Treatment of 2, 6-bis-aminomethyladamantane-2, 6-diol (II) with nitrous acid gives tricyclo[4.4.1.13, 8]dodecane-4, 9-dione (III), which is converted into the tricyclo[4.4.1.13, 8]dodeca-4, 9-diene (V) by reduction to the corresponding diols (IV, R = H) and pyrolysis of their O-4-methylphenyl thiocarbonate O-esters. II is accessible from 2, 6-adamantanedione by reaction with diethyl aluminium cyanide and subsequent reduction with lithiumaluminium hydride. The physical properties of the new compounds including 13C NMR. spectra of III, V and tricyclo-[4.4.1.13, 8]dodecane are given.  相似文献

A number of 8-hydroxy-6-methyl-1,6-naphthyridin-5(6H)-one-7-carboxylic acid alkyl esters 3 and the isomeric 5-hydroxy-7-methyl-1,7-naphthyridin-8(7H)-one-6-carboxylic acid alkyl esters 4 were synthesized from acyclic precursors obtained starting from quinolinic anhydride 5. Thus, methanolysis of 5 afforded the hemiester 6 which treated with oxalyl chloride and sarcosine ethyl ester gave 3-(N-ethoxycarbonylmethyl-N-methylcarbamoyl)pyridine-2-carboxylic acid methyl ester 8. Compound 8 was cyclized to naphthyridines 3a-e with sodium alkoxides. The isomeric naphthyridines 4a-c were obtained by cyclization of the open intermediary 2-(N-ethoxycarbonylmethyl-N-methylcarbamoyl)pyridine-3-carboxylic acid methyl ester 9 obtained by a route that involves treatment of 5 with sarcosine ethyl ester and esterification with diazomethane. Spectroscopic properties (1H nmr, uv, ir) of compounds 3 and 4 are discussed and confirmed the proposed structures.  相似文献

A series of new tricyclic fluoroquinolones was prepared by replacing fluorine atoms in derivatives of 2-R-8-Y-7-oxo-9,10-difluoro-7H-1,3,4-thiadiazino[6,5,4-i,j]quinoline-6-carboxylic acids. In acids and esters containing a hydrogen atom in position 8 occurred replacement of F10 by amine rests, and in compounds with a fluorine in position 8 was substituted either F8or F10 and F8depending on the amine character.  相似文献

A series of new 3-(aminomethyl)pyrazoles and 3-(aminomethyl)isoxazoles was synthesized along a route involving the formation as key intermediates of esters of 5-substituted 1H-pyrazole-3-carboxylic and 1H-isoxazole-3-carboxylic acids. N‐(3‐Ferrocenyl‐2‐naphthoyl) dipeptide ethyl esters 1–4 and N‐(6‐ferrocenyl‐2‐naphthoyl) dipeptide ethyl esters 5–8 were prepared by coupling either 3‐ferrocenylnaphthalene‐2‐carboxylic acid or 6‐ferrocenylnaphthalene‐2‐carboxylic acid to the dipeptide ethyl esters GlyGly(OEt) (1, 5), AlaGly(OEt) (2, 6), GlyPhe(OEt) (3, 7) and GlyLeu(OEt) (4, 8), using the standard N‐(3‐dimethylaminopropyl)‐N'‐ethylcarbodiimide hydrochloride, 1‐hydroxybenzotriazole protocol. Electrospray ionization mass spectrometry (ESI‐MS) and laser desorption ionization mass spectrometry (LDI‐MS) were employed in conjunction with tandem mass spectrometry in the analysis of N‐(3‐ferrocenyl‐2‐naphthoyl) dipeptide ethyl esters 1–4 and N‐(6‐ferrocenyl‐2‐naphthoyl) dipeptide ethyl esters 5–8. Radical cations, [M]+? and [M + H]+ species were both observed in the mass spectra. Intense sodium [M + Na]+ and potassium [M + K]+ adducts were also present. An important diagnostic ion at m/z [M–65]+ was observed in both the MS and MS/MS spectra of the N‐(3‐ferrocenyl‐2‐naphthoyl) dipeptide derivatives. Sequence‐specific ions were generally not observed in the MS/MS spectra of the N‐(3‐ferrocenyl‐2‐naphthoyl) series due to formation of the diagnostic [M–65]+ ion. Sequence‐specific ions were observed in the MS/MS spectra of the N‐(6‐ferrocenyl‐2‐naphthoyl) dipeptide esters with charge retention on the derivatized N‐terminal of the dipeptide. The 13C NMR spectra of a number of pyridazine derivatives have been recorded in DMSO-d6 solution and analysed. Examination of the most diagnostic resonances, with particular emphasis on those arising from the pyridazine ring system, enabled the ready establishment of the presence of a ring-chain tautomerism in 5-(o-aminophenylcarbamoyl)pyridazine-4-carboxylic acid, methyl 5-(o-aminophenylcarbamoyl)pyridazine-4-carboxylate, 5-(o-aminophenylcarbamoyl)-3,6,-dimethylpyridazine-4-carboxylic acid and 5-(2-amino-1,2-dicyanovinylenecarbamoyl)pyridazine-4-carboxylic acid. This gave rise to 3′,4′-dihydro-3′-oxospiro[pyridazine-5(2H),2′(1H)-quinoxaline]-4-carboxylic acid, methyl 3′,4′-dihydro-3′oxospiro[pyridazine-5(2H),2′(1′H)-quinoxaline]-4-carboxylate, 3′,4′-dihydro-3′-oxo-3,6-dimethylspiro[pyridazine-5(2H), 2′(1′H)-quinoxaline]-4-carboxylic acid and 5-oxo-2,3-dicyano-1,4,8,9-tetraazaspiro[5.5]undeca-2,7,10-triene-11-carboxylic acid, respectively.  相似文献

A synthesis of 12-oxa-3,5,9,10-tetrachlorohexacyclo[5.4.1.02,6.03,10.05,9.08,11]dodecane-4-one (6) from 4,4-dimethoxy-2,3,5,6-tetrachloropentacyclo [[5.4.0.02,6.03,10.05,9]undecane-8,11-dione (1) is described. Reaction of 6 with sodium hydroxide in refluxing benzene, toluene, or tetrahydrofuran affords 11-oxa-3,4,5-exo-6-tetrachloropentacyclo [[6.2.1.02,7.04,10.05,9]undecane-endo-3-carboxylic acid (7a, 80·2% yield). The corresponding reaction of 6 with refluxing aqueous sodium hydroxide solution affords 4,12-dioxa-8,11-dichlorohexacyclo-[5.4.1.02,6.03,10.05,9.08,11]dodecane-1-carboxylic acid (8a, 66·5% yield). A mechanism which accounts for the formation of 7a and 8a from 6 is presented.  相似文献

The Gewald syntheses were employed to prepare a series of 2-amino-3-carboethoxythiophenes, and the syntheses of two of these, namely, the 3,4-trimethylene ( 1f ) and 3,4-tetramethylene ( 1g ) derivatives, were examined in detail. In two preparations of 1f , octahydro-6a-(4-morpholinyl)-2-thioxocyclopenta[b]pyrrole-3-carboxylic acid ( 7 ) was a co-product. The structure of 7 was ascertained from its 300 MHz 1H nmr and 13C nmr spectra, and by its conversion to 1,4,5,6-tetrahydro-2-mercaptocyclopenta[b]pyrrole-3-carboxylic acid ethyl ester ( 8 ). Isolation of 7 and other observations led to postulated mechanisms for three of the Gewald thiophene syntheses.  相似文献

Starting from 2-bromo-9-oxo-fluorene-1-carboxylic acid the biangular bis-indenofluorene 14, 15-dihydro-13H-diindeno[2, 1-a; 1′, 2′-1]fluorene (VIII) and the monoangular 14, 15-dihydro-8H-diindeno[2, 1-a; 2′, 1′-h]fluorene (XI) have been synthesised in 6 resp. 7 steps (overall yield 22% resp. 18%). A series of thidiazole derivatives (4, 7) from pyrazole-3-carboxylic acid chloride (2) and pyrazole-3,4-dicarboxylic acid chloride derivatives (6) were synthesized and characterized. The structures of the new compounds were confirmed byelemental analysis, NMR (1H and 13C) and IR spectra. The molecular and crystal structure of 4-benzoyl-N-[5-(methylthio)-1,3,4-thiadiazol-2-yl]-1,5-di-phenyl-1H-pyrazole-3-carboxamide(4d)was determined by single crystal X-ray diffraction method.  相似文献

A new procedure for the cyclization of 2-indolecarbohydrazones (5) to 1,2,3,4-tetrahydro-4-oxo-5H-pyridazino[4,5-b]indoles (6) and for the cyclization of 3-indolecarbohydrazones (7) to 1-oxo-1,2,3,4-tetrahydro-5H-pyridazino[4,5-b]indoles (8 and 9) is described. The hydrazones (5 or 7) were treated with an acyl halide (acetyl or benzoyl chlorides) and triethylamine in ethyl acetate of chloroform as solvents to give the compounds 6 (20–70%) from the compounds 5 , and the compounds 8 (20–60%) from the compounds 7 . Through refluxing with ethanol-hydrochloric acid the compounds 8a-8f selectively separate the acetyl group on N5 to give the respective compounds, 9a-9f. The ir and 1H-nmr spectra of all the compounds 5, 6, 7, 8 and 9 and the uv, mass and 13C-nmr spectra of the compounds 7h, 7i, 8h and 8i are discussed.  相似文献

The condensation of the acetylmethylene group in the tert-butyl esters of 7Z-acetylmethylene-3-methyl-3-cephem-4-carboxylic acid and 7Z-acetylmethylene-3-methyl-1,1-dioxo-3-cephem-4-carboxylic acid and in 7Z-acetylmethylene-3-methylene-1,1-dioxo-3-cephem with arylmethoxyamines and O-alkylation of the tert-butyl ester of 7Z-(2-hydroxyimino)propylidene-3-methyl-1,1-dioxo-3-cephem-4-carboxylic acid using substituted benzyl bromides as well as pyridylmethyl chlorides gave arylmethoxyimino and pyridylmethoxyimino derivatives of these compounds in the syn and anti isomeric forms. The Vilsmaier reagent was used to introduce the N,N-dimethylaminomethylene group at C-2 of the cephem system in the tert-butyl esters of 7Z-[2-(arylmethoxyimino)propylidene]-3-methyl-1,1-dioxo-3-cephem-4-carboxylic acid. Subsequent transformation of the N,N-dimethylaminomethylene cephems using hydroxylamine led to 3Z-[2-(anti-arylmethoxyimino)propylidene]-tert-butoxycarbonylmethyl-4-(5-methyl-4-isoxazolylsulfonyl)- azetidin-2-ones. Condensation of the acetyl group in the tert-butyl ester of 7Z-acetylmethylene- 3-methyl-1,1-dioxo-3-cephem-4-carboxylic acid with 4-bromophenylhydrazine gave a cephem with a 2-(4-bromophenylhydrazono)propylidene group at C-7. Acylation of the tert-butyl ester of 7Z-(2-hydroxyimino)propylidene-3-methyl-1,1-dioxo-3-cephem-4-carboxylic acid by 2-bromobenzoyl chloride gave a cephem with a 2-(2-bromo-benzoyloxyimino)propylidene group at C-7. Biological screening of these products towards to malignant and normal cells in vitro showed that their antitumor activity and cytotoxic selectivity towards to malignant and normal cells depend on the structure and configuration of the arylmethoxyimino and pyridylmethoxyimino groups in the 7-alkylidene substituent as well as on the presence or absence of N,N-dimethylaminomethylene and carboxyl groups, respectively, at C-2 and C-4 of the cephem system.  相似文献

The synthesis of a new series of P-heterocyclic compounds, substituted 2-aminomethyl-2-oxo-2λ5-perhydro-[1,3,2]oxazaphospholo[3,4-a]pyridine derivatives 8(a-j), was accomplished. A key intermediate, 2-(chloromethyl)-2-oxo-2λ5-perhydro-[1,3,2]oxazaphospholo[3,4-a]pyridine (6) was primarily synthesized by the condensation of (±)-2-piperidinemethanol (4) and chloromethylphosphonic dichloride (5); subsequently, it was treated with various heterocyclic amines/benzylamines/aminoacid esters, 7(a-j) to obtain the desired products. The structures of the newly synthesized compounds were elucidated by 1H, 13C, and 31P NMR spectroscopy, mass spectra and elemental analyses. The biological potency of title products was investigated by screening in vitro antimicrobial activity. The bio-screening data revealed that most of the synthesized derivatives showed potent growth of inhibition against fungi while compared with bacteria. Particularly, compounds 8c and 8i against bacterial strains, and 8a and 8f against fungi exhibited promising activity.  相似文献

A new series of t-4-aryl-3,c-6-dihydroxy-6-methyl-4,5,6,7-tetrahydro-1H-indazole-r-5-carboxylic acid isopropyl esters has been synthesized by adopting a conventional method from cyclic β-keto esters. 1H, 13C NMR, and IR spectra for all the compounds were investigated. HMBC, HSQC, COSY, and NOESY spectra of the representative compounds were studied. The stereochemistry of a six-membered ring of the fused indazoles resembled that of keto esters. From the HMBC correlations the indazole structure was confirmed as 1H-indazole.  相似文献

3-exo-Aminobicyclo[2.2.1]hept-5-ene-2-exo-carboxylic acid and ethyl 3-endo-aminobicyclo[2.2.1]heptane-2-endo-carboxylate ( 6 ) were reduced with lithium aluminum hydride to the corresponding bicyclic aminoalcohols 3 and 4 . These and the saturated endo-endo and exo-exo N-methylaminoalcohols 16 and 22 , respectively, were converted to methylene-bridged tetrahydro- ( 11 ) and hexahydro-3,1-benzoxazin-2-ones 12, 17, 23 and 3,1-benzoxazin-2-thiones 13, 14, 18, 24 . The exo-exo 3 and endo-endo 4 aminoalcohols were cyclized by means of ethyl arylimidates to tricyclic dihydro-1,3-oxazines 7a-d, 8a-d . The structures were confirmed by ir, 1H and 13C nmr spectroscopy.  相似文献

Ethyl esters of 1-(7-Z-1-ethyl-6-fluoro-4-oxo-1,4-dihydroquinoline-3-carbamoyl)-5-X-6,7,8-trifluoro-4-oxo-1,4-dihydroquinoline-3-carboxylic acids (X = H, F; Z = pyrrolidino-, piperidino-, hexamethylenimino-, morpholino-, thiomorpholino-) have been synthesized by the interaction of quinolone-3-carboxylic acid hydrazides with ethyl esters of 3-ethoxy-2-(polyfluorobenzoyl)acrylic acid . It was shown possible to cyclize intramolecularly the esters obtained with the formation of 1,3,4-oxadiazino[6,5,4-i,j]quinoline derivatives.  相似文献
