4-[(1E)-3-(biphenyl-4-yl)buta-1,3-dien-1-yl]phenyl prop-2-enoate (ACH) and 4-[(1E)-3-(biphenyl-4-yl)buta-1,3-dien-1-yl]phenyl 2-methylprop-2-enoate (MCH) was synthesized from biphenyl in three steps and their structures were confirmed by elemental analysis, IR, NMR (1H, 13C, DEPT135, 1H-1H COSY, 1H-13C HSQC and 1H-13C HMBC) spectroscopic techniques. In this present study, various physicochemical characteristics we demonstrate solubility, color, absorbance and fluorescence property of novel biphenyl based acrylate and methacrylate measured in different solvents like benzene, dichloromethane, tetrahydrofuran, acetonitrile, dimethylsulfoxide and ethanol.  相似文献

Volatile fatty acids (VFAs) are produced in the human colon by the bacterial breakdown of carbohydrates that escape digestion and absorption in the small intestine. They have important local and systemic effects on gastrointestinal and nutritional functions. Measuring their production is difficult because of inaccessibility of sampling sites and low circulating concentrations. Stable isotope tracer techniques are a way to measure VFA production but require measurement of isotope dilution in blood and other biological fluids. We have developed a streamlined and robust method to measure the concentration and enrichment of [(2)H]-labelled VFAs by gas chromatography/mass spectrometry (GC/MS) and [(13)C]-labelled VFAs by gas chromatography/combustion/isotope ratio mass spectrometry (GC/C/IRMS). Both types of analysis were carried out on the same samples allowing multiple tracer studies to be conducted. Good accuracy and repeatability were found for GC/MS analysis of [(2)H]-labelled VFAs. Careful handling of the background contribution, especially acetate, allowed quantitation of concentration and enrichment within the analysis. GC/C/IRMS analysis of [(13)C] VFAs was also achieved with good accuracy and repeatability. This methodology was used to determine whole-body acetate production in two subjects using multiple tracers ([(2)H(3)]- and [1-(13)C]acetate) and blood and urine sampling. Whole-body acetate flux was similar when measured either with [(2)H(3)]- or [1-(13)C]acetate, and when flux was determined from plasma or urine tracer enrichment. This new method will permit rapid and accurate measurement of VFA flux using [(2)H]- and/or [(13)C]-labelled VFAs as tracers. Measurements of the contribution of colonic VFA production to whole-body VFA flux are now possible.  相似文献

The methanesulfonic acid catalyzed reaction of 1-(4-chloro- and 2,4-dichlorophenyl)-2-(1-methyl-2-imida-zolyl)ethanones 1a and 1b with glycerol produced cis- and trans-{2-haloaryl-2-[(1-methyl-2-imidazolyl)methyl]-4-hydroxymethyl}-1,3-dioxolanes 2a and 2b with a 2:1 cis/trans ratio. Besides these five-membered ketals, the reaction of 1a with glycerol afforded a small amount of trans-{2-(4-chlorophenyl)-2-[(1-methyl-2-imidazolyl)methyl]-5-hydroxy}-1,3-dioxane ( 3a , 7%). The reaction of methanesulfonyl chloride with cis-1 formed the corresponding methanesulfonates, cis- 4 , which rapidly cyclized to the title compounds 5 . Base-catalyzed ring opening of 5 furnished 1-methyl-5,6-dihydro-6-hydroxymethyl-8-(4-chloro- and 2,4-dichlorophenyl)-1H-imidazo[3,2-d][1,4]oxazepinium methanesulfonates 7 . Acid-catalyzed hydrolyses of 5 or 7 provided 1-methyl-2-[(4-chloro- and 2,4-dichloro)phenacyl]-3-[(2,3-dihydroxy)-1-propyl]imidazolium salts 12 . Structure proofs were based on extensive 1H and 13C chemical shifts and coupling constants and structures of 3a and 5a were confirmed by single crystal X-ray crystallography.  相似文献

We report the complete (1)H and (13)C NMR assignment of impurities of six Lopinavir (2S)-N-[(2S, 4S, 5S)-5-{[2-(2,6-dimethylphenoxy)acetyl]amino}-4-hydroxy-1,6-diphenyl hexan-2-yl]-3-methyl-2-(2-oxo-1,3-diazinan-1-yl)butan- amide. Two of the impurities are regioisomers and GCOSY used to differentiate the two structures. The spectral assignments for all six impurities were achieved by concerted application of one and two-dimensional NMR techniques ((1)H NMR, (13)C NMR, DEPT, GCOSY, GHSQC and GHMBC).  相似文献

The coupling between tetramethylguanidine, HN=C(NMe2)2, and coordinated organonitriles in the platinum(II) complexes cis/trans-[PtCl2(RCN)2] (R = Me, Et, Ph) proceeds rapidly under mild conditions to afford the diimino compounds containing two N-bound monodentate 1,3-diaza-1,3-diene ligands [PtCl2{NH=C(R)N=C(NMe2)2}2] (R = Et, trans-1; R = Ph, trans-2; R = Me, cis-3; R = Et, cis-4), and this reaction is the first observation of metal-mediated nucleophilic addition of a guanidine to ligated nitrile. Complexes 1-4 were characterized by elemental analyses (C, H, N), X-ray diffraction, FAB mass spectrometry, IR, and 1H and 13C{1H} NMR spectroscopies; assignment of signals from E/Z-forms of 1,3-diaza-1,3-diene ligands and verification of routes for their Z right harpoon over left harpoon E isomerization in solution were performed using 2D 1H,1H-COSY, 1H,13C-HETCOR, and 1D NOE NMR experiments. The newly formed and previously unknown 1,3-diaza-1,3-dienes NH=C(R)N=C(NMe2)2 were liberated from the platinum(II) complexes [PtCl2{NH=C(R)N=C(NMe2)2}2] (1-3) by substitution with 2 equiv of 1,2-bis-(diphenylphosphino)ethane (dppe) to give the uncomplexed HN=C(R)N=C(NMe2)2 species (5-7) in solution and the solid [Pt(dppe)2](Cl)2. The former were utilized in situ, after filtration of the latter, in the reaction with 1,3-di-p-tolylcarbodiimide, (p-tol)N=C=N(tol-p), in CDCl3 to generate (6E)-N,N-dimethyl-1-(4-methylphenyl)-6-[(4-methylphenyl)imino]-1,6-dihydro-1,3,5-triazin-2-amines) (8-10) due to the [4 + 2]-cycloaddition accompanying elimination of HNMe2. The formulation of 8-10 is based on ESI-MS, 1H, 13C{1H} NMR, and X-ray crystal structures determined for 9 and 10. The reaction of 1,3-diaza-1,3-dienes with 1,3-di-p-tolylcarbodiimide, described in this article, constitutes a novel synthetic approach to a useful class of heterocyclic species like 1,6-dihydro-1,3,5-triazines.  相似文献

The aliphatic, phosphine-based pincer complex [(C(10)H(13)-1,3-(CH(2)P(Cy(2))(2))Pd(Cl)] (1) is a highly active Negishi catalyst, enable to quantitatively couple various electronically activated, non-activated, deactivated, sterically hindered and functionalized aryl bromides with various diarylzinc reagents within short reaction times and low catalyst loadings. Experimental observations strongly indicate that a molecular mechanism is operative with initial chloride dissociation of 1 and formation of the cationic T-shaped 14e(-) complex [(C(10)H(13)-1,3-(CH(2)P(C(6)H(11))(2))(2))Pd](+) (B), which undergoes oxidative addition of an aryl bromide (Ar'Br) to yield the cationic, penta-coordinated aryl bromide pincer complexes of type [(C(10)H(13)-1,3-(CH(2)P(Cy(2))(2))Pd(Br)(aryl')](+) (C) with the metal center in the oxidation state of +IV and the aryl unit in cis position relative to the aliphatic pincer core. Subsequent transmetalation with Zn(aryl)(2) result in the cationic diaryl pincer complexes of type [(C(10)H(13)-1,3-(CH(2)P(Cy(2))(2))Pd(aryl)(aryl')](+) (D), which reductively eliminate the coupling products, thereby regenerating the catalyst. The neutral square planar aryl pincer complex--a possible key intermediate in the catalytic cycle--was found to be reversibly formed in the reaction mixture but is not involved in the catalytic mechanism. Similarly, palladium nanoparticles as the catalytically active form of 1 could have been excluded.  相似文献

Two new polyketides: 2Z-(heptadec-12-enyl)-4-hydroxy-3,4,7,8-tetrahydro-2H-chromen-5(6H)-one (1) and 2-(heptadec-12-enyl)-5-hydroxy-5,6,7,8-tetrahydrochromen- 4-one (2), together with eleven known compounds: 4-hydroxy-2-[(3,4-methylenedioxy- phenyl)tridecanoyl] cyclohexane-1,3-dione (3), oleiferinone (4), 4-hydroxy-2-[(3,4- methylenedioxyphenyl)undecanoyl]cyclohexane-1,3-dione (5), 4-hydroxy-2-[(11-phenyl- undecanoyl)cyclohexane-1,3-dione (6), proctorione C (7), surinone C (8), 5-hydroxy- 7,8,4'-trimethoxyflavone (9), 5-hydroxy-7,8,3',4'-tetramethoxyflavone (10), 5-hydroxy- 7,3',4'-trimethoxyflavone (11), 5,8-dihydroxy-7,3',4'-trimethoxyflavone (12) and cepharanone B (13) were isolated from the whole plant of Peperomia dindygulensis Miq. Their structures were elucidated by spectroscopic methods, including 2D-NMR techniques. Compounds 2, 3, 5 and 8 inhibited human umbilical vein endothelial cell (HUVEC) proliferation and compounds 5 and 8 sharply suppressed HUVEC tube formation.  相似文献

The electronic properties of cation radical salts derived from organometallic mixed-ligand complexes [(ppy)Au(S-S)](ppy- = C-dehydro-2-phenylpyridine(-); S-S(2-) = dithiolene ligand) with Au(III)-C sigma-bond were investigated. A 2:1 salt complex [(ppy)Au(C8H4S8)]2[PF6] (C8H4S8(2-) = 2-((4,5-ethylenedithio)-1,3-dithiole-2-ylidene)-1,3-dithiole-4,5-dithiolate(2-)) exhibited semiconductive behavior under ambient pressure (rho rt = 2.6 Omega cm, Ea = 0.03 eV). Magnetic measurements show that it is a Mott insulator close to the metal-insulator boundary. Raman and infrared spectra have revealed that the complex has a quasi-one-dimensional dimeric structure consisting of uniformly charged donor molecules. The complex exhibits metallic behavior at pressures above 0.8 GPa. In contrast, a similar compound [(ppy)Au(C8H4S6O2)]2[BF4] (C8H4S6O2(2-) = 2-((4,5-ethylenedioxy)-1,3-dithiole-2-ylidene)-1,3-dithiole-4,5-dithiolate(2-)) is a band insulator.  相似文献

Aryl 2-[(2-imidazolyl)ethyl or 3-(2-imidazolyl)propyl]ketones were ketalized by glycerol or 3-mercapto-1,2-propanediol in boiling benzene in the presence of 4-toluenesulfonic acid to provide the title compounds. The aryl substituents are 4-chloro-, 4-bromo-, 4-fluoro-, or 2,4-dichlorophenyl. While aryl (2-imidazolyl)methyl ketones condensed with glycerol to form cis- and trans-{2-aryl-2-[(2-imidazolyl)methyl]-4-(hydroxymethyl)}-1,3-dioxolanes, related condensations with 3-mercapto-1,2-propanediol, under similar, or even more stringent reaction conditions, produced no 1,3-oxathiolane analogs, with the starting ketones being recovered. Separation and structure determination of these racemic cis and trans isomeric products are described. The structure of these stereoisomers was established by means of 1H and 13C nmr correlation and nOe experiments. Selective methylation of the N-unsubstituted 2-imidazolyl alcohols with one equivalent sodium hydride and methyl iodide provided the corresponding N-methyl alcohols in excellent yields. With excess benzoyl chloride, N-unsubstituted 2-imidazolyl alcohols were initially converted to O, N-dibenzoates from which the N-benzoyl group was easily cleaved by ammonium hydroxide in ethanol to provide benzoate esters.  相似文献

A significant contaminant of the antimalarial drug piperaquine (1,3-bis-[4-(7-chloroquinolyl-4)-piperazinyl-1]propane) has been identified using liquid chromatography-mass spectrometry (LC-MS) and 2D NMR spectroscopy (1H-1H COSY, 1H-13C HSQC, 1H-13C HMBC). The impurity was identified as the positional isomer 1-[(5-chloroquinolin-4)-piperazinyl]-3-[(7-chloroquinolin-4)-piperazinyl]propane. The impurity is formed because of contamination of batches of 4,7-dichloroquinoline (a precursor in the synthesis of piperaquine) with 4,5-dichloroquinoline. The amount of impurity (peak area impurity/peak area piperaquine using LC-UV at 347 nm) in old batches of piperaquine and in Artekin (the combination of dihydroartemisinin-piperaquine) ranged from 1.5 to 5%.  相似文献

Biological magic angle spinning (MAS) solid-state nuclear magnetic resonance spectroscopy has developed rapidly over the past two decades. For the structure determination of a protein by solid-state NMR, routinely (13)C,(13)C distance restraints as well as dihedral restraints are employed. In protonated samples, this is achieved by growing the bacterium on a medium which contains [1,3]-(13)C glycerol or [2]-(13)C glycerol to dilute the (13)C spin system. Labeling schemes, which rely on heteronuclei, are insensitive both for detection and in terms of quantification of distances, since they are relying on low-γ nuclei. Proton detection can in principle provide a gain in sensitivity by a factor of 8 and 31, compared to the (13)C or (15)N detected version of the experiment. We report here a new labeling scheme, which enables (1)H-detection of aliphatic resonances with high resolution in MAS solid-state NMR spectroscopy. We prepared microcrystals of the SH3 domain of chicken α-spectrin with 5% protonation at nonexchangeable sites and obtained line widths on the order of 25 Hz for aliphatic (1)H resonances. We show further that (13)C resolved 3D-(1)H,(1)H correlation experiments yield access to long-range proton-proton distances in the protein.  相似文献

The precise assignments of cross polarization/magic angle spinning (CP/MAS) (13)C NMR spectra of cellulose I(alpha) and I(beta) were performed by using (13)C labeled cellulose biosynthesized by Acetobacter xylinum (A. xylinum) ATCC10245 strain from culture medium containing D-[1,3-(13)C]glycerol or D-[2-(13)C]glucose as a carbon source. On the CP/MAS (13)C NMR spectrum of cellulose from D-[1,3-(13)C]glycerol, the introduced (13)C labeling were observed at C1, C3, C4, and C6 of the biosynthesized cellulose. In the case of cellulose biosynthesized from D-[2-(13)C]glucose, the transitions of (13)C labeling to C1, C3, and C5 from C2 were observed. With the quantitative analysis of the (13)C transition ratio and comparing the CP/MAS (13)C NMR spectrum of the Cladophora cellulose with those of the (13)C labeled celluloses, the assignments of the cluster of resonances which belong to C2, C3, and C5 of cellulose, which have not been assigned before, were performed. As a result, all carbons of cellulose I(alpha) and I(beta) except for C1 and C6 of cellulose I(alpha) and C2 of cellulose I(beta) were shown in equal intensity of doublet in the CP/MAS spectrum of the native cellulose, which suggests that two inequivalent glucopyranose residues were contained in the unit cells of both cellulose I(alpha) and I(beta) allomorphs.  相似文献

The homonuclear dipolar coupling of a directly bonded (13)C-(13)C pair has been used to create a dipolar double-quantum filter (D-DQF) to remove the natural-abundance (13)C background in (13)C[(2)H] rotational-echo double-resonance (REDOR) experiments. The most efficient version of this experiment has the D-DQF excitation and reconversion preceding the REDOR evolution period. Calculated and observed (13)C[(2)H]D-DQF-REDOR dephasings were in agreement for a test sample of mixed recrystallized labeled alanines.  相似文献

A series of new cycloalkanespiro-5-hydantoin phosphonic acids have been synthesized and characterized. The mixture of [(2,4-dioxo-1,3-diazaspiro-alkane-3-yl)-methyl]phosphonic acids and [(2,4-dioxo-1,3-diazaspiro-alkane-1,3-diyl)dimethyl]diphospho-nic acids was obtained from cycloalkanespiro-5-hydantoins, formaldehyde, and phosphorus trichloride in a molar ratio of 1:2:2, by a procedure modified by us. Their structures were proved by means of IR, 1H, 13C{1H} and 31P NMR spectroscopy.  相似文献

Two novel anellated N-heterocyclic carbenes (NHC), 1,3-dineopentylnaphtho[2,3-d]imidazol-2-ylidene, and 1,3-dineopentyl-2-ylido-imidazolo[4,5-b]pyridine were obtained by reduction of the respective thiones with potassium, the former also by deprotonation of the corresponding naphthimidazolium hexafluorophosphate by using excess KH in THF. The use of equimolar amounts of KH provided an unexpected formal addition product of this NHC with KOH. X-ray crystal structure analysis of the adduct provided evidence for a distorted tetrameric N-heterocyclic alkoxide, stabilized by two THF molecules. In C(6)D(6) the compound undergoes disproportionation. Transition-metal complexes [(NHC)AgCl], [(NHC)Rh(cod)Cl], and (E)-[(NHC)(2)PdCl(2)] of the novel naphthimidazol-2-ylidene were synthesized. X-ray crystal structures and (1)H and (13)C NMR spectroscopic data provided detailed structural information. Comparing characteristic data with those of nonanellated and differently anellated NHCs or their complexes provides information on the influence of the extended anellation.  相似文献

The total synthesis of 6-[(1S,3S)-1,3-dimethyl-2-oxopentyl]-4-hydroxy-3,5-dimethyl-2H-pyran-2-one (22), the enantiomer of the natural product micropyrone (1), was achieved in 9 linear steps (10% overall yield), from Evans auxiliary (R)-12 with key coupling of the dianion of dione 17 and aldehyde 11. Formation of the pyrone ring and subsequent oxidation at C7 was achieved without epimerization of the sensitive position α to both the pyrone ring and the carbonyl. The same sequence using the alternate dione 24 achieved the total synthesis of 6-[(1S,3S)-1,3-dimethyl-2-oxopentyl]-4-hydroxy-3-methyl-2H-pyran-2-one (28), the (+)-enantiomer of the natural product, ascosalipyrone (2). In both cases diastereomeric aldehydes 11 and 16 were taken through the synthetic sequence to give two possible diastereomers of the natural products. Comparison of the (1)H and (13)C NMR data for the synthetic isomers with that reported for the natural products determined their relative stereochemistry. Comparison of the optical rotation obtained for 22 established it to be the enantiomer of micropyrone.  相似文献

The Staudinger reaction of the imidazolin-2-ylidenes, 1,3-di-tert-butylimidazolin-2-ylidene (1a), 1,3-diisopropylimidazolin-2-ylidene (1b), 1,3-diisopropyl-4,5-dimethylimidazolin-2-ylidene (1c), 1,3-bis(2,4,6-trimethylphenyl)imidazolin-2-ylidene (1d) and 1,3-bis(2,6-diisopropylphenylimidazolin-2-ylidene (1e), with trimethylsilyl azide furnishes the corresponding N-silylated 2-iminoimidazolines 2a-e, which react with [(eta-C5H5)TiCl3] to afford half-sandwich cyclopentadienyl titanium complexes of the type [CpTi(L)Cl2] (3) (L = imidazolin-2-iminato ligand). Similarly, the reactions of 1,3-di-tert-butyl-2-(trimethylsilylimino)imidazoline (2a) with [(eta-tBuC5H4)TiCl3] results in the formation of [(eta-tBuC5H4)Ti(L)Cl2] (4) (L = 1,3-di-tert-butylimidazolin-2-imide). Bis(1,3-di-tert-butylimidazolin-2-iminato)titanium dichloride (5) is obtained from the reaction of two eq. of 2a with TiCl4. Treatment of 5 with methyllithium results in the formation of the corresponding dimethyl complex [L2Ti(CH3)2] (6), whereas [CpTi(L)(CH3)2] (7) is similarly obtained from 3a. The molecular structures of 3a, 3b, 3c, 3e x C7H8, 4 and 7 are reported revealing linearly coordinated imidazolin-2-iminato ligands together with very short Ti-N bond distances. All dichloro complexes (3a-e, 4 and 5) can be activated with methylaluminoxane (MAO) to give active catalysts for ethylene homopolymerization. In most cases, moderate to high activities are observed together with the formation of high (HMWPE) or even ultra high molecular weight polyethylene (UHMWPE).  相似文献

Isotopic substitution is known to affect kinetic rate constants and equilibrium constants in chemistry. In this study, we have used tritium substitution and high pH to probe the glucose left harpoon over right harpoon glucose(-) + H(+) equilibrium. Passing partially ionized mixtures of [(3)H]- and [(14)C]glucose over anionic exchange resin has permitted the detection of subtle differences in pK(a). We have found that, at pH 11.7 in an anionic exchange system, [(3)H]glucose always elutes ahead of the [(14)C]glucose, and we report isotope effects of 1.051 +/- 0.0007, 1.012 +/- 1.0005, 1.014 +/- 0.0004, 1.024 +/- 0.0003, 1.014 +/- 0.0004, and 1.015 +/- 0.0014 for [1-(3)H]-, [2-(3)H]-, [3-(3)H]-, [4-(3)H]-, [5-(3)H]-, and [6,6-(3)H(2)]glucose, respectively, as compared to either [2-(14)C]-or [6-(14)C]glucose. The elevated isotope effects at H1 and H4 imply unusual charge sharing in anionic aqueous glucose. Base titration of (13)C-chemical shift changes demonstrates the dominance of pyranose forms at elevated pH, and ab initio isotope effect computations on gas-phase glucose anions are presented.  相似文献
