摘要: 金属有机框架材料(MOFs)具有均匀的孔隙率和大的比表面积,可作为固定化酶的载体。然而,固定化酶由于较长响应时间或酶易泄漏的缺点阻碍了其应用。本研究选取类过氧化物酶MIL-101为载体,戊二醛(GA)为交联剂,通过交联法将葡萄糖氧化酶(GOx)固定在载体上,建立了模拟多酶体系GOx@GA@MIL-101。该复合物的制备可进一步高效催化级联反应检测葡萄糖。GOx@GA@MIL-101具有更快的催化变色效果(30秒)。该策略成功地结合了天然酶与模拟酶的优点,为研究化学催化剂和天然酶催化剂之间的协同效应提供了机会。

Abstract: Metal-organic frameworks (MOFs)with uniform porosity and large surface areas can function as carriers of immobilized enzymes. However, drawbacks including long response times or enzyme leakage have hindered their applicability. In this study, we selected a peroxidase-like MIL-101as the carrier, glutaraldehyde (GA) as cross-linking agent, glucose oxidase (GOx) was immobilized on the carrier by cross-linking method, and a simulated multi-enzyme system GOx@GA@MIL-101was established. The preparation of this composite can further efficiently catalyze the cascade reaction to detect glucose. The method GOx@GA@MIL-101 has faster catalytic discoloration effect (30 seconds). It is noteworthy that the strategy successfully combines the advantages of natural enzymes and enzyme mimics and provides an opportunity to study the synergistic effect between chemical catalysts and natural enzymes.
