To improve the biological anticancer activity of 20(S)-camptothecin, a novel class of 20(S)-20-O-camptothecin β-aminopropionates were designed and synthesized with camptothecins as the starting materials, through acylation with acryloyl chloride followed by Michael′s addition. Twelve esters, 1a-1d, 4a-4d and 5a-5d, were synthesized and evaluated by using MTT assay method. The results demonstrate that all these compounds show a potential cytotoxicity on KB, HT-29, HCT-8, and Bel7402 tumor cell lines. Some compounds, 1d, 4c, 5b, 5c and 5d, show a higher cytoto-xicity on KB and HCT-8 compared with camptothecin.  相似文献

Racemic 1,1′-methylene[(1RS,1′RS,3RS,3′RS,5RS,5′RS)-8-oxabicyclo[3.2.1]oct-6-en-3-ol] ((±)-6) derived from 2,2′-methylenedifuran has been resolved kinetically with Candida cyclindracea lipase-catalysed transesterification giving 1,1′-methylenedi[(1R,1′R,3R,3′R,5R,5′R)-8-oxabicyclo[3.2.1]oct-6-en-3-ol] (−)-6 (30% yield, 98% ee) and 1,1′-methylenedi[(1S,1′S,3S,3′S,5S,5′S)-8-oxabicyclo[3.2.1]oct-6-en-3-yl] diacetate (+)-8, (40% yield, 98% ee). These compounds have been converted into 1,1′-methylenedi[(4S,4′S,6S,6′S)- and (4R,4′R,6R,6′R)-cyclohept-1-en-4,6-diyl] derivatives.  相似文献

The P–Ph cleavage of phenyldibenzophosphole (1) with lithium in THF gives lithium dibenzophospholide (2). Reaction of 2 with ethyleneglycol ditosylate produces the known chelate ligand 1,2-bis(dibenzophospholyl)ethane (3) in good yield. Similarly, 2 and (2R,3R)-butanediol ditosylate give the new chiral chelate ligand (2S,3S)-bis(dibenzophospholyl)butane (4). Ligand exchange of [CpRu(PPh3)2Cl] with 3 or 4 yields the halfsandwich complexes [CpRu(C12H8PC2H4PC12H8)Cl] (5) and [CpRu((S,S)-C12H8PCHMeCHMePC12H8)Cl] (6). Complex 6 was characterized crystallographically (monoclinic, space group P21 (no. 4), a=820.6(4), b=1501.0(3), c=1172.8(6) pm, β=108.87(2)°, V=1.367(1)×109 pm3, Z=2). The most conspicuous feature of the structure of 6 is the perfect coplanarity of the two dibenzophosphole moieties imposed by their steric interaction with the Cp ligand. Complex 6 and the thiophene complex [CpRu((S,S)-C12H8PCHMeCHMePC12H8)(SC4H4)]BF4 (7) derived therefrom are remarkably unreactive with regard to ligand substitutions. A possible explanation is the lack of intramolecular –C stabilization en route to the transition state of ligand substitution. The enantiomeric purity of 6 and 7 could nevertheless be demonstrated by conversion to diastereomerically pure [CpRu((S,S)-C12H8PCHMeCHMePC12H8)((S)-CNCHMePh)]BF4 (8).  相似文献

Vinyl substituted (1R,2S)-amino alcohols 5 were obtained by addition of vinyl magnesium bromide to the corresponding cyanohydrin O-trimethylsilyl ethers (R)-2. The O- and N-protected vinyl amino alcohols 6 were ozonized at −78°C in methanol yielding (1R,2S)-2-amino-1,3-diols7 in high enantiomeric and diastereomeric excesses. For purification, compounds 7 in some cases were acetylated to give the derivatives (1R,2S)-8. Racemic 6a was converted by oxidative ozonolysis at −78°C in methanolic NaOH solution to the corresponding methyl N-acetyl-β-hydroxy propanoate 9a. The configuration of (1R,2S)-8a was confirmed by x-ray crystallographic analysis.  相似文献

Several (3S,4S)- and (3S,4R)-statine derivatives have been prepared by attack of nucleophiles on crystalline, epimeric N-BOC-lactams 7a and 7b. The key step in the synthesis of the lactams was the TiCl4-catalyzed coupling reactions of acetals derived from (R)-1,3-butanediol with allyltrimethylsilane.

Several enantiometrically pure (3S,4S- and (3S, 4R)-statine derivatives were made by sodium cyanide-catalyzed reaction of nucleophiles with the lactams 3a and 3b which were synthesized by the scheme 1 → 2 → 3.  相似文献

The Reformatsky reagent tert-butoxycarbonylmethylzinc bromide adds in high yields to N-sulfonylimines, e.g. 1a–1d, derived by condensation of benzaldehyde dimethyl acetal with methanesulfonamide, toluene-4-sulfonamide, 4-(methoxycarbonyl)benzenesulfonamide and sulfamide: the products are protected β-amino acids 2a–2d. N-Deprotection occurs reductively (Na-naphthalene; low yields) for 2b and 2c or hydrolytically (refluxing aq. pyridine; 76% yield of amino acid 3a after acid hydrolysis of the t-butyl ester) for the sulfamide derivatives 2d. Anthracene-9-sulfonamide (6) is readily available by sulfonation and chlorination of anthracene, and condenses with aldehydes [RCHO; R = Ph, 4-FC6H4, 4-MeOC6H4, 4-NCC6H4, 2-furyl, (E)-styryl], e.g. in the presence of TiCl4/Et3N, to yield imines 7a–7f, which after addition of tert-butoxycarbonylmethylzinc bromide give protected amino acids 8a–8f; however, 8f cyclizes to the sultam 9 via a spontaneous intramolecular Diels-Alder reaction. Reductive cleavage of the N-anthracene-9-sulfonyl group is much easier than for traditional N-sulfonyl protecting groups, as demonstrated by the deprotection of 8a and 8c using aluminium amalgam.  相似文献

Described herein is a versatile approach to (i) (2S,3S,4S)-3-hydroxy-4-methylproline 3, a constituent of echinocandins and related oligopeptide antibiotics; (ii) (2S,3S)-3-hydroxyproline 1; (iii) (2R,3S)-3-hydroxyprolinol 5, and (iv) 4′-tert-butoxyamido-2′-deoxythymidine 6b. The key steps of this synthesis are the stereoselective Michael addition of 2-lithio-1,3-dithiane to 3-O-benzyl-5,6-dideoxy-1,2-O-isopropylidene-6-nitro--d-xilo-hex-5-enofuranose followed by the enantioselective two-step transformation of 3-O-benzyl-5,6-dideoxy-5-C-(1,3-dithian-2-yl)-6-nitro-β-l-idofuranose into (1S,2S,3S,4S,5S,6R)-5-benzyloxy-6-hydroxy-3,4-(isopropylidendioxy)-2-nitro-cyclohexanecarbaldehyde propylene dithioacetal, which was finally converted into the target compound.  相似文献

A series of furo[3,2-g]chromone derivatives were synthesized via the reaction of furochromone carbaldehyde 1 with amines 3a - d and thioglycolic acid to give thiazolidinones 4a - d . The later react with benzaldehyde/thiourea or hydroxylamine and DMF-DMA in glacial acetic acid to give thiazolopyrimidines 5a - d or thiazoloisoxazoles 6a - d , respectively. Also, the synthesis of α-aminophosphonates via the one-pot reaction of 1 and amines 3a , b were trapped by dialkylphosphites 7a - c afforded the corresponding α-aminophosphonates 8a - f . Applying hexaalkyltriamidophosphites 9a , b to 1 gives alkylidenephosphorane ylides 11a , b in an open structure form. In the present investigation, the in vitro inhibition capacity of compounds ( 4a , 4c , 5b , 5c , 6b - d , 8a - f , and 11a ) was screened in three human cancer cell lines HCT-116, MCF-7, and HepG2. The anticancer activity results revealed that 8b and 8e had more potent cytotoxic inhibition activity against HCT-116 cell line; however, all the tested compounds had obviously less cytotoxic activity against MCF-7 cell line, while 5b , 5c , and 6d were potent against HepG2 cell line compared with that of doxorubicin.  相似文献

Structure—gas chromatographic retention time models were developed for 26 tetra-n-alkylsilanes, 26 tetra-n-alkylgermanes, and the mixed set of silanes and germanes using topological indexes. The topological indexes used were molecular connectivity indexes, the new Kier—Hall total topological indexes, and the new Kier—Hall electrotopological state atom index, S, for the silicon and germanium atoms. For the mixed data set, a model based on 1χ, 3χc, and S gave r = 0.999 and s = 0.033. In two cases the residual was more than twice the standard error. Replacement of 1χ by TTV, the new Kier—Hall total topological index using δv values for the silicon and germanium atoms, also gave r = 0.999 and s = 0.033 but in no case was there a residual greater than twice the standard error. A two-variable model based on 0χr, which uses δr values for the metal atoms, and 4χpcv, which uses δv values for the metal atoms gave r = 0.999 and s = 0.035. The results demonstrate the adequacy of the Kier—Hall electrotopological state atom index for encoding the atomic characteristics of silicon and germanium in a mixed data set. The results show that the Kier—Hall total topological indexes can be used in place of 1χ to encode the size and skeletal branching of silanes and germanes.  相似文献
