon. A bidimensional supramolecular array is formed by interlinked DAP–TEO tetramers, producing a two‐dimensional sheet.  相似文献

Two tricarbonyl complexes of rhenium(I) and manganese(I) coordinated by the ligand 2‐{[2‐(1H‐imidazol‐4‐yl)ethyl]iminomethyl}‐5‐methylphenolate are reported, viz. fac‐tricarbonyl(2‐{[2‐(1H‐imidazol‐4‐yl‐κN3)ethyl]iminomethyl‐κN}‐5‐methylphenolato‐κO)rhenium(I) methanol monosolvate, [Re(C16H14N3O4)(CO)3]·CH3OH, (I), and fac‐tricarbonyl(2‐{[2‐(1H‐imidazol‐4‐yl‐κN3)ethyl]iminomethyl‐κN}‐5‐methylphenolato‐κO)manganese(I), fac‐[Mn(C16H14N3O4)(CO)3], (II), display facial coordination in a distorted octahedral environment. The crystal structure of (I) is stabilized by O—H...O, N—H...O and C—H...O hydrogen‐bond interactions, while that of (II) is stabilized by N—H...O hydrogen‐bond interactions only. These interactions result in two‐dimensional networks and π–π stacking for both structures.  相似文献

The complexes [2‐(1H‐imidazol‐4‐yl‐κN3)ethylamine‐κN]bis(tri‐tert‐butoxysilanethiolato‐κS)cobalt(II), [Co(C12H27O3SSi)2(C5H9N3)], and [2‐(1H‐imidazol‐4‐yl‐κN3)ethylamine‐κN]bis(tri‐tert‐butoxysilanethiolato‐κS)zinc(II), [Zn(C12H27O3SSi)2(C5H9N3)], are isomorphous. The central ZnII/CoII ions are surrounded by two S atoms from the tri‐tert‐butoxysilanethiolate ligand and by two N atoms from the chelating histamine ligand in a distorted tetrahedral geometry, with two intramolecular N—H...O hydrogen‐bonding interactions between the histamine NH2 groups and tert‐butoxy O atoms. Molecules of the complexes are joined into dimers via two intermolecular bifurcated N—H...(S,O) hydrogen bonds. The ZnII atom in [(1H‐imidazol‐4‐yl‐κN3)methanol]bis(tri‐tert‐butoxysilanethiolato‐κ2O,S)zinc(II), [Zn(C12H27O3SSi)2(C4H6N2O)], is five‐coordinated by two O and two S atoms from the O,S‐chelating silanethiolate ligand and by one N atom from (1H‐imidazol‐4‐yl)methanol; the hydroxy group forms an intramolecular hydrogen bond with sulfur. Molecules of this complex pack as zigzag chains linked by N—H...O hydrogen bonds. These structures provide reference details for cysteine‐ and histidine‐ligated metal centers in proteins.  相似文献

In the benzene and phenol solvates of (S)‐4‐{3‐[2‐(dimethylamino)ethyl]‐1H‐indol‐5‐ylmethyl}oxazolidin‐2‐one, viz. C16H21N3O2·C6H6, (I), and C16H21N3O2·C6H5OH, (II), the host molecule has three linked residues, namely a planar indole ring system, an ethylamine side chain and an oxazolidinone system. It has comparable features to that of sumatriptan, although the side‐chain orientations of (I) and (II) differ from those of sumatriptan. Both (I) and (II) have host–guest‐type structures. The host molecule in (I) and (II) has an L‐shaped form, with the oxazolidinone ring occupying the base and the remainder of the molecule forming the upright section. In (I), each benzene guest molecule is surrounded by four host molecules, and these molecules are linked by a combination of N—H...N, N—H...O and C—H...O hydrogen bonds into chains of edge‐fused R44(33) rings. In (II), two independent molecules are present in the asymmetric unit, with similar conformations. The heterocyclic components are connected through N—H...N, N—H...O and C—H...O interactions to form chains of edge‐fused R64(38) rings, from which the phenol guest molecules are pendent, linked by O—H...O hydrogen bonds. Polymorph (Ia) of eldoral [5‐ethyl‐5‐(piperidin‐1‐yl)barbituric acid or 5‐ethyl‐5‐(piperidin‐1‐yl)‐1,3‐diazinane‐2,4,6‐trione], C11H17N3O3, displays a hydrogen‐bonded layer structure parallel to (100). The piperidine N atom and the barbiturate carbonyl group in the 2‐position are utilized in N—H...N and N—H...O=C hydrogen bonds, respectively. The structure of polymorph (Ib) contains pseudosymmetry elements. The two independent molecules of (Ib) are connected via N—H...O=C(4/6‐position) and N—H...N(piperidine) hydrogen bonds to give a chain structure in the [100] direction. The hydrogen‐bonded layers, parallel to (010), formed in the salt diethylammonium 5‐ethyl‐5‐(piperidin‐1‐yl)barbiturate [or diethylammonium 5‐ethyl‐2,4,6‐trioxo‐5‐(piperidin‐1‐yl)‐1,3‐diazinan‐1‐ide], C4H12N+·C11H16N3O3, (II), closely resemble the corresponding hydrogen‐bonded structure in polymorph (Ia). Like many other 5,5‐disubstituted derivatives of barbituric acid, polymorphs (Ia) and (Ib) contain the R22(8) N—H...O=C hydrogen‐bond motif. However, the overall hydrogen‐bonded chain and layer structures of (Ia) and (Ib) are unique because of the involvement of the hydrogen‐bond acceptor function in the piperidine group.  相似文献

The ion pairs are linked into a three‐dimensional framework structure by three independent C—H...O hydrogen bonds, augmented by C—H...π(arene) hydrogen bonds and an aromatic π–π stacking interaction. In flunarizinediium bis(4‐toluenesulfonate) dihydrate {systematic name: 1‐[bis(4‐fluorophenyl)methyl]‐4‐[(2E)‐3‐phenylprop‐2‐en‐1‐yl]piperazine‐1,4‐diium bis(4‐methylbenzenesulfonate) dihydrate}, C26H28F2N22+·2C7H7O3S·2H2O, (II), one of the anions is disordered over two sites with occupancies of 0.832 (6) and 0.168 (6). The five independent components are linked into ribbons by two independent N—H...O hydrogen bonds and four independent O—H...O hydrogen bonds, and these ribbons are linked to form a three‐dimensional framework by two independent C—H...O hydrogen bonds, but C—H...π(arene) hydrogen bonds and aromatic π–π stacking interactions are absent from the structure of (II). In the inner‐salt zwitterion of 3,6‐bis(pyridin‐2‐yl)pyrazine‐2,5‐dicarboxylic acid, (I), namely 5‐carboxy‐3‐(pyridin‐1‐ium‐2‐yl)‐6‐(pyridin‐2‐yl)pyrazine‐2‐carboxylate, [C16H10N4O4, (Ia)], the pyrazine ring has a twist–boat conformation. The opposing pyridine and pyridinium rings are almost perpendicular to one another, with a dihedral angle of 80.24 (18)°, and are inclined to the pyrazine mean plane by 36.83 (17) and 43.74 (17)°, respectively. The carboxy and carboxylate groups are inclined to the mean plane of the pyrazine ring by 43.60 (17) and 45.46 (17)°, respectively. In the crystal structure, the molecules are linked via N—H...O and O—H...O hydrogen bonds, leading to the formation of double‐stranded chains propagating in the [010] direction. On treating (Ia) with aqueous 1 M HCl, the diprotonated dihydrate form 2,2′‐(3,6‐dicarboxypyrazine‐2,5‐diyl)bis(pyridin‐1‐ium) dichloride dihydrate [C16H12N4O42+·2Cl·2H2O, (Ib)] was obtained. The cation lies about an inversion centre. The pyridinium rings and carboxy groups are inclined to the planar pyrazine ring by 55.53 (9) and 19.8 (2)°, respectively. In the crystal structure, the molecules are involved in N—H...Cl, O—H...Owater and Owater—H...Cl hydrogen bonds, leading to the formation of chains propagating in the [010] direction. When (Ia) was recrystallized from dimethyl sulfoxide (DMSO), the DMSO disolvate 3,6‐bis(pyridin‐2‐yl)pyrazine‐2,5‐dicarboxylic acid dimethyl sulfoxide disolvate [C16H10N4O4·2C2H6OS, (Ic)] of (I) was obtained. Here, the molecule of (I) lies about an inversion centre and the pyridine rings are inclined to the planar pyrazine ring by only 23.59 (12)°. However, the carboxy groups are inclined to the pyrazine ring by 69.0 (3)°. In the crystal structure, the carboxy groups are linked to the DMSO molecules by O—H...O hydrogen bonds. In all three crystal structures, the presence of nonclassical hydrogen bonds gives rise to the formation of three‐dimensional supramolecular architectures.  相似文献

The crystal structure of N‐[(1‐{2‐oxo‐2‐[2‐(pyrazin‐2‐ylcarbonyl)hydrazin‐1‐yl]ethyl}cyclohexyl)methyl]pyrazine‐2‐carboxamide monohydrate (Pyr‐Gpn‐NN‐NH‐Pyr·H2O), C19H23N7O3·H2O, reveals an unusual trans–gauche (tg) conformation for the gabapentin (Gpn) residue around the Cγ—Cβ1) and Cβ—Cα2) bonds. The molecular conformation is stabilized by intramolecular N—H...N hydrogen bonds and weak C—H...O interactions. The packing of the molecules in the crystal lattice shows a network of strong N—H...O and O—H...O hydrogen bonds together with weak C—H...O and π–π inteactions.  相似文献

In the crystal structure of the benzoate salt (systematic name: (3R)‐1‐{3‐[(2‐cyanophenyl)methyl]‐1‐methyl‐2,6‐dioxo‐1,2,3,6‐tetrahydropyrimidin‐4‐yl}piperidin‐3‐aminium benzoate), C18H22N5O2+·C7H5O2, the NH3+ group of the cation is engaged in three intermolecular N—H...O hydrogen bonds to yield a hydrogen‐bonded layer structure. The compounds N‐[2‐(4‐cyano‐5‐dicyanomethylene‐2,2‐dimethyl‐2,5‐dihydrofuran‐3‐yl)vinyl]‐N‐phenylacetamide, C20H16N4O2,(I), and 2‐{3‐cyano‐5,5‐dimethyl‐4‐[2‐(piperidin‐1‐yl)vinyl]‐2,5‐dihydrofuran‐2‐ylidene}malononitrile 0.376‐hydrate, C17H18N4O·0.376H2O, (II), are novel push–pull molecules. The significant bonding changes in the polyene chain compared with the parent molecule 2‐dicyanomethylene‐4,5,5‐trimethyl‐2,5‐dihyrofuran‐3‐carbonitrile are consistent with the relative electron‐donating properties of the acetanilido and piperidine groups. The packing of (I) utilizes one phenyl–cyano C—H...N and two phenyl–carbonyl C—H...O hydrogen bonds. Compound (II) crystallizes with a partial water molecule (0.376H2O), consistent with cell packing that is dominated by attractive C—H...N(cyano) interactions. These compounds are precursors to novel nonlinear optical chromophores, studied to assess the impact of donor strength and the extent of conjugation on bond‐length alternation, crystal packing and aggregation.  相似文献

In both 1‐(2‐cyano­ethyl)thymine [systematic name: 3‐(5‐methyl‐2,4‐dioxo‐1,2,3,4‐tetra­hydro­pyrimidin‐1‐yl)propane­nitrile], C8H9N3O2, (I), and 1‐(3‐cyano­propyl)thymine [systematic name: 4‐(5‐methyl‐2,4‐dioxo‐1,2,3,4‐tetra­hydro­pyrimidin‐1‐yl)butane­nitrile], C9H11N3O2, (II), the core of the supra­molecular structure is formed by centrosymmetric dimers generated by N—H⋯O hydrogen bonds. Further weak hydrogen bonds of C—H⋯O and C—H⋯N types generate mol­ecular tapes and sheets that resemble those in uracil and its methyl derivatives. The steric hindrance that arises from the cyano­alkyl substituents perturbs the conformations of the tapes and sheets.  相似文献

In each of 6‐amino‐3‐methyl‐2‐(morpholin‐4‐yl)‐5‐nitrosopyrimidin‐4(3H)‐one, C9H13N5O3, (I), morpholin‐4‐ium 4‐amino‐2‐(morpholin‐4‐yl)‐5‐nitroso‐6‐oxo‐1,6‐dihydropyrimidin‐1‐ide, C4H10NO+·C8H10N5O3, (II), and 6‐amino‐2‐(morpholin‐4‐yl)‐5‐nitrosopyrimidin‐4(3H)‐one hemihydrate, C8H11N5O3·0.5H2O, (III), the bond distances within the pyrimidine components are consistent with significant electronic polarization, which is most marked in (II) and least marked in (I). Despite the high level of substitution, the pyrimidine rings are all effectively planar, and in each of the pyrimidine components, there are intramolecular N—H...O hydrogen bonds. In each compound, the organic components are linked by multiple N—H...O hydrogen bonds to form sheets of widely differing construction, and in compound (III) adjacent sheets are linked by the water molecules, so forming a three‐dimensional hydrogen‐bonded framework. This study also contains the first direct geometric comparison between the electronic polarization in a neutral aminonitrosopyrimidine and that in its ring‐deprotonated conjugate anion in a metal‐free environment.  相似文献

A concise and efficient synthesis of 6‐benzimidazolyl‐5‐nitrosopyrimidines has been developed using Schiff base‐type intermediates derived from N4‐(2‐aminophenyl)‐6‐methoxy‐5‐nitrosopyrimidine‐2,4‐diamine. 6‐Methoxy‐N4‐{2‐[(4‐methylbenzylidene)amino]phenyl}‐5‐nitrosopyrimidine‐2,4‐diamine, (I), and N4‐{2‐[(ethoxymethylidene)amino]phenyl}‐6‐methoxy‐5‐nitrosopyrimidine‐2,4‐diamine, (III), both crystallize from dimethyl sulfoxide solution as the 1:1 solvates C19H18N6O2·C2H6OS, (Ia), and C14H16N6O3·C2H6OS, (IIIa), respectively. The interatomic distances in these intermediates indicate significant electronic polarization within the substituted pyrimidine system. In each of (Ia) and (IIIa), intermolecular N—H…O hydrogen bonds generate centrosymmetric four‐molecule aggregates. Oxidative ring closure of intermediate (I), effected using ammonium hexanitratocerate(IV), produced 4‐methoxy‐6‐[2‐(4‐methylphenyl‐1H‐benzimidazol‐1‐yl]‐5‐nitrosopyrimidin‐2‐amine, C19H16N6O2, (II) [Cobo et al. (2018). Private communication (CCDC 1830889). CCDC, Cambridge, England], where the extent of electronic polarization is much less than in (Ia) and (IIIa).
