An efficient and versatile synthesis of aminophosphine phosphinite (AMPP) ligands derived from ephedrine, with possible stereogenic P(III)-center(s) is described, using the borane complex methodology. The reaction of oxazaphospholidine borane with an organolithium reagent, leads to the formation of the ring-opened product, which is trapped by a chlorophosphine (borane), to afford the corresponding aminophosphine phosphinite boranes in good yields. Treatment of the borane complexes with dabco, gives the corresponding aminophosphine phosphinite ligands in 70–90% yield. These ligands are used for the preparation of Rh catalysts applied to the asymmetric hydrogenation of methyl α-acetamidocinnamate yielding the phenylalanine derivative with (R) 22% to (S) 99% e.e. These results show the importance of the structural modification at the P-stereogenic center(s), which could either amplify or cancel out the asymmetric induction resulting from the ephedrine backbone, for enantioselective catalysis.  相似文献

The efficient synthesis of chiral or achiral tertiary phosphines bearing an o-bromo (or iodo)aryl substituent is described. The key step of this synthesis is based on the reaction of a secondary phosphine borane with the 1,2-dibromo (or diiodo)arene, owing to the formation in situ of an aryne species in the presence of n-butyllithium. When P-chirogenic secondary phosphine boranes were used, the corresponding o-halogeno-arylphosphine boranes were obtained without racemization in moderate to good yields and with ee up to 99%. The stereochemistry of the reaction, with complete retention of the configuration at the P atom, has been established by X-ray structures of P-chirogenic o-halogenophenyl phosphine borane complexes. The decomplexation of the borane was easily achieved without racemization using DABCO to obtain the free o-halogeno-arylphosphines in high yields.  相似文献

The stereoselective synthesis of P-chirogenic chlorophosphine boranes 4 was investigated by HCl acidolysis of the corresponding aminophosphine boranes 10. The reaction afforded the P-N bond cleavage with inversion of the configuration at the phosphorus center, leading to the chlorophosphine boranes 4 with high to excellent enantiomeric purities (80-99% ee), except in the case of the chloro-1-naphthylphenylphosphine borane 4d. Reaction conditions and workup significantly influence the enantiomeric purity of the product, with the exception of the o-anisyl- and o-tolylchlorophenylphosphine boranes, 4b and 4c, which were found to be particularly stable even after purification by chromatography on silica gel. Reaction of the chlorophosphine boranes 4 with various nucleophiles, such as carbanions, phenolates, thiophenolates, or amides, afforded the corresponding organophosphorus borane complexes via P-C, P-O, P-S, and P-N bond formation, respectively, in 34-93% yield and with up to 99% ee. This work demonstrates the importance of chlorophosphine boranes 4 as new and powerful electrophilic building blocks for the highly stereoselective synthesis of P-chirogenic organophosphorus compounds.  相似文献

An efficient enantioselective strategy for the synthesis of variously substituted phosphine oxides has been developed, incorporating the use of (1S,2S)‐2‐aminocyclohexanol as the chiral auxiliary. The method relies on three key steps: 1) Highly diastereoselective formation of PV oxazaphospholidine, rationalized by a theoretical study; 2) highly diastereoselective ring‐opening of the oxazaphospholidine oxide with organometallic reagents that takes place with inversion of configuration at the P atom; 3) enantioselective synthesis of phosphine oxides by cleavage of the remaining P?O bond. Interestingly, the use of a PIII phosphine precursor afforded a P‐epimer oxazaphospholidine. Hence, the two enantiomeric phosphine oxides can be synthesized starting from either a PV or a PIII phosphine precursor, which constitutes a clear advantage for the stereoselective synthesis of sterically hindered phosphine oxides.  相似文献

The novel P-chirogenic anisylphenylMOP derivatives (R,R) and (R,S)-2-(anisylphenylphosphino)-2′-methoxy-1,1′-binaphthyl (10a and b) have been synthesized and their corresponding oxides characterised by X-ray crystallography. The results of a parallel screening regimen with various reducing agents highlight the sensitivity of the tertiary phosphine oxides to epimerisation and, interestingly, reveal that the PO, O-CH3 and P-C6H5 bonds can all be cleaved selectively depending on the reducing agents employed. An alternative synthesis was provided by direct coupling of the secondary phosphine with (R)-methoxytriflate 4, which led to the isolation of the optically pure P-chirogenic phosphines via their borane adducts. A brief study of the coordination chemistry of 10a with different rhodium precursors, relevant to the catalytic asymmetric addition of boronic acids to aldehydes is also reported.  相似文献

Following our recent total synthesis of the biologically potent natural products otteliones A and B in racemic form, we have now accomplished the total synthesis of both the enantiomers of otteliones A and B through an enantiodivergent strategy emanating from the readily available Diels-Alder adduct of cyclopentadiene and p-benzoquinone. These endeavors have led to the elucidation of the absolute configuration of naturally occurring otteliones A and B.  相似文献

Oxadiazinanones derived from (1R,2S)-ephedrine and (1R,2S)-norephedrine were employed in the asymmetric α-halo aldol reaction. The optimized yields and diastereoselectivities for the ephedrine based oxadiazinanone aldol reaction ranged from fair to good. The ephedrine based aldol adducts were hydrolyzed to afford the α-bromo-β-hydroxycarboxylic acids. The absolute stereochemistry and enantiomeric purity of these products were determined by chiral HPLC and specific rotation measurements.  相似文献

The lipase-promoted kinetic resolution of a series of alkoxy(hydroxymethyl)phenylphosphine P-boranes proceeded with moderate stereoselectivity to give both the unreacted substrates and their O-acetyl derivatives. The absolute configurations of the products, which were earlier ascribed on the basis of the stereoselective reduction of the corresponding phosphine oxides with borane and comparison with the literature data concerning bicyclic phosphine oxides, were disputed by theoretical calculation. Some additional studies were carried out, including theoretical calculations and more accurate chemical correlation, which proved that the borane reduction of acyclic phosphine oxides proceeded with inversion of configuration at the phosphorus center and, therefore, the former assignment of the absolute configurations was incorrect. On this basis, the stereochemistry of the enzymatic reaction was ultimately determined. A mechanism of the borane reduction of acyclic phosphine oxides explaining inversion of configuration at phosphorus is proposed.  相似文献

The first asymmetric synthesis of P-stereogenic 2-hydroxyarylphosphine ligands is described, using borane complexation methodology. This synthesis is based on the highly stereoselective preparation of bromoarylphosphinite boranes, leading to the 2-hydroxyarylphosphine derivatives, by an intramolecular ortho Fries-like rearrangement mediated in basic conditions. The o-anisyl-2-hydroxynaphthylphenylphosphine borane has been decomplexed in EtOH, affording the P(III)-stereogenic hydroxyarylphosphine ligand with 84% yield. The interest of the hydroxyarylphosphine borane is also demonstrated by the preparation of a new class of phosphine-phosphinite ligands, by trapping the rearrangement products first with chlorodiphenylphosphine, Ph2PCl, then with borane. The corresponding phosphine-phosphinites are obtained and purified as diborane complexes, with the decomplexation of these borane complexes being achieved by heating with dabco, to afford the free hybrid ligands with retention of the configuration at the P-atom (isolated yield up to 53%).  相似文献

Abstract

The synthesis and the complex formation of chiral heterotopic ligands with two different binding sites each capable of bonding a different type of metal are described. The action of bis(dimethylamino) aryl phosphine on (S)-(+)-prolinol gives rise to the kinetic stereoisomer (2S,4S)-2-phenyl-1,3,2-oxazaphospholidine 1 and the thermodynamic stereoisomer (2R,4S)-2-phenyl-1,3,2-oxazaphospholidine 2. 1 is then totally converted into 2 at the end of the reaction. The Michaelis Arbuzov reaction of 2 with benzylbromide affords (Rp)-benzylaryl-(2-(S)-bromomethyl pyrrolidine-1-yl) phosphine oxide 3 in 80% yield. Compound 3 is an ideal chiral precursor for the synthesis of chiral hybrid phosphine-phosphine oxide ligands. The bromide atom is smoothly displaced by lithium diphenylphosphide to afford in 80% yield (Rp)-benzylaryl-(2-(S)-diphenylphosphinomethylpyrrolidine-1-yl) phosphine oxide 5. These reaction are proved to be totally stereoselective : the Michaelis Arbuzov reaction does not change the configuration at C2 in the proline ring. The relative configuration (Rp) of 5 (aryl = phenyl) was determinated by X-ray diffraction, from the known configuration of (S)-(+)-prolinol.  相似文献

(E)-Arylaldehyde oxime ethers bearing a (1S)-2-hydroxy-1-phenylethyl or (2R)-1-hydroxy-2-phenylethyl group as a chiral auxiliary, both derived from a single precursor, methyl (R)-mandelate, underwent nucleophilic addition with organolithium reagents via six-membered chelates to give the diastereomerically enriched (R)- and (S)-adducts, respectively, which, after chiral auxiliary removal by reductive N-O bond cleavage, led to the corresponding (R)- and (S)-1-(aryl)ethylamines. This organolithium addition protocol using methyllithium was applied in an enantiodivergent fashion to the preparation of both enantiomers of 1-(2-hydroxyphenyl)ethylamine, which has been previously used as an efficient chiral auxiliary for the synthesis of natural products in this laboratory. The synthetic utility of this methodology involving diastereoselective methyl addition was demonstrated by further application to the asymmetric synthesis of a new type of calcium receptor agonist (calcimimetics), (R)-(+)-NPS R-568 and its thio analogue. Furthermore, diastereoselective vinylation was accomplished by application of the hydroxy oxime ether-based protocol using vinyllithium, which allowed the development of the enantioselective synthesis of the NK-1 receptor antagonists, (+)-CP-99,994 and (+)-CP-122,721.  相似文献

The synthesis of P-benzylic products by reaction of anions stabilised by N-phosphorylphosphazenyl, N-methoxycarbonylphosphazenyl, phosphine borane complex, and phosphine oxide groups by displacement of hydrogen of a variety of electron-deficient benzene derivatives is described. Lithium phosphazenes were the most suitable nucleophiles for the substitution of hydrogen in nitrobenzene and some ortho-, meta-, and para- substituted nitrobenzenes. Lithiated phosphine borane complexes produced efficiently the substitution of the hydrogen at the para position of a cyano group in cyanobenzenes, whereas the anion of ethyldiphenylphosphine oxide lead to complex mixtures with all electrophiles assayed. The method reported here represents a convenient alternative to the vicarious nucleophilic substitution for the synthesis of benzylic phosphorus derivatives using phosphorus-stabilised anions that do not bear a leaving group at the carbanionic centre.  相似文献

Some aromatic ketones have been reduced by borane or by catecholborane using oxazaborolidine as a catalyst. It has been found that, when borane is used, the enantiomeric excess of alcohol produced decreases as the substitution on the ortho position of benzene ring increases. The diastereoselective synthesis of a P-chirogenic β-aminophosphine ligand with carbon–carbon bond formation of the ethano bridge in a 3:1 ratio via reaction of an α-metallated P-chirogenic phosphine borane with a benzaldimine is described. The diastereoselectivity is attributed to a transition state where the lithium cation chelates the phosphine borane carbanion and the nitrogen of the imine and the attack of the CN occurs on the face opposite to the P–B bond, due to its interaction with the antibonding P–B bond. The major diastereoisomeric β-aminophosphine borane was then separated and decomplexed into the corresponding β-aminophosphine under neutral conditions and without epimerization by heating at reflux in EtOH. This synthesis offers a short hemisynthetic route to the P-chirogenic β-P,N-ligands, by bridge formation starting from methylphosphine boranes.  相似文献

The first total syntheses of (−)-dysibetaine CPa and the antipode have been achieved using enantioselective solvolysis of meso-cyclic anhydride mediated by quinine derivative as an organocatalyst. The synthesis features a demonstration of an enantiodivergent organic synthesis of both enantiomers of dysibetaine CPa whereby the absolute configurations of natural product were elucidated as (3R,4R). The synthesis of benzylisopropylphenylphosphine by reaction between the benzylphenylphosphide anion and isopropyl chloride and its subsequent resolution by means of an optically active palladium metallacycle is reported. The synthesis of the organometallic complex [Pd(η3-2-MeC3H4)Cl(PBniPrPh)] is also described, as well as the assignment of the absolute configuration of the coordinated phosphine by mono- and bidimensional proton NMR spectra, using the homochiral palladacycle as a reference point. In order to estimate the height of the energy barrier corresponding to the rotation of the phosphine ligand around the PdP bond, several calculations were performed at the semiempirical PM3(tm) level.  相似文献
