The antioxidant, antiproliferative and cytotoxic activities against different human cancer cells were investigated in local and recently introduced plants of Mentha sp., Rosmarinus officinalis L. (ROL) and Origanum majorana L. (OML). ROL exhibited the highest antioxidant activities (IC50 8.4 ± 0.2 μg/mL) followed by OML and mint species such as Mentha suaveolens ‘apple mint’ and Mentha longifolia L. exhibiting moderate antioxidant activities. HPLC analysis of leaf extract revealed that rosmarinic acid is the main component followed by caffeic acid. Herbal leaf extracts varied in their proliferation inhibition and cytotoxicity against HeLa, MCF-7 and Jurkat cancer cells in a dose-dependent matter. The highest antiproliferative inhibition and cytotoxic activity were detected in ROL and OML followed by mint. Local herbs might have a potential role as anticancer natural medicines in addition to their high antioxidant activities due to the presence of different phenolics in their aqueous tea extracts.  相似文献

In the study, anti-Candida activity and phenol contents of Lythrum salicaria L. calli and wild species have been evaluated. The seeds of L. salicaria (Lythraceae), collected from Lahidjan City in the north of Iran, were cultured in Murashige and Skoog medium (MSM) with a supplement, gibberellin, to germinate. Callus inductions were performed from segments of seedling on MSM containing different concentrations of plant growth regulators, 2,4-dichlorophenoxyacetic acid (2,4-D) and 6-benzylaminopurine (BAP). The activity of calluses extracts, wild plant, gallic acid, and 3,3′,4′-tri-O-methylellagic acid-4-O-β-d-glucopyranoside (TMEG) as the main phenolic compounds against Candida albicans was assessed using cup plate diffusion method. The total phenols contents of calli and wild plant extracts were analyzed using Folin–Ciocalteu reagent. The callus formation in MSM supplemented with various concentrations of 2,4-D and BAP were 0–100 %. Anti-Candida activity of callus extract which obtained from MSM supplemented with 2,4-D and BAP (1 mg?dm?3) was similar to the wild plant extract. Minimum inhibitory concentration values of gallic acid and TMEG were obtained as 0.312 and 2.5 mg?cm?3, respectively. Gallic acid equivalent values in all treatments were from 0 to 288 μg GAE mg?1. Phenolic contents of plant aerial parts (331?±?3.7 μg GAE mg?1) and the callus, which developed in MSM including 1 mg?dm?3 of both 2,4-D and BAP, showed the same phenolic value and exhibited anti-Candida extract activity.  相似文献

This study aims to investigate whether the in vitro-cultured L. pumila var. alata has higher antioxidant activity than its wild plant. An 8-week-old L. pumila var. alata nodal segment and leaf explants were cultured onto Murashige and Skoog (MS) medium supplemented with various cytokinins (zeatin, kinetin, and 6-benzylaminopurine (BAP)) for shoot multiplication and auxins (2,4-dichlorophenoxyacetic acid (2,4-D) and picloram) for callus induction, respectively. The results showed that 2 mg/L zeatin produced the optimal results for shoot and leaf development, and 0.5 mg/L 2,4-D produced the highest callus induction results (60%). After this, 0.5 mg/L 2,4-D was combined with 0.25 mg/L cytokinins and supplemented to the MS medium. The optimal results for callus induction (100%) with yellowish to greenish and compact texture were obtained using 0.5 mg/L 2,4-D combined with 0.25 mg/L zeatin. Leaves obtained from in vitro plantlets and wild plants as well as callus were extracted and analyzed for their antioxidant activities (DPPH and FRAP methods) and polyphenolic properties (total flavonoid and total phenolic content). When compared with leaf extracts of in vitro plantlets and wild plants of L. pumila var. alata, the callus extract displayed significantly higher antioxidant activities and total phenolic and flavonoid content. Hence, callus culture potentially can be adapted for antioxidant and polyphenolic production to satisfy pharmaceutical and nutraceutical needs while conserving wild L. pumila var. alata.  相似文献

Ethanol extracts of Stachys glutinosa L. (Lamiaceae) were investigated for antioxidative properties, as well as antiproliferative action on various cell lines. The antioxidant activities were investigated by ABTS (2,2′-azinobis-3-ethylbenzothiazoline-6-sulphonic acid) assay, DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging, β-carotene/linoleic acid assay, scavenging of hydrogen peroxide (horseradish peroxidase test), superoxide anion scavenging, and hypochlorous acid scavenging (taurine test). The antioxidant activity was reported as IC50 and reveals antioxidant effects. Antiproliferative effects were measured in vitro on three cell lines: HepG2 (human hepatocarcinoma), MCF7 (breast human adenocarcinoma) and C2C12 (mouse myoblast) cell lines by MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay. The ethanol extract induced variations in cell viability on all cell lines tested. At 200 μg/mL, the effects on cell viability were ? 23%, ? 27% and ? 37%, respectively, for C2C12, MCF7 and HepG2.  相似文献

An improved procedure has been developed for efficient somatic embryogenesis in Anethum graveolens. Green friable embryogenic callus was obtained from hypocotyl segments on medium augmented with 2,4-dichlorophenoxyacetic acid (2,4-D). The highest embryogenic callus induction frequency of 87 % was obtained on Murashige and Skoog (MS) medium containing 1.13 μM 2,4-D. At lower concentration of 2,4-D (0.34 μM) callus turned dark in color and slow growing. Embryogenic cultures (76 %) responded with a mean number of 43 globular and 18 heart stage embryos. Somatic embryo maturation and subsequent conversion into plantlets took place on MS lacking growth regulators. Maximum number of somatic embryos developed on MS medium was 128.3 (per flask) and a plantlet conversion of 82 % was observed. Calcium alginate beads were produced by encapsulating somatic embryos. Highest percent germination (83 %) was observed on 0.8 % agar solidified MS medium with the plantlets acquiring an average length of 2.1 cm. Encapsulated somatic embryos could be stored at 4 °C up to 60 days with a conversion frequency of 49.3 %. Highest protein and proline content has been observed in embryogenic callus with small globular embryos. During morphological differentiation of the somatic embryos, changes in the antioxidant enzymatic system were observed. Superoxide dismutase (SOD) activity increased during initial stages and decreased catalase (CAT), peroxidase (POD), and ascorbate peroxidase (APX) activities were detected.  相似文献

Protocols for regeneration and Agrobacterium-mediated transformation of the apomictic species Eulaliopsis binata were developed. Initially, seeds of four genotypes of E. binata were incubated on a callus induction Murashige and Skoog (MS) basal medium supplemented with three concentrations of 2,4-dichlorophenoxyacetic acid (2,4-D). It was found that 36.2 % of explants developed highly friable callus on medium containing 3.0 mg l?1 2,4-D. Based on frequency of callus induction, the genotype Neixiang was selected for regeneration and transformation. Callus incubated on MS basal medium supplemented with 0.2 mg l?1 α-naphthalene acetic acid and 6.0 mg l?1 6-furfuryl-aminopurine developed shoots. Subsequently, Agrobacterium tumefaciens strain EHA105—harboring a plasmid pCAMBIA1381 carrying a hygromycin phosphotransferase (hpt) resistance gene and a synthetic green fluorescent protein (GFP) gene, both driven by the cauliflower mosaic virus 35S promoter—was used for transformation system. Putative transgenic callus was obtained following two cycles of hygromycin selection. Expression of the transgene(s) in putative transgenic callus was analyzed using the GFP detection. Molecular identification of putative transformed shoots was performed by polymerase chain reaction and Southern blot analysis to confirm presence and integration of the hpt gene.  相似文献

Nerium odorum, Linn. (Apocynaceae) is an important evergreen shrub. It is heat, salinity and drought tolerant. Plants with milky sap have medicinal value, mainly cardenolides, flavonoids and terpenes. It is used for wastewater purification and for restoration of riparian woodlands. In view of these facts, the study was conducted for micropropagation of N. odorum. Murashige and Skoog (MS) media supplemented with different concentrations (0.5–10.0 mg/l) of 1-naphthaleneacetic acid (NAA), 2,4-dichlorophenoxyacetic acid (2,4-D), 6-benzylaminopurine (BAP) and kinetin (Kin) were used singly and in combinations. Among all the growth hormones, 2,4-D was the best for callus induction (75 % in stem and 79 % in leaf) and in combination 2,4-D and BAP (78 % in stem and 81 % in leaf). The day of callus induction started from the 19th to the 37th day. This variation is due to the differences in culture conditions and the age of explants. The fresh and dry weight and moisture content showed good growth of callus, which is used in further studies of alkaloid production. Micropropagation of this plant allows the production of clones at a fast rate and in continuous manner. This work can lead to the development of an efficient protocol for callus induction and other issues.  相似文献

Coreopsis tinctoria flowering tops (CTFs) from the Kunlun Mountains in Xinjing (north-western China) have been used for tea production for about a century. This study aims to assess the antioxidant activities and total phenolic, flavonoid and proanthocyanidin contents of various solvent extracts of CTF. CTF was extracted using n-hexane, chloroform, ethyl acetate, n-butanol, 75% aqueous ethanol (AEE) and water. The antioxidant activities of the CTF extracts were investigated through DPPH, ABTS, *OH, *O2, total antioxidant capacity and reducing power assays. The results showed that n-butanol extract showed the highest contents of total phenols and flavonoids, with DPPH, ABTS and *OH radical-scavenging activities with IC50 values of 134, 90.72 and 13.8 μg mL? 1, respectively. The AEE demonstrated the strongest DPPH and ABTS radical-scavenging activities, with IC50 values of 103 and 75.16 μg mL? 1, respectively. Given its high antioxidant effect, CTF is a good source of natural antioxidants or functional food materials.  相似文献

Plants have been found useful in treating many human diseases caused by bacteria and viruses. The ability to synthesize compounds by plant secondary metabolism makes them an invaluable source of pharmaceutical and therapeutic products. The present study was designed to evaluate the phytochemical constituents, antioxidant, and anticancer activities of Tribulus terrestris seed extracts on HepG2 cell lines. TPC and TFC contents were 51 ± 0.7 mg GAE/g and 66.5 ± 0.4 mg QE/g, respectively. The antioxidant profile of the T. terrestris revealed that all the extracts have antioxidant potential and display the highest antiradical behavior in the pattern of methanolic > acetonic > chloroform > n-hexane, through DPPH, FRAP, OH radical scavenging, and NO radical scavenging assays. The antioxidant activity explored at the cellular level against H2O2-induced DNA damage showed a dose-dependent antioxidant effect of T. terrestris. Moreover, the methanolic extracts of all plant extracts showed notable thrombolytic potentials, the percentage of clot lysis accounted for T. terrestris was 33%, 27%, 17%, and 6% which indicated the significant clot lysis of methanolic and acetonic extracts in contrast to positive and negative standards. The genotoxicity was assessed through comet assay which exposed that T. terrestris at a low dose (0.5 mg/mL) is considered to be safe for effective treatment. MTT assay using HepG2 cell lines revealed that the highest tested concentration i.e., 100 μg/mL of the methanolic extract resulted in 86% cell viability compared to the control group. In silico study, from 14 selected compounds, three compounds, Heptacosane, Apiol, and Palmitic acid showed an affinity with target protein 51X0. The present findings may serve as a guideline for the standardization and validation of natural drugs containing the T. terrestris as an ingredient.  相似文献

Abstract

A novel neodymium(III) complex containing tungstogermanate and 5-fluorouracil, K11(C4H4FN2O2)2Nd(GeW11O39)2·15H2O (FNdGeW), was synthesized and its structure was characterized by 1H NMR, IR, XRD, and TG-DSC. The results indicated that FNdGeW had Keggin structure of hetereopolyanion and structure of 5-fluorouracil. The thermal analysis showed that FNdGeW decomposed at 255.3 °C. MTT tests were performed to study the anticancer activities against SGC-7901 and HepG2 cells. The results showed that FNdGeW and FGeW possessed higher anticancer activities than 5-fluorouracil. FNdGeW exhibited the highest anticancer activities against SGC-7901 cells (IC50 = 0.79 μmol?L-1) and HepG2 cells (IC50 = 0.37 μmol?L-1).  相似文献

An improved ion-pair HPLC method was developed for the simultaneous determination of ribonucleoside triphosphates and their corresponding deoxyribonucleoside triphosphates in HepG2 cell extracts. HPLC conditions, flow rate and column temperature were optimized and good linearity (r 2  > 0.9993) was obtained over the investigated concentration ranges. Reproducibility was evaluated by intra- and inter-day assays and RSD values were below 5.39%. Recoveries ranged from 98.2 ± 3.49% to 103.1 ± 1.75%, respectively. Finally, the method was successfully applied to the analysis of eight compounds present in HepG2 cell extracts.  相似文献

Taking into account the importance of natural antioxidants in the preservation of oils and fats, the present study evaluated the antioxidant action of five plant extracts in the control of soybean oil stability, by means of the accelerated techniques Rancimat and PDSC. These plants are rosemary (Rosmarinus officinalis L.), chamomile (Matricaria recutita L.), coriander (Coriandrum sativum L.), fennel (Foeniculum vulgare), and senna (Cassia angustifolia Vahl). The plant extracts and also the synthetic antioxidant BHT were added to the samples of crude soybean oil at the concentration of 1,000 mg kg?1. The values of total phenolic contents ranged from 8.7 ± 0.4 to 63.0 ± 2.3 mg GAE g?1 extract and a strong positive correlation was observed between the total phenolic contents and the overall antioxidant activity of the plant extracts. Such high values indicate a good protection of the analyzed soybean oil, moreover for the Rosemary extract that was superior to the remaining extracts. In the Rancimat technique the rosemary extract was more effective than the synthetic BHT antioxidant. The OIT values of Rosemary extract and the BHT antioxidant were equivalent, and the former, showed the highest phenolic contents among the extracts, for all the performed tests, confirming that it is a powerful natural source of antioxidants.  相似文献

This study aimed to compare phytochemical profiles and antioxidant capacity of various extracts including crude extracts, aqueous- and saponin-enriched butanol fractions prepared from the stems and leaves of Helicteres hirsuta Lour. The results revealed that all the three powdered extracts from the leaves and the stems possessed high levels of phenolics (177.07–241.03 mg GAE g?1), flavonoids (158.03–280.06 mg CE g?1) and saponins (165.77–1035.33 mg ESE g?1) and exhibited strong antioxidant capacity. HPLC analysis identified nine major compounds in the leaf powder crude extract; however, the leaf aqueous fraction had three extra compounds; whereas, the saponin-enriched butanol leaf fraction had seven extra compounds. For the stems, twelve main compounds were evident in either the powdered crude extract or the aqueous fraction, and five new compounds were revealed in the saponin-enriched butanol fraction. The findings revealed that the powdered aqueous fractions and saponin-enriched butanol fractions are potential sources of biologically active compounds for further investigation and industrial utilisation.  相似文献

Ficus palmata Forssk. (Moraceae family) is medicinally valuable plant that is mostly used as folk medicine for the treatment of different diseases. Phytochemical composition was evaluated by preliminary phytochemical investigation, GCMS analysis, and total bioactive contents (TPC and TFC). The antioxidant, enzyme inhibition, antimicrobial, thrombolytic and anticancer activities were performed for biological evaluation. The extract exhibited the maximum total phenolic (49.24 ± 1.21 mg GAE/g) and total flavonoid contents (29.9 ± 1.13 mg QE/g) which may be correlated to higher antioxidant potential of extract. The GCMS investigation identified the presence of 27 phytocompounds of different classes related to aldehydes, esters of fatty acids, triterpenes, steroids, triterpenoid. The extract possessed the strong α-glucosidase (73.4 ± 4.65 %) and moderate α-amylase inhibition activity (47.1 ± 3.29 %). Significant results were observed in case of antiviral, antifungal, and antibacterial activities. F. palmata extract inhibited the growth of HepG2 cancer cells in a dose-dependent manner. The extract also exhibited moderate in vitro thrombolytic activity. In addition, the phytocompounds identified by GCMS were subjected to in silico molecular docking studies to analyze the binding affinity between phytocompounds and enzymes (α-glucosidase and α-amylase). Moreover, the best docked compounds were selected for ADMET studies which provide information about pharmacokinetics, physicochemical properties, drug-likeness, and toxicity of identified phytocompounds. The outcome of our research revealed that ethanolic extract of F. palmata possessed good antidiabetic, antimicrobial, thrombolytic and anticancer potential. This plant should be further explored to isolate the bioactive compounds for new drug development.  相似文献

This study investigated the in vitro antioxidant and anticancer properties of the Fomitopsis pinicola extract (EMFP). The antioxidant activity of EFMP was analysed via free radical scavenging (DPPH, ABTS and Hydroxyl radicals) assay and a protein oxidation assay. EFMP effectively scavenged free radicals and exhibited remarkable protection against protein oxidation. The proliferation of EMFP-treated HepG2 cells was remarkably decreased. EMFP effectively increased the reactive oxygen species (ROS) production, depleted the mitochondrial membrane potential (MMP) and promoted the apoptosis of HepG2 cells. In addition, EMFP increased the malondialdehyde (MDA) content and reduced the activities of SOD, CAT and GPx in HepG2 cells. Using UPLC-Triple-TOF-MS, 2 phenolic compounds and 14 triterpenes were identified. These compounds may be the primary contributors to the antioxidant and anticancer capacities of EMFP. Together, these findings highlight the possibility of exploiting EMFP for its desired pharmaceutical ingredients.  相似文献

Purpose of studyOtostegia limbata (Benth.) Boiss. (Family: Lamiacae) is an important underexplored ethnomedicinal plant that has been used as antinflammatory, anticancer and antibacterial herbal remedy previously. The present work was aimed to evaluate the antioxidant, antimicrobial, antileishmanial, and anticancer prospective of O. limbata stem and leaf extracts.ResultsThe highest amount of phenolic and flavonoid content was obtained in the methanol-acetone and methanol stem extracts i.e., 53.29 ± 1.33 and 28.64 ± 1.16, respectively with highest DPPH scavenging in MeH stem extract (IC50 = 34.5 ± 1.34 μg/ml). Significant amount of catechin, gallic acid, apigenin and rutin was quantified. A moderate antibacterial and substantial antifungal activity was observed. Cytotoxicity against brine shrimps categorized 21% of stem (3 out of 14 extracts) and 57% (8 out of 14 extracts) of leaf extracts as potent. Substantial cytotoxicity against THP-1 cell line (IC50 = 3.46 ± 0.25 μg/ml) and Leishmania (IC50 = 1.50 ± 0.23 μg/ml) was exhibited by methanol-distilled water leaf extract while noteworthy antiproliferative activity against Hep-G2 (IC50 = 0.44 ± 0.45 μg/ml) was manifested by n-hexane stem extract. Absence of hemolysis in normal RBCs signified plant’s selective cytotoxicity. Methanol-distilled water and chloroform stem extracts displayed prominent protein kinase inhibition and antidiabetic potential of plant.ConclusionThe results of present study recommend O. limbata as a potential source of antifungal, antileishmanial, anticancer, and α-amylase inhibitory agents.  相似文献

Previous studies have shown that the ethanolic extracts from Sophora moorcroftiana seeds (ee-Sms) have in vitro anticancer properties. The anti-proliferation effects of ee-Sms on HepG2 cells were assessed by MTT assay and cell cycle analysis. Total cell proteins were separated by two-dimensional electrophoresis (2-DE), and protein spots with more than two-fold difference were analysed by MALDI-TOF/TOF-MS. MTT assay showed that the anti-proliferation of ee-Sms demonstrates dose- and time dependently. HepG2 cells were treated with ee-Sms at 1.30 mg/mL for 48 h induced cell cycle arrest in S phase. The differentially-expressed proteins were involved in DNA repair, cell proliferation, cell metabolism and immunoreaction. This study sheds new insights into the molecular mechanisms underlying the anti-proliferation properties of ee-Sms in HepG2 cells.  相似文献

Callus culture of Artemisia absinthium L. was established for enhanced production of phenolics and higher antioxidant activity. Callus was induced from seed-derived leaf explants, incubated on to MS media supplemented with thidiazuron (TDZ; 0.5–5.0 mg/l) either alone or in combination with α-naphthalene acetic acid (NAA; 1.0 mg/l). These callus cultures were investigated for their growth kinetics, total phenolic content, and antioxidant activity on weekly basis for a period of 49 days. Maximum dry biomass accumulation of 8.73 g/l was observed on day 42 in response to 1.0 mg/l TDZ and 1.0 mg/l NAA. Furthermore, maximum level of total phenolic content of 8.53 mg GAE/g DW and highest 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity of 72.6 % were observed in calli formed in response to 1.0 mg/l TDZ on day 42. The results showed a positive correlation of total phenolic content and DPPH radical scavenging activity in most of the callus cultures of A. absinthium L.  相似文献

The use of natural antioxidants has been widely promoted in the food industry, because it is economically viable and very attractive with consumers. In this study, we determined the extractable total phenolic content (FET) of crude ethanol extracts of the leaves (EL), flowers, and seed pods of Moringa oleifera Lam. We also evaluated the antioxidant effect of the extracts on oxidative stability of soybean oil, through the accelerated PDSC and Rancimat® techniques using the synthetic antioxidants butylated hydroxytoluene (BHT) and tert-butylhydroquinone (TBHQ) as positive control. The values of FET in the extracts ranged from 41.75 ± 3.35 to 53.69 ± 1.00 mg GAE g?1, and EL extract exhibited best result. The results of oxidative stability in both techniques showed that the EL extract provided greater protection to the oil, indicating a correlation between the amount of FET and the protective effect. Comparison made between the synthetic antioxidants and extracts revealed that in the Rancimat technique, extracts were less effective than the synthetic antioxidants TBHQ and BHT, however, in the PDSC technique the EL extract was more effective than BHT, proving to be a good alternative for applications in the soybean oil, replacing this synthetic antioxidant.  相似文献

Dioscorea alata var. purpurae (Indian purple yam) is an important source of diosgenin, a triterpenoid that is used as a raw material in the synthesis of corticosteroid hormones. These drugs are used for the treatment of pharmacological conditions such as arthritis. This paper reports in vitro propagation of explants of various parts of Dioscorea—tuber, leaves, and nodes. Murashige and Skoog media supplemented with hormones and additives was used to get maximum callus initiation and shoot/root induction. All the cultures were maintained at 25?±?2 °C under cool-white fluorescent tubes with 16-h photoperiod. Callus initiation was observed from 8th to 11th day of inoculation, and subsequent root/shoot was initiated in nodal callus after 21 days. Hormones such as kinetin, indole-3-acetic acid, indole-3-butyric acid, α-naphthalene acetic acid, and thiadizuron did not show significant enhancement. Also, there was no need for supplementing additives (silver nitrate, glutamine, l-asparagine monohydrate, polyethylene glycol). Combination of 6-benzylaminopurine (0.2 ppm) and 2,4-dichlorophenoxyacetic acid (2 ppm) hormones gave the best results, and all parts of the plants gave similar callus induction.  相似文献
