Standard Reference Material 968e Fat-Soluble Vitamins, Carotenoids, and Cholesterol in Human Serum provides certified values for total retinol, γ- and α-tocopherol, total lutein, total zeaxanthin, total β-cryptoxanthin, total β-carotene, 25-hydroxyvitamin D3, and cholesterol. Reference and information values are also reported for nine additional compounds including total α-cryptoxanthin, trans- and total lycopene, total α-carotene, trans-β-carotene, and coenzyme Q10. The certified values for the fat-soluble vitamins and carotenoids in SRM 968e were based on the agreement of results from the means of two liquid chromatographic methods used at the National Institute of Standards and Technology (NIST) and from the median of results of an interlaboratory comparison exercise among institutions that participate in the NIST Micronutrients Measurement Quality Assurance Program. The assigned values for cholesterol and 25-hydroxyvitamin D3 in the SRM are the means of results obtained using the NIST reference method based upon gas chromatography-isotope dilution mass spectrometry and liquid chromatography-isotope dilution tandem mass spectrometry, respectively. SRM 968e is currently one of two available health-related NIST reference materials with concentration values assigned for selected fat-soluble vitamins, carotenoids, and cholesterol in human serum matrix. This SRM is used extensively by laboratories worldwide primarily to validate methods for determining these analytes in human serum and plasma and for assigning values to in-house control materials. The value assignment of the analytes in this SRM will help support measurement accuracy and traceability for laboratories performing health-related measurements in the clinical and nutritional communities.  相似文献

 Proficiency testing is a means of assessing the ability of laboratories to competently perform specific tests and/or measurements. It supplements a laboratory's own internal quality control procedure by providing an additional external audit of their testing capability and provides laboratories with a sound basis for continuous improvement. It is also a means towards achieving comparability of measurement between laboratories. Participation is one of the few ways in which a laboratory can compare its performance with that of other laboratories. Good performance in proficiency testing schemes provides independent evidence and hence reassurance to the laboratory and its clients that its procedures, test methods and other laboratory operations are under control. For test results to have any credibility, they must be traceable to a standard of measurement, preferably in terms of SI units, and must be accompanied by a statement of uncertainty. Analytical chemists are coming to realise that this is just as true in their field as it is for physical measurements, and applies equally to proficiency testing results and laboratory test reports. Recent approaches toward ensuring the quality and comparability of proficiency testing schemes and the means of evaluating proficiency test results are described. These have led to the drafting of guidelines and subsequently to the development of international requirements for the competence of scheme providers. In proficiency tests the selection of an appropriate assigned value is one of the most critical points. Determination of the assigned value for proficiency test can be done in 2 ways: based on reference value and consensus value. The use of such a consensus value as an assigned value is a commonplace in proficiency testing, because there is seldom at present a cost effective alternative. However, the consensus value has a weakness in that it could be significantly different from the true value due largely inexperienced laboratories. Therefore in this work, the reference values were used as the assigned values for all analytes of interest. The reference values in spiked drinking water samples were produced by the Metrology in Chemistry Laboratory of Research Center for Chemistry - Indonesian Institute of Sciences (RCChem-LIPI) with traceable to SI units through standard solutions from the National Institute Standard and Technology, USA. The reference values of cobalt and manganese were obtained by measurement using two from three comparable methods of Graphite Furnace-Atomic Absorption Spectrometry (GF-AAS), Inductively Coupled Plasma-Optical Emission Spectrometry (ICP-OES) and Inductively Coupled Plasma-Mass Spectrometry (ICP-MS) while that for cadmium was obtained by a primary method of ICP-Isotope Dilution Mass Spectrometry (ICP-IDMS).The obtained reference values were used to evaluate the laboratory performance result in a proficiency testing scheme of cadmium, cobalt and manganese in drinking water which was provided in 2014. All fifty nine laboratories reported cadmium and cobalt at low concentration levels which were about more than 82% showed satisfactory performance results. Similarly, the results of manganese were also comparable at high level concentrations.  相似文献

High-quality and reliable laboratory services are important components of effective and well-functioning health systems. Accurate, reliable and timely medical laboratory testing is crucial to patient care and disease surveillance. Unfortunately, in many sub-Saharan African countries, medical laboratory systems are adversely affected by the unavailability of medical laboratories, poor laboratory infrastructure and lack of well-trained personnel [1]. Quality in the laboratory is only achieved in a systematic way through the implementation of a quality management system. The results of the study showed that approximately 60?% of the 78 respondents were unaware of the requirements of ISO 15189:2007. A trial of proficiency testing, termed ??blind proficiency testing??, was carried out in which 19 laboratories determined the concentrations of urea and cholesterol in a proficiency testing material. Of the 19 laboratories that determined the concentration of urea, 63?% produced satisfactory results with scores between ?2 and +2. Similarly, 63?% of the participating laboratories obtained satisfactory z scores for cholesterol determination. Some of the laboratories that obtained satisfactory scores for urea determination had unsatisfactory scores for cholesterol determination and vice versa. It is recommended that the Ghanaian government pass a law and establish a standard to regulate medical laboratories in Ghana in order to improve quality in a significant way.  相似文献

The two most important concepts in metrology are certainly “traceability to standards” and “measurement uncertainty evaluation”. So far the questions related to these concepts have been reasonably solved in the metrology of “classical quantities”, but for the introduction of metrological concepts in new fields, such as chemistry and biology, a lot of problems remain and must be solved in order to support international arrangements. In this presentation, the authors want to develop the strategy implemented at Laboratoire national de métrologie et d’essais (LNE) in metrology in chemistry and biology. The strategy is based on: (1) pure solutions for calibration of analytical instruments, (2) use of certified reference materials (matrix reference materials), and (3) participation to proficiency testing schemes. Examples will be presented in organic and inorganic chemistry. For laboratory medicine, proficiency testing providers play an important role in the organization of External Quality Assessment Schemes. For the time being, the reference value or the assigned value of the comparison is calculated with the results obtained by the participants. This assigned value is not often traceable to SI units. One of the methods suggested by LNE is to ensure the metrological traceability to SI units of the assigned value for the more critical quantities carried on analytes by implementing the Joint committee for traceability in laboratory medicine reference methods.  相似文献

Proficiency testing as a means of external quality assessment plays the role of independent evidence of laboratories’ performance. To enable laboratories to fulfil the requirements stated in legislation, methodology for evaluation of laboratories’ performance in proficiency testing schemes should incorporate principles of measurement results which are fit for intended use and incorporate evaluation of laboratories’ performances based on independent reference value. A proficiency testing scheme was designed to support Drinking Water Directive (98/83/EC) specifically. The methodology for performance evaluation, which takes into account a “fitness for purpose”-based standard deviation for proficiency assessment, is proposed and discussed in terms of requirements of the Drinking Water Directive. A ζ′-score, modified by application of target uncertainty was developed in a way that fulfils requirements defined in the legislation. As an illustration, results are reported for nitrate concentration in water. The approach presented can also be applied to other fields of measurements.  相似文献

Proficiency testing (PT) is an essential tool used by laboratory accreditation bodies to assess the competency of laboratories. Because of limited resources of PT providers or for other reasons, the assigned reference value used in the calculation of z-score values has usually been derived from some sort of consensus value obtained by central tendency estimators such as the arithmetic mean or robust mean. However, if the assigned reference value deviates significantly from the ‘true value’ of the analyte in the test material, laboratories’ performance will be evaluated incorrectly. This paper evaluates the use of consensus values in proficiency testing programmes using the Monte Carlo simulation technique. The results indicated that the deviation of the assigned value from the true value could be as large as 40%, depending on the parameters of the proficiency testing programmes under investigation such as sample homogeneity, number of participant laboratories, concentration level, method precision and laboratory bias. To study how these parameters affect the degree of discrepancy between the consensus value and the true value, a fractional factorial design was also applied. The findings indicate that the number of participating laboratories and the distribution of laboratory bias were the prime two factors affecting the deviation of the consensus value from the true value.  相似文献

Many proficiency tests are operated with a consensus value derived from the participants’ results. Apart from technical issues, one of the reasons often mentioned is that proficiency tests operated with consensus values would be cheaper than those using reference values obtained from a priori characterisation measurements. The economy of a proficiency test must of course be balanced by the need of the participants, and the quality of the comparison in general. The proficiency tests selected in this study had both a reference value and a consensus value, one of which was used for assessing the performance of the participating laboratories. In this work, both a technical and an economical assessment of how the comparisons were operated is made. From the evaluation, it follows that usually the use of consensus values does not necessarily reduce the costs of a proficiency test. Data from proficiency testing can be used to increase our knowledge of the performance of populations of laboratories, individual laboratories and different measurement methods. To support the evaluation and interpretation of results from proficiency testing an error model containing different random and systematic components is presented. From a single round of a proficiency testing scheme the total variation in a population of laboratories can be estimated. With results from several rounds the random variation can be separated into a laboratory and time component and for individual laboratories it is then also possible to evaluate stability and bias in relation to the population mean. By comparing results from laboratories using different methods systematic differences between methods may be indicated. By using results from several rounds a systematic difference can be partitioned into two components: a common systematic difference, possibly depending on the level, and a sample-specific component. It is essential to distinguish between these two components as the former may be eliminated by a correction while the latter must be treated as a random component in the evaluation of uncertainty.  The aim of the International Measurement Evaluation Programme (IMEP) is to give an objective picture of the state-of-the-practice (SoP) of chemical measurements in field laboratories by comparing them to a reference range that contains a value that is as traceable to the SI system of measurements (in this case to the Avogadro Constant, one of the best realizations of the mole so far), as can presently be achieved, but which in any case is independent of human or political decision. Thus, a large-scale field test has been made to realize (international) comparability of these measurements by providing them with an independent scientific common basis. In the third measurement round, IMEP-3, ten trace elements, B, Ca, Cd, Cu, Fe, K, Li, Pb, Rb and Zn were measured in a synthetic and in a natural water by about 155 participating laboratories using their routine methods. The (coded) results are graphically reported and compared to certified reference values, established by IRMM and NIST, using isotope specific methods (isotope dilution mass spectrometry and neutron activation analysis), wherever possible. One of these methods (IDMS) has recently been defined as a primary method of measurement by the Consultative Committee on Amount of Substance (Comité Consultatif pour la Quantité de Matière: CCQM) in its founding meeting in April 1995 at BIPM, Paris. Results indicate a spread of more than ±50%, asymmetrically distributed around the reference range, although the declared accuracy was 5–10%. Self-assessment by participants of their analytical capabilities does not show a high correlation between a self-rating ("more experienced" or "less experienced") and actual performance. In the way they have been applied, all methods seem to produce results of approximately the same quality. There is little reduction in the spread of the measurements if the results obtained for one element in the natural water B are divided by the results obtained for the same element in the synthetic water A (which was unknowingly, a reference material). The candidate reference material (RM) was prepared by freeze-drying pooled urine samples obtained from healthy persons occupationally exposed to toluene. Homogeneity testing yielded no significant differences of urinary concentrations of o-cresol and phenol in 15 randomly selected units of RM. Stability testing showed no significant changes of concentrations of the above analytes over a period of 6 months. Property (pre-certified) values of phenol and o-cresol concentrations and their uncertainties (9.06±0.47 mg l−1 and 1.03±0.06 mg l−1, respectively) were estimated from results of a small-scale interlaboratory comparison in which two laboratories participated using four different analytical methods.  相似文献

The Institute for Chemical Processing of Coal in Zabrze, Poland, organized a proficiency testing round to determine the mass fraction of ash, total sulfur and total carbon and the value of gross calorific value. Forty-four participants from Polish heat and power plants, research entities and independent laboratories reported results. The performance of the participants was evaluated using z and E n scores. Over 85 % of the participants reported satisfactory results indicating the high level of proficiency of the Polish laboratories in the field.  相似文献

Four proficiency tests of environmentally significant elements in soil and water are described. The effects of sample matrix, analyte, sample preparation procedure, extraction method efficiency and instrumental technique on laboratory performance were assessed. Participants’ performance was evaluated using z scores and E n scores. One study included dried sediment and sediment digest as test samples. By testing both dried sediment and sediment digest in the same study, differences in laboratories’ results due to the extraction method or instrumental technique were evaluated. Cadmium and nickel were the elements with the most unsatisfactory z scores for dried sediment compared to sediment digest. The second study had as test samples river water and river water fortified with biosolid. For an evaluation of the efficiency of participants’ extraction method, the results reported for the analysis of river water were compared to the results for analysis of the same river water fortified with biosolid. No consensus could be found between participants’ results for Cr and Ni in this study as the extraction method employed by them produced results that were too variable. The last two studies included saline and fresh water samples. The analysis of fresh and saline water samples with similar analyte concentrations assisted laboratories in developing their methods for trace elements in saline water. For Cr, Pb and Se, the assigned values were reference values traceable to SI, measured using isotope dilution inductively coupled plasma mass spectrometry. Saline water containing low concentrations of analytes presented difficulties to some laboratories. Arsenic and selenium were the analytes most difficult to analyse in all types of environmental samples.  相似文献

Since 1994, when the NIST/NOAA Quality Assurance Program in Chemical Measurements was discussed in Queretaro, CENAM, the National Measurement Institute (NMI) of Mexico, has become involved in the development of reference materials. In the field of biological and environmental reference materials, in particular, the NORAMET collaboration program with NIST and NRC, and the North-American Environmental Cooperation signed among three free-trade treaty organizations, have greatly helped the development of the materials metrology program in the newly established CENAM. This paper describes some particularly significant efforts of CENAM in the development of biological and environmental reference materials, on the basis of inter-comparison studies organized with local and governmental environmental agencies of Mexico. In the field of water pollution CENAM has developed a practical proficiency testing (PT) scheme for field laboratories, as a part of registration by local government in the metropolitan area, according to the Mexican Ecological Regulation. The results from these eight PTs in the last 5 years have demonstrated that this scheme has helped ensure the reliability of analytical capability of more than 50 field laboratories in three states, Mexico, D.F., and the States of Mexico and Queretaro. Similar experience has been obtained for more than 70 service units of stack emission measurements in the three states in 1998 and 1999, as a result of the design of a PT scheme for reference gas mixtures. This PT scheme has been accomplished successfully by 30 analytical laboratories who provide monitoring services and perform research on toxic substances (Hg, methylmercury, PCB, etc.) in Mexico. To support these activities, reference samples have been produced through the NIST SRMs, and efforts have been made to increase CENAM's capability in the preparation of primary reference materials in spectrometric solutions and gas mixtures. Collaboration among NMIs has also successfully overcome the inability of CENAM to prepare biological tissue for mercury assessment and marine sediments for analysis of trace metals. The importance of international collaboration is stressed not only in the NORAMET region but also in the SIM, to provide help for each other and achieve mutual recognition among member countries of the region.  相似文献

A proficiency testing (PT) scheme was developed for a limited number of analytical laboratories participating in the analysis of natural water in Israel. Three fit-for-intended-use reference materials (RMs) were prepared for a pilot PT from natural water: RM-blank and two in-house reference materials (IHRMs) with seven analytes fortified to achieve different levels of analyte concentrations. The measurands for the PT participants were the traceable spike values certified in the IHRMs, i.e., added mass concentrations of the analytes. The RM-blank and IHRMs were found to be homogeneous and stable over 11 days, the time necessary for the experimental part of the PT. The RMs were distributed to the PT participants as unknown test items similar to routine samples. The test results were evaluated using their deviations from the IHRM-certified values. Eight Israeli laboratories took part in the interlaboratory comparison. Individual laboratory performance and metrological compatibility of the PT results of the participants, as a local group of laboratories, were evaluated for every analyte.  相似文献

Current methods of testing laboratories for their proficiency in reporting correct measurement results are liable to substantial errors of the second kind. This means that laboratories with deflated uncertainties are accepted as proficient, even though their reported measurement results pave the way for erroneous conclusions. Only by using E n numbers based on an accepted reference value with the lowest possible uncertainty can the risk of recognising incorrect measurement results be kept at an acceptable level. Based on an actual set of proficiency test (PT)-data for the concentration of Pb in water, this paper compares PT results obtained by methods using E n numbers with methods based on z-scores. Immunochemical methods (in particular immunoassays) have been applied to spring and surface water samples, respectively, which were set-up as reference materials (RM) within two proficiency testing campaigns. For the first set of proficiency tests (PTs) described here (which were actually the second round of PTs organized, spring 2005), three ELISAs (enzyme-linked immunosorbent assays) were employed in the enzyme tracer format for isoproturon, diuron, and atrazine, respectively. Results were evaluated in comparison with conventional reference methods (LC, GC). Based on their Z-score laboratory performances, the results for isoproturon and diuron were satisfactory, both for fortified spring water and for the blind solution. The results for atrazine were strongly influenced by other triazines present and needed detailed interpretation. For the second set of PTs described here (which were actually the third round of PTs organized, spring 2006), two ELISAs in the coating antigen format were used for isoproturon and diuron, and the result was included with the results obtained by conventional methods during the PTs. The results (the Z-scores) for isoproturon were again classified as satisfactory, in both fortified surface water and blind solution. The results for diuron in ELISA showed an influence of the water matrix, while the analysis of the blind solution was satisfactory. In addition, an ELISA in the enzyme tracer format was applied to analyze isoproturon, diuron, and atrazine in surface water samples, which had been set-up and spiked during a field trial (tank experiment) at the Maas River at Eijsden, The Netherlands. The immunoassay results were compared with those from an in-house on-line SPE LC/MS–MS used as reference. Although the immunochemical results were sometimes higher than those determined in the reference analysis, the general concentration trends in the samples were similar. The contribution of immunochemical methods to the implementation of the European Water Framework Directive is also discussed.  相似文献

 This paper presents the result of an investigation concerning which areas of EN 45001 are considered as especially problematic by small laboratories (<10 employees). The investigation was performed by distributing questionnaires to European laboratories. To be able to differentiate between areas considered as problematic only by small laboratories and areas considered as problematic by laboratories in general, some questionnaires sent in by larger (>10 employees) laboratories were used. The determination of measurement uncertainties within reasonable efforts and the requirements to take part in round robin and proficiency testing were considered problematic by laboratories of all sizes. Training is an area where small laboratories have specific problems and so are the requirements for the identification of all equipment. Other problematic areas for small laboratories are areas were there are needs for personnel, e.g. appointing supervising personnel and expenditure for internal audits. The last area to be considered as problematic is matters concerning documentation. Finally some advice concerning assessment of small laboratories is given. The interlaboratory variability in the quantification of adsorbable organically bound halogens (AOX) in industrially contaminated soil is presented. Three consecutive rounds of a proficiency testing scheme, in which between 88 and 119 routine laboratories participated, yielded relative reproducibility standard deviations between 7 and 20% at AOX contents between 10.9 and 268 mg kg−1. Nineteen laboratories with established proficiency were invited to participate in the certification of the AOX content in three soil reference materials meant for the internal quality control in analytical laboratories. The certified values are (1349 ± 59) mg kg−1, (80 ± 7) mg kg−1 and (102 ± 8) mg kg−1, respectively.  相似文献
