从顺磁共振波谱检测到通过竹红菌素的光诱导还原和基态竹红菌素跟脂肪胺的电子转移两条途径所得到的自由基信号, 并由电化学还原得到同一自由基, 确认它为半醌负离子自由基。竹红菌素溶液中加入芳香胺观察不到ESR信号。它的3,10位上与醌基相邻近的羟基离解以后, 无论在基态还是在激发态都观察不到ESR信号。此外, 还从吸收光谱观察到半醌负离子自由基吸收以及竹红菌素跟脂肪胺之间的相互作用。  相似文献

The electroreduction of eosin Y on a platinum electrode in deaerated slightly acidic aqueous 0.1 mol L(-1) potassium chloride medium is followed in situ by electron spin resonance (ESR) spectroscopy and UV/Vis spectroscopy. The electrochemical formation of a semiquinone radical is proved by both the appearance of an absorbing band at 408 nm, and a strong ESR signal observed during a negative-going scan. The system is also studied in the presence of Zn(II) ions due to its importance for understanding the growth mechanism of nanostructured ZnO/Eosin Y hybrid films by electrodeposition; under such conditions the ESR and UV/Vis response of the semiquinone radical is not observed. Zinc (II) ions form a complex with the dye, which is reduced by a fast two-electron process.  相似文献

In order to discuss the free radicals formation mechanism of Hypocrellin A(HA)with amino derivatives,the electron-spin resonance( ESR) spectroscopy was adopted to study the photochemistry on HA with dibenzyl amine(DBA)and N-methyl benzyl amine(NMBA),respectively. When HA with DBA or NMBA in chloroform solution was illuminated with visible light,singlet oxygen,semiquinone radical and oxynitride radical were formed depending on the condition of the solvent system containing the amino-substituted and solved oxygen. The signal intensity of oxynitride radical decreased with increasing the illumination time,and the signal intensity of semiquinone radical increased with increasing the illumination time. The oxynitride radical content was in inverse ratio with the semiquinone radical generated by being irradiated. In the aerobic system of chloroform solution containing DBA/HA,smiquinone radical was the main radical irradiated. The results indicated that HA induced amino derivatives into HA semiquinone radical.  相似文献

Abstract The photoallergens bithionol (BT) and fentichlor (FT) generated free radical photoproducts upon UV photolysis which were observable by direct electron spin resonance (ESR). Both the yield and the type of free radical photoproducts were affected by pH, and to some extent, concentration of oxygen and concentrations of the photosensitizers. At pH 8.5, bithionol (0.9 mM) generated a semiquinone type free radical (BI) via a mechanism which probably involves substitution of the 4-chlorine by hydroxyl to form the corresponding hydroquinone followed by oxidation. The photolysis of 4-chlorophenol, 4-chlorocatechol and 2,2'-methylene-bis(4-chlorophenol) also generated the corresponding semiquinone radicals, suggesting that this mechanism is shared by other 4-chlorophenols. At pH 8.5, only photoproduct BI was observed during the irradiation of BT; FT related photoproducts were not observed at this pH. However, at higher pH values (pH 10.7 or pH 12), FT photoproducts were also observed in addition to BI upon prolonged irradiation. Moreover, the yield of BI increased drastically at higher pH. Oxygen did not play any role at pH 10.7, although it enhanced the yield of BI at pH 8.5. At pH 8.5, irradiated fentichlor generated, in roughly equal amounts, a semiquinone radical (Fla) and an unidentified species which contained two inequivalent protons (FII). At higher pH values (pH 10.7 and pH 12), at least four species were observed. All of the species are believed to be semiquinone radicals and two have been unambiguously identified. The yield of FI increased by a factor of 50 as the pH was increased from 8.5 to 12. Oxygen played only a minor role at pH 10.7 and above. However, at pH 8.5, it also enhanced the yield of FI.  相似文献

Polychlorinated biphenyls (PCBs) comprise a group of persistent organic pollutants that differ significantly in their physicochemical properties, their persistence, and their biological activities. They can be metabolized via hydroxylated and dihydroxylated metabolites to PCB quinone intermediates. We have recently demonstrated that both dihydroxy PCBs and PCB quinones can form semiquinone radicals (SQ(*-)) in vitro. These semiquinone radicals are reactive intermediates that have been implicated in the toxicity of lower chlorinated PCB congeners. Here we describe the synthesis of selected PCB metabolites with differing degrees of chlorination on the oxygenated phenyl ring, e.g., 4,4'-dichloro-biphenyl-2,5-diol, 3,6,4'-trichloro-biphenyl-2,5-diol, 3,4,6,-trichloro-biphenyl-2,5-diol, and their corresponding quinones. In addition, two chlorinated o-hydroquinones were prepared, 6-chloro-biphenyl-3,4-diol and 6,4'-dichloro-biphenyl-3,4-diol. These PCB (hydro-)quinones readily react with oxygen or via comproportionation to yield the corresponding semiquinone free radicals, as detected by electron paramagnetic resonance spectroscopy (EPR alias ESR). The greater the number of chlorines on the (hydro-)quinone (oxygenated) ring, the higher the steady-state level of the resulting semiquinone radical at near neutral pH.  相似文献

Exposure to asbestos and air pollution particles can be associated with increased human morbidity and mortality. However, the molecular mechanism of lung injuries remains unknown. It has been postulated that the in vivo toxicity results from the catalysis of free radical generation. Using electron spin resonance (ESR) in conjunction with the spin trap alpha-(4-pyridyl-1-oxide)-N-tert-butylnitrone (4-POBN) we previously investigated in vivo free radical production by rats treated with intratracheal instillation of asbestos (crocidolite fibers) and an emission source air pollution particle (oil fly ash). In this report we compare the effect of two different exposures on the type of free radicals they induce in in vivo animal model. Twenty-four hours after the exposure, ESR spectroscopy of the chloroform extract from lungs of animals exposed to either asbestos or oil fly ash gave a spectrum consistent with a carbon-centered radical adduct (aN = 15.01 G and aH = 2.46 G). To test whether free radical formation occurred in vivo and not in vitro, a number of control experiments were performed. Combinations (both individually and together) of asbestos or oil fly ash and 4-POBN were added to lung homogenate of unexposed rats prior to chloroform extraction. No detectable ESR signal resulted. To exclude the possibility of ex vivo free radical generation, asbestos or oil fly ash was added to lung homogenate of an animal treated with 4-POBN. Also, 4-POBN was added to lung homogenate from rats instilled with asbestos or oil fly ash. Neither system produced radical adducts, indicating that the ESR signal detected in the lung extracts of the treated animals must be produced in vivo and not ex vivo or in vitro. In conclusion, ESR analysis of lung tissue demonstrated that both exposures produce lipid-derived radical metabolites despite their different composition and structure. Analogously, both exposures provide evidence of in vivo enhanced lipid peroxidation. Furthermore, it is concluded that without the presence of a spin-trapping agent, no free radical metabolites could be detected directly by ESR in either exposure.  相似文献

Electron spin resonance (ESR) techniques have been applied to a detailed study of batch and semicontinuous emulsion polymerization of methyl methacrylate; butyl acrylate and styrene have been briefly studied. Quenching of samples from the polymerization reactor followed by ESR analysis are useful, but we have developed continuous flow techniques which are preferable in many cases. ESR techniques can provide valuable information relating to the nature of free radical reactions, the concentration of propagating free radicals, and the kinetic processes in these reactions. Direct ESR analysis is most valuable but is not applicable to all systems. Spin trapping techniques can be useful for systems not accessible by direct analysis.  相似文献

A novel method has been employed to prepare 2-butylamino-2-demethoxy hypocrellin A (BADMHA) and 2-butylamino-2-demethoxy hypocrellin B (BADMHB). Both compounds exhibit stronger absorption at the phototherapeutic window (600-900 nm). The spin trapping and spin counteraction studies have shown that they are both efficient generators of the active oxygen (1O2, O2*-) in the aerobic condition. Under the anaerobic condition they generate non-oxygen free radical (semiquinone radical anion), and the active oxygen mechanism of photosensitization can be converted into non-oxygen free radical mechanism with the depletion of oxygen. The quantum yields of 1O2 generation of BADMHA and BADMHB are 0.46 and 0.44, respectively. Both are lower than those of their parent compounds HA and HB. But the productions of superoxide anion are enhanced significantly compared with HA and HB, indicating they are both favorable Type I phototherapeutic agents.  相似文献

The ESR signals of bilirubin-IXα were studied including the samples treated with free radical generating and inhibiting systems, i.e.X-X0., Fe/EDTA, SOD, mannitol/ascorbate, DTPA, KCN et al.These stable signals all comprise those originated from a semiquinone radical(g=2.0012)and superoxide radical(g11=2.041, g=2.0040).The latter is shown to be bound with metal ionsespecially iron, chelated by bilirubin.The iron probably comes from bilirubin precursor---hemootobin.Active oxygen free radical scavengers may destroy these radicals.Kinetic curves of regeneration of the bilirubin radicals have been determined.Bilirubin is discussed as"active oxygen trap"in mammatians.  相似文献

Fast-flow electron spin resonance (ESR) spectroscopy has been used to detect a free radical formed from the reaction of l-tryptophan with Ce (4+) in an acidic aqueous environment. Computer simulations of the ESR spectra from l-tryptophan and several isotopically modified forms strongly support the conclusion that the l-tryptophan radical cation has been detected by ESR for the first time. The hyperfine coupling constants (HFCs) determined from the well-resolved isotropic ESR spectra support experimental and computational efforts to understand l-tryptophan's role in protein catalysis of oxidation-reduction processes. l-Tryptophan HFCs facilitated the simulation of fast-flow ESR spectra of free radicals from two related compounds, tryptamine and 3-methylindole. Analysis of these three compounds' beta-methylene hydrogen HFC data along with equivalent l-tyrosine data has led to a new computational method that can distinguish between these two amino acid free radicals in proteins without dependence on isotope labeling, electron-nuclear double resonance, or high-field ESR. This approach also produces geometric parameters (dihedral angles for the beta-methylene hydrogens) that should facilitate protein site assignment of observed l-tryptophan radicals as has been done for l-tyrosine radicals.  相似文献

The formation of free radical anions of 18 various indandione-1,3 derivatives and their aza analogues, I–III, during the process of electrochemical reduction on a mercury electrode in N,N-dimethylformamide has been proved by ESR spectroscopy.In the case of 2-substituted indandione-1,3 derivatives of non-fixed diketo structure, ESR spectra of the one-electron reduction products have been registered, while in the case of 2,2-substituted compounds, ESR spectra of both one- and three-electron reduction products have been obtained. Unpaired electron distribution in free radical anions and the relative electrophilic values in the corresponding positions of the phthaloyl ring have been evaluated on the basis of hyperfine structure (HFS) of the ESR spectral obtained.In the case of compounds I (X ≠ NO2), the unpaired spin is mainly localized (~ 0.8 of an electron) on the carbonyls, but a fairly high unpaired spin density is created in the 5- and 6-positions of the phthaloyl ring whilst that in the 4- and 7-positions is insignificant. A high unpaired electron density is created on the nitrogen atom of compound III, whilst the unpaired electron density on the hetero-atom in compound II is insignificant.Free radical products have also been found in the reduction process by alkaline metals in dimethoxyethane. However, in this case a mixture of radical ions is formed, their composition being difficult to identify from the HFS of the ESR spectra.  相似文献

Unlike direct ESR, spin trap methodology depends on the absolute fidelity of the spin trap reaction. Two alternative reactions of 5,5-dimethyl-1-pyrroline N-oxide (DMPO) leading to radical adduct artifacts have been discovered and investigated: inverted spin trapping and the Forrester-Hepburn nucleophilic mechanism. These two alternate pathways to radical adducts are a combination of one-electron oxidation and nucleophilic addition, in either order. In biological systems, serious artifacts have been reported due to the Forrester-Hepburn mechanism, which is initiated by the addition of a nucleophile to DMPO. It has recently been demonstrated that (bi)sulfite (hydrated sulfur dioxide) can react with DMPO via a nonradical, nucleophilic reaction, and it has been further proposed that DMPO/(?)SO(3)(-) formation in biological systems is an artifact and not the result of spin trapping of sulfur trioxide anion radical ((?)SO(3)(-)). The one-electron oxidation of (bi)sulfite catalyzed by horseradish peroxidase (HRP)/hydrogen peroxide (H(2)O(2)) has been reinvestigated by ESR spin trapping with DMPO and oxygen uptake studies to obtain further evidence for the radical reaction mechanism. In the absence of DMPO, the initial rate of (bi)sulfite-dependent oxygen and H(2)O(2) consumption was determined to be half of the initial rate of DMPO/(?)SO(3)(-) radical adduct formation as determined by ESR, demonstrating that, under our experimental conditions, DMPO exclusively forms the radical adduct by trapping the (?)SO(3)(-).  相似文献

Nitroxide free radicals are the most commonly used source for dynamic nuclear polarization (DNP) enhanced nuclear magnetic resonance (NMR) experiments and are also exclusively employed as spin labels for electron spin resonance (ESR) spectroscopy of diamagnetic molecules and materials. Nitroxide free radicals have been shown to have strong dipolar coupling to (1)H in water, and thus result in large DNP enhancement of (1)H NMR signal via the well known Overhauser effect. The fundamental parameter in a DNP experiment is the coupling factor, since it ultimately determines the maximum NMR signal enhancements which can be achieved. Despite their widespread use, measurements of the coupling factor of nitroxide free radicals have been inconsistent, and current models have failed to successfully explain our experimental data. We found that the inconsistency in determining the coupling factor arises from not taking into account the characteristics of the ESR transitions, which are split into three (or two) lines due to the hyperfine coupling of the electron to the (14)N nuclei (or (15)N) of the nitric oxide radical. Both intermolecular Heisenberg spin exchange interactions as well as intramolecular nitrogen nuclear spin relaxation mix the three (or two) ESR transitions. However, neither effect has been taken into account in any experimental studies on utilizing or quantifying the Overhauser driven DNP effects. The expected effect of Heisenberg spin exchange on Overhauser enhancements has already been theoretically predicted and observed by Bates and Drozdoski [J. Chem. Phys. 67, 4038 (1977)]. Here, we present a new model for quantifying Overhauser enhancements through nitroxide free radicals that includes both effects on mixing the ESR hyperfine states. This model predicts the maximum saturation factor to be considerably higher by the effect of nitrogen nuclear spin relaxation. Because intramolecular nitrogen spin relaxation is independent of the nitroxide concentration, this effect is still significant at low radical concentrations where electron spin exchange is negligible. This implies that the only correct way to determine the coupling factor of nitroxide free radicals is to measure the maximum enhancement at different concentrations and extrapolate the results to infinite concentration. We verify our model with a series of DNP experimental studies on (1)H NMR signal enhancement of water by means of (14)N as well as (15)N isotope enriched nitroxide radicals.  相似文献

Oxidation of catechols and catecholamines by horseradish peroxidase (HRP) and lactoperoxidase (LPO) has been studied by electron spin resonance (ESR) and electronic spectroscopies. The ESR technique has been used as an ESR spin stabilization approach by complexation of o-semiquinone free radicals with ZnII ions. ESR spectra and parameters of these free radical complexed forms are in good agreement with those obtained previously for complexed and uncomplexed species. The Km values obtained with the two methods show stereoselective effects towards the chiral substrates l- and d-dopa from HRP and LPO. Furthermore, these enzymes display opposite stereochemical interactions, in agreement with the analogous effects observed on l- and d-tyrosine by electronic and NMR binding studies.  相似文献

In vivo copper-mediated free radical production: an ESR spin-trapping study   总被引:2,自引:0,他引:2  
Copper has been suggested to facilitate oxidative tissue injury through a free radical-mediated pathway analogous to the Fenton reaction. By applying the electron spin resonance (ESR) spin-trapping technique, evidence for hydroxyl radical formation in vivo was obtained in rats treated simultaneously with copper and ascorbic acid or paraquat. A secondary radical spin-trapping technique was used in which the hydroxyl radical formed the methyl radical upon reaction with dimethylsulfoxide. The methyl radical was then detected by ESR spectroscopy as its adduct with the spin trap phenyl-N-t-butyl- nitrone (PBN). In contrast, lipid derived radical was detected in vivo in copper-challenged, vitamin E and selenium-deficient rats. These findings support the proposal that dietary selenium and vitamin E can protect against lipid peroxidation and copper toxicity. Since copper excreted into the bile from treated animals is expected to be maintained in the Cu(I) state (by ascorbic acid or glutathione), a chelating agent that would redox-stablilize it in the Cu(I) state was used to prevent ex vivo redox chemistry. Bile samples were collected directly into solutions of bathocuproinedisulfonic acid, a Cu(I)-stabilizing agent, and 2,2'-dipyridyl, a Fe(II)-stabilizing agent. If these precautions were not taken, radical adducts generated ex vivo could be mistaken for radical adducts produced in vivo and excreted into the bile.  相似文献
