用膜片钳研究CuCl2对萝卜液泡膜SV通道的影响   总被引:3,自引:0,他引:3  
用膜片钳全液泡记录方式研究了CuCl2对萝卜SV通道的影响.研究表明,SV通道为阳离子选择性通道,细胞质钙可激活此通道,且其表现出钙依赖性.细胞质钙达2mmol/L时,SV通道电流基本上达到饱和.当在外液中加入不同浓度的CuCl2时,SV通道电流有明显的变化.高浓度的铜离子对通道电流有抑制作用,而低浓度(<0.1mmol/L)则增强通道电流.电极内液中的CuCl2对SV通道电流也有抑制作用.这为进一步研究铜离子对植物生理活动的影响以及铜肥对农作物的作用在通道水平上提供重要的依据.  相似文献

The effects of BaCl2 on slow vacuolar (SV) currents of radish are studied by using the whole-vacuolar patch-clamp recording mode. The Ca2+-dependent SV channel can be activated by cytosolic Ca2+. When 1 mmol/L BaCl2 is added into pipette solution, SV currents are suppressed remarkably. Then adding BaCl2 of different concentrations into the bath solution, SV currents reflect different effects. The results show that BaCl2 with a lower concentration (<3 mmol/L) promotes the channel currents and the currents are saturated when BaCl2 concentrations are between 1 μmol/L and 1 mmol/L, but BaCl2 with higher concentration (≥ 3 mmol/L) inhibits SV currents.  相似文献

用膜片钳全液泡记录方式研究了Mg2+对萝卜(Raphanus sativus L.)液泡膜上SV通道电流的影响. 结果表明: 用EGTA配位Ca2+后, 胞质Mg2+不能够代替Ca2+来激活SV通道; 外液中不同浓度的Mg2+对通道电流有抑制作用, 并且呈一定的浓度依赖性, 用Hill方程拟合浓度依赖性曲线, 得抑制常数Ki=(1.94±0.11) mmol/L, 而内液中的Mg2+不影响通道电流. 这一结果为进一步研究镁对植物生理活动的影响从通道水平提供了重要依据.  相似文献

The paper presents results of investigation of exchange of the clinoptilolite tuff cations with hydrogen ions from HCl solution of concentration 0.1 mmol cm(-3) and ammonium ions solutions of concentrations 0.0071 to 2.6 mmol cm(-3). Molal concentrations, x (mmol g(-1)) of cations exchanged in acid solution and in ammonium ions solutions were compared with molal concentrations of cations obtained by determination of the cation-exchange capacity of clinoptilolite tuff. The obtained results show that at ammonium ion concentrations lower than 0.1 mmol cm(-3), with regard to exchange capacity for particular ions, best exchanged are Na+ ions, followed by Mg2+ and Ca2+ ions, while exchange of K+ ions is the poorest (Na+ > Mg2+ > Ca2+ > K+). At ammonium concentrations from 0.2 to 1 mmol cm(-3) the order is Na+ > Ca2+ > Mg2+ > K+. At concentrations higher than 1 mmol cm(-3) the order is Na+ > Ca2+ > K+ > Mg2+. The results are a consequence of the uptake of hydrogen ions by zeolite samples in ammonium ions solutions at concentrations lower than 1 mmol cm(-3) and indicate the importance of Mg2+ (besides Na+ ions) for the exchange between clinoptilolite cations and H+ ions, in contrast to K+ ions, whose participation in the reaction with H+ ions is the lowest. During decationization of the clinoptilolite in acid solution, best exchanged are Na+, Mg2+, and Ca2+ ions, while exchange of K+ ions is the poorest. Due to poor exchange of K+ and H+ ions and good exchange of Na+, Mg2+, and Ca2+ ions, it is to be assumed that preservation of stability of the clinoptilolite structure is caused by K+ ions present in the channel C. Clinoptilolite is dissolved in the clinoptilolite A and B channels where Na+, Mg2+, and Ca2+ ions are present. On the acid-modified clinoptilolite samples, exchange of ammonium ions is poorer than on natural zeolite. The longer the contact time of the zeolite and acid solution, the worse ammonium ions exchange. It can be assumed that H+ ions exchanged with zeolite cations are consumed for solution of aluminum in the clinoptilolite structure; therefore the concentration of H+ ions as exchangeable cations decreases. In the ammonium ion solution at a concentration of 0.0065 mmol cm(-3), from the acid-modified zeolite samples, Al3+ ions are exchanged best, followed by Na+, Mg2+, Ca2+, and K+ ions. Further to the results, it is to be assumed that exchangeable Al3+ ions available from clinoptilolite dissolution are best exchanged with H+ ions in acid solution.  相似文献

Whether La3+ can enter human peripheral blood lymphocytes by the Na+/Ca2+ exchanger or not and the effect of La3+on the Na+/Ca2+ exchanger activity are examined by fura-2 technique. And that whether La3+ is sequestered by intracellular organelles (mainly endoplasmic reticulum and mitochondria) is studied by this method. La3+uptake is obviously stimulated by pre-treating the cells with ouabain and by removing extracellular Na+, and intracellular La3+concentration ([La3+]i) is directly proportional to its extracellular concentration ([La3+]o). But when [La3+]o exceeds 0.4 mmol/L, the 340/380 nm ratio of fluorescence is no longer varied and the maximum [La3+], is 1.5×10-12 mol · L-1. The higher concentration of La3+ (0.1 mmol/L) increases Na+/Ca2+ exchange-mediated calcium influx, but lower concentration (10 μmol/L) appears to block calcium influx. The results also suggest that cytosolic La3+ is transported by the ATP-dependent Ca2+ pump. Intracellular Ca2+ stores are depleted by ionomycin, and then ion  相似文献

At the same time, the time constants of activation become lesser after adding Cu2 . By means of the two-compartment system. PTL-channels, the first lectin channels formed on planar lipid bilayers by Pinellia ternata lectin (PTL) have been studied. The results show (i) PTL-channels are voltage-independent and have apparent subunits; (ii) in 50 mmol/L KC1 and 25 mmol/L BaCl2 solutions, a single channel has unit conductance of 21 pS and 42 pS, respectively; (iii) the channel exhibits a slightly higher permeability to divalent than monovalent cation (PBa/PK=4.1), and (iv) the selectivity among divalent or monovalent cations is poor. The cation selectivity sequence for the channel will follow PBa(7. 0)-Psr (6. 4)~PMg(6. 4)>PK(1. 7)>PN.(1. 0)~PLi(l. 0).Moreover, these data also give explanation to the facilitatory action of PTL on the release of acetyl-choline from motor nerve terminals.  相似文献

DSB显著提高羧酸盐驱油体系抗钙镁离子能力的研究   总被引:2,自引:0,他引:2  
测定了工业品级的天然混合长链烷基羧酸盐(SDC)以及与3-(N,N-二甲基十二烷基胺)-2-羟基-丙磺酸(DSB)复配驱油体系的界面张力(ITFmin), 分别得出其抗钙镁离子的能力为400和5000 mg/L. 选择试剂级十二烷基羧酸钠与DSB复配, 测定了不同配比溶液的表面张力值和临界胶束浓度cmc, 结合长链烷基脂肪酸与钙离子的溶度积Ksp, 分析了对不溶性长链烷基羧酸盐形成的影响. 根据现场驱油体系配方, 计算了两者在溶液中的摩尔配比为4:1时的十二烷基羧酸盐在胶束中的摩尔分数xm1为0.51, 相互作用参数βm值为-3.11, 反映了两者有较强的相互作用. 采用量子化学方法, 对由1个十二烷基羧酸分子、1个DSB分子及1个二价钙离子组成的模型复合物进行了能量计算和电荷分布计算, 得出在长链烷基羧酸盐和DSB两者混合胶束的界面层中存在负电荷空穴, 提出二价金属离子被络合的模型, 合理地解释了实验事实.  相似文献

A magnetic sensor for detection of Pb~(2+) has been developed based on Fe/Fe_3O_4 nanoparticles modified by3-(3,4-dihydroxyphenyl)propionic acid(DHCA). The carboxyl groups of DHCA have a strong affinity to coordination behavior of Pb~(2+) thus inducing the transformation of Fe/Fe_3O_4 nanoparticles from a dispersed to an aggregated state with a corresponding decrease, then increase in transverse relaxation time(T_2) of the surrounding water protons. Upon addition of the different concentrations of Pb~(2+) to an aq. solution of DHCA functionalized Fe/Fe_3O_4 nanoparticles(DHCA-Fe/Fe_3O_4 NPs)([Fe] = 90 mmol/L), the change of T_2 values display a good linear relationship with the concentration of Pb~(2+) from 40 μmol/L to 100 μmol/L and from 130 μmol/L to 200 μmol/L, respectively. Owing to the especially strong interaction between DHCA and Pb~(2+), DHCA-Fe/Fe_3O_4 NPs exhibited a high selectivity over other metal ions.  相似文献

Capillary electrophoresis (CE) with tris(2,2'-bipyridyl) ruthenium (II) (Ru(bpy)3(2+)) electrochemiluminescence (ECL) detection technique was developed for the analysis of four polyamines (putrescine (Put), cadaverine (Cad), spermidine (Spd), and spermine (Spm)) analysis. The four polyamines contain different amine groups, which have different ECL activity. There are several parameters which influence the resolution and ECL peak intensities, including the buffer pH and concentrations, separation voltage, sample injection, electrode materials, and Ru(bpy)3(2+) concentrations. Polyamines are separated by capillary zone electrophoresis in an uncoated fused-silica capillary (50 cmx25 micro m (ID) filled with acidic phosphate buffer (200 mmol/L phosphate, pH 2.0) - 1mol/L phosphoric acid (9:1 v/v) and a separation voltage of 5 kV (25 micro A), with end-column Ru(bpy)3(2+) ECL detection. A 5 mmol/L Ru(bpy)3(2+) solution plus 200 mmol/L phosphate buffer (pH 11.0) is added into the reagent reservoir. The calibration curve is linear over a concentration range of two or three orders of magnitude for the polyamines. The analysis time is less than 25 min. Detection limits for Put and Cad are 1.9x10(-7) mol/L and 7.6x10(-9) mol/L for Spd and Spm, respectively. Intraday and interday relative standard deviations of ECL peak intensities are less than 8%. The main advantages of this CE-ECL detection technique for polyamines analysis presented herein are the omission of chemical derivatization of the analytes and the high selectivity.  相似文献

研究了镧(La^3+)和铕(Eu^3+)对蚕豆保卫细胞外向钾通道(Kout^+)电流的作用.在细胞外,La^3+对Kout^+电流是抑制的,半抑制浓度EC50为81μmol·L^-1;Eu^3+对Kout^+电流表现为低浓度(10和50μmol·L^-1)激活,高浓度(≥1mmol·L^-1)抑制.La^3+在细胞内对Kout^+电流也表现为低浓度(1.13×10^-14 mol·L^-1)激活,高浓度(5.86×10^-14 mol·L^-1)抑制现象.稀土离子La^3+和Eu^3+对保卫细胞Kout^+电流的作用,可能和植物调节水分有关,这可能是稀土生物效应的机制之一.  相似文献

建立了一种用于海水中K+、Na+、Ca2+、Cl-同时快速、自动测定的流动注射离子选择电极自动方法(FI-ISE-M),优化了方法的最佳条件:总离子强度调节液(TISAB)的组成为0.3 mmol/L K+、6.0 mmol/L Na+、1.4 mmol/L Ca2+、21.0 mmol/L Cl-及pH8.0的80 mmol/L的tris-H3BO3缓冲溶液,其流量为1.25mL/min;每次测定的样品用量仅为160μL,载流流量为1.42 mL/min,混合盘管长度为30cm(I.D.0.5 mm)。本方法的分析速度为100样/h,相对标准偏差为2.0%(n=11),标准添加回收率为94.9%~104.3%,测定范围分别为1.0~8.0 mmol/L K+、50~290 mmol/L Na+、3.2~16.0 mmol/L Ca2+、70~430 mmol/LCl-。本方法已应用于反渗透海水淡化系统的水质检测。  相似文献

通过1-(3-二甲氨基丙基)-3-乙基碳二亚胺(EDC)与N-羟基琥珀酰亚胺(NHS)活化反应将多肽偶联到金纳米粒子表面,采用荧光光谱研究其形成的酰胺键的反应偶联效率.考察了实验条件,包括缓冲液的种类(HEPES、Tris-HCl、硼酸、PBS缓冲液)、pH值(pH 6.5~9.0)、缓冲液浓度(10, 25, 40和50 mmol/L)、NHS和EDC的浓度(NHS 0.2~1.0 mol/L,EDC 0.01~0.5 mol/L)及二者比例(0,0.5,1.0,2.0和2.5)、偶联反应时间(4, 8, 12, 24和36 h)等对偶联效率的影响,筛选出最佳实验条件.实验结果表明, 25 mmol/L 4-羟乙基哌嗪乙磺酸(HEPES)缓冲液(pH 7.0), NHS/EDC浓度为0.4 mol/L/0.2 mol/L和24 h的反应时间为EDC-NHS活化反应将多肽偶联到金纳米粒子上的最佳实验条件.本研究结果可为相关研究提供技术参考.  相似文献

研究了经聚苯乙烯氯乙酰树脂(PS-Ace-Cl)制得的PS手性苯丙氨酸树脂(PS-Ace-Phe)对Cu2 的吸附性能,即通过pH、时间、Cu2 浓度等条件的改变,研究了PS-Ace-Phe对Cu2 的吸附,得到了Cu2 的最佳吸附条件:Cu2 溶液浓度为100 mmol/L,pH为6.0~8.5,吸附时间5h,此时吸附效率可达98%,树脂的最高吸附量为2.03 mmol/g,且不同Phe担载量的树脂对Cu2 的吸附率都可大于90%,PS-Ace-Phe对Cu2 的吸附可达近1∶1,说明PS-Ace-Phe对Cu2 的吸附属络合作用.同时得到吸附树脂的稳定存在条件:Cu2 溶液浓度为10mmol/L,pH为2.0~8.0时,该树脂对Cu2 的吸附稳定性可达24h.  相似文献

通过研究异烟肼在多壁碳纳米管修饰碳糊电极上的电化学行为建立了测定异烟肼的电化学方法。在pH6.8的磷酸氢二钠–柠檬酸缓冲溶液中,异烟肼在–1.114 V处能产生一明显还原峰,异烟肼在修饰电极上的反应是受吸附控制的2质子、2电子电极反应过程。研究发现还原峰峰电流大小与异烟肼浓度在5.00×10~(-3)~5.00 mmol/L内呈良好的线性关系,相关系数r=0.997 0,方法检出限为1.70×10~(-3) mmol/L,测定结果的相对标准偏差为3.11%(n=5),回收率为97.6%~103.6%。该方法简单、灵敏,精密度高,可应用于样品中异烟肼的测定。  相似文献

首次研究了狼磷脂-水脂质体中不同种类羧酯钾对草酸钙晶体生长的调控作用 。加入一元DAc只诱导一水草酸钙(COM)生成。二元K_2tart在其浓度大于1. 0mmol/L时可以诱导三水草酸钙(COT)生成。而加入三元的K_3cit和四元的 K_2edta后, 在不同的浓度下,可以分别诱导COM,二水草酸钙(COD)和COT的生 成。在低浓度(—3.3-17mmol/L)范围,, COD含量达到100%;而在较高浓度(> 17mmol/L)时,COD减少,COT含量增加。在不同的浓度区间,无论是COM含量减少, 还是COT含量增加,或者是COD含量的先增加后减少,均与该羧酸钾浓度的对数呈线 性关系。不同羧酯钾抑制COM生长并诱导COD形成的能力顺序为:K_3cit>K_2edta >>K_2tart-KAC,诱导COT生长的能力顺序为:K_2tart>>K_3cit>K_2edta>> KAc.由此推测抑制草酸钙结石形成的潜在效率依次为:K_3cit>K_2edta>> K_2tart>>KAc.  相似文献
