Ultrasound-assisted enzymatic maceration (UAEM) has gained considerable interest in the fruit juice industry, owing to its potential to increase juice yield and content of polyphenols while simultaneously saving time and energy. In this study, the effects of UAEM (ultrasonic probe, 20 kHz, 21 W*cm−2 and 33 W*cm−2) on pectin degradation in a continuous circulation system were investigated over 60 and 90 min. Main pectinolytic enzymes activities of (polygalacturonase, pectin lyase and pectin methylesterase) of a commercial enzyme preparation were examined for individual synergistic effects with US. Pectin hydrolysis by UAEM differed significantly compared to treatment with ultrasound or enzymes alone regarding the profile of degradation products compared to treatment with ultrasound or enzymes alone. Ultrasound fragmented pectin to less branched oligomers of medium molecular weight (Mp approx. 150 kDa), which were further degraded by pectinolytic activities. The low molecular weight fraction (<30 kDa), which is known to be beneficial for juice-quality by adding nutritional value and stabilizing polyphenols, was enriched in small oligomers of homogalacturonan-derived, rhamnogalacturonan I-derived, and rhamnogalacturonan II-derived residues. Synergistic effects of ultrasound application enhanced the effective activities of polygalacturonase and pectin lyase and even prolonged their performance over 90 min, whereas the effective activity of pectin methylesterase was not affected. Final marker concentrations determined by each enzyme assay revealed a considerable higher total process output after UAEM treatment at reduced temperature (30 °C) comparable to the output after conventional batch maceration at 50 °C. The obtained results demonstrate the high potential of UAEM to produce high-quality juice by controlling pectin degradation while reducing process temperature and equally highlight the matrix and enzyme specific effects of a simultaneous US treatment.  相似文献

This study investigated the effects of high-power ultrasound (HPU, 0–45 °C, 242–968 W/cm2, 2–16 min) on the rheological properties of strawberry pulp. Following the HPU treatment, the strawberry pulp exhibited an increase in apparent viscosity, storage modulus (G′), and loss modulus (G″). The water-soluble pectin (WSP), pectin methylesterase (PME) activity, and free calcium ions (Ca2+) of the strawberry pulp after HPU treatment were investigated to determine a possible reason for this phenomenon. HPU caused a significant decrease in the degree of esterification (DE), molecular weight (Mw), and particle size of strawberry WSP, but no significant changes were evident in the galacturonic acid (GalA) content and the zeta (ζ)-potential (P > 0.05), resulting in decrease in the apparent viscosity. Moreover, the largest reduction of PME activity was 22.6% after HPU treatment at 605 W/cm2 and 45 °C for 16 min, indicating that the PME was resistant to the HPU treatments. The free Ca2+ content in the strawberry pulp was significantly decreased after exposure to HPU (P < 0.05). The maximal reduction of 52.01% in the free Ca2+ was achieved at 605 W/cm2 and 45 °C for 16 min. The overall results indicated that the high residual activity (RA) of PME after HPU might induce the low esterification of WSP, while HPU promoted the interaction of free Ca2+ and low-methylated pectin, to form the network structure of Ca2+-low-methylated pectin, resulting in an increase in viscosity in the complex strawberry system.  相似文献

The combination of ultrasound and pressurized liquid extraction (UAPLE) was evaluated for the extraction of phenolic compounds from pomegranate peels (Punica granatum L.). The influence of several variables of the process on extraction yield, including solvent type (water, ethanol + water 30, 50 and 70% v:v), temperature (50–100 °C), ultrasound power (0–800 W at the generator, or 0–38.5 W at the tip of the probe), mean particle size (0.68 and 1.05 mm), and number of cycles (1–5), were analyzed according to the yield of 20 different phenolic compounds. The most suitable temperatures for the extraction of phenolic compounds using water were from 70 to 80 °C. In general, 100 °C was not adequate since the lowest extraction yields were observed. Results suggested that ultrasound had a greater impact on extraction yields using large particles and that intermediate ultrasound power (480–640 W at the generator, or 23.1–30.8 W at the tip of the probe) produced the best results. Using small particles (0.68 mm) or large particles (1.05 mm), extraction with ultrasound was 1 cycle faster. Ultrasound may have offset the negative effect of the use of large particles, however, did not increase the yield of phenolic compounds in any of the cases studied after five cycles. Additionally, the continuous clogging problems observed with small particles were avoided with the use of large particles, which combined with ultrasound allowed consistent operation with good intra and inter-day reproducibility (>95%). Using samples with large particle size, the best extraction conditions were achieved with water extraction solvent, 70 °C extraction temperature, ultrasound power at 480 W, and 3 cycles, yielding 61.72 ± 7.70 mg/g. UAPLE demonstrated to be a clean, efficient and a green alternative for the extraction of phenolic compounds from pomegranate peels. These findings indicate that UAPLE has a great potential to improve the extraction of bioactive compounds from natural products.  相似文献

We investigated the effect of ultrasound on plasma membrane (PM) Ca2+-ATPase activity of Aloe arborescens callus cells in solid culture. The calluses were exposed by a 20 kHz digital sonifier at the powers of 2 and 10 W from the effective exposure times of 2-10 s. PM Ca2+-ATPase activity was almost significantly higher at 2 W both in continuous wave and 10% duty cycle than that of the control (no ultrasound) at effective exposure times of 5 and 10 s. However, its activity decreased at 10 W in continuous wave exposure. It is possible that the PM Ca2+-ATPase configuration or structure may be partly damaged by high-energy ultrasound at 10 W. Our results showed that low-energy ultrasound exposure was a useful physical field to stimulate A. arborescens callus cells to adapt environmental stress through PM Ca2+-ATPase activity increase.  相似文献

The present study presents an attempt to modify the surface properties of macroporous resins (MRs) in order to improve anthocyanin adsorption and desorption from Pyrus communis var Starkrimson fruit peel extract. A number of MRs were tested to optimise the ultrasonic-assisted adsorption (UAA) conditions; including ultrasonic power (100–400 W), resin-to-extract ratio (1–3 g/50 mL) and temperature (20–40 °C). Similarly, varying ultrasonic-assisted desorption (UAD) conditions were optimised; including ultrasonic power (200–600 W), resin-to-solvent ratio (1–4 g/50 mL), ethanol concentration (60–90% v/v) and temperature (20–40 °C). The Amberlyst 15 (H) cationic resin was found to be superior to the other tested resins. The maximum adsorption capacity (659 µg/g) of cyanidin 3-galactoside (Cy 3-gal) was achieved under the optimised UAA conditions (400 W, 20 °C and 1 g/50 mL), while 616 µg/g of Cy 3-gal was recovered under the optimised UAD conditions (582 W, 1 g/50 mL, 60% and 20 °C). Moreover, titratable-acid and total-sugar contents were found to be significantly lower under UAA than under conventional-assisted adsorption (CAA). ANOVA revealed that process factors had significant effects on the Cy 3-gal purification, as depicted by their linear, quadratic and interactive effects. While anthocyanin adsorption was found to be significantly improved at lower ultrasonic power, higher power promoted the desorption process. Adsorption under optimized UAA conditions followed pseudo second-order kinetics and multilayer adsorption (Freundlich isotherm) onto the Amberlyst 15 (H) resin surface was observed. The particle-size distribution curve and scanning electron microscopic images also revealed higher resin-surface roughness, peeling and the appearance of pores on the surface under ultrasonication. This is the first study to use ultrasonication to modify a cationic exchange resin for the improvement of Cy 3-gal purification from a fruit extract. This study can recommend the use of ultrasonication as a low-cost green technique that can improve macroporous resin characteristics for better purification of compounds from an extract.  相似文献

An efficient cold-mechanical/sonic-assisted extraction technique was developed for extraction of genipin from genipap (Genipa americana) peel. Ultrasound assisted extraction (285 W, 24 kHz) was performed at 5, 10 and 15 °C for 5, 10 and 15 min. After cold-extraction, genipin was separated from pectin and proteins by aid of fungal pectinesterase. The maximum yield of non-cross-linked genipin was 7.85 ± 0.33 mg/g, at 10 °C for 15 min by means of ultrasound extraction. The protein amount in extracts decreased in all samples. If mechanical process is combined with ultrasound assisted extraction the yield is increased by 8 times after the pectinesterase-assisted polyelectrolyte complex formation between pectic polysaccharides and proteins, avoiding the typical cross-linking of genipin. This novel process is viable to obtain non-cross-linked genipin, to be used as a natural colorant and cross-linker in the food and biotechnological industries.  相似文献

Burning of coal accounts for an enormous proportion of the current energy supply, especially in developing countries. Burning of coal produces large amounts of coal fly ash, which causes serious environmental problems unless it is managed properly. Using chemical analysis, we found that coal fly ash could be a promising source of Si, Al, Ca and some rare earth elements, especially with the assistance of some measures such as ultrasound. In this study, we extracted silicon from coal fly ash using an alkaline dissolution strategy and investigated the effects of temperature and ultrasonic power on the efficiency of silicon extraction. During a 70 min reaction, the efficiency of silicon extraction increased markedly, from 9.41% to 34.96%, as the reaction temperature increased from 70 °C to 110 °C. With ultrasound assistance, ultrasonic waves enhanced the extraction of silicon at both 80 °C and 110 °C at 720 W ultrasound, increasing the efficiency of silicon extraction from 6.01% to 15.36% and from 34.96% to 54.42%, respectively. However, at 900 W ultrasonic power, extraction was slightly inhibited at both temperatures, causing a little decrease in efficiency.  相似文献

Ultrasound-assisted approach was successfully applied for the synthesis of mayenite from calcium and aluminum hydroxides and then subsequently impregnated with Ni by the wet impregnation method. The synthesis was performed with a 13 mm probe-type ultrasound, operating under an acoustic power of 30.5 W and a frequency of 20 kHz. Ultrasound application was studied in detail from a 3k experimental design, where the variables studied were ultrasound time (10–50 min) and calcination temperature (900–1200 °C). Ultrasound promoted an effective dispersion of the precursors in a short time of 10 min leading to a high conversion to mayenite after calcination at 1200 °C. Ultrasound treatment also had a positive effect on Ni impregnation, increasing the dispersion of the metal in the support and leading to a stronger interaction of nickel-containing species with mayenite support. The use of ultrasound application has proved to be attractive both for catalyst properties and for facilitating catalyst synthesis.  相似文献

The present study deals with the effect of ultrasound on the swelling and dissolution behavior of the cellulose pulp. Hardwood, acid sulfite cellulose pulp sheets were sonicated at different temperatures, operated at a fixed frequency (37 kHz) and power (320 W) to break the intermolecular forces and hydrogen bonds of crystalline region. The obtained samples were evaluated for crystallinity, intrinsic viscosity, molecular weight (MW), molecular weight distribution (MWD) and surface morphology. It was observed that the crystallinity reduced from 61.9 to 18.9% after 20 min of ultrasound treatment at 30 °C, which was equivalent to the swelling at 75 °C for 20 min without the use of ultrasound frequency. It was also found that ultrasound pre-treatment significantly decreased the particle size of the slurry and shortened the dissolution time and temperature requirement without affecting the cellulose solution quality.  相似文献

Concept of waste to wealth is a hot topic with research ongoing globally to reduce carbon footprint. In an effort to follow up this cause present study focused on tomato industry waste specifically the peel of tomatoes for extraction of pectin. Pectin extraction was performed using five different extraction techniques (Ultrasound assisted extraction (UAE); microwave assisted extraction (MAE); ohmic heating assisted extraction (OHAE); ultrasound assisted microwave extraction (UAME) and ultrasound assisted ohmic heating extraction (UAOHE) at different power levels to study its extraction and degradation kinetics and in turn to optimize the extraction process. The extracted pectin yield ranged from 9.30% for OHAE to 25.42% for MAE. Also, there was very less difference in the yield of MAE and UAME extracted pectin, but at the cost of major difference in degree of esterification 59.76 ± 0.70 and 73.33 ± 1.76%, respectively. In addition, all the pectin extracted under optimized conditions was having acceptable purity, [Galacturonic acid (GalA) content ranged from 675.8 ± 11.31 to 913.3 ± 20.50 g/kg of pectin]. FTIR analysis confirmed the presence of functional groups in the finger print region of identification for polysaccharide in all the extracted pectin. According to obtained results, UAME can be considered as better green extraction technology in terms of extraction yield as well as in quality of pectin compared to the other treatments used. Therefore, results suggest that UAME can be used as an efficient pectin extraction method from tomato processing waste.  相似文献

Modified pectin (MP) was reported to have increased bioactivities compared with the original one. However, traditional modification methods such as using an acidic solvent with heating are not only costly but causing severe pollution as well. In this study, manothermosonication (MTS) with a continuous-flow system was utilized to modify citrus pectin. The citrus pectin (5 g/L) treated by MTS (3.23 W/mL, 400 kPa, 45 °C) exhibited lower molecular weight (Mw, 248.17 kDa) and PDI (2.76). The pectin treated by MTS (400 KPa, 45 °C, 5 min) exhibited a narrower Mw distribution and lowered more Mw (48.8%) than the ultrasound(US)-treated (23.8%). Pectin degradation data fitted well to kinetic model of 1/Mwt −1/Mw0 = kt (45–65 °C). A lower activation energy of 13.33 kJ/mol was observed in the MTS treatment compared with the US-treated (16.38 kJ/mol). The MTS-treated pectin lowered the degree of methoxylation (DM), mol% of rhamnose and galacturonic acid (GalA) while increased mol% of galactose (Gal), xylose (Xyl), and arabinose (Ara). The 1H and 13C nuclear magnetic resonance showed that MTS could not alter the primary structures of citrus pectin. However, an elevated (Gal + Ara)/Rha and reduced GalA/(Rha + Ara + Gal + Xyl) molar ratios after MTS suggested that MTS resulted in more significant degradation on the main chains and less on the side chains of pectin, in agreement with the result of atomic force microscope. Moreover, the MTS-treated pectin exhibited a higher 1,1-diphenyl-2picryl hydrazyl radical scavenging capacity compared with original pectin.  相似文献

We have conducted experiments to study the behavior of W5%Re–W26%Re (type C) and Pt10%Rh–Pt (type S) thermocouples under high pressure in a multi-anvil apparatus. The electromotive force (emf) between four different or three identical thermocouple wires was measured up to 15?GPa and 2100?°C. Mechanical and chemical stability of the thermocouples was examined during and after the experiments. Due to the effect of pressure on the emf/temperature relation, the temperature reading of the type C minus that of the type S thermocouple rises to +5?°C then falls to ?15?°C between room temperature and 1500?°C at 5?GPa, and to +25?°C and then ?35?°C between room temperature and 1800?°C at 15?GPa. In addition, we observed variations in the emf/temperature relation caused by uncertainties in the position and geometry of hot junctions in a steep temperature gradient, and by variable distribution of pressure gradient and non-hydrostatic stress on the thermocouple wires. These errors are estimated at 1.6% for the type S thermocouple up to 1700?°C, and 0.8% for the type C thermocouple up to 2100?°C. Self-diffusion and chemical contamination of the thermocouples by high-purity insulating ceramics appear negligible for the type S thermocouple at 1700?°C for one hour, and for the type C thermocouple at 2100?°C for half an hour. In contrast, large-scale displacement of the hot junction due to dislocation of the type C thermocouple wires and plastic deformation of the type S thermocouple wires may lead to large errors in temperature measurement (±200?°C).  相似文献

Four factors three level face centered central composite response surface design was employed in this study to investigate and optimize the effect of process variables (liquid-solid (LS) ratio (10:1–20:1 ml/g), pH (1−2), sonication time (15–30 min) and extraction temperature (50–70 °C)) on the maximum extraction yield of pectin from waste Artocarpus heterophyllus (Jackfruit) peel by ultrasound assisted extraction method. Numerical optimization method was adapted in this study and the following optimal condition was obtained as follows: Liquid-solid ratio of 15:1 ml/g, pH of 1.6, sonication time of 24 min and temperature of 60 °C. The optimal condition was validated through experiments and the observed value was interrelated with predicted value.  相似文献

Paracetamol (acetaminophen) is one of the most frequently used analgesic and antipyretic drugs. This work deals with ultrasound assisted synthesis (UAS) of paracetamol from hydroquinone using ammonium acetate as an amidating agent. The optimization of various reaction and ultrasound parameters was performed to minimize the energy and time requirement. UAS of paracetamol was achieved at a lower temperature (60 °C) and the time (150 min) without formation of salt as a byproduct, making reaction green and inherently safer. On the other hand, the conventional process requires high reaction temperature (220 °C) and time (15 h). The quantification of the product was done by using high performance liquid chromatography (HPLC). Optimization of parameters revealed that the percent yield of 57.72% can be obtained in 150 min by performing the reaction in the ultrasound bath at 22 kHz frequency, 60 °C temperature, hydroquinone to ammonium acetate and acetic acid in a molar ratio of 1:6:5, 125 W power, 50% duty cycle and agitation speed of 300 RPM. Hence, ultrasound assisted synthesis can be considered as a process intensification tool for the synthesis of paracetamol and possibly other pharmaceutical compounds.  相似文献

The effects of thermosonication processing (TS, 90 °C, ultrasound powers of 200, 400, and 600 W) on the quality parameters of Jamun fruit dairy dessert compared to conventional heating processing (high-temperature short time, (HTST), 90 °C/20 s) were evaluated. Microbiological inactivation and stability, rheological parameters, physical properties, volatile and fatty acid profiles, and bioactive compounds were assessed. TS provided more significant microbial inactivation (1 log CFU mL−1) and higher microbial stability during storage (21 days) than HTST, with 3, 2, and 2.8 log CFU mL−1 lower counts for yeasts and molds, aerobic mesophilic bacteria, and lactic acid bacteria, respectively. In addition, TS-treated samples showed higher anti-hypertensive (>39%), antioxidant (>33%), and anti-diabetic (>27%) activities, a higher concentration of phenolic compounds (>22%), preservation of anthocyanins, and better digestibility due to the smaller fat droplet size (observed by confocal laser scanning microscopy). Furthermore, lower TS powers (200 W) improved the fatty acid (higher monounsaturated and polyunsaturated fatty acid contents, 52.78 and 132.24%) and volatile (higher number of terpenes, n = 5) profiles and decreased the atherogenic index. On the other hand, higher TS powers (600 W) maintained the rheological parameters of the control product and contributed more significantly to the functional properties of the products (antioxidant, anti-hypertensive, and anti-diabetic). In conclusion, TS proved to be efficient in treating Jamun fruit dairy dessert, opening space for new studies to define process parameters and expand TS application in other food matrices.  相似文献

This research investigated the effects of ultrasound application (192 ± 6 W/L) on the microstructure of vegetables/fruits with different porosities, cell sizes and patterns (eggplants, beetroots, and apples), submitted to an immersion treatment in different liquids: distilled water, citric acid (1% w/v), and the vegetable/fruit juice, at 25 °C during 5 min. The ultrasound application did not significantly (p > 0.05) affect the size of the cells of the most porous material (eggplant) compared to the samples immersed without ultrasound assistance. The apple samples (with a middle-high porosity and the largest cells) were the most affected by ultrasound application. The median cell areas of samples treated with ultrasound in water and apple juice were 26 and 20% larger than those of samples treated without ultrasound, mainly because of cell wall disruption which caused the cells to merge into bigger clusters, but no effect was observed with the citric acid. Ultrasound application significantly (p < 0.05) increased the median cell area of the less porous raw matter (beetroot) only when the treatment was carried out in the vegetable juice (cells were 26% larger after treatment assisted with ultrasound than without it). Thus, the effects of ultrasound differ in materials with initially different characteristics.  相似文献

Mo films deposited by DC sputtering are widely used as back contact in CIGS and CZTS based thin film solar cells. However, there have been only a few studies on the deposition of Mo films by RF sputtering method. In this context, Mo films on SLG substrates were prepared as a function of deposition pressure and power by using RF magnetron sputtering method to contribute to this shortcoming. Mo films were deposited at 250 °C substrate temperature by using 20, 15, 10 mTorr Ar pressures at 120 W RF power and 10 mTorr Ar pressure at 100 W RF power. Structural, morphological and reflectivity properties of RF-sputtered Mo films were clarified by XRD, AFM, FE-SEM and UV–Vis measurements. In addition, due to sodium incorporation from SLG substrate to the absorber layer through Mo back contact layer is so essential in terms of improving the conversion efficiency values of CIGS and CZTS thin film solar cell devices, the effects of Na diffusion in the films were analyzed with SIMS depth profile. The electrical properties of the films such as mobility, carrier density and resistivity were determined by Hall Effect measurements. It was found that Mo films prepared at 120 W, 10 mtorr and 250 °C substrate temperature and then annealed at 500 °C for 30 min, had resistivity as low as 10−5 Ω cm, as well as higher amount of Na incorporation than other films.  相似文献

Summary A water/butoxyethanol solution has been investigated by means of several techniques (viscosity, ultrasound and hypersound) as a function of concentration and temperature. The butoxyethanol molar fraction range investigated was situated between 0.015 and 0.09, while temperature ranged between −10°C and +45°C. We observed the building-up of large structures lowering the temperature, in particular below +15°C. All data coming from different techniques seem to confirm such a picture and indicate that this aggregation phenomenon is a percolation-like process. Bacterial cellulose (BC) film formation could be a critical issue in nanotechnology applications such as biomedical or smart materials products. In this research, purified pretreated BC was subjected to high intensity ultrasound (HIUS) and was investigated for the development of BC films. The morphological, structural and thermal properties of the obtained films were studied by using FE-SEM, AFM, FT-IR, XRD, TGA and DSC characterizations. Results showed that the most favorable purification treatment was the 0.01 M NaOH at 70 °C for 2 h under continuous stirring. The most suitable ultrasound operating conditions were found to be, 1 cm distance of ultrasonic probe from the bottom of the beaker, submerged in cold water bath cooling around 12 ± 2 °C. The power (25 W/cm2), time (30 min), BC concentration (0.1% w/w), amplitude (20 μm) and frequency (20 kHz) were maintained constant.  相似文献

An investigation was conducted into the utilization of treatments combining ultrasound and lysozyme (US + Lys) to deactivate Salmonella typhimurium (S. typhimurium) in the liquid whole egg (LWE). Furthermore, US + Lys and heat treatment (HT) with a similar microbial inactivation effect were comparatively evaluated by examining their impact on the quality attributes of LWE. The LWE was treated with US at 35–45 °C and 605–968 W/cm2 for 5–35 min, and with HT at 58–64 °C for 3–4 min. Lysozyme (Lys) alone achieved a minimal degree of inactivation in S. typhimurium, while it was enhanced with the application of US alone when the treatment temperature, time, and energy were increased. Furthermore, US and US + Lys caused a reduction of 3.31 and 4.26 log10 cycles in S. typhimurium, respectively at 968 W/cm2 and 35 °C for 20 min, indicating a synergistic relationship between US and Lys for the effective inactivation of S. typhimurium. Similarly, HT and HT + Lys achieved a reduction of 4.10 and 4.75 log10 cycles at 64 °C/3 min, respectively. The L* and b* values of the LWE following US and US + Lys application were significantly higher than untreated and heat-treated LWE, indicating that US treated LWE had a brighter and yellower appearance. The protein solubility (PS) slightly decreased after all treatments, while the pH increased. Furthermore, the foaming capacity (FC) and foam stability (FS) were decreased, revealing that LWE had a lower FC and unstable foam after all treatments. Therefore, US and US + Lys could increase the viscosity and gelation temperature (Tg) of LWE, indicating that LWE exhibited higher heat resistance after US treatment. These results indicated that US + Lys might be a promising pasteurization technology in the processing of LWE.  相似文献
