Abstract

A simple and sensitive method for the simultaneous determination of five fluoroquinolones (ciprofloxacin, enrofloxacin, danofloxacin, difloxacin, and sarafloxacin) in bovine milk was developed. Protein precipitation from milk samples was achieved by the addition of acetonitrile and o‐phosphoric acid. Acetonitrile was removed with dichloromethane, leaving the fluoroquinolones in the acid aqueous extract. The aqueous extract was analyzed by liquid chromatography with fluorescence detection (LC–FD). The mobile phase was composed of acetonitrile and 10 mM citrate buffer solution of pH 4.5, with an initial composition of acetonitrile‐water (12∶88, v/v) and using linear gradient elution. Norfloxacin was used as an internal standard. The limits of detection found ranged from 1 to 6 ng · mL?1 and were below the maximum residue limits (MRLs) established by the European Union. The proposed method was applied to the determination of these compounds in different bovine milk samples. Method validation was carried out by a recovery assay.  相似文献

Abstract

A sensitive analytical method for the determination of clarithromycin in biological fluids with electrochemiluminescence detection after capillary electrophoresis separation was proposed in this paper, based on the enhancing effect of clarithromycin on electrochemiluminescence of tris(2,2-bipyridyl) ruthenium(II) (Ru(bpy)3 2+). Various factors influencing the separation and detection of clarithromycin were examined to establish the detection system. Under the optimized conditions, the electrochemiluminescence intensity is proportional with the concentration of the analyte over the range of 0.1–10 µM with a detection limit of 30 nM (S/N = 3). The proposed method has been successfully applied for clarithromycin content determination in spiked human plasma and urine samples.  相似文献

Abstract

A highly selective and effective method for the purification and preconcentration of norfloxacin (NFX) in seawater samples was developed based on molecularly imprinted solid-phase extraction (MISPE). The molecularly imprinted polymer was synthesized by precipitation polymerization. Methacrylic acid (MAA) and ethylene glycol dimethacrylate (EGDMA) were used as the functional monomer and crosslinker, respectively. The resulting molecularly imprinted polymer (MIP) showed high adsorption for NFX and was selective for its solid-phase extraction. An offline MISPE method followed by high performance liquid chromatography with diode array detection was established for the determination of NFX in seawater. The recoveries of spiked seawater samples using the MISPE columns were satisfactorily higher than 77.6%. The relative standard deviation was less than 5.60%, and the limit of detection was 0.027?μg L?1. Four seawater samples obtained from the Bohai Sea were analyzed, and NFX was found only at one location at a concentration of 0.280?μg L?1.  相似文献

Abstract

A new method has been developed for field screening of copper (II) in water samples, which is based on an enzyme inhibition reaction between copper (II) and nitrate reductase. The concentration of copper (II) was acquired by indirect determination the reaction product (nitrite) with a mini optical reflection sensor. Under the optimum conditions, the calibration graph was linear in the range of 5.0–50 ng mL?1. The limit of detection was 0.5 ng mL?1. This method has been used for the field screening of copper (II) with satisfactory results.  相似文献

Abstract

A fast, efficient and low-cost high performance liquid chromatography–tandem mass spectrometry methodology was developed and validated for the simultaneous determination of free urinary cortisone, cortisol and their tetrahydro-metabolites. The developed method comprises a simple liquid-liquid extraction with CH2Cl2, followed by reversed-phase liquid chromatography–tandem mass spectrometry (LC–MS/MS) with electrospray ionization (ESI) in positive mode. The baseline chromatographic separation of the analytes, including the stereoisomers tetrahydrocortisol (THF) and allo-THF, was achieved on a Hypersil Gold C18 column with a mobile phase consisting of 0.05%v/v formic acid in water—acetonitrile, using a gradient elution program. The influence of the mobile phase composition and the ESI parameters on the sensitivity of the method was extensively studied. Sample preparation was also optimized, testing two techniques: solid phase extraction (SPE) and liquid-liquid extraction (LLE). Recoveries ranged from 74.7% (a-THF) to 93.5% (cortisol) and the method limits of detection (MLD) ranged from 0.34?ng mL?1 (cortisol) to 1.37?ng mL?1 (THF). Intra- and inter-day coefficient of variation of the assay varied from1.5% (allo-THF) to 13% (tetrahydrocortisone) and from 3.6% (allo-THF) to 14.9% (tetrahydrocortisone), respectively. The method was applied for the analysis of urine samples from 53 healthy individuals with a mean age of 13.96?years in order to estimate the concentration of the five corticosteroids and the ratio of the metabolites. Associations between urinary cortisol/cortisone and serum cortisol/cortisone values were also characterized.  相似文献

Abstract

The polarographic behavior of the complex of iron–4– (2–pyridylazo) resorcin(PAR) was studied. In HAc– NaAc– EDTA buffer solution, the complex can be adsorped on a hanging mercury drop electrode giving a sensitive adsorptive complex reduction peak with a peak potential at -0.36V(vs. SCE). Optimum experimental conditions were found by the use of 0.08mol/L HAc, 0.06mol/L NaAc, 5.0 × 10?3mol/L EDTA and 1.0 × 10?5mol/L PAR. With preconcentration for 60s, the derivative peak height of the complex compound is linearly proportional to the concentration for Fe in the range from 1.0 × 10?9mol/L to 1.0 × 10?7mol/L. For a 2–min pre–concentration time, the detection limit found was 2.0 × 10?10mol/L. This method has high sensitivity and selectivity. It has been applied to the determination of trace iron in food and water samples without any pre–separation step.  相似文献

A reliable and robust analytical method based on solid phase extraction (SPE) and high performance liquid chromatography (HPLC) with ultraviolet (UV) detector was developed for the simultaneous determination of five cephalosporin antibiotics (Ceftazidime, Cefradine, Cefaclor, Cefotaxime and Cefoperazone) in various water samples. Under optimised conditions, it was applicable to preconcentrate up to 500?ml of water samples in the OASIS HLB cartridges with reasonable recoveries for all the cephalosporin antibiotics tested. Recoveries were as follows: deionised water, tap water and groundwater, between 84.2 and 98.9%; surface water, between 71.2 and 81.0%; influent and effluent of wastewater treatment plant (WWTP), between 56.9 and 72.1%. The method detection limits (MDLs) for different water samples were in the range of 26 to 59?ng?l?1. Real water samples were analysed using the proposed approach to demonstrate the applicability and validation. Negative results were obtained for the tap water and groundwater. However, all the selected cephalosporin antibiotics were identified in the influent and effluent of a local WWTP at ng?l?1–µg?l?1 level. In addition, Ceftazidime was found in surface water with a concentration of 0.75–2.60?µg?l?1. The results indicate that the ‘pseudo-persistent’ contamination of cephalosporin antibiotics in the water environment could not be neglected.  相似文献

A modified hollow-fiber-supported dispersive liquid-liquid microextraction (HF-DLLME) method was developed for the determination of aflatoxins and ochratoxin A in food samples. The various parameters affecting the efficiency of extraction, such as pH, salt addition, extraction time, stirring rate, desorption time, type and volume of extractant and disperser solvents were carefully studied and optimized using two step strategies. The linearity of the evaluated results was 0.1 to 30?μg L?1 for aflatoxins and 0.1 to 20?μg L?1 for ochratoxin A, with regression coefficients (R2) exceeding 0.9990. The precision was satisfactory with relative standard deviation values less than 11%. The method accuracy was within the recommended range from 70% to 120% and analyte accuracy between 83% and 101%. The limits of detection and quantification were in the range from 0.04 to 0.06?μg L?1 and 0.08 to 0.13?μg L?1, respectively, for multi-aflatoxins, and 0.02 to 0.04?µg L?1 and 0.08 to 0.10?µg L?1, respectively, for ochratoxin A. The developed method was successfully applied for the determination of mycotoxins in food samples.  相似文献

A voltammetric procedure in the flow system for determination of traces of Cr(VI) in the presence of Cr(III) and humic acid is presented. The calibration graph is linear from 5×10−10 to 1×10−7 mol l−1 for an accumulation time of 120 s. The R.S.D. for 1×10−8 mol l−1 Cr(VI) is 5.3% (n=5). The detection limit estimated from 3σ for a low concentration of Cr(VI) and accumulation time of 120 s is 2×10−10 mol l−1. The method can be used for Cr(VI) determination in the presence of up to 50 mg l−1 of humic acid. The validation of the method was carried out by studying the recovery of Cr(VI) from spiked river water and by the comparison of the results of determination of Cr(VI) in a soil sample. The method cannot be used for analysis of samples containing high concentrations of chloride ions such as seawater and estuarine water.  相似文献

Abstract

A Partial Least Squares (PLS) calibration method was applied to the simultaneous determination of iprodione, procymidone and chlorothalonil in mixtures, by uv-vis absorption spectrophotometry and by high performance liquid chromatography (HPLC). Signals and first-derivative (1D) signals were used to optimize the calibration matrices by the PLS-1 method. Quantitative results are presented for synthetic mixtures and for extracts from soil and groundwater samples. Significant improvements were achieved by using the PLS-1 method built with first-derivative chromatograms, in the determination of iprodione, procymidone and chlorothalonil in environmental samples.  相似文献

The method relies on selective complexation of As(III) with a suitable chelating agent followed by dispersive liquid–liquid microextraction (DLLME) method. Flame atomic absorption spectrometry (FAAS) equipped with microsample introduction system was utilised for determination of As(III). 1-Undecanol and acetone were used as extraction solvent and disperser solvent respectively. Some effective parameters on complex formation and extraction have been optimised. Under the optimum conditions, the enrichment factor of 108 for As(III) was obtained from 9.8?mL of water samples. The calibration graph was linear in the range of 2–15?µg?L?1 with detection limits of 0.60?µg?L?1 for As(III). The relative standard deviation (R.S.D.) for ten replicate measurements of 5.00?µ?gL?1 of As(III) was 6.2%. Operation simplicity and high enrichment factors are the main advantages of DLLME for the determination of As(III) without necessity for hydride generation in water samples.  相似文献

Abstract

The reaction between lead(II) and 2-(2-Thiazolylazo)-p-Cresol(TAC) in the presence of TERGITOL NPX (4 mg/ml) at an apparent pH 9.0–10.0 results in an intensely colored complex which is stable for at least 4 hr.

The composition of the complex is 1:2 cation: TAC and the log of the formation constant is 11.92 ± 0. 40. Beer's law is obeyed up to 6.0μg.ml?1 of lead(II) at 650nm.

The apparent molar absorptivity at 650 nm is 2.07 × 104 1. mole?1.cm?1 and the detection limit was obtained as 10.0 ng.ml?1 of lead(II).

Abstract

An indirect photometric method with a continuous-flow analysis is presented for the determination of trace amounts of vanadium(IV). It is based on the redox reaction of copper(II) with vanadium(1V) in the presence of neocuproine. In the presence of neocuproine, copper(I1) is reduced easily by vanadium(I V) to a copper(1)-neocuproine complex, which shows a n absorption maximum at 454 nm. By measuring t h e absorbance of the complex at this wavelength, vanadium(1V) in t h e range 2×10?6 - 8 × mol dm?5 mol dm?3 can be determined at a rate of 120 samples h?1. A new and highly sensitive spectrophotometric method is developed for the determination of sub ppm levels of the widely used organophosphorus insecticide dichlorvos. The method is based on alkaline hydrolysis of dichlorvos to dichloroacetaldehyde followed by coupling with diphenyl semicarbazide (DPC) in alkaline medium. The absorption maxima of the wine red dye compound formed is measured at 490 nm. Beer's law is obeyed over the concentration range of 4.3 to 34 μg in a final solution volume of 25 mL (0.18–1.36 ppm). The molar absorptivity, Sandell's sensitivity and correlation coefficient were found to be 2.9 × 105 l mole?1 cm?1, 0.013 μg cm?2 and 0.9999, respectively. The standard deviation and relative standard deviation were found to be ± 0.007 and 1.90%, respectively. The lower limit of detection is 0.04 μg. The method is simple, sensitive and free from interferences of other pesticides and diverse ions. Other organophosphorous pesticides do not interfere with the proposed method. The method has been satisfactorily applied to the determination of dichlorvos in environmental and agricultural samples.  相似文献

Novel and fast deep eutectic solvent (DES)-based homogeneous liquid–liquid microextraction (HLLME) was applied for the extraction of copper from vegetable samples followed by flame atomic absorption spectrometry (FAAS). 1,5-diphenyl carbazone (DPC) was used as the chelating agent, and a DES was used as the extraction media. The utilized DES was based on benzyl triphenyl phosphonium bromide and ethylene glycol in a 1:8?mole ratio. The phase separation phenomenon was occurred by changing of sample temperature. Several factors influencing the extraction efficiency were investigated and optimized. Under the optimized conditions, an enhancement factor of 64 was obtained. The limit of detection, based on three signal-to-noise ratio, and limit of quantification were found to be 0.13?µg L?1 and 5.0?µg L?1, respectively. The calibration curve was linear within the range of 5.0–250?µg L?1 with r2 > 0.9957. Intra- and inter-day relative standard deviations (%) of 2.1% and 2.6% were obtained at the concentration of 25?µg L?1. The accuracy of the proposed method was evaluated by analyzing a tomato leaves certified reference material and the results were to be in agreement with the certified value. Finally, the feasibility of the method was successfully confirmed by determination of copper in spinach, lettuce, broccoli, potato, carrot and parsley samples.  相似文献

A new method using differential pulse adsorptive stripping voltammetry for the determination of atrazine (ATZ) in natural water samples using a bismuth film electrode (BiFE) is proposed. The calibration curve was linear in the atrazine concentration range from 6.7×10?7 to 2.0×10?5 mol L?1, with a limit of detection (LOD) of 1.4×10?7 mol L?1. Abstract

A simple, rapid, injection chemiluminescence method is described for the determination of prulifloxacin, a commonly used antibiotic. A strong chemiluminescence signal was detected when a mixture of the analyte and tris-(4,7-diphenyl-1,10-phenanthrolinedisulfonic acid)ruthenium(II) was injected into cerium(IV) sulfate. The chemiluminescence signal is proportional to the concentration of prulifloxacin in the range 4.0 × 10?8–9.0 × 10?6 mol L?1. The detection limit is 1.0 × 10?8 mol L?1, and the relative standard deviation is 2.2% (n = 11) for the determination of 8.0 × 10?7 mol L?1 prulifloxacin. The proposed method was successfully applied to the determination of prulifloxacin in pharmaceutical preparations in capsules, spiked serum, and urine samples.  相似文献

Abstract

A new chemiluminescence (CL) reaction was observed when cloperastine hydrochloride was injected into the reaction mixture after the CL reaction of Ce(IV) and sodium sulfite finished. A new flow injection CL method for the determination of cloperastine hydrochloride was established based on the CL reaction. The relative standard deviation (RSD) for the determination of cloperastine hydrochloride was 1.3% (n=11, c=1.0×10?6 g/mL). The CL intensity responded linearly to the concentration of cloperastine hydrochloride in the range 2.0×10?7~2.0×10?5 g/mL (r=0.9962). The detection limit was 5×10?8 g/mL cloperastine hydrochloride. A new method for the simultaneous determination of 12 volatile organic compounds (trans-1,2-dichloroethene, 1,1,1-trichloroethane, benzene, 1,2-dichloroethane, trichloroethene, toluene, 1,1,2-trichloroethane, tetrachloroethene, ethylbenzene, m-, p-, o-xylene) in water samples by headspace solid phase microextraction (HS–SPME)–gas chromatography mass spectrometry (GC–MS) was described, using a 100?µm PDMS (polydimethylsiloxane) coated fibre. The response surface methodology was used to optimise the effect of the extraction time and temperature, as well as the influence of the salt addition in the extraction process. Optimal conditions were extraction time and temperature of 30?min and ?20°C, respectively, and NaCl concentration of 4?mol?L?1. The detection limits were in the range of 1.1?×?10?3–2.3?µg?L?1 for the 12 volatile organic compounds (VOCs). Global uncertainties were in the range of 4–68%, when concentrations decrease from 250?µg?L?1 down to the limits of quantification. Abstract

Based on the fact that nonionic surfactant polyethylene glycol-400 (PEG-400) catalyzed the chemiluminescence of luminol-H2O2 system, a novel and simple chemiluminescence method has been developed for the determination of PEG-400. Under the optimal conditions, the standard curve was Y = 198835X–2091.8, where the correlation coefficient (R) was 0.9999. The detection limit was 4 × 10?5 g·ml?1 PEG-400 and the linear range was 1.0 × 10?4–4.0 × 10?2g·ml?1. The relative standard deviation was 3.2% at 2.0 × 10?3 g·ml?1 PEG-400 (n = 7). This method has been applied to the determination of PEG-400 in cosmetic samples with satisfactory results. Furthermore, the dynamics characteristic curve of PEG-400 in luminol-H2O2 system was compared to typical metallic ion and other surfactants. Moreover, the mechanism of the luminol-H2O2-PEG-400 chemiluminescence system was studied, assisted by fluorescence spectra and UV spectra.  相似文献
